Neutrophil swarming delays the growth of clusters of pathogenic fungi.

Neutrophils employ several mechanisms to restrict fungi, including the action of enzymes such as myeloperoxidase (MPO) or NADPH oxidase, and the release of neutrophil extracellular traps (NETs). Moreover, they cooperate, forming "swarms" to attack fungi that are larger than individual neutrophils. Here, we designed an assay for studying how these mechanisms work together and contribute to neutrophil's ability to contain clusters of live Candida. We find that neutrophil swarming over Candida clusters delays germination through the action of MPO and NADPH oxidase, and restricts fungal growth thr…

[Candida auris: The Recent Emergence of a Multiresistant Pathogenic Fungi].

Candida auris was first described as a new species back in 2009. Although it differs markedly from other Candida species, this species can be misidentified as other yeasts in the routine microbiology laboratories. Therefore, its identification to species level should be confirmed by reference laboratories. Candida auris exhibits potential to cause invasive infections, and frequently shows a multidrugresistance pattern, and it is associated with high mortality rates. Outbreaks caused by Candida auris and associated with health care institutions have been reported in several countries around the

[Infections of the central nervous system by protozoa, helminths and fungi].

A plethora of different parasites and fungi can lead to infections of the central nervous system (CNS) and cause different clinical symptoms and outcomes depending on the pathogen and the anatomic location of the infection. The diagnosis and treatment of these eukaryotic infections is challenging. The prevalence of CNS infections depends on many factors, including geographical location, living conditions, genetic background and the immune status of the individual. In Germany, infections of the CNS by fungi and parasites are rare but can lead to considerable morbidity. Some parasitic and fungal…

Recent Advances in Applications of Acidophilic Fungi to Produce Chemicals.

Processing of fossil fuels is the major environmental issue today. Biomass utilization for the production of chemicals presents an alternative to simple energy generation by burning. Lignocellulosic biomass (cellulose, hemicellulose and lignin) is abundant and has been used for variety of purposes. Among them, lignin polymer having phenyl-propanoid subunits linked together either through C-C bonds or ether linkages can produce chemicals. It can be depolymerized by fungi using their enzyme machinery (laccases and peroxidases). Both acetic acid and formic acid production by certain fungi contrib…

Role of FKS Gene in the Susceptibility of Pathogenic Fungi to Echinocandins.

Echinocandins are antifungal agents that specifically inhibit the biosynthesis of 1,3-β-D-glucan, a major structural component of fungal cel … es to not only acquired resistance in Candida spp., but also inherent resistance in some pathogenic fungi. An understanding of the echinocandin resistance mechanism is important to develop both effective diagnosis and treatment options for echinocandin-resistant fungal diseases.

The impact of the postharvest environment on the viability and virulence of decay fungi.

Postharvest decay of fruits, vegetables, and grains by fungal pathogens causes significant economic losses. Infected produce presents a potential health risk since some decay fungi produce mycotoxins that are hazardous to human health. Infections are the result of the interplay between host resistance and pathogen virulence. Both of these processes, however, are significantly impacted by environmental factors, such as temperature, UV, oxidative stress, and water activity. In the present review, the impact of various physical postharvest treatments (e.g., heat and UV) on the viability and virul…

Mechanisms of copper and zinc homeostasis in pathogenic black fungi.

Black fungi comprise a diverse group of melanized microorganisms, many of which are able to infect humans. One of the recognized diseases that arise with black fungi infection is chromoblastomycosis, a neglected implantation mycosis. Considering their ecology, black fungi may face conditions with distinct metal availability. Zinc and copper are essential transition metals, which become toxic in excess. During the interaction with host, fungi may face either metal deprivation or poisoning. Here we report an in silico analysis of four black fungi genomes concerning zinc and copper homeostasis. O…

Parasites and fungi as a threat for prenatal and postnatal human development.

Recent literature data reveals the most common etiological agents of congenital parasitoses to be Toxoplasma gondii, Trypanosoma cruzi, Leishmania donovani and Plasmodium falciparum. An analysis of clinical data indicates that parasitic congenital infections are often asymptomatic, whereas symptomatic newborns usually display nonspecific symptoms, which greatly hinders correct diagnosis. The long-term consequences of prenatal infections are serious clinical problems. This article presents the possible routes of vertical transmissions (mother-to-child) of pathogens including prenatal, perinatal

Distribution of yeast-like fungi at a university hospital in Turkey.

BACKGROUND: The increased life span has led to application of more invasive procedures for diagnosis and treatment of particularly immunosuppressed individuals. This situation drew more attention to fungal infections due to existence of yeast-like fungi. Candida infections have increased due to transplant in patients, prolonged intensive care unit (ICU) stays, and invasive procedures. Recently, identification of yeast-like fungi as well as antifungal susceptibility test has been gaining more importance. OBJECTIVES: In our study, we aimed to evaluate the distribution of yeast-like fungi strain…

[Infections of finger and toe nails due to fungi and bacteria].

Infections of the finger and the toe nails are most frequently caused by fungi, primarily dermatophytes. Causative agents of tinea unguium are mostly anthropophilic dermatophytes. Both in Germany, and worldwide, Trichophyton rubrum represents the main important causative agent of onychomycoses. Yeasts are isolated from fungal nail infections, both paronychia and onychomycosis far more often than generally expected. This can represent either saprophytic colonization as well as acute or chronic infection of the nail organ. The main yeasts causing nail infections are Candida parapsilosis, and Can…

Role of fungi in pathogenesis of chronic rhinosinusitis: the hypothesis rejected.

PURPOSE OF REVIEW: Fungi have been suggested to play an important role in the pathogenesis of chronic rhinosinusitis (CRS). This review describes the recent knowledge concerning the role of fungi in the pathogenesis of CRS and allergic fungal rhinosinusitis (AFRS) and the clinical implications for treatment. RECENT FINDINGS: Recent studies show that, although there are several potential deficits in the innate and potentially also in the acquired immunity of CRS patients that might reduce or change their ability to react to fungi, there are not many arguments to suggest a causative role for fu…

Bioreduction of alpha-chloroacetophenone by whole cells of marine fungi.

The asymmetric reduction of 2-chloro-1-phenylethanone (1) by seven strains of marine fungi was evaluated and afforded (S)-(-)-2-chloro-1-phenylethanol with, in the best case, an enantiomeric excess of 50% and an isolated yield of 60%. The ability of marine fungi to catalyse the reduction was directly dependent on growth in artificial sea water-based medium containing a high concentration of Cl(-) (1.2 M). When fungi were grown in the absence of artificial sea water, no reduction of 1 by whole cells was observed. The biocatalytic reduction of 1 was more efficient at neutral rather than acidic p…

Intracellular pathogenic bacteria and fungi--a case of convergent evolution?

The bacterium Yersinia pestis and the fungus Cryptococcus neoformans are the causative agents of human plague and cryptococcosis, respectively. Both microorganisms are facultatively intracellular pathogens. A comparison of their pathogenic strategies reveals similar tactics for intracellular survival in Y. pestis and C. neoformans despite their genetic unrelatedness. Both organisms can survive in environments where they are vulnerable to predation by amoeboid protozoal hosts. Here, we propose that the overall similarities in their pathogenic strategies are an example of convergent evolution th

Physical and biochemical interactions of soil fungi with asbestos fibers.

Iron, a structural component of most asbestos, is thought to play a crucial role in asbestos toxicity. Surface iron favors fiber uptake by mammalian cells and promotes the generation of free radicals with consequent DNA and lipid damage. In some cases, chemical removal of iron from asbestos fibers has reduced several cellular responses to asbestos. Since iron represents an essential element for soil microorganisms, many of them have developed mechanisms to scavenge this element from poorly soluble forms. We have investigated the ability of some soil fungi to produce iron chelators capable of e…
