Sharp increase in illicit drug use in Greece: trends from a general population survey on licit and illicit drug use.

The study aims to follow trends in the prevalence of licit and illicit substance use between 1984 and 1998 in the Greek general population. Data are drawn from two nationwide cross-sectional household surveys. Probability samples consisted of 4,297 (1984) and 3,759 (1998) 12- to 64-year-old respondents. Regular tobacco smoking in the total sample increased in women and decreased in men while alcohol consumption remained at similar levels. The use of illicit drugs showed a 3-fold increase since 1984 reaching a lifetime prevalence of 19% in males and 6% in females. In contrast, a significant dec

Successful Management of Generalized Drug-Influenced Gingival Enlargement: A Clinical Case Report.

Drug-influenced gingival enlargement (DIGE) is a clinically significant adverse effect of medications such as phenytoin and may be exacerbated by plaque-induced inflammation. The interaction between drug-related alterations in gingival connective-tissue metabolism and local inflammatory factors can result in marked gingival enlargement, compromising oral hygiene and periodontal health. We present a case of a 40-year-old man with a long history of phenytoin use who developed generalized, bulbous, and fibrotic gingival enlargement associated with periodontal pocketing. The management of DIGE inc

TechBio 3.0 closes the drug discovery loop with multimodal AI and generative chemistry.

Artificial intelligence (AI) has transformed molecular prediction and design, yet these advances remain only partially integrated with experimental discovery. We define TechBio 3.0 as an emerging paradigm for closed-loop drug discovery that connects multimodal molecular representations, generative chemistry, and automated experimentation. We examine its early preclinical and clinical evidence, identify remaining technical and translational barriers, and argue that this convergence is beginning to define biotechnology's next era.

Thermoplastic elastomer based microfluidic gradient generator for cell culture and drug testing.

Microfluidic systems are increasingly utilized in preclinical research, however, developing platforms that are compatible with large-scale manufacturing remains a critical challenge. In this regard, we show that thermoplastic elastomer (TPE)-based microfluidic chips could be a viable alternative to traditional poly(dimethylsiloxane) (PDMS) devices for drug testing applications. Microfluidic concentration gradient generators are essential components in these systems, enabling local spatial and temporal control of biochemical stimuli for applications such as cellular response studies and drug sc

Topoisomerase I/II inhibitors: from established drugs to next-generation therapeutics.

Topoisomerases are crucial enzymes that control the DNA topology in replication, transcription, recombination, and chromosomal segregation, and therefore are validated as the targets of anticancer therapy. Topoisomerase-targeting agents are mechanistically divided into poison-type and catalytic-type inhibitors. Poison-type inhibitors stabilize covalent enzyme-DNA cleavage complexes, whereas catalytic inhibitors prevent enzymatic turnover without stabilizing cleavage complexes. Both classes may selectively target either topoisomerase I or topoisomerase II isoforms. Established and clinically us

Barrier-Oriented Design of Next-Generation Polymeric Nanocarriers for Targeted Drug Delivery.

Targeted drug delivery remains difficult because multiple biological barriers interfere with the stable transport of therapeutics to the site of action. Polymeric nanocarriers have gained broad attention as delivery platforms since their composition and surface properties can be adjusted to improve circulation behavior and cellular delivery. This review discusses the major biological barriers involved in targeted drug delivery and describes how polymeric nanocarriers are engineered to overcome them. Major carrier types, including polymeric nanoparticles and micelles, are considered with emphas

Ultrahigh-DAR antibody-drug conjugates: advancing the next generation of targeted therapy.

Conventional antibody-drug conjugates (ADCs) are typically limited to drug-to-antibody ratios (DARs) of ≤8 because of restricted conjugation sites and unfavorable physicochemical properties at higher DARs, including increased hydrophobicity, aggregation, and rapid systemic clearance. Consequently, most clinically approved ADCs rely on highly potent cytotoxic payloads (e.g. IC50 < 1 nM), which can result in significant off-target toxicities. This mini-review summarizes recent advances in next-generation ultrahigh-DAR antibody therapeutics, with a particular focus on two emerging platforms: anti

Insights on the Generation of Polymeric Drug Carriers via Supercritical Fluid-Assisted Technologies.

Polymeric drug carriers have emerged as versatile platforms for improving therapeutic efficacy, drug stability and controlled and sustained release of active pharmaceutical ingredients. Conventional manufacturing techniques often rely on high processing temperatures, utilization of organic solvents and consequent purification procedures that may compromise morphology, encapsulation efficiency and carrier biocompatibility. Supercritical fluid technologies overcome the limitations of conventional methods and emerge as environmentally friendly, sustainable and flexible alternatives to produce adv

SynFrag: Synthetic Accessibility Predictor Based on Fragment Assembly Generation in Drug Discovery.

AI-driven molecular generation encounters a "generation-synthesis gap": most computationally designed molecules cannot be synthesized in laboratories, limiting AI-assisted drug design (AIDD) applications. Current approaches to assess synthetic accessibility (SA) include computer-aided synthesis planning (CASP) tools that perform retrosynthetic searches and machine learning-based SA prediction models that provide rapid scoring. CASP tools are computationally expensive for high-throughput screening, while existing SA prediction models may lack chemical synthesis logic or exhibit variable perform

Protocols for MCF-7 derived mammosphere generation and anti-cancer drug testing.

Cancer research is a rapidly expanding field, requiring new methods and techniques to investigate the complex biology of the disease and its characteristics. A pivotal aspect of this research is the evaluation of the effects and efficacy of therapies. Here, we describe a simple protocol to generate mammospheres from MCF-7 cells, a useful model of breast cancer stem cells to evaluate cell death after therapy treatment. This subpopulation of cells, found in all types of tumors, is critically involved in therapeutic response as well as in disease recurrence and relapse, and therefore requires par

Generational shifts in thiolated cyclodextrins: Toward more efficient drug delivery systems.

Thiolated cyclodextrins represent a rapidly evolving class of multifunctional excipients that extend the role of cyclodextrins beyond solubilization toward active participation in mucosal drug delivery. By integrating the inclusion complexation capacity of cyclodextrins with the reactivity of thiol groups, these systems can enhance drug residence time on target tissues, epithelial permeation, and cellular uptake. This review discusses the generational evolution of thiolated cyclodextrins, from first-generation derivatives bearing free sulfhydryl groups to advanced S-protected systems designed

Next-generation drug delivery systems for aspergillosis: Overcoming barriers in antifungal therapy.

Aspergillosis is a group of diseases caused primarily by Aspergillus fumigatus, an opportunistic fungal pathogen recently classified as a critical priority by the World Health Organization (WHO) due to its global impact on morbidity and mortality. The respiratory tract, particularly the nose, sinuses, and lungs, is the primary site of colonization and infection, where aspergillosis manifests in a spectrum of clinical forms, from allergic reactions to invasive disease. Steroidal drugs are commonly used to treat allergic forms, while azole antifungals remain the first-line therapy for invasive a

[Prescription of Benzodiazepines and Z-Drugs for Sleep Disorders in General Practice: A Qualitative Study].

INTRODUCTION: Sleep disorders, particularly insomnia, are a common reason for consultation in general practice. Among the approved treatment options are benzodiazepines (BZ) and benzodiazepine receptor agonists (BZRA). However, these medications carry a risk of dependence. The extent to which the risk of medication dependence influences the treatment decisions of general practitioners (GPs) is unclear. This study aimed to explore the spectrum of awareness regarding dependency potential of these drugs and prescribing practices. METHODS: As part of an exploratory qualitative interview study, 17

PED: a novel predictor-encoder-decoder model for Alzheimer drug molecular generation.

Alzheimer's disease (AD) is a gradually advancing neurodegenerative disorder characterized by a concealed onset. Acetylcholinesterase (AChE) is an efficient hydrolase that catalyzes the hydrolysis of acetylcholine (ACh), which regulates the concentration of ACh at synapses and then terminates ACh-mediated neurotransmission. There are inhibitors to inhibit the activity of AChE currently, but its side effects are inevitable. In various application fields where Al have gained prominence, neural network-based models for molecular design have recently emerged and demonstrate encouraging outcomes. H

Drug-induced change in transmitter identity is a shared mechanism generating cognitive deficits.

Cognitive deficits are a long-lasting consequence of drug use, yet the convergent mechanism by which classes of drugs with different pharmacological properties cause similar deficits is unclear. We find that both phencyclidine and methamphetamine, despite differing in their targets in the brain, cause the same glutamatergic neurons in the medial prefrontal cortex to gain a GABAergic phenotype and decrease their expression of the vesicular glutamate transporter. Suppressing the drug-induced gain of GABA with RNA-interference prevents the appearance of memory deficits. Stimulation of dopaminergi

Crosstalk between GABAA receptors in astrocytes and neurons triggered by general anesthetic drugs.

General anesthetic drugs cause cognitive deficits that persist after the drugs have been eliminated. Astrocytes may contribute to such cognition-impairing effects through the release of one or more paracrine factors that increase a tonic inhibitory conductance generated by extrasynaptic γ-aminobutyric acid type A (GABAA) receptors in hippocampal neurons. The mechanisms underlying this astrocyte-to-neuron crosstalk remain unknown. Interestingly, astrocytes express anesthetic-sensitive GABAA receptors. Here, we tested the hypothesis that anesthetic drugs activate astrocytic GABAA receptors to in

Effects of the second-generation antipsychotic drugs aripiprazole and olanzapine on human adipocyte differentiation.

Second-generation antipsychotics (SGAs), used as the cornerstone treatment for schizophrenia and other mental disorders, can cause adverse metabolic effects (e.g. obesity and type 2 diabetes). We investigated the effects of SGAs on adipocyte differentiation and metabolism. The presence of therapeutic concentrations of aripiprazole (ARI) or its active metabolite dehydroaripiprazole (DARI) during human adipocyte differentiation impaired adipocyte glucose uptake while the expression of gene markers of fatty acid oxidation were increased. Additionally, the use of a supra-therapeutic concentration
