Cross-modality word matching in letter-by-letter readers.

Letter-by-letter readers can match spoken words to printed words they cannot read. Good word-matching ability may be attributable to a priming effect. However, since letter-by-letter readers have intact spelling ability, an alternative hypothesis is that word matching depends on a strategy of scanning printed words for recognizable letters known to be in the target items. In the present study, experimental manipulations that taxed the ability of the two subjects to process key letters (those that distinguished target words from foils) or scan for them caused a decline in word-matching performa

Musical Emotion Modulates Concrete and Abstract Word Processing: Evidence from ERPs.

Background/Objectives: Cross-modal emotional priming effects of music on language have been widely demonstrated, but how musical emotion interacts with different types of conceptual representation remains unclear. This study examined whether emotional musical contexts exert differential influences on the processing of concrete and abstract words, whose processing is thought to depend to different degrees on sensorimotor and affective experience. Methods: Two ERP experiments were conducted using a long stimulus-onset asynchrony priming paradigm with two independent groups of participants. In Ex

Reserpine enhances dendritic cell cross-presentation and strengthens CD8+ T cell-mediated antitumor immunity.

Dendritic cell (DC) cross-presentation is essential for priming tumor-reactive CD8+ T cells, but small-molecule approaches to enhance this process remain limited. A flow-cytometry-based natural-product screen identified reserpine (Res) as a candidate enhancer of antigen cross-presentation. Res increased surface H-2Kb-SIINFEKL presentation in MutuDC cells and Flt3L-derived bone marrow DCs, and Res-conditioned DCs promoted OT-I CD8+ T-cell IFN-γ production and antigen-specific B16-OVA tumor cell killing. In vivo, Res reduced B16 melanoma and MC38 colon tumor growth and improved anti-PD-1 respons

Dynamic remodeling of the transcriptional landscape drives Merkel cell specification and mechanosensory differentiation.

Merkel cells (MCs) are specialized mechanosensory receptor cells essential for touch sensation, exhibiting both epithelial and neuroendocrine characteristics. However, the regulatory processes that shape MC fate remain poorly defined. Using single-cell RNA sequencing, we construct a conserved, triphasic atlas of MC ontogeny comprising an epithelial-like stage, a transitional intermediate phase, and a terminal neuroendocrine state. Importantly, we identify JunB as a priming factor essential for initiating MC differentiation, while Pou4f1 acts as a driver of epithelial-neuroendocrine conversion

An Open Resource for Non-human Primate Imaging.

Non-human primate neuroimaging is a rapidly growing area of research that promises to transform and scale translational and cross-species comparative neuroscience. Unfortunately, the technological and methodological advances of the past two decades have outpaced the accrual of data, which is particularly challenging given the relatively few centers that have the necessary facilities and capabilities. The PRIMatE Data Exchange (PRIME-DE) addresses this challenge by aggregating independently acquired non-human primate magnetic resonance imaging (MRI) datasets and openly sharing them via the Inte

ERα blockade in dendritic cells enhances antigen cross-presentation and induces antitumor CD8+ T cell immunity.

Dendritic cells (DC) dysfunction impedes antitumor immunity, so understanding intrinsic regulators of DC antigen presentation and the resulting activation of antitumor CD8+ T cells could benefit therapy. Here, we use a genome-wide CRISPR screen for antigen presentation-regulating genes in DCs, and identify estrogen receptor α (ERα) as an intrinsic checkpoint inhibiting antigen cross-presentation. ERα genomic deficiency or pharmacological degradation enhances antigen presentation and CD8+ T cell priming in mouse and human DCs. Mechanistically, independently of canonical estrogen signaling, ERα

Chimeric TIM-4 receptor-modified T cells targeting phosphatidylserine mediates both cytotoxic anti-tumor responses and phagocytic uptake of tumor-associated antigen for T cell cross-presentation.

To leverage complementary mechanisms for cancer cell removal, we developed a novel cell engineering and therapeutic strategy co-opting phagocytic clearance and antigen presentation activity into T cells. We engineered a chimeric engulfment receptor (CER)-1236, which combines the extracellular domain of TIM-4, a phagocytic receptor recognizing the "eat me" signal phosphatidylserine, with intracellular signaling domains (TLR2/TIR, CD28, and CD3ζ) to enhance both TIM-4-mediated phagocytosis and T cell cytotoxic function. CER-1236 T cells demonstrate target-dependent phagocytic function and induce

Effective PD-1 checkpoint blockade in bladder cancer model requires tumor-draining lymph nodes and lymphocyte trafficking.

INTRODUCTION: Tumor-draining lymph nodes (tdLNs) are key hubs for antitumor T-cell priming. However, their contribution to PD-1 checkpoint blockade in bladder cancer remains poorly defined. In this study, we investigated whether tdLNs and lymphocyte trafficking are required for effective anti-PD-1 therapy in an orthotopic preclinical model of bladder cancer. METHODS: We used C57BL/6 mice bearing intravesical MB49-luciferase tumors. Lymphocyte trafficking was blocked pharmacologically using the sphingosine-1-phosphate receptor modulator FTY720, or surgically by iliac/obturator and lumbar lymph

Mutation of conserved MHC class I cytoplasmic tyrosine affects CD8+ T cell priming, effector function, and memory response.

The cytoplasmic domain of MHC class I (MHC-I) molecules contains a single, highly conserved tyrosine residue (Y320). In previous work, we found that mice expressing a Y320F-mutated form of H-2Kb had reduced capacity to generate Kb-restricted cytotoxic T lymphocyte (CTL) responses following viral infection, due at least in part to defects in endolysosomal trafficking of H-2Kb and antigen cross-presentation by dendritic cells (DCs). In this study, we investigated whether there are additional, post-presentation dependencies on Y320 for T cell priming. We engineered both human- and mouse-derived a

Intranasal Boosting with Spike Fc-RBD of Wild-Type SARS-CoV-2 Induces Neutralizing Antibodies against Omicron Subvariants and Reduces Viral Load in the Nasal Turbinate of Mice.

The emergence of new immune-evasive severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants and subvariants outpaces the development of vaccines specific against the dominant circulating strains. In terms of the only accepted immune correlate of protection, the inactivated whole-virion vaccine using wild-type SARS-CoV-2 spike induces a much lower serum neutralizing antibody titre against the Omicron subvariants. Since the inactivated vaccine given intramuscularly is one of the most commonly used coronavirus disease 2019 (COVID-19) vaccines in developing regions, we tested the hyp

Expression of inflammation in myocardial tissue of rabbits: comparison of two miniaturized heart-lung machines.

The majority of cases involving the surgical treatment of congenital heart disease require implementation of cardiopulmonary bypass (CPB). However, neonates and infants are particularly prone to serious complications associated with CPB as a result of capillary leak due to cardiovascular failure. These complications are related to the transfusion of foreign blood, the disproportionately large area of contact between the patient's blood and foreign material, as well as the systemic inflammatory response induced by hemolysis. To attenuate these risks, we developed a novel, highly integrative, mi

A temporal role of type I interferon signaling in CD8+ T cell maturation during acute West Nile virus infection.

A genetic absence of the common IFN-α/β signaling receptor (IFNAR) in mice is associated with enhanced viral replication and altered adaptive immune responses. However, analysis of IFNAR(-/-) mice is limited for studying the functions of type I IFN at discrete stages of viral infection. To define the temporal functions of type I IFN signaling in the context of infection by West Nile virus (WNV), we treated mice with MAR1-5A3, a neutralizing, non cell-depleting anti-IFNAR antibody. Inhibition of type I IFN signaling at or before day 2 after infection was associated with markedly enhanced viral

CTL responses to H2-M3-restricted Listeria epitopes.

Cytotoxic T cells (CTL) play a critical role in the murine immune response to Listeria monocytogenes (Listeria). Bacterial antigens are presented to Listeria-specific CTL by products of both conventional, polymorphic MHC class Ia and non-polymorphic MHC class Ib alleles. The H2-M3 class Ib gene product, M3, preferentially presents formylmethionine-initiating (fMet) peptides derived from the N termini of bacterial and mitochondrial proteins. Thus, M3 signals the presence of bacterial invaders to CTL effectors. Listeria-encoded fMet peptide epitopes for H2-M3-restricted CTL have recently been id

Cellular mechanisms underlying beneficial versus detrimental effects of bacterial antitumor immunotherapy.

Bacteria engineered to express tumor antigens as cancer vaccines have produced mixed results. Here, we used an attenuated strain of Listeria monocytogenes (ΔactA, Lm) that lacks tumor antigens to examine the immune response to Lm itself in tumor-bearing mice following intravenous (i.v.), intratumoral (i.t.), or combined i.v. + i.t. Lm delivery. Unexpectedly, i.t. Lm alone recruited neutrophils to tumors, which became immunosuppressive, provided an intracellular reservoir for long-term persistence of Lm in tumors, and promoted tumor growth. In contrast, prior i.v. Lm administration generated an

Endotoxin Tolerance Impinges on T Cell Activation and Chemoattraction in Autoimmune Diabetes.

Gut microbiota may affect the development of autoimmune (type 1) diabetes by differential potency of intestinal species to induce endotoxin tolerance (ET). However, where ET impinges on immune mechanisms underlying autoimmune diabetes is yet incompletely understood. We investigated the effects of lipopolysaccharide (LPS) from E. coli and B. vulgatus, two common intestinal species dominating either in low- or high-incidence countries, on activation and chemoattraction of islet-specific T cells in non-obese diabetic (NOD) mice. Intraperitoneal (i.p.) injection of E. coli LPS induced costimulator

Thiopental--ketamine association and low dose priming with rocuronium for rapid sequence in duction of anaesthesia for elective cesareum section.

Obstetric patients undergoing caesarean section under general anaesthesia require rapid induction due to high risk of aspiration. Rocuronium provides the shortest onset of action of nondepolarizing blocking agents. Onset time can be shortened by the priming principle. Ketamine has been shown to improve intubating conditions when used in association with rocuronium. Even if ketamine crosses the placenta rapidly, it does not produce neonatal depression unless used in doses above 1-1.5 mg x kg(-1). We present a case of elective caesarean section due to pelvic disproportion managed in general anae
