Hybridization Approach Applied to Umbelliferon and Vanilloids toward New Inhibitors of Carbonic Anhydrases IX and XII with In Vitro Antiproliferative and Anti-inflammatory Activities.

Human carbonic anhydrases (hCAs) IX and XII have emerged as promising therapeutic targets and are overexpressed in hypoxic tumors. Leveragin … ounds induced G1 cell cycle arrest, reduced expression of the metastasis-associated markers CD9 and epithelial cell adhesion molecule, and exhibited cytoprotective and anti-inflammatory effects in BEAS-2B cells. Collectively, these findings identify UMB-vanilloid hybrids as promising candidates for the development of novel therapeutics for nonsmall cell lung cancer.

Immunopathology of oral mucosal ulcerative, desquamative, and bullous diseases. Selective review of the literature.

Cell/cell and cell/matrix adhesion proteins are responsible for maintaining the integrity of the mucosal lining of the oral cavity. Disease processes that destroy keratinocytes or adversely affect their adhesion to one another or to the subjacent basement membrane will result in erosions, ulcerations, and desquamations. Immunologic processes that have a deleterious effect on the integrity of the epithelial/basement membrane/submucosa complex are reviewed, and current research findings with respect to pathogenesis are discussed. In particular, T-cell-mediated hypersensitivity is involved in rec

A comparative study of circulating tumor cell isolation and enumeration technologies in lung cancer.

Circulating tumor cells (CTCs) have potential as diagnostic, prognostic, and predictive biomarkers in solid tumors. Despite Food and Drug Administration (FDA) approval of CTC devices in various cancers, the rarity and heterogeneity of CTCs in lung cancer make them technically challenging to isolate and analyze, hindering their clinical integration. Establishing a consensus through comparative analysis of different CTC systems is warranted. This study aimed to evaluate seven different CTC enrichment methods across five technologies using a standardized spike-in protocol: the CellMag™ (EpCAM-dep

Lung epithelium is the major source of IL-33 and is regulated by IL-33-dependent and IL-33-independent mechanisms in pulmonary cryptococcosis.

Inhalation of the fungus Cryptococcus neoformans (C. neoformans) results in pulmonary cryptococcosis associated with IL-33-dependent type 2 immunity. Lung epithelium represents the initial contact site of infection. The role of IL-33 in type 2 immunity has been analyzed, but the source of this cytokine and its effect on lung epithelial cell function in pulmonary cryptococcosis remained unclear. In mice infected with C. neoformans, we identified alveolar type 2 epithelial cells as major source of IL-33. On both, IL-33-positive and IL-33-negative epithelial cells, IL-33 receptor expression was d

In situ expression of cell adhesion molecules in chronic gastritis with Helicobacter pylori infection.

Helicobacter pylori infection of the stomach results in acute inflammation followed by chronic inflammation, but the mechanism is unknown. Adhesion molecules such as ICAM-1, Mac-1, and LFA-1 may help regulate interactions of immune cells and inflammatory cells. We used immunohistochemistry to locate these molecules in the gastric mucosa of patients with chronic gastritis arising from H. pylori infection. Biopsy specimens were taken from five H. pylori-negative healthy volunteers and 20 H. pylori-positive patients with chronic gastritis for immunohistochemical studies of adhesion molecules. In

Logic-Gated Dual-Aptamer Proximity AlphaLISA for Rapid and Sensitive Detection of Tumor-Derived Small Extracellular Vesicles.

Small extracellular vesicles (sEVs) have emerged as promising biomarkers for liquid biopsy; however, the limited specificity of single-marker detection and the intrinsic heterogeneity of circulating sEVs hinder their reliable clinical application. Herein, we develop a programmable dual-aptamer amplified luminescent proximity homogeneous assay (AlphaLISA) for rapid and wash-free detection of tumor-derived sEVs. In this strategy, EpCAM and PD-L1 aptamers simultaneously recognize two membrane proteins on the same vesicle, enabling proximity-induced interaction between donor and acceptor beads to

The Management of Oral Pemphigus Vulgaris in a Hypertensive Patient: A Case Report.

Pemphigus vulgaris (PV) is a chronic autoimmune disorder that causes painful blisters on the skin and mucosa along with erosions due to intra-epithelial acantholysis. This acantholysis is mainly due to an immune reaction against desmoglein (an adhesion glycoprotein molecule) by IgG autoantibodies, which causes loss of cell-to-cell adhesion. The treatment consists of systemic corticosteroids such as tablet prednisolone along with topical steroids such as 0.1% triamcinolone paste. But steroids are known for their side effects, one of which is hypertension. It is vital for a dentist to curate the

Role of JAM-C in lens cell adhesion, calcium homeostasis, and cataract pathogenesis.

Junctional adhesion molecule C (JAMC) mutations are associated with autosomal recessive forms of cataracts. Our previous studies characterized the features of nuclear cataracts and revealed abnormal lens development and differentiation in Jamc knockout (Jamc-KO) mice. However, the pathogenic mechanisms underlying cataract formation following JAMC deletion remain largely unclear. In this study, we further investigated lens impairments in Jamc-KO mice. We found that JAM-C plays a critical role in maintaining lens cell adhesion and preserving the integrity of ball-and-socket junctions. Moreover,

Transgenic overexpression of ITGB6 in intestinal epithelial cells exacerbates dextran sulfate sodium-induced colitis in mice.

Integrins, as a large family of cell adhesion molecules, play a crucial role in maintaining intestinal homeostasis. In inflammatory bowel disease (IBD), homeostasis is disrupted. Integrin αvβ6, which is mainly regulated by the integrin β6 subunit gene (ITGB6), is a cell adhesion molecule that mediates cell-cell and cell-matrix interactions. However, the role of ITGB6 in the pathogenesis of IBD remains elusive. In this study, we found that ITGB6 was markedly upregulated in inflamed intestinal tissues from patients with IBD. Then, we generated an intestinal epithelial cell-specific ITGB6 transge

Calprotectin expression inhibits bacterial binding to mucosal epithelial cells.

Squamous mucosal epithelial cells constitutively express calprotectin in the cytoplasm. To study how this antimicrobial protein complex confers epithelial resistance to invading bacteria, an epithelial cell line was stably transfected to express the calprotectin complex. Cells expressing calprotectin resist invasion by Listeria monocytogenes and Salmonella enterica serovar Typhimurium. Calprotectin expression was accompanied by altered actin organization, increased alpha3 integrin expression, and spreading cell morphology. In this study, we assessed whether calprotectin expression affects bact

Design and evaluation of an EpCAM-specific peptide-diphtheria toxin fusion (DT-SNFYMPL) for targeted epithelial cancer treatment.

Epithelial cell adhesion molecule (EpCAM, CD326) is a transmembrane glycoprotein that plays important roles in both normal epithelial tissues and epithelial malignancies. Its frequent overexpression in several cancers, including pancreatic, colorectal, and prostate carcinomas, has made it an attractive therapeutic target. In this study, we designed and evaluated a novel EpCAM-binding diphtheria toxin fusion construct, DT-SNFYMPL, in which the EpCAM-binding peptide SNFYMPL was fused to a truncated diphtheria toxin. The recombinant construct was cloned, expressed in Escherichia coli BL21 cells, …

Adherence of neutrophils to human peritoneal mesothelial cells: role of intercellular adhesion molecule-1.

Peritonitis is accompanied by a massive influx of polymorphonuclear leukocytes (PMN) into the peritoneal cavity, little is known, however, about the process of neutrophil transmigration across the peritoneal membrane. The study presented here, therefore, investigates the adherence of human PMN to human peritoneal mesothelial cell monolayers and examines the importance of intercellular adhesion molecule-1 (ICAM-1) in the process. Human peritoneal mesothelial cells (HPMC) constitutively expressed ICAM-1 protein and mRNA. Stimulation with IL-1 beta or TNF alpha resulted in time- and dose-dependen

Electrochemical immunoplatform for the quantification of epithelial extracellular vesicles applied to prostate cancer diagnosis.

Prostate cancer (PCa) is the second most commonly diagnosed cancer in men worldwide, and its early detection is critical for improving patient outcomes through timely and effective treatment. In this work, we present the first electrochemical immunoplatform based on magnetic microbeads (MBs) for the determination of epithelial extracellular vesicles (EpEVs), which are emerging as promising biomarkers for PCa diagnosis and prognosis. The immunoplatform employs MBs functionalized with anti-EpCAM antibodies to selectively capture EpEVs, forming sandwich-type immune complexes that are detected via

Slug-positive circulating tumor cells in mesenchymal malignancies: what remains to validate.

Circulating tumor cells show promise as noninvasive response biomarkers, but their utility in mesenchymal malignancies requires further validation. This commentary examines He and colleagues' study proposing Slug-positive circulating tumor cells as predictors of neoadjuvant chemotherapy response. While the study provides valuable exploratory data, three considerations warrant careful interpretation: (1) Response Evaluation Criteria in Solid Tumors criteria versus histopathological assessment, the preferred sarcoma reference standard; (2) potential Epithelial Cell Adhesion Molecule-enrichment l…

Suramin affects differentiated and undifferentiated human thyroid epithelial cells in vitro.

In the last decade, suramin has become known for its antiproliferative, differentiation-inducing effects on cells and has been successfully used in the therapy of cancer patients. The present study was undertaken to investigate the effects of suramin on normal human thyroid cells in primary monolayer culture and to analyse whether it also affected cells from thyroid carcinomas. The results show that suramin, at concentrations similar to serum levels obtainable during therapy, inhibited the proliferation of thyroid cells as well as the secretion of thyroglobulin. It suppressed the activation of

Molecular mechanisms involved in intraepithelial lymphocyte migration: a comparative study in skin and tonsil.

Intraepithelial lymphocyte migration is a biological process frequently observed in skin and tonsil. Using immunohistochemistry, we have studied the molecular bases of this process in seven skin biopsies involved by mycosis fungoides (MF) and in 12 tonsils, four involved by B-chronic lymphocytic leukaemia (B-CLL) and eight by lymphoid follicular hyperplasia (LH). In the skin, intraepidermal T-lymphocyte infiltration was associated with narrowing and fragmentation of the basement membrane, as shown by an anti-collagen type IV antibody. Immunostaining of serial sections with an anti-collagenase
