Excitonic Structure and Dynamics in Two-Dimensional Halide Perovskites.

Layered two-dimensional halide perovskites (2D HPs) have emerged as a versatile platform for studying strongly bound excitons in the presence of spin-orbit coupling, structural symmetry breaking, and pronounced coupling to lattice vibrations. In this Mini-Review, we focus on the monolayer limit of this class of materials and highlight recent progress in understanding excitonic fine structure and optical selection rules, chiral and spin-dependent optical response, exciton-phonon coupling and exciton relaxation pathways, and the breakdown of simple band-edge pictures in 2D HPs in which organic a

From Undruggable to Actionable: Redefining Druggability Through Emerging Therapeutic Modalities.

The concept of "undruggable" targets has long represented a major limitation in medicinal chemistry, referring to proteins and biomolecules that lack well-defined binding pockets or exhibit highly dynamic conformational behavior. However, advances in structural biology, chemical biology, and computational methodologies are rapidly expanding the scope of tractable therapeutic space. Emerging modalities, including targeted protein degradation, molecular glues, macrocycles, RNA-targeting small molecules, covalent ligands, and artificial intelligence-assisted molecular design, enable pharmacologic

Transformation of Dewar Metallabenzenes: Valence Bond Isomerization to Metallabenzenes and Beyond.

The study of valence isomerism in aromatic systems has long provided fundamental insights into molecular stability, reactivity, and electronic structure. While the mechanism and possibility of the interconversion between benzene and its Dewar isomer are well established in organic chemistry, analogous transformations in metallaaromatic systems remain largely unexplored. Here, we report the first experimental observations with well-defined Dewar metallabenzenes that are consistent with their rearrangement into metallabenzenes, a process previously considered only computationally. Structurally c

Entropy-weighted topological descriptors in QSPR modeling of antituberculosis drugs.

In this study, we develop quantitative structure-property relationship (QSPR) model using entropy-weighted topological descriptors to predict the physicochemical properties of antituberculosis drugs. The molecular structures of fifteen drugs used to treat tuberculosis were taken from PubChem, and their conventional degree-based descriptors were enhanced via Shannon entropy to account for both structural magnitude and distributional irregularity. These entropy-augmented indices were incorporated into quadratic, cubic, exponential, and logarithmic regression models to describe structure-property

Alternative and Sustainable Route to Explore a New Class of Amidines by Photochemical Synergistic Effect of Copper/Nitroxyl Radical Catalysis via Halogen-Atom Transfer.

Amidines are a vital class of bioactive compounds and often necessitate multiple components for their synthesis. Therefore, exploring efficient and sustainable methodologies for their synthesis is indispensable. Herein, we disclose an alternative and greener method for synthesizing an unexplored new class of amidines through the photochemical synergistic effect of copper/nitroxyl radical catalysis. This approach facilitates site-selective radical amination of unactivated imine C(sp2)-H bond in C,N,N-cyclic imines over favored selectivity via halogen-atom transfer (XAT). This greener method tic

Data-driven machine learning models for predicting the maximum absorption and emission wavelengths of single benzene fluorophores.

Single benzene fluorophores (SBFs) have garnered significant research attention due to their ease of preparation, seamless diffusion into biological samples, and low molecular weight. Accurately predicting the molecular photophysical properties, specifically the maximum absorption and emission wavelengths, is pivotal in advancing functional SBFs. In this study, we introduce a machine-learning model to estimate the maximum absorption and emission wavelengths of SBFs precisely. This model leverages a Full Connect Neural Network and computational chemistry and is tailored to address the challenge

Photoinduced Copper-Catalyzed Asymmetric C(sp3 )-H Alkynylation of Cyclic Amines by Intramolecular 1,5-Hydrogen Atom Transfer.

The development of a mild and general method for C(sp3 )-H functionalization of cyclic amines has been an ongoing challenge. In this work, we describe the copper-catalyzed enantioselective C(sp3 )-H alkynylation of unactivated cyclic 2-iodo-benzamide under photo-irradiation by intramolecular 1,5-hydrogen atom transfer (HAT). The employment of a new bisoxazoline diphenylamine ligand, in conjunction with 1,1'-bi-2-naphthol, which significantly improved the reduction potential of the copper complex, was the key to success of this chemistry. Mechanistic and computational studies supported that the

Syntheses, biological activities and SAR studies of novel carboxamide compounds containing piperazine and arylsulfonyl moieties.

A series of novel carboxamide compounds 19a-19j, 20a-20j and 22a-22d containing piperazine and arylsulfonyl moieties have been synthesized. The bioassay results showed that some compounds exhibited favorable herbicidal activities against dicotyledonous plants and many of them possessed excellent antifungal activities. Among 24 novel compounds, some showed superiority over the commercial fungicides Chlorothalonil, Dimethomorph, Thiophanate-methyl, Iprodione, and Zhongshengmycin at 500 mg/L concentration. Some compounds also exhibited high KARI inhibitory activity at 100 μg/mL concentration and

Computational Study of Gas-Phase Reactions to Produce Sulfite Esters: A New Class of Sulfur Containing Molecules in the Atmosphere?

We use computational quantum chemistry to study gas-phase reactions of aldehydes with water and sulfur dioxide. We find that a new reaction leading to the formation of previously nearly unstudied sulfite esters is possible with a comparatively low barrier. The reaction can be catalyzed by the presence of additional hydrogen bonding molecules, including water, nitric acid, and sulfuric acid. Our calculations also show that the binding energy between nitrate ions and sulfite esters should be high enough to enable their detection by nitrate ion chemical ionization mass spectrometry (CIMS). New mechanistic frontiers in carbohydrate-active enzymes revealed by QM/MM simulations.

Carbohydrate-active enzymes (CAZymes) catalyze reactions central to glycan degradation, biosynthesis, and remodeling. Although their mechanisms have traditionally been described using a limited set of canonical models, recent studies are revealing a broader and more dynamic mechanistic landscape. We discuss how QM/MM-based enhanced-sampling methods have expanded our understanding of CAZyme catalysis by uncovering substrate-assisted pathways, complex proton-transfer processes, unconventional catalytic residues, and the roles of substrate distortion and enzyme dynamics in shaping reactivity. OM-sqPCR enables one-step multiplex quantification of circulating small-RNA subclasses for colorectal cancer classification.

Accurate multiplex quantification of circulating small RNAs (sRNAs) enables comprehensive evaluation of their diagnostic potential. Here, we develop OM-sqPCR, a closed-tube, one-step multiplex RT-qPCR platform that simultaneously quantifies microRNAs, transfer RNA-derived sRNAs (tsRNAs), and ribosomal RNA-derived sRNAs (rsRNAs). By integrating optimized reaction chemistry for short RNA templates with computational primer-probe design, OM-sqPCR enables coordinated amplification of multiple sRNA subclasses in a single reaction. Applied to a multicenter cohort of 651 participants, OM-sqPCR identi

Unveiling the Dual Nature of the Smallest Aqueous Ferrocene Complex.

The structure of the ferrocene-water ((Cp)2Fe:H2O) complex has been characterized by combining state of the art rotational spectroscopy and computational chemistry. Two structures have been retrieved experimentally where water interacts with two distinguished binding sites, the exo Cp π cloud and the planetary-like planar orbit around iron. The spectra of both conformers exhibit significant averaging effects due to the nearly free rotational dynamics of water. This is in good agreement with the exploration of the potential energy surface of (Cp)2Fe:H2O. For water interacting with the exo Cp π

Microwave-assisted synthesis of new triangulenium dyes for lifetime imaging microscopy (FLIM).

We introduce a novel microwave-assisted method for the rapid and efficient synthesis of triangulenium dyes, enabling the creation of a broader range of these fluorophores, which are characterized by very long fluorescence lifetimes. We prepared different azadioxatriangulenium (ADOTA) and diazaoxatriangulenium (DAOTA) derivatives and thoroughly characterized their photophysical properties. We investigated their solvatochromic properties and the effects of the solvent, aided by computational chemistry results. The new ADOTA and DAOTA dyes were applied in multiple in cellulo microscopy modalities

ChemBounce: a computational framework for scaffold hopping in drug discovery.

SUMMARY: Scaffold hopping is a critical strategy in medicinal chemistry for generating novel and patentable drug candidates. Here, we present ChemBounce, a computational framework designed to facilitate scaffold hopping by generating structurally diverse scaffolds with high synthetic accessibility. Given a user-supplied molecule in SMILES format, ChemBounce identifies the core scaffolds and replaces them using a curated in-house library of over 3 million fragments derived from the ChEMBL database. The generated compounds are evaluated based on Tanimoto and electron shape similarities to ensure

SLICE (SMARTS and Logic In ChEmistry): fast generation of molecules using advanced chemical synthesis logic and modern coding style.

While virtual libraries of synthetically accessible compounds have exploded in size to many billions, our capacity to extract valuable drug leads from these vast databases remains limited by computational resources. To overcome this, we developed SLICE SMARTS and Logic In ChEmistry), a powerful new tool designed for the agile exploration of massive chemical spaces. SLICE enables the fast, "à la carte" generation of virtual compound libraries through chemist-defined reaction chemistries and readily available building blocks. Its user-friendly, no-code graphical interface, the SLICE Designer, al

Comparative study of Sombor index and its various versions using regression models for top priority polycyclic aromatic hydrocarbons.

The aromatic compounds having structural configurations with two or more fused benzene rings are the polycyclic aromatic hydrocarbons (PAHs). Topological indices are valuable tools for studying the structure property relationships of PAHs and also helps in predicting various properties and activities. They find applications widely in computational chemistry, drug design and QSPR studies. This article focuses on analysing the potential predictive index for Sombor index (SO), elliptic Sombor index (ESO), Euler Sombor index (EU), reverse Sombor index (RSO), reverse elliptic Sombor index (RESO) an

Protoglobin-Catalyzed Formation of cis-Trifluoromethyl-Substituted Cyclopropanes by Carbene Transfer.

Trifluoromethyl-substituted cyclopropanes (CF3 -CPAs) constitute an important class of compounds for drug discovery. While several methods have been developed for synthesis of trans-CF3 -CPAs, stereoselective production of corresponding cis-diastereomers remains a formidable challenge. We report a biocatalyst for diastereo- and enantio-selective synthesis of cis-CF3 -CPAs with activity on a variety of alkenes. We found that an engineered protoglobin from Aeropyrnum pernix (ApePgb) can catalyze this unusual reaction at preparative scale with low-to-excellent yield (6-55 %) and enantioselectivit

Harnessing Shape Complementarity for Upgraded Cyclohexane Purification through Adaptive Bottlenecked Pores in an Imidazole-Containing MOF.

Shape complementarity is a biological craft for precisely binding substrates at protein-protein interfaces. An analogy to such a function can be drawn conceptually for crystalline porous solids; yet the manifested entities are rare in reticular chemistry. The bottleneck-shaped pores carved out of a metal-organic framework, Zn(MIBA)2 (aka. MAF-stu-13), can perfectly accommodate benzene molecules. Remarkably, its framework adapts to the optimal guest binding-the enhanced host-guest interactions in the neck in turn minimize the guest-guest repulsion in the pore to the extent it turns into attract

Bimetallic Cooperative Catalysis for Decarbonylative Heteroarylation of Carboxylic Acids via C-O/C-H Coupling.

Cooperative bimetallic catalysis is a fundamental approach in modern synthetic chemistry. We report bimetallic cooperative catalysis for the direct decarbonylative heteroarylation of ubiquitous carboxylic acids via acyl C-O/C-H coupling. This novel catalytic system exploits the cooperative action of a copper catalyst and a palladium catalyst in decarbonylation, which enables highly chemoselective synthesis of important heterobiaryl motifs through the coupling of carboxylic acids with heteroarenes in the absence of prefunctionalization or directing groups. This cooperative decarbonylative metho
