In vitro activation and resultant binding of benzo[a]pyrene to DNA by microsomes from rats fed corn and menhaden oils.

Dietary unsaturated fat is required for maximum induction of the hepatic mixed function oxidases (MFO) responsible for activating carcinogens to forms that may bind covalently to DNA. The aim of this study was to assess the influence of dietary fat type and content on the activities of some enzymes involved in activation and detoxification of the carcinogen benzo[a]pyrene (B[a]P). Modification of these changes by pretreatment with phenobarbital (PB) was also evaluated. Male rats were fed diet devoid of fat or containing 20% corn oil (CO) or 20% menhaden fish oil (MO) for 4 days. PB induced sol

Expression of kisspeptin and its receptor in different functional classes of ovarian follicle in the buffalo (Bubalus bubalis).

Recently, we reported the differential expression of kisspeptinergic system in the bubaline hypothalamus and corpus luteum. Here, we document the expression of kisspeptin (Kp) and its receptor (Kiss1r) in the ovarian follicles of the buffalo with respect to the functional status. Follicles of ≥10 to ≤13 mm diameter (n = 45) were retrospectively categorized into active (n = 18), intermediate (n = 16) and atretic (n = 11) follicles based on the concentrations of intrafollicular progesterone (P4) and estradiol (E2). The P4:E2 ratio was significantly lower in the active follicle (0.43 ± 0.08) than

Critical role of the β isoform of protein kinase C (PKCβ) in angiotensin II-induced oxidative stress in vascular smooth muscle cells.

The aim of this study was to elucidate the role of protein kinase Cβ (PKCβ) in subacute angiotensin II (Ang II)-induced oxidative stress and the mechanism of its effects on vascular smooth muscle cells (VSMCs) using a PKCβ-knockout strategy. Both short-term (30 min) and prolonged (24 h) treatment with Ang II increased reactive oxygen species (ROS) production through NADPH oxidase (NOX) activation, with increased phosphorylation of PKCβ at active site Ser660 in rat primary VSMCs from mesenteric artery. The increases in ROS production and NOX activity were completely abolished in VSMCs of PKCβ-k

Diversity of Babesia spp. in cervid ungulates based on the 18S rDNA and cytochrome c oxidase subunit I phylogenies.

Free ranging ungulates, represented in Europe mostly by several deer species, are important hosts for ticks and reservoirs of tick-borne infections. A number of studies have focused on the prevalence of tick borne pathogens in deer chiefly with the aim to determine their potential role as reservoir hosts for important human and livestock pathogens. However, genetic similarity of Babesia spp. forming a group commonly termed as a clade VI that accommodates the deer piroplasms, complicates this task and has led to the description of a bewildering array of poorly characterised strains. This study

Altered expression of intestinal duodenal cytochrome b and divalent metal transporter 1 might be associated with cardio-renal anemia syndrome.

The interaction among heart failure (HF), chronic kidney disease (CKD), and anemia is called cardio-renal anemia syndrome. The mechanism of anemia in cardio-renal anemia syndrome is complex and remains completely unknown. We have previously reported that impaired intestinal iron transporters may contribute to the mechanism of anemia in HF using in vivo HF model rats. In this study, we assessed intestinal iron transporters in CKD model rats to investigate the association of intestinal iron transporters in the mechanism of cardio-renal anemia syndrome. CKD was induced by 5/6 nephrectomy in Sprag

Babesia banethi sp. nov. in red foxes.

BACKGROUND: Babesia spp. are widespread tick-borne intraerythrocytic protozoa, infecting a broad range of vertebrate hosts. Red foxes are reservoirs of Babesia vulpes, belonging to the Babesia microti-like group (clade I), and play an important role in the epidemiology of canine and wildlife babesiosis. Besides B. vulpes, another species of this genus was molecularly reported in red foxes from Israel and Iraq and provisionally named "Babesia sp. MML-2014"; however, no morphological description of this small Babesia species was provided, preventing a proper species naming. METHODS: Infection w

Humans and great apes cohabiting the forest ecosystem in central african republic harbour the same hookworms.

BACKGROUND: Hookworms are important pathogens of humans. To date, Necator americanus is the sole, known species of the genus Necator infecting humans. In contrast, several Necator species have been described in African great apes and other primates. It has not yet been determined whether primate-originating Necator species are also parasitic in humans. METHODOLOGY/PRINCIPAL FINDINGS: The infective larvae of Necator spp. were developed using modified Harada-Mori filter-paper cultures from faeces of humans and great apes inhabiting Dzanga-Sangha Protected Areas, Central African Republic. Ameliorative impact of aprepitant on cisplatin testicular toxicity: Dual regulation of PI3K/AKT pro-survival signaling and Bax/cytochrome c/caspase-3 apoptotic signaling.

AIM: Cisplatin is a powerful antineoplastic agent utilized for the management of several cancers. Due to its significant deleterious impact on the testis, manifested as long-term infertility and irreversible azoospermia in 50% of individuals, its clinical application is markedly restricted. Therefore, this study focused on the investigation of aprepitant's potential effectiveness against cisplatin-induced testicular injury due to its previously reported efficacy in modulation of inflammatory, pro-survival, and apoptotic signaling. MAIN METHODS: In a 10-day experimental protocol, male Wistar r

Detection and characterization of a compound 1 species from the c-type heme enzyme cytochrome c'β.

Compound 1 is a key intermediate in a large group of enzymes containing heme b including cytochromes (cyt) P450, peroxidases and catalases. The extreme reactivity of this highly oxidized species has made its trapping and characterization challenging. Cyt c'β of the ammonia-oxidizing bacterium Nitrosomonas europaea is an enzyme containing heme c and a member of the P460 superfamily of cytochromes. Herein, a compound 1 species within the P460 superfamily was trapped and spectroscopically characterized with stopped-flow absorption, EPR and variable-field Mössbauer spectroscopy. Artificial cytochrome b: computer modeling and evaluation of redox potentials.

We generated atomic coordinates of an artificial protein that was recently synthesized to model the central part of the native cytochrome b (Cb) subunit consisting of a four-helix bundle with two hemes. Since no X-ray structure is available, the structural elements of the artificial Cb were assembled from scratch using all known chemical and structural information available and avoiding strain as much as possible. Molecular dynamics (MD) simulations applied to this model protein exhibited root-mean-square deviations as small as those obtained from MD simulations starting with the crystal struc

Discovery of new group I-D introns leads to creation of subtypes and link to an adaptive response of the mitochondrial genome in fungi.

Group I catalytic introns are widespread in bacterial, archaeal, viral, organellar, and some eukaryotic genomes, where they are reported to provide regulatory functions. The group I introns are currently divided into five types (A-E), which are themselves distributed into several subtypes, with the exception of group I type D intron (GI-D). GI-D introns belong to the rarest group with only 17 described to date, including only one with a putative role reported in fungi, where it would interfere with an adaptive response in the cytochrome b (COB) gene to quinone outside inhibitor (QoI) fungicide

Sequence variability in four mitochondrial genes among rabbit pinworm (Passalurus ambiguus) isolates from different localities in China.

Passalurus ambiguus is a common pinworm which parasitizes in the caecum and colon of rabbits. This study examined genetic variability among P. ambiguus isolated from naturally infected rabbits in four different provinces in China. The partial mitochondrial (mt) cytochrome c oxidase subunit 1 (pcox1), cytochrome b (pcytb) and NADH dehydrogenase subunits 1 and 5 (pnad1 and pnad5) were amplified separately from individual nematodes by PCR and sequenced. The results showed that pcox1, pcytb, pnad1 and pnad5 were 714, 663, 645 and 546 bp in length, respectively. The intra-specific sequence variatio

Assessing the diversity and distribution of potential intermediate hosts snails for urogenital schistosomiasis: Bulinus spp. (Gastropoda: Planorbidae) of Lake Victoria.

BACKGROUND: The Lake Victoria basin is one of the most persistent hotspots of schistosomiasis in Africa, the intestinal form of the disease being studied more often than the urogenital form. Most schistosomiasis studies have been directed to Schistosoma mansoni and their corresponding intermediate snail hosts of the genus Biomphalaria, while neglecting S. haematobium and their intermediate snail hosts of the genus Bulinus. In the present study, we used DNA sequences from part of the cytochrome c oxidase subunit 1 (cox1) gene and the internal transcribed spacer 2 (ITS2) region to investigate Bu
