Burrowing mayfly Ephemera orientalis (Ephemeroptera: Ephemeridae) as a new test species for pesticide toxicity.

The potential of mayfly Ephemera orientalis McLachlan eggs and first-instar larvae in ecotoxicological testing was investigated. Both stages of E. orientalis showed high tolerance to various environmental variables, such as water temperature, pH, water hardness, and dissolved organic carbon. Toxicological assays were conducted with three insecticides (emamectin benzoate, endosulfan, and cypermethrin), one fungicide (mancozeb), and one herbicide (paraquat dichloride). The two toxicity endpoints for the assay were the 14-day egg median hatching rate (EHC50) in static and renewal exposure systems

Toxicity of seven priority hazardous and noxious substances (HNSs) to marine organisms: Current status, knowledge gaps and recommendations for future research.

Shipping industry and seaborne trade have rapidly increased over the last fifty years, mainly due to the continuous increasing demand for chemicals and fuels. Consequently, despite current regulations, the occurrence of accidental spills poses an important risk. Hazardous and noxious substances (HNSs) have been raising major concern among environmental managers and scientific community for their heterogeneity, hazardous potential towards aquatic organisms and associated social-economic impacts. A literature review on ecotoxicological hazards to aquatic organisms was conducted for seven HNSs: a

Assessment of RNA extraction protocols from cladocerans.

The usage of cladocerans as non-model organisms in ecotoxicological and risk assessment studies has intensified in recent years due to their ecological importance in aquatic ecosystems. The molecular assessment such as gene expression analysis has been introduced in ecotoxicological and risk assessment to link the expression of specific genes to a biological process in the cladocerans. The validity and accuracy of gene expression analysis depends on the quantity, quality and integrity of extracted ribonucleic acid (RNA) of the sample. However, the standard methods of RNA extraction from the cl

Assessment of the environmental impact of an abandoned mine using an integrative approach: A case-study of the "Las Musas" mine (Extremadura, Spain).

The mine abandonment is generally associated with the release of potentially toxic metals into the environment, which may depend on metals speciation, soil properties and climate conditions. The goal of the present work was to assess the environmental impact of the abandoned Pb-Zn mine "Las Musas" (Spain) using an integrative approach. The impact on soils and surface waters was performed using: chemical parameters, quantification of potentially toxic metals (Cd, Cu, Pb and Zn), and ecotoxicological responses using lethal and sub-lethal bioassays with organisms' representative of different trop

Attenuation of MEHP-induced hepatotoxicity and gut microbiota dysbiosis by Lactobacillus brevis Lb-0401 in Carassius auratus.

Mono-2-ethylhexyl phthalate (MEHP) is one of the major biologically active metabolites of phthalate plasticizers that pose a health threat to aquatic animals. In this study, we investigated the ecotoxicological effects of MEHP on liver damage and gut microbiota in crucian carp (Carassius carassius) and the mitigating effects of Lactobacillus brevis Lb-0401 as a dietary supplement. The results showed that MEHP induced hepatotoxicity and increased serum AST, ALT and triglyceride levels, while L. brevis Lb-0401 alleviated abnormalities in serum metabolic parameters and increased antioxidant enzym

Ecotoxic emissions generated by illegal burning of household waste.

The practice of burning household waste including different types of plastic is illegal in Hungary, still an existing problem. As environmental consequences are hardly known, this study attempts to give an initial estimation of the ecotoxicity generated during controlled combustion of different waste types. These samples included polystyrene (PS), polyethylene (PE), polypropylene (PP), polyvinyl chloride (PVC), polyurethane (PU), oriented strand board (OSB) and rag (RAG). Ecotoxicological profiling was completed using the following test battery: Vibrio fischeri bioluminescence inhibition assay

The State-of-the Art of Environmental Toxicogenomics: Challenges and Perspectives of "Omics" Approaches Directed to Toxicant Mixtures.

The last decade witnessed extraordinary advances in "omics" methods, particularly transcriptomics, proteomics and metabolomics, enabling toxicologists to integrate toxicokinetics and toxicodynamics with mechanistic insights on the mode-of-action of noxious chemicals, single or combined. The toxicology of mixtures is, nonetheless, a most challenging enterprise, especially for environmental toxicologists and ecotoxicologists, who invariably deal with chemical mixtures, many of which contain unknowns. Despite costs and demanding computations, the systems toxicology framework, of which "omics" is

Corticosterone in American alligator (Alligator mississippiensis) tail scutes: Evaluating the feasibility of using unconventional samples for investigating environmental stressors.

Baseline plasma corticosterone (CORT) concentrations have been widely used to investigate the effects of stressors in wild and captive crocodilians. However, collecting baseline plasma CORT samples from wild crocodilians may be particularly difficult due to the capture and handling protocols used for large individuals. Thus, it may prove beneficial to use recently modified techniques for extracting CORT deposited in keratinized and non-keratinized tissues to better quantify the effects of long-term stress in crocodilians. In this study, we investigated the feasibility of using American alligat

Genotoxic and teratogenic effect of freshwater sediment samples from the Rhine and Elbe River (Germany) in zebrafish embryo using a multi-endpoint testing strategy.

The embryotoxic potential of three model sediment samples with a distinct and well-characterized pollutant burden from the main German river basins Rhine and Elbe was investigated. The Fish Embryo Contact Test (FECT) in zebrafish (Danio rerio) was applied and submitted to further development to allow for a comprehensive risk assessment of such complex environmental samples. As particulate pollutants are constructive constituents of sediments, they underlay episodic source-sink dynamics, becoming available to benthic organisms. As bioavailability of xenobiotics is a crucial factor for ecotoxico

An aquatic toxicology primer for the experimental biologist and guidance for conducting robust stressor exposures.

The field of aquatic toxicology has undergone a paradigm shift in recent decades. The movement away from observational data detailing the concentrations that produce deleterious effects and towards a mechanistic understanding of how toxicants affect organismal physiology has significant advantages for the field. This mechanistic approach allows for the prediction of toxicant effects across species and among toxicants with similar modes of action. Critically, knowledge of the pathways by which toxicants exert their impacts can provide insight into how environmental factors such as temperature,

Ecotoxicity of Folsomia candida induced by 1,2,4-TCB: Insights from multi-level experiments and molecular simulations.

The large-scale production and application of industrial chemicals and pesticides have intensified soil contamination by chlorobenzenes particularly 1,2,4-trichlorobenzene (1,2,4-TCB), thereby posing a serious threat to the safety of soil-dwelling organisms. However, the toxicological impacts of 1,2,4-TCB on soil biota remain poorly characterized. In the present study, the effects of 1,2,4-TCB on soil invertebrates specifically the springtail Folsomia candida were investigated by integrating multiple biomarkers, transcriptomic profiling, and molecular docking analyses. The results of the study

Ferroptosis Induced by Pollutants: An Emerging Mechanism in Environmental Toxicology.

Environmental pollutants have been recognized for their ability to induce various adverse outcomes in both the environment and human health, including inflammation, apoptosis, necrosis, pyroptosis, and autophagy. Understanding these biological mechanisms has played a crucial role in risk assessment and management efforts. However, the recent identification of ferroptosis as a form of programmed cell death has emerged as a critical mechanism underlying pollutant-induced toxicity. Numerous studies have demonstrated that fine particulates, heavy metals, and organic substances can trigger ferropto

Oxidation of metazachlor herbicide by ozone in the gas and aqueous phases: mechanistic, kinetics, and ecotoxicity studies.

The atmospheric and aqueous ozonolysis of metazachlor (MTZ) is investigated using high-level quantum chemical and kinetic calculations (M06-2X/6-311 +  + G(3df,3pd)//M06-2X/6-31 + G(d,p) level of theory). The ozone (O3)-initiated degradation pathways of MTZ under three different mechanisms, namely cycloaddition, oxygen-addition, and single electron transfer (SET), are explored in the temperature range of 283-333 K and 1 atm pressure. As a result, the cycloaddition reaction at the C16C18 double bond of the benzene ring of MTZ is found to be the most dominant channel in the atmosphere with the s

In situ experiments to assess effects of constraints linked to caging on ecotoxicity biomarkers of the three-spined stickleback (Gasterosteus aculeatus L.).

The aim of this study was to evaluate the effects of caging constraints on multiple fish biomarkers used during ecotoxicological studies (biometric data, immune and antioxidant systems, and energetic status). Two of these constraints were linked to caging: starvation and fish density in cages, and one in relation to the post-caging handling: a short transport. Three in situ experiments were conducted with three-spined sticklebacks (Gasterosteus aculeatus). The first experiment compared the effects of three densities (low, medium, and high). The second experiment compared effects of starvation

Development of Comparative Toxicity Potentials of 14 cationic metals in freshwater.

Site-dependent and site-generic Comparative Toxicity Potentials (CTPs) (also known as Characterization Factors (CFs)) were calculated for 14 cationic metals (Al(III), Ba, Be, Cd, Co, Cr(III), Cs, Cu(II), Fe(II), Fe(III), Mn(II), Ni, Pb, Sr and Zn), to be applied in Life Cycle Impact Assessment. CTPs were calculated for 7 EU-archetypes, taking bioavailability and speciation pattern into account. The resulting site-dependent CTPs showed up to 2.4-6.5 orders of magnitude variation across archetypes for those metals that form stable hydroxyl compounds in slightly alkaline waters (Al(III), Be, Cr(I

Ecotoxicity and analysis of nanomaterials in the aquatic environment.

Nanotechnology is a major innovative scientific and economic growth area. However nanomaterial residues may have a detrimental effect on human health and the environment. To date there is a lack of quantitative ecotoxicity data, and recently there has been great scientific concern about the possible adverse effects that may be associated with manufactured nanomaterials. Nanomaterials are in the 1- to 100-nm size range and can be composed of many different base materials (carbon, silicon and metals, such as gold, cadmium and selenium) and they have different shapes. Particles in the nanometer s

Evaluation of aspartame effects at environmental concentration on early development of zebrafish: Morphology and transcriptome1.

The use of aspartame as an artificial sweetener is prevalent in a wide range of everyday food products, potentially leading to health complications such as obesity, diabetes mellitus, autism spectrum disorders, and neurodegeneration. Aspartame has also been detected in natural water bodies at a concentration of 0.49 μg/L, yet research on its ecotoxicological effects on aquatic life remains scarce. This study aimed to investigate the potential negative effects of environmentally relevant concentrations of aspartame on the development of various tissues and organs in zebrafish embryos. We used a

Is blood a reliable indicator of trace metal concentrations in organs of small mammals?

In wildlife ecotoxicology, the rationale for using blood rather than other body fluids or tissues is that sampling blood is a minimally invasive technique without animal mortality, providing both ethical and scientific benefits. To date, few studies are available on the relationships between blood and organ metal concentrations of small mammals living in contaminated sites. The present work aimed to study the relationships between the concentrations of 18 essential and nonessential metals in blood and their concentrations in the liver and kidneys, two accumulation and target organs, in wood mi…
