In silico biophysics and rheology of blood and red blood cells in Gaucher Disease.

Gaucher Disease (GD) is a rare genetic disorder characterized by a deficiency in the enzyme glucocerebrosidase, leading to the accumulation of glucosylceramide in various cells, including red blood cells (RBCs). This accumulation results in altered biomechanical properties and rheological behavior of RBCs, which may play an important role in blood rheology and the development of bone infarcts, avascular necrosis (AVN) and other bone diseases associated with GD. In this study, dissipative particle dynamics (DPD) simulations are employed to investigate the biomechanics and rheology of blood and

[The blood rheological indices of patients with necrobiosis lipoidica].

Rheologic parameters of the blood were examined in 17 patients with necrobiosis lipoidica before and over the course of trental (pentoxyphyllin) therapy. The study has revealed an unchanged hematocrit in the presence of significantly increased blood viscosity, red cell aggregation and stability of red cell aggregates. Another finding was a deterioration of the red cell deformability; red cell mechanical resistance was found liable to alter. Trental therapy has resulted in positive shifts in respect of the skin symptoms; a tendency to normalization of the examined blood rheologic parameters was

Relationship between blood viscosity and existence and severity of carotid artery plaque.

BACKGROUND: Accumulating evidence shows that the increase in blood viscosity (BV) is an independent risk factor for atherosclerosis and its … was close to statistically significant (F = 2.438, p = 0.089). The values of parameters describing erythrocyte aggregation and rigidity increased among the mild, moderate, and severe groups. The difference was statistically significant in RBC rigidity "k" and ER of RBC (F = 3.863, p = 0.022; F = 5.897, p = 0.003, respectively). CONCLUSION: Increased blood viscosity is …

Potentiation by red blood cells of shear-induced platelet aggregation: relative importance of chemical and physical mechanisms.

Evidence has been reported to indicate that red blood cells (RBCs) may potentiate platelet adherence and platelet aggregation (PAG) in different flow systems in vitro as well as hemostatic platelet plug formation in response to vascular injury. In this study, we demonstrate that RBCs enhance PAG induced by well-defined, low-intensity, uniform, laminar shear stress. Potentiation by RBCs of shear-induced PAG was associated with appreciable loss of adenine nucleotides from 14C-adenine-labeled RBCs, the extent of which increased with increasing RBC concentration. The concentrations of RBC-derived

Binding of two spin-labelled derivatives of chlorpromazine to human erythrocytes.

The binding to human intact erythrocytes of two different spin-labelled derivatives of chlorpromazine has been studied. The influence of the positively charged side chain of the drug has been the focus of our attention. The positively charged amphiphilic compound (spin derivative I) is water-soluble up to 80 microM at pH values below 5.9. The apolar analogue (spin derivative II) aggregates in aqueous buffer from the lowest concentration tested. Both spin derivatives undergo a slow reduction inside the erythrocyte. The reduced nitroxides are readily reoxidized by adding a low, non-quenching, co

Interaction of miltefosine with the lipid and protein components of the erythrocyte membrane.

Miltefosine (MT) is an alkylphospholipid that has been approved for the treatment of breast cancer metastasis and visceral leishmaniasis, although its mechanism of action remains poorly understood. Electron paramagnetic resonance spectroscopy of a spin-labeled lipid and a thiol-specific spin label showed that MT causes an increase in the molecular dynamics of erythrocyte ghost membranes and detergent-resistant membranes (DRMs) prepared from erythrocyte ghosts. In the vesicles of lipid raft constituents, it was shown that 20 mol % sphingomyelin could be replaced by 20 mol % MT with no change in

Biofilm is associated with chronic streptococcal meningoencephalitis in fish.

Biofilms are aggregates of attached microbial organisms whose existence on tissues is often recognised as a mechanism for the establishment of most chronic diseases. Herein we investigated the ability of piscine Streptococcus agalactiae, an important aquatic pathogen, for adaptation to this sessile lifestyle in vitro and in the brain of a tilapia fish model. Piscine S. agalactiae exhibited a weak attachment to polystyrene plates and expressed a low biofilm phenotype under the study conditions. Furthermore, fluorescent in situ hybridization and confocal laser scanning microscopy revealed discre

Stroke propensity in the Th3+/ mouse model of β-thalassemia intermedia.

β-thalassemia is associated with multiple hematological and cerebrovascular symptoms linked to a hypercoagulable state that has not been fully replicated in animal models for the development of stroke treatments. Herein we compared the physiological properties and responses to transient cerebral hypoxia-ischemia (tHI) between six-month-old wildtype and heterozygous Th3/+ mice, a model of non-transfusion-dependent β-thalassemia intermedia (β-TI). We found that Th3/+ mice developed microcytic anemia, splenomegaly, higher platelet counts, and increased platelet-erythrocyte plus erythrocyte-leukoc

Inhalation of methane preserves the epithelial barrier during ischemia and reperfusion in the rat small intestine.

BACKGROUND: Methane is part of the gaseous environment of the intestinal lumen. The purpose of this study was to elucidate the bioactivity of exogenous methane on the intestinal barrier function in an antigen-independent model of acute inflammation. METHODS: Anesthetized rats underwent sham operation or 45-min occlusion of the superior mesenteric artery. A normoxic methane (2.2%)-air mixture was inhaled for 15 min at the end of ischemia and at the beginning of a 60-min or 180-min reperfusion. The integrity of the epithelial barrier of the ileum was assessed by determining the lumen-to-blood c

Delayed Rewarming Thrombocytopenia Not Disseminated Intravascular Coagulation, is the Cause of Rewarming Deaths in Neonatal Cold Injury.

Disseminated intravascular coagulation (DIC) was suspected of being the cause of fatal rewarming deaths in neonatal cold injury (NCI) in the absence of any other known explanation for this complication. It was associated with thrombocytopenia, bleeding, and abnormal clotting studies. Subsequently, this suspicion was questioned because of the reversibility of thrombocytopenia and more recently because of the delineation of a new entity DRT (delayed rewarming thrombocytopenia) that better explained the clinical and laboratory features of this condition. The sudden bleeding that occurs after 24 h

Protein quality control during erythropoiesis and hemoglobin synthesis.

Erythrocytes must regulate hemoglobin synthesis to limit the toxicities of unstable free globin chain subunits. This regulation is particularly relevant in β-thalassemia, in which β-globin deficiency causes accumulation of free α-globin, which forms intracellular precipitates that destroy erythroid precursors. Experimental evidence accumulated over more than 40 years indicates that erythroid cells can neutralize moderate amounts of free α-globin through generalized protein quality control mechanisms, including molecular chaperones, the ubiquitin-proteasome system, and autophagy. In many ways,

[Intravital microscopy and post mortem histology of burn shock in rabbits (author's transl)].

This paper describes results of intravital microscopic observations of the mesenteric blood and lymph vessels of the rabbit. 20% of the surface of the skin of these rabbits were burned. Immediately after producing the shock, general unspecific symptoms were seen: decrease of circulatory flow, aggregates of erythrocytes. Specific alterations consisted in leukocyte sticking, platelet aggregation, hemolysis and stasis. Vascular spasm and hemorrhage were conspicuously absent. These microcirculatory features were compared with those of different forms of shock. There was a striking similarity with

Prolonged alterations in red blood cell rheology following mild SARS-CoV-2 infection: Implications for microvascular health.

The COVID-19 pandemic has prompted widespread investigation into its systemic effects, focusing on the consequences of even mild SARS-CoV-2 infections. This study explores the impact of mild COVID-19 on red blood cell (RBC) rheology, specifically deformability and aggregation, both critical for efficient microvascular perfusion and oxygen delivery. Using advanced microfluidic techniques, blood samples from 23 individuals recovering from mild COVID-19 were analyzed and compared to 21 healthy controls. Results revealed significant reductions in RBC deformability and increased aggregation tendenc
