The branched respiratory system of photosynthetically grown Rhodopseudomonas capsulata.

Various respiratory electron transport activities of Rhodopseudomonas capsulata were studied in membrane fragments prepared from photosynthetically grown cells of a parental strain and two terminal oxidase-defective mutant strains. The NADH and succinate oxidase activities of the mutant having a functional N,N,N1,N1-tetramethyl-p-phenylenediamine oxidase, M6, were consideraly more sensitive to inhibition by either antimycin A or cyanide than the corresponding activities of the mutant lacking a functional N,N,N1,N1-tetramethyl-p-phenylenediamine oxidase, M7. The parental strain, Z-1, but not th

APOE expression and secretion are modulated by mitochondrial dysfunction.

Mitochondria influence cellular function through both cell-autonomous and non-cell autonomous mechanisms, such as production of paracrine and endocrine factors. Here, we demonstrate that mitochondrial regulation of the secretome is more extensive than previously appreciated, as both genetic and pharmacological disruption of the electron transport chain caused upregulation of the Alzheimer's disease risk factor apolipoprotein E (APOE) and other secretome components. Indirect disruption of the electron transport chain by gene editing of SLC25A mitochondrial membrane transporters as well as direc

Cytochrome oxidase activity as a measure of synaptogenesis by multifunctional neuron L7 of Aplysia.

Neuron L7 of the marine mollusc, Aplysia californica, is unique in that it innervates five different target tissues in the animal. We show that when L7 is grown in vitro with two of these targets, that is, muscle cells isolated from the auricle or the gill vein, newly formed L7 neurites contact the muscle cells. Chemical synapses are formed since intracellular stimulation of L7 elicits contraction of individual muscle cells. Interestingly, auricle muscles are also innervated by neuron RBhe and co-cultures of RBhe and auricle muscle cells also exhibit synapse formation. To explore the molecular

Ischemic pre-conditioning preserves brain mitochondrial functions during the middle cerebral artery occlusion in rat.

It is well known that a brief period of ischemia increases tolerance to a subsequent severe ischemic episode. In the present study, bilateral carotid arteries occlusion (BCAO) was applied as pre-conditioning to testify whether this kind of ischemia could preserve the function of mitochondria with the impairment induced by middle cerebral artery occlusion (MCAO) in brain. The activities of respiratory enzyme complex I to IV, mitochondria swelling, membrane potential, and membrane fluidity were investigated. The results showed that the percentage of infarct area decreased greatly due to the isch

[Ultrastructural immunocytochemical localization of electron-transport enzymes in mitochondrial myopathy].

Ultrastructural localization of mitochondrial electron transport enzymes in biopsied muscle from a patient with mitochondrial myopathy was studied. We applied the immunoelectron microscopic technique using colloidal-gold labeled antibodies. The results demonstrated a very distinct and dense labeling by gold particles of complexes I, III, and IV in the inner mitochondrial membrane. The density of gold particles reacted to complex IV was cleanly decreased. This result was correlated with the decreased biochemical activity of complex IV. Labeling on the paracrystalline inclusions was substantiall

Ultrastructure of mouthpart and terminalia in Musca crassirostris (Diptera: Muscidae) with molecular confirmation from Thailand.

Musca crassirostris Stein, 1903 (Diptera: Muscidae) is a medically and veterinary important hematophagous fly whose feeding activity causes significant irritation and blood loss in livestock and is occasionally associated with human myiasis. Although this species is widely distributed and epidemiologically relevant, its feeding-related functional morphology and genital diagnostic characters have remained insufficiently documented at the ultrastructural level. Here, we present the first comprehensive scanning electron microscopy (SEM)-based characterization of the labellar feeding apparatus and

A novel system for assigning the mode of inheritance in mitochondrial disorders using cybrids and rhodamine 6G.

When normal human cultured skin fibroblasts were treated with the fluorescent dye rhodamine 6G (R6G), there was a drastic reduction in numbers of intact mitochondria and electron transport chain enzyme activities, despite the fact that mitochondrial DNA (mtDNA) was still present in treated cells. We used this observation to develop a novel system for generating cybrids. When cultured skin fibroblast cells from a patient with the mitochondrial encephalopathy and ragged-red fibers (MERRF) syndrome harboring the A8344G mtDNA mutation and which showed a severe reduction in cytochrome c oxidase act

Gas regulation of complex II reversal via electron shunting to fumarate in the mammalian ETC.

The electron transport chain (ETC) is a major currency converter that exchanges the chemical energy of fuel oxidation to proton motive force and, subsequently, ATP generation, using O2 as a terminal electron acceptor. Discussed herein, two new studies reveal that the mammalian ETC is forked. Hypoxia or H2S exposure promotes the use of fumarate as an alternate terminal electron acceptor. The fumarate/succinate and CoQH2/CoQ redox couples are nearly iso-potential, revealing that complex II is poised for facile reverse electron transfer, which is sensitive to CoQH2 and fumarate concentrations. A novel mechanism causing imbalance of mitochondrial fusion and fission in human myopathies.

Mitochondrial dynamics play an important role in cellular homeostasis and a variety of human diseases are linked to its dysregulated function. Here, we describe a 15-year-old boy with a novel disease caused by altered mitochondrial dynamics. The patient was the second child of consanguineous Jewish parents. He developed progressive muscle weakness and exercise intolerance at 6 years of age. His muscle biopsy revealed mitochondrial myopathy with numerous ragged red and cytochrome c oxidase (COX) negative fibers and combined respiratory chain complex I and IV deficiency. MtDNA copy number was el

Severe Hyperhomocysteinemia Decreases Respiratory Enzyme and Na(+)-K(+) ATPase Activities, and Leads to Mitochondrial Alterations in Rat Amygdala.

Severe hyperhomocysteinemia is caused by increased plasma levels of homocysteine (Hcy), a methionine derivative, and is associated with cerebral disorders. Creatine supplementation has emerged as an adjuvant to protect against neurodegenerative diseases, due to its potential antioxidant role. Here, we examined the effects of severe hyperhomocysteinemia on brain metabolism, and evaluated a possible neuroprotective role of creatine in hyperhomocysteinemia, by concomitant treatment with Hcy and creatine (50 mg/Kg body weight). Hyperhomocysteinemia was induced in young rats (6-day-old) by treatmen

Separating Faces in ARMS Metabarcoding Improves Marine Biodiversity Monitoring: A Comparison Across Protocols, Experimental Designs and Photographic Surveys.

Monitoring marine biodiversity requires approaches capable of capturing its spatial and temporal complexity. DNA metabarcoding coupled with Autonomous Reef Monitoring Structures (ARMS) is increasingly used for this purpose, yet most applications still pool all sessile fractions and rarely benchmark molecular outputs against photographic observations. Here, we combined photographic analysis with cytochrome c oxidase I (COI) metabarcoding across 10 north-western Mediterranean sites to compare and refine ARMS-based monitoring protocols. We first optimized laboratory procedures (DNA extraction and

The predicted interactome of the human mitochondrial proteome.

Despite the fundamental importance of mitochondria in cellular metabolism, the molecular function(s) of many mitochondrial proteins remain unknown. Since protein function can be inferred from their interacting partners, we repurpose the protein structure prediction algorithm AlphaFold Multimer (AFM) as a classification model to predict protein-protein interactions of the entire human mitochondrial proteome. By screening 630,003 protein pairs, we create a compendium of 2,895 previously known and newly observed interactions, which include the interacting partner(s) of 85 uncharacterized mitochon

Potential Inhibitory Effect of LED-Sourced Red Light Therapy on Ocular Growth in Normal and Myopic Chicks.

Repeated low-level red light (RLRL) has been reported to control myopia progression clinically. Given safety concerns with laser sources, light-emitting diodes (LED)-sourced red light represents a promising alternative. This study investigated the effects of LED-sourced red light (RL) on cellular response in vitro and ocular growth in normal and lens-induced myopic chicks. In vitro, the mouse photoreceptor 661W cell line was exposed to 625 and 664 nm LED-sourced RL (3 min, twice daily) for 3 days, and cytochrome c oxidase (CCO) activity and cell viability were assessed. In vivo, chicks were ra

Genotypic Analysis of Enterobius vermicularis (Rhabditida: Oxyuridae, Linnaeus, 1758) Among Infected Individuals in Bulgaria: A First Phylogenetic Study.

Enterobiasis, caused by the nematode Enterobius vermicularis, remains a widespread public health issue, yet data regarding its genetic structure in Southeast Europe are scarce. This study presents the first molecular and phylogenetic characterization of E. vermicularis isolates from Bulgaria. Between 2022 and 2025, perianal tape test samples were collected from 128 individuals (92.2% of whom were children) with enterobiasis from 17 regions of the country. Molecular identification was performed via nested PCR targeting a 324 bp fragment of the mitochondrial cytochrome c oxidase subunit 1 (cox1)

Genetic confirmation ends a diagnostic odyssey in rare Bertiella studeri infection.

In this study, we identified a case of Bertiella studeri infection in a child using etiological and molecular biological methods, which provided clear guidance for subsequent diagnostic work. Cytochrome c oxidase subunit 1 gene (COX1) was amplified and the sequence is closely related to Bertiella studeri (OP474069.1) The patient had been treated with Praziquantel 1 pill for one time and took it continuously for 6 days as a course of treatment. After six months, the patient did not find any more proglottids in the feces and returned to normal. The patient was a 2 years and 7 months boy, who liv

Welcome back to life: Validating the genus Illiosentis Van Cleave and Lincicome, 1939 (Leptorhynchoididae: Palaeacanthocephala), with the description of a new species from the pigfish (Orthopristis chrysoptera: Haemulidae) in Yucatán Peninsula, Mexico.

The haemulid fish Orthopristis chrysoptera commonly known as pigfish is distributed in the Western Atlantic coast, Caribbean Sea and Gulf of Mexico. Specimens of O. chrysoptera sampled off the coast of Sisal, Yucatán, in the Gulf of Mexico were infected with two species of acanthocephalans from the family Leptorhynchoididae. One of them was identified as Dollfusentis bravoae and the other was allocated into the genus Illiosentis. However, the taxonomic status of Illiosentis has been controversial since it has been proposed as synonym of Tegorhynchus. Here, sequences the small (SSU) and large (

Occurrence of Anisakis pegreffii (Nematoda: Anisakidae) larvae in Larimus pacificus (Eupercaria: Sciaenidae) from northern Peruvian waters.

Anisakis pegreffii Campana-Rouget & Biocca, 1955 (Nematoda: Anisakidae) is a zoonotic nematode of increasing public health concern, particularly in regions where raw or lightly processed fish is traditionally consumed. This study provides the first molecular evidence of A. pegreffii infection in the Pacific drum, Larimus pacificus Jordan & Bollman, 1890 (Eupercaria: Sciaenidae), a neritic fish species endemic to the eastern Pacific and frequently sold in local markets along the northern Peruvian coast. A total of 78 specimens of L. pacificus were collected from northern Peruvian waters and exa

Apristurus iterum, a new shark discovered based on egg case morphology and sequence data obtained from a formalin-fixed specimen.

Apristurus iterum, a new species of deepwater catshark, is described from northeastern Australia. The new species is diagnosed through examination of its unique egg case morphology based on a single whole gravid female specimen and 10 egg cases deposited in different Australian ichthyological collections. This discovery prompted a more detailed investigation of Apristurus specimens collected off Queensland, northeastern Australia. The voucher of the gravid female with two egg cases, previously identified as 'Apristurus', collected off southeast Queensland, was designated as the holotype and us

Myiasis by Cordylobia anthropophaga and C. rodhaini (Diptera: Calliphoridae) in Polish travelers to Africa with new molecular data.

Myiasis is a parasitic infestation of soft vertebrate tissues by larval stages of Diptera. We briefly described the lesion-causing genus Cordylobia Grünberg (Diptera: Calliphoridae). Three Polish travelers to Uganda, Gambia, and Senegal returned with furuncular myiasis. To identify the third-instar larvae removed from their skin, we examined the morphological features of the 3 specimens and sequenced a 5' barcoding fragment of the cytochrome c oxidase subunit I gene (COI-5P). One larva was identified as C. rodhaini Gedoelst, and 2 larvae were identified as C. anthropophaga (Blanchard). We were
