Comparison of the Life Cycles of Genetically Distant Species C and Species D Human Adenoviruses Ad6 and Ad26 in Human Cells.

UNLABELLED: Our understanding of adenovirus (Ad) biology is largely extrapolated from human species C Ad5. Most humans are immune to Ad5, so lower-seroprevalence viruses like human Ad6 and Ad26 are being tested as therapeutic vectors. Ad6 and Ad26 differ at the DNA level by 34%. To better understand how this might impact their biology, we examined the life cycle of the two viruses in human lung cells in vitro. Both viruses infected A549 cells with similar efficiencies, executed DNA replication with identical kinetics within 12 h, and began killing cells within 72 h. While Ad6-infected cells re

Synthetic Integration of an FCS into Coronaviruses-Hype or an Unresolved Biorisk? An Integrative Analysis of DNA Repair, Cancer Research, Drug Development, and Escape Mutant Traits.

A 19 nt fragment that spans the SARS-CoV-2 furin cleavage site (FCS) is identical to the reverse complement of a proprietary human DNA repair gene sequence. Rather than interpreting this overlap as evidence of a laboratory event, this article uses it as a theoretical springboard to explore underappreciated biorisk concerns, specifically in the context of cancer research. Although they are RNA viruses, coronaviruses are capable of hijacking host DNA damage response (DDR) pathways, exploiting nuclear functions to enhance replication and evade innate immunity. Under selective pressures (antiviral

Tripartite Motif 22 (TRIM22) protein restricts herpes simplex virus 1 by epigenetic silencing of viral immediate-early genes.

Intrinsic resistance is a crucial line of defense against virus infections, and members of the Tripartite Ring Interaction Motif (TRIM) family of proteins are major players in this system, such as cytoplasmic TRIM5α or nuclear promyelocytic leukemia (PML/TRIM19) protein. Previous reports on the antiviral function of another TRIM protein, TRIM22, emphasized its innate immune role as a Type I and Type II interferon-stimulated gene against RNA viruses. This study shows that TRIM22 has an additional intrinsic role against DNA viruses. Here, we report that TRIM22 is a novel restriction factor of HS…

Blowing Off Steam: Virus Inhibition of cGAS DNA Sensing.

Detection of viral DNA is essential for eliciting mammalian innate immunity. However, viruses have acquired effective mechanisms for blocking host defense. Indeed, in this issue of Cell Host & Microbe, Wu et al. (2015) discover a herpesviral strategy for inhibiting the prominent host sensor of viral DNA, cGAS.

[Expression of orthopoxvirus DNA sequences in Escherichia coli cells].

We observed the expression of recombinant plasmids genes containing ectromelia virus DNA fragments in E. coli minicells. Using plasmids with vaccinia or ectromelia viruses DNA fragments inserted upstream of lacZ gene we showed that certain orthopoxvirus genome fragments carry out a promoter-like function in bacterial cells.

Two hytrosaviruses, MdSGHV and GpSGHV, induce distinct cytopathologies in their respective host insects.

Recently, a new virus family (Hytrosaviridae) was proposed for double-stranded DNA viruses that cause salivary gland hypertrophy in their dipteran hosts. The two type species, MdSGHV and GpSGHV, induce similar gross pathology and share several morphological, biological, and molecular characteristics. This histological study revealed profound differences in the cytopathology of these viruses supporting their previously proposed placement in different genera.

Electron microscopic autoradiographic studies on dynamics and location of DNA synthesis of duck plague virus.

Electron microscopic autoradiographic studies on the dynamics and location of DNA synthesis by means of incorporation of 8H-thymidine during the replication of duck plague virus (DPV) revealed that the duration of DNA synthesis of DPV was rather long. The replication of viral DNA occurred simultaneously with the assembly procedure of nucleocapsids, the maturation and release of viruses. DNA synthesis of DPV occurred in the matrix with lower electron density in the nucleus. The replicated viral DNA accumulated in the viroplast with high electron density where the assembly of nucleocapsids occur

ICTV Virus Taxonomy Profile: Bacilladnaviridae 2026.

Bacilladnaviridae is a family of viruses with single-stranded, circular DNA genomes that range from 3.4 to 6 kb and contain a short double-stranded region of 0.5-1 kb. The genome is replicated by the rolling-circle mechanism. Bacilladnavirids encode a jelly-roll capsid protein which forms non-enveloped icosahedral capsids with a T=3 symmetry. All cultured bacilladnavirids infect diatoms, a group of unicellular algae (Stramenopiles), and cause lysis of the host cells. This is a summary of the International Committee on Taxonomy of Viruses (ICTV) Report on the family Bacilladnaviridae, which is

Looking at protists as a source of pathogenic viruses.

In the environment, protozoa are predators of bacteria and feed on them. The possibility that some protozoa could be a source of human pathogens is consistent with the discovery that free-living amoebae were the reservoir of Legionella pneumophila, the agent of Legionnaires' disease. Later, while searching for Legionella in the environment using amoeba co-culture, the first giant virus, Acanthamoeba polyphaga mimivirus, was discovered. Since then, many other giant viruses have been isolated, including Marseilleviridae, Pithovirus sibericum, Cafeteria roenbergensis virus and Pandoravirus spp. T

Real-time microchip PCR for detecting single-base differences in viral and human DNA.

This report describes real-time 5' nuclease PCR assays to rapidly distinguish single-base polymorphism using a battery-powered miniature analytical thermal cycling instrument (MATCI). Orthopoxviruses and the human complement component C6 gene served as targets to demonstrate the feasibility of using the MATCI for diagnosis of infectious diseases and genetic disorders. In the Orthopoxvirus assay, consensus Orthopoxvirus PCR primers were designed to amplify 266-281 base-pair (bp) segments of the hemagglutinin (HA) gene in camelpox, cowpox, monkeypox, and vaccinia viruses. A vaccinia virus-specif

Virus Size Analysis by Gas-Phase Mobility Measurements: Resolution Limits.

Electrospraying (ES) dissolved viral particles, followed by charge reduction and size analysis with a differential mobility analyzer (DMA), offers a flexible size-analysis tool for small particles in solution. The technique relies on pioneering work by Kaufman and colleagues, commercialized by TSI, and often referred to as GEMMA. However, viral studies with TSI's GEMMA have suffered from limited resolving power, possibly because of imperfections in either the instrument (DMA or charge reduction) or the sample solution preparation. Here, we explore the limits of the resolution achievable by GEM

Involvement of Follicular Patency in the Ovarian Developmental Block in Virus-infected, MdSGHV, House Flies, Musca domestica (Diptera: Muscidae).

The Musca domestica salivary gland hypertrophy virus (MdSGHV) is known to have marked effects on the female Musca domestica L. (or common house fly) reproductive system, particularly regarding the size and functionality of the ovaries. Examination of the terminal ovarian follicles can help determine if and how MdSGHV mechanistically causes the block in ovarian development. In this study, terminal ovarian follicle lengths were measured and monitored for patency using Trypan blue dye staining. We examined the effect of MdSGHV infection on female house fly ovarian follicles and attempted to rescu

Prevalence of SGHV among tsetse species of economic importance in Tanzania and their implication for SIT application.

Sterile Insect technique is an important component in area-wide integrated tsetse control. The presence of the salivary glands hypertrophy virus (SGHV) in the wild tsetse, which are the seeds for colony adaptations in the laboratory has become a stumbling block in establishing and maintaining colonies in the laboratory. The virus is transmitted both vertically (in the wild) and horizontally (in the laboratory). However, its prevalence is magnified in the laboratory as a result of the use of in vitro membrane feeding regimen. Fly species of Glossina fuscipes fuscipes, G. pallidipes, G. morsitan

[Detection of herpesvirus markers in patients with precancerous lesions of the lungs and stomach].

Blood-serum concentrations of antibodies to such viruses as herpes simplex virus, cytomegalovirus and Epstein--Barr virus have been assayed versus frequency of viral DNA detection in blood and saliva suspensions from patients with lung and stomach pre-cancers. Condition of local immunity of oral mucosa was assessed on the basis of saliva levels of secretory IgA and lysozyme. It was found that as local immunity of oral mucosa deteriorated, irrespective of pre-cancer localization, high titers of antiviral antibodies (class G) were established. The frequency of herpes DNA was much higher in lymph

Role of TLR9 in Oncogenic Virus-Produced Cancer.

Nucleic acid recognition by toll-like receptor 9 (TLR9) initiates signaling pathways that regulate the production of proinflammatory cytokines or type I interferons, as well as many other molecules required to initialize the immune response. The use of synthetic oligodeoxynucleotides (ODNs) has been crucial to emulate the recognition of DNA sequences by TLR9. Furthermore, ODN administration to mice has shown to confer protection against a wide range of viral, bacterial, and parasitic pathogens. In contrast, oncogenic DNA viruses like hepatitis B virus, Epstein-Barr virus, and human papilloma v…
