Lactic acid bacteria-derived biosurfactants: emerging strategies for biofilm control, virulence attenuation, and food safety.

Biofilm-associated infections caused by multidrug-resistant bacteria and fungi impose a substantial global health burden, and conventional antibiotics are increasingly ineffective at eradicating them due to poor penetration of the extracellular polymeric substance (EPS) matrix and escalating resistance. Biosurfactants produced by lactic acid bacteria (LAB) have attracted research interest as alternatives, owing to the GRAS status of their producing organisms and their broad mechanistic repertoire. This review critically evaluates the structural diversity of LAB-derived biosurfactants, covering

An operon encoding enzymes for synthesis of a putative extracellular carbohydrate attenuates acquired vancomycin resistance in Streptomyces coelicolor.

Actinomycete bacteria use polyprenol phosphate mannose as a lipid-linked sugar donor for extra-cytoplasmic glycosyl transferases that transfer mannose to cell envelope polymers, including glycoproteins and glycolipids. Strains of Streptomyces coelicolor with mutations in the gene ppm1, encoding polyprenol phosphate mannose synthase, and in pmt, encoding a protein O-mannosyltransferase, are resistant to phage ϕC31 and have greatly increased susceptibility to some antibiotics, including vancomycin. In this work, second-site suppressors of the vancomycin susceptibility were isolated. The suppress

Multiscale elucidation of reactive black 5 biodegradation by a synergistic bacterial consortium biofilm: experimental validation and molecular dynamics insights.

Reactive Black 5 (RB5) is a widely used azo dye in the textile industry that is highly toxic and harms both human health and the environment. The biofilm is a complex matrix comprising microorganisms and extracellular polymeric substances (EPS). Biofilm-based bioremediation is one of the most effective approaches for removing toxic RB5 from water. In this study, a biofilm-forming bacterial consortium comprising Bacillus tequilensis BS007 and Enterobacter mori BS008 was evaluated for treated RB5 dye. The consortium exhibited a specific growth rate (μmax) of 0.265 h⁻1 and efficient biofilm forma

Large-scale screening identifies enzyme combinations that remove in situ grown oral biofilm.

Bacteria in the oral cavity are responsible for the development of dental diseases such as caries and periodontitis, but it is becoming increasingly clear that the oral microbiome also benefits human health. Many oral care products on the market are antimicrobial, killing a large part of the oral microbiome but without removing the disease-causing biofilm. Instead, non-biocidal matrix-degrading enzymes may be used to selectively remove biofilm without harming the overall microbiome. The challenge of using enzymes to degrade biofilms is to match the narrow specificity of enzymes with the large

Helicobacter pylori Biofilm Formation and Its Potential Role in Pathogenesis.

Despite decades of effort, Helicobacter pylori infections remain difficult to treat. Over half of the world's population is infected by H. pylori, which is a major cause of duodenal and gastric ulcers as well as gastric cancer. During chronic infection, H. pylori localizes within the gastric mucosal layer, including deep within invaginations called glands; thanks to its impressive ability to survive despite the harsh acidic environment, it can persist for the host's lifetime. This ability to survive and persist in the stomach is associated with urease production, chemotactic motility, and the

Spatial distribution and structural discrimination of quorum sensing metabolites in Pseudomonas aeruginosa by MALDI-MSI.

Bacterial biofilms are complex bacterial communities embedded within an extracellular polymeric substance (EPS) that protects the cells and increases survival. Once established on an orthopaedic implant, a biofilm can cause difficult-to-treat and severe infections. Pseudomonas aeruginosa is of particular concern due to its strong biofilm-forming capability. Within biofilm communities, quorum sensing (QS) networks regulate biofilm formation, virulence, and intercellular communication. This is achieved through the production of signalling molecules, QS metabolites. In P. aeruginosa, quinolones s

Bacteria-polysaccharide interfacial regulation by calcium alginate hydrogel capsules supports sulfate reduction in acidic water.

Calcium alginate hydrogels are widely used for microbial encapsulation, yet their role in regulating bacteria-polysaccharide interfaces under acidic sulfate conditions remains unclear. Here, calcium alginate hydrogel capsules (SAC) were used to encapsulate a functional sulfate-reducing consortium and evaluate its performance in acidic sulfate-containing water. At pH 4, SAC maintained live-cell densities 147-185 times higher than those of free cells and achieved higher sulfate removal rates on days 7 and 14 (19.9% and 21.3%) than free cells (3.7% and 10.0%). Inactivated SAC showed limited sulfa

Tunneling nanotube-mediated intercellular filamin A transport mechanosensitively programs osteoclastogenesis in myeloma bone disease.

Intercellular communication between multiple myeloma (MM) cells and osteoclast precursor cells (pre-OCs) contributes extensively to the occurrence and development of myeloma-related bone destruction. However, key interacting modes and the exchanged substances involved in this communication remain unclear. In this study, we discover that tunneling nanotubes (TNTs) directly connect MM and pre-OCs. Using the stable isotope labeling with amino acids in cell culture assay and a positive-negative double selection strategy, we identify filamin A (FLNA) as a major protein transported from MM to pre-OC

Removal Mechanism of Cutibacterium acnes Biofilms: A Comparison between Synthetic Surfactants and Rhamnolipid Biosurfactants.

Biofilms are composed of autogenously secreted extracellular polymeric substances (EPS) and affect nearly all aspects of human activity. Biofilms with negative health and economic effects can be controlled through the integrated use of surfactants and mechanical force. However, the relationships between the surfactant molecular structures and biofilm removal pathways have not been sufficiently elucidated. Additionally, Cutibacterium acnes (C. acnes), a skin commensal bacterium with biofilm-forming capabilities that can exacerbate acne vulgaris, has been rarely investigated. In this study, we e

Quantitative and chemical adaptation of exopolymeric substances formed by a river microbial consortium during exposure to the antibiotic trimethoprim.

In natural environments, most microorganisms reside attached to a surface growing as a biofilm, which is a universal microbial strategy for their survival. As a result, several studies have focused on the role of the biofilm matrix (made of extracellular polymeric substances (EPS)) in tolerance to antimicrobials. However, few studies have focused directly on the characterization of EPS as a response to antibiotic stress. In this study, we analyzed the impact of trimethoprim (TMP) on the production and characteristics of EPS from four natural biofilm-producing river bacterial strains. Extractio

Extracellular DNA enhances biofilm integrity and mechanical properties of mucoid Pseudomonas aeruginosa.

Pseudomonas aeruginosa is one of the most common biofilm-forming pathogens responsible for lung infections of individuals with cystic fibrosis (CF). P. aeruginosa becomes tolerant to antimicrobials in the biofilm state and is difficult to treat. Production of extracellular polymeric substances (EPS), such as alginate and extracellular DNA (eDNA), can allow adherence to abiotic and biotic surfaces, antimicrobial evasion, and resilience to environmental pressures. Alginate-producing mucoid variants of P. aeruginosa are frequently isolated from CF airway samples and are associated with worsening

Extracellular polymeric substances of bacteria and their potential environmental applications.

Biopolymers are considered a potential alternative to conventional chemical polymers because of their ease of biodegradability, high efficiency, non-toxicity and non-secondary pollution. Recently, extracellular polymeric substances (EPS, biopolymers produced by the microorganisms) have been recognised by many researchers as a potential flocculent for their applications in various water, wastewater and sludge treatment processes. In this context, literature information on EPS is widely dispersed and is very scarce. Thus, this review marginalizes various studies conducted so far about EPS nature

Long-term effects of zinc oxide nanoparticles on soluble microbial product composition and transformation in submerged membrane bioreactors.

The increasing release of engineered nanoparticles into wastewater systems has raised concerns regarding their long-term effects on biological treatment processes and soluble microbial product (SMP) formation. However, the effects of zinc oxide nanoparticles (ZnO NPs) on the composition, diversity, and persistence of low-molecular-weight (low-MW) SMPs in membrane bioreactors (MBRs) remain poorly understood. In this study, the long-term effects of ZnO NP exposure on SMP characteristics were investigated in submerged anoxic-aerobic MBRs operated with and without ZnO NP addition. SMP production,

Black Cumin Seed Oil Disrupts Structural Biomass and Attenuates Porphyrin-Associated Maturation in Mature Oral Microcosm Biofilms: An In Vitro Study.

Background/Objective: Mature oral biofilms resist conventional antiseptics via dense extracellular polymeric substance (EPS) matrices. Consequently, the prolonged use of broad-spectrum agents like chlorhexidine (CHX) raises profound ecological concerns. Using a mature oral microcosm biofilm model, we evaluated the multifaceted efficacy of 0.5% black cumin seed oil (BCSO) in disrupting structural biomass, suppressing aciduric bacterial viability, and attenuating porphyrin-associated anaerobic biofilm maturation. Methods: Saliva-derived microcosm biofilms were cultivated on hydroxyapatite discs

Enhanced nitrogen and manganese removal in a manganese redox-driven moving bed biofilm reactor: role of extracellular polymeric substances.

In this study, the continuous flow moving bed biofilm reactor (MBBR) was developed for conversion of nitrogen and manganese including MBBR1 (R1, with manganese redox cycling function) and MBBR2 (R2, control group). The result suggested that NH4+-N removal reached 95±2% and 78±1% by R1 and R2 with NO3--N effluent concentrations of 2.1±0.4 and 7.1±0.6 mg·L-1, and TN removal through R1 increased compared with R2. Fluorescence characteristics and concentration of extracellular polymeric substances (EPS) were explored, and reactive oxygen species, apoptosis and enzyme activity of biofilm including

Thiosulfate-Driven redox buffering enables efficient nitrogen removal and norfloxacin degradation in mixed denitrifying systems.

Wastewaters often contain both conventional pollutants and recalcitrant antibiotics, posing challenges to biological treatment. This study investigated a mixed autotrophic-heterotrophic denitrification system driven by sodium acetate and sodium thiosulfate for simultaneous nitrate and norfloxacin removal. A sequencing batch reactor was operated in four stages, culminating in norfloxacin exposure (0.5 mg/L). Results showed stable nitrogen removal (>95%) and norfloxacin degradation (>90%) under sustained antibiotic stress. Batch tests confirmed that the co-presence of thiosulfate and acetate enh

Biogas production and biomolecule degradation from Chlorella vulgaris using a lagoon-digester granular sludge versus a silty sludge adapted to microalgae.

Anaerobic digestion of microalgal biomass is a promising route for biogas production, but its efficiency is constrained by algal properties and by the biomolecule degradation kinetics of the microbial consortia used as inoculum. This study provides the first evaluation of biogas production and biomolecule degradation kinetics by a spontaneously formed granular sludge from an industrial lagoon digester, using Chlorella vulgaris biomass as substrate, in comparison with a silty sludge naturally adapted to the presence of microalgae (chlorophytes). Granular sludge initiated methanogenesis without

Antibiofilm Mechanisms and Applications of Natural Compounds.

Biofilms formed on food-contact surfaces and processing equipment serve as persistent bacterial reservoirs that lead to recurrent food contamination and foodborne illnesses. Conventional sanitizers and antimicrobial treatments often fail to remove these structured microbial communities, which are protected by matrixes formed from extracellular polymeric substances (EPSs), the presence of metabolically dormant persister cells, and various adaptive resistance mechanisms. Natural compounds (NCs) are increasingly viewed as alternative antibiofilm agents, but evidence supporting their effectiveness

Detection of group A Streptococcus in tonsils from pediatric patients reveals high rate of asymptomatic streptococcal carriage.

BACKGROUND: Group A Streptococcus (GAS) causes acute tonsillopharyngitis in children, and approximately 20% of this population are chronic carriers of GAS. Antibacterial therapy has previously been shown to be insufficient at clearing GAS carriage. Bacterial biofilms are a surface-attached bacterial community that is encased in a matrix of extracellular polymeric substances. Biofilms have been shown to provide a protective niche against the immune response and antibiotic treatments, and are often associated with recurrent or chronic bacterial infections. The objective of this study was to test
