Multi-trait and cross-population genome-wide association studies across autoimmune and allergic diseases identify shared and distinct genetic component.

OBJECTIVES: Autoimmune and allergic diseases are outcomes of the dysregulation of the immune system. Our study aimed to elucidate differences or shared components in genetic backgrounds between autoimmune and allergic diseases. METHODS: We estimated genetic correlation and performed multi-trait and cross-population genome-wide association study (GWAS) meta-analysis of six immune-related diseases: rheumatoid arthritis, Graves' disease, type 1 diabetes for autoimmune diseases and asthma, atopic dermatitis and pollinosis for allergic diseases. By integrating large-scale biobank resources (Bioban

Comparison of life history parameters of two different genetic clusters of Bemisia tabaci MED (Hemiptera: Aleyrodidae) through single and cross mating.

Bemisia tabaci Mediterranean (Gennadius) (Hemiptera: Aleyrodidae) is an economically important insect pest worldwide. Previously, we have reported that most B. tabaci Mediterranean (MED) populations occurring in greenhouse tomatoes in Korea have been displaced from well-differentiated two genetic clusters (C1 and C2) to one (C2) during one-year period. To elucidate factors responsible for this phenomenon, we compared life history parameters of these two different genetic clusters through single and cross mating experiments on two different host plants, cucumber and tobacco, at 26°C. Intrinsic

Cross-resistance, genetics, and realized heritability of resistance to fipronil in the house fly, Musca domestica (Diptera: Muscidae): a potential vector for disease transmission.

Houseflies, Musca domestica (L.), are ubiquitous pests that have the potential to spread a variety of pathogens to humans, poultries, and dairies. Pesticides are commonly used for the management of this pest. Fipronil is a GABA-gated chloride channel-inhibiting insecticide that has been commonly used for the management of different pests including M. domestica throughout the world. Many pests have developed resistance to this insecticide. A field-collected strain of M. domestica was selected with fipronil for continuous 11 generations to assess the cross-resistance, genetics, and realized heri

X-ray repair cross-complementing group 1 (XRCC1) genetic polymorphisms and risk of childhood acute lymphoblastic leukemia: a meta-analysis.

BACKGROUND: Recently, there have been a number of studies on the association between XRCC1 polymorphisms and childhood acute lymphoblastic leukemia (ALL) risk. However, the results of previous reports are inconsistent. Thus, we performed a meta-analysis to clarify the effects of XRCC1 variants on childhood ALL risk. METHODS: A meta-analysis was performed to examine the association between XRCC1 polymorphisms (Arg399Gln, Arg194Trp, and Arg280His) and childhood ALL risk. We critically reviewed 7 studies with a total of 880 cases and 1311 controls for Arg399Gln polymorphism, 3 studies with a tot

Epigenetic signatures and genetic variants associated with muscle strength in postmenopausal women: potential bone muscle cross talk via BMP1 mechanisms.

Muscle strength decline is a hallmark of aging and contributes to frailty and bone deterioration, yet the genomic and epigenomic mechanisms predicting functional strength remain unclear. We applied a multiomics approach to identify genetic and epigenetic signatures of muscle strength variability in postmenopausal women. A total of 141 women aged 50-70 yr underwent functional tests, biochemical analysis, anthropometry, blood pressure assessment, and dual-energy X-ray absorptiometry. Participants were classified into higher- and lower-strength groups based on validated upper and lower limb tests

Shared genetic architecture of psychoactive substance use and pan-cancer: insights from a large‑scale genome‑wide cross‑trait analysis.

BACKGROUND: Psychoactive substance use (PSU) and cancer are frequently observed comorbidities that have reciprocal influences and shared behavioral traits of the affected patients. While, e.g., nicotine and alcohol are major carcinogens in the etiology of lung and head and neck cancers, little is known about a shared overarching genetic architecture of PSU and cancer that may predispose individuals to both illnesses. METHODS: Large-scale genome-wide association study (GWAS) summary data revealed shared genetic architecture between cancer and PSU, including alcohol use dependence (AlcUD) and n

Neuroimmune Genetic Overlap Between Chronic Pruritic Skin Diseases and Major Depressive Disorder: Insights from Cross-Trait GWAS Analysis.

BACKGROUND: Psoriasis (PSO), atopic dermatitis (AD), and urticaria (URT) are chronic inflammatory skin diseases marked by severe pruritus. These conditions are frequently comorbid with psychiatric disorders, especially major depressive disorder (MDD). Despite this clinical overlap, the underlying shared genetic mechanisms driving these comorbidities remain largely unclear. METHODS: We integrated large-scale European-ancestry genome-wide association study (GWAS) summary statistics for PSO, AD, URT, and MDD. Genetic overlap was assessed using linkage disequilibrium score regression (LDSC), high

The shared genetic etiology of autoimmune disorders and interstitial lung disease: insights from large-scale genome-wide cross-trait analysis.

Autoimmune diseases often co-occur with interstitial lung disease (ILD), and ILD is associated with patient prognosis. Research has demonstrated a relationship between autoimmune diseases and ILD; however, the genetic basis underlying this connection is frequently overlooked. Linkage disequilibrium score regression and high-definition likelihood methods were applied to large-scale genome-wide association studies summary-level data sets to assess genetic correlations between 17 autoimmune disorders and ILD. Several functional annotations and tissue-specific analyses were performed to determine

Novel Genetic Variants of CDC25A Significantly Increase Risk of Spermatogenesis Arrest in Men from Bengali Population, India: A Cross-Sectional Study.

BACKGROUND: Idiopathic azoospermia is one of the most common reasons for male infertility, but little is known about its genetic origins. The CDC25A gene, a meiotic core regulator, encodes a phosphatase that triggers the G1/S transition of meiosis. It dephosphorylates and activates CDK2, as well as enhances CDC2-cyclin E, CDK2-cyclin A and CDK1-cyclin B complex formation, which is crucial for chromosome condensation and progression of meiosis. AIM: The aim of this study was to identify individual variants of the CDC25A gene that make men susceptible to idiopathic azoospermia. SETTING AND DES

Investigating the shared genetic architecture between post-traumatic stress disorder and neurodegenerative diseases: a large-scale genomewide cross-trait analysis.

BACKGROUND: Post-traumatic stress disorder (PTSD), the most prevalent psychopathological consequence following traumatic events, profoundly impacts human life amidst global societal pressures. Emerging twin and family studies suggest that individuals with stress-related disorders face an elevated risk of neurodegenerative diseases, yet the genetic underpinnings of this association remain poorly understood. METHODS: Here, we present a comprehensive framework elucidating this genetic basis. Using bivariate causal mixture models (MiXeR), we quantified polygenic overlap between PTSD (N = 1,280,93

Investigating Genetic Overlap between Alzheimer's Disease, Lipids, and Coronary Artery Disease: A Large-Scale Genome-Wide Cross Trait Analysis.

There is evidence to support a link between abnormal lipid metabolism and Alzheimer's disease (AD) risk. Similarly, observational studies suggest a comorbid relationship between AD and coronary artery disease (CAD). However, the intricate biological mechanisms of AD are poorly understood, and its relationship with lipids and CAD traits remains unresolved. Conflicting evidence further underscores the ongoing investigation into this research area. Here, we systematically assess the cross-trait genetic overlap of AD with 13 representative lipids (from eight classes) and seven CAD traits, leveragi

A large-scale genome-wide cross-trait analysis reveals shared genetic architecture between Alzheimer's disease and gastrointestinal tract disorders.

Consistent with the concept of the gut-brain phenomenon, observational studies suggest a relationship between Alzheimer's disease (AD) and gastrointestinal tract (GIT) disorders; however, their underlying mechanisms remain unclear. Here, we analyse several genome-wide association studies (GWAS) summary statistics (N = 34,652-456,327), to assess the relationship of AD with GIT disorders. Findings reveal a positive significant genetic overlap and correlation between AD and gastroesophageal reflux disease (GERD), peptic ulcer disease (PUD), gastritis-duodenitis, irritable bowel syndrome and diver

Genetic studies in inbred BB/Wor rats. Analysis of progeny produced by crossing lymphopenic diabetes-prone rats with nonlymphopenic diabetic rats.

BB/Wor diabetes-prone (DP) rats are lymphopenic and frequently develop insulin-dependent diabetes. Diabetes-resistant (DR) BB/Wor rats are not lymphopenic and become diabetic rarely and at a significantly younger age. To examine the genetic basis for diabetes, lymphopenia, and age at onset of diabetes among inbred BB/Wor rats, we crossed nonlymphopenic diabetic rats with lymphopenic DP animals and studied F1, F2, and backcross progeny. F1 rats were neither diabetic nor lymphopenic. Diabetes (both types) and lymphopenia reappeared among F2 rats, confirming the permissive association of diabetes

Parent-of-origin has no detectable effect on survival days of Marek's disease virus infected White Leghorns.

Marek's Disease (MD) is a neoplastic disease of chickens and remains as a chronic infectious disease that threatens the poultry industry. Improving genetic resistance to MD in poultry is an important long-term goal, which would significantly augment the current control measures against MD and eventually reduce the annual economic loss. In this study, survival patterns of F2 birds from 2 reciprocal crosses were compared to examine possible difference in survival between the reciprocal crosses in response to MD virus (MDV) challenge. A total of 246 and 224 F2 chicks derived from reciprocal cross

Hippocampal commissure defects in crosses of four inbred mouse strains with absent corpus callosum.

It is known that four common inbred mouse strains show defects of the forebrain commissures. The BALB/cJ strain has a low frequency of abnormally small corpus callosum, whereas the 129 strains have many animals with deficient corpus callosum. The I/LnJ and BTBR T+ tf/J strains never have a corpus callosum, whereas half of I/LnJ and almost all BTBR show severely reduced size of the hippocampal commissure. Certain F1 hybrid crosses among these strains are known to be less severely abnormal than the inbred parents, suggesting that the parent strains have different genetic causes of commissure def
