The use of electrodermal activity in pulpal diagnosis and dental pain assessment.

AIMS: To explore whether electrodermal activity (EDA) can serve as a complementary tool for pulpal diagnosis (Aim 1) and an objective metric to assess dental pain before and after local anaesthesia (Aim 2). METHODOLOGY: A total of 53 subjects (189 teeth) and 14 subjects (14 teeth) were recruited for Aim 1 and Aim 2, respectively. We recorded EDA using commercially available devices, PowerLab and Galvanic Skin Response (GSR) Amplifier, in conjunction with cold and electric pulp testing (EPT). Participants rated their level of sensation on a 0-10 visual analogue scale (VAS) after each test. We

Effect of Orthodontic Tooth Movement on Pulp Vitality in Previously Traumatized Teeth: A Prospective Cohort Study.

INTRODUCTION: Dental trauma commonly affects the anterior teeth of young individuals, often leading to pulpal compromise through neurovascular damage. Orthodontic tooth movement (OTM) may further stress these teeth, thereby increasing the risk of loss of vitality. This prospective cohort study aimed to assess the impact of OTM on pulp vitality in previously traumatized permanent anterior teeth compared to non-traumatized contralateral controls. MATERIALS AND METHODS: Sixty patients (mean age: 16.5 ± 3.2 years) participated, each providing one traumatized anterior tooth (traumatized group) and

Artificial intelligence in the detection of dental pulp calcifications: a scoping review.

BACKGROUND: Dental pulp calcifications, including pulp stones and diffuse calcific changes, can complicate endodontic access, canal negotiation, and treatment planning. Artificial intelligence may support radiographic detection of these findings, but the available evidence remains limited and methodologically heterogeneous. This scoping review mapped peer-reviewed studies that applied artificial intelligence to detect, classify, or segment dental pulp calcifications on two-dimensional radiographs and cone-beam computed tomography. METHODS: A scoping review was conducted in accordance with the

Direct capping with four different materials in humans: histological analysis of odontoblast activity.

Pulp inflammation in restored teeth is mainly due to the presence of bacteria or bacterial products introduced by microleakage around the restoration or to the material toxicity. Recent knowledge has permitted a precise identification of the risks for pulpal irritation associated with adhesive materials and procedures. The purpose of this work was to evaluate the cellular events that occur in direct pulp exposure capped using different materials. Twenty-four vital teeth without caries, scheduled for extraction for orthodontic reasons, were selected. After a control of the hemostasis, each pulp

Lipopolysaccharide-induced DNA damage response activates nuclear factor κB signalling pathway via GATA4 in dental pulp cells.

AIM: To investigate the role of GATA-binding protein 4 (GATA4) in the inflammatory response induced by DNA double-strand breaks (DSBs) in human dental pulp cells (hDPCs). METHODOLOGY: Lipopolysaccharide (LPS) was used for stimulating inflammation in dental pulp tissue in vivo and hDPCs in vitro. Expression levels of GATA4 and γ-H2A.X (a marker for DSBs) were detected at different stages of pulpitis in a rat model and human pulp tissues by immunohistochemistry. Real-time quantitative polymerase chain reaction and Western blot were performed to assess expression of GATA4 and γ-H2A.X and the act

Mesenchymal stem cells derived from inflamed dental pulpal and gingival tissue: a potential application for bone formation.

BACKGROUND: Chronic periodontal disease is an infectious disease consisting of prolonged inflammation of the supporting tooth tissue and resulting in bone loss. Guided bone regeneration procedures have become common and safe treatments in dentistry, and in this context dental stem cells would represent the ideal solution as autologous cells. In this study, we verified the ability of dental pulp mesenchymal stem cells (DPSCs) and gingival mesenchymal stem cells (GMSCs) harvested from periodontally affected teeth to produce new mineralized bone tissue in vitro, and compared this to cells from he

Pilot spatial transcriptomics of dental pulpitis suggests immune-fibroblast profiling linked to reversibility.

BACKGROUND: Diagnostic protocols in endodontics rely heavily on subjective pain assessments and sensibility testing, which may not reflect the underlying histopathological and molecular state of the dental pulp. This limitation can contribute to discordance between clinical diagnosis (reversible (RP) vs. irreversible (IP) pulpitis) and tissue biology, potentially leading to overtreatment of teeth that might respond to conservative, pulp-preserving approaches. Spatial transcriptomics offers an opportunity to explore the cellular and molecular heterogeneity of inflamed pulp in a spatially resolv

[Orthognathic surgery. Study of nerve injuries].

The correction of oral and maxillo-facial mal-formations causes lesions to sensitive nerves due to the invasive and traumatic surgery techniques required. Generally neuro-sensory and cutaneous-mucosal defects are reversible, with full recovery within 6-12 months. In a follow-up study (2 and 8 years) we found altered tactile and pain sensitivity in 43% of patients at subjective evaluation, with objective alteration in 17% of the group. Dental sensitivity evaluated by a pulp test showed an increase in for all teeth tested; values were higher for superior than for inferior teeth. Despite the incr

Ultrastructural evaluation of adverse effects on dentine formation from systemic fluoride application in an experimental mouse model.

AIM: Fluoride is widely used in dentistry for its caries prevention. To reduce dental caries, the optimal fluoride concentration of public water supplies in the United States is 0.7 ppm. However, excessive systemic fluoride consumption can lead to dental/enamel fluorosis. Numerous studies have explored the effects of fluoride on enamel and enamel-forming cells. However, research on systemic fluoride's impact on dentine is limited, particularly the effect of fluoride on the structure of the dentine-pulp complex. Therefore, this study aimed to identify how excessive fluoride affects dentine micr

Endodontic and Orthodontic Management of a Fused Mandibular Incisor and Supernumerary: A Case Report.

Dental fusion is a rare anomaly that can present unique challenges with clinical treatment. This report outlines a case of an 11-year-old who presented with fusion of a mandibular lateral incisor with a supernumerary. At presentation, the teeth were fully mature and normally responsive to pulp sensibility testing. Assessment with CBCT confirmed that the majority of the root canal system and pulp chambers were separate, with a single point of communication between both tooth units in the mid-crown. Removal of one of the teeth was requested to facilitate the patient's orthodontic treatment plan,
