Components of a new gene family of ferroxidases involved in virulence are functionally specialized in fungal dimorphism.

Mucormycosis is an emerging angio-invasive infection caused by Mucorales that presents unacceptable mortality rates. Iron uptake has been related to mucormycosis, since serum iron availability predisposes the host to suffer this infection. In addition, iron uptake has been described as a limiting factor that determines virulence in other fungal infections, becoming a promising field to study virulence in Mucorales. Here, we identified a gene family of three ferroxidases in Mucor circinelloides, fet3a, fet3b and fet3c, which are overexpressed during infection in a mouse model for mucormycosis,

Fluconazole and Voriconazole Resistance in Candida parapsilosis Is Conferred by Gain-of-Function Mutations in MRR1 Transcription Factor Gene.

Candida parapsilosis is the second most prevalent fungal agent causing bloodstream infections. Nevertheless, there is little information about the molecular mechanisms underlying azole resistance in this species. Mutations (G1747A, A2619C, and A3191C) in the MRR1 transcription factor gene were identified in fluconazole- and voriconazole-resistant strains. Independent expression of MRR1 genes harboring these mutations showed that G1747A (G583R) and A2619C (K873N) are gain-of-function mutations responsible for azole resistance, the first described in C. parapsilosis.

Misregulation of a DDHD Domain-containing Lipase Causes Mitochondrial Dysfunction in Yeast.

The DDHD domain-containing proteins, which belong to the intracellular phospholipase A1 (iPLA1) family, have been predicted to be involved in phospholipid metabolism, lipid trafficking, membrane turnover, and signaling. Defective cardiolipin (CL), phosphatidylethanolamine, and phosphatidylglycerol remodeling cause Barth syndrome and mitochondrial dysfunction. Here, we report that Yor022c is a Ddl1 (DDHD domain-containing lipase 1) that hydrolyzes CL, phosphatidylethanolamine, and phosphatidylglycerol. Ddl1 has been implicated in the remodeling of mitochondrial phospholipids and CL degradation.

Diversity of Secondary Metabolism in Aspergillus nidulans Clinical Isolates.

The filamentous fungus Aspergillus nidulans has been a primary workhorse used to understand fungal genetics. Much of this work has focused on elucidating the genetics of biosynthetic gene clusters (BGCs) and the secondary metabolites (SMs) they produce. SMs are both niche defining in fungi and of great economic importance to humans. Despite the focus on A. nidulans, very little is known about the natural diversity in secondary metabolism within this species. We determined the BGC content and looked for evolutionary patterns in BGCs from whole-genome sequences of two clinical isolates and the A

Absence of a gene encoding cytosine deaminase in the genome of the agaricomycete Coprinopsis cinerea enables simple marker recycling through 5-fluorocytosine counterselection.

Coprinopsis cinerea is a model species for molecular genetics studies of sexual development in agaricomycetes or homobasidiomycetes. Recently, efficient gene targeting was established in this fungus by generating Cc.ku70 or Cc.lig4 disruptants. To determine the molecular mechanisms underlying sexual development, which involves many genes, generating multiple gene disruptants is required. However, the number of transformation markers available for C. cinerea is limited. This problem would be solved by establishing marker recycling. In this study, we found that C. cinerea lacks a gene encoding a

Slt2 kinase of the cell wall integrity pathway is required for Fks2-specific echinocandin resistance in Candida glabrata.

Candida glabrata is an opportunistic yeast that causes invasive infections in immunocompromised individuals. Echinocandin antifungals are first-line agents in the treatment of invasive candidiasis and act by inhibiting fungal cell wall synthesis by targeting β-1,3-glucan synthase. Echinocandin resistance is primarily associated with mutations in FKS1 or FKS2, which encode for the catalytic subunit (Fks1 or Fks2) of the targeted glucan synthase. The cell wall integrity (CWI) pathway, including Slt2 kinase, is known to mediate tolerance to antifungal drugs, but its role in resistance is not comp

The global regulator LaeA controls expression of genes involved in diverse primary and secondary metabolites biosynthesis and their secretion in fungi.

LaeA is a global transcriptional regulator that differentially modulates expression of an impressive variety of secondary metabolite gene clusters in fungi, acting as a positive regulator fo some while repressing others. This regulator controls the production of fungal cellulolytic/ligninolytic enzymes and citric acid secretion in Ascomycetes. Also, it regulates the biosynthesis of siderophores and the antitumor agent ganoderic acid in Basidiomycetes. This regulator contains an S-adenosylmethionine-binding motif, suggesting a function as a methyltransferase, although its specific methylation s

F2F-202, a Selective Histone Deacetylase 6 (HDAC6) Inhibitor, Behaves as an Arrow with Multiple Tips against Azole-Resistant C. albicans: Modulation of Yeast-to-Hyphae Transition, Trailing Effect, and Oxidative Stress.

Fungal infections are increasingly threatening public health due to the onset of multidrug-resistant fungal strains. Accordingly, the incidence of chemoresistant C. albicans strains has steadily increased, along with associated mortality, thus highlighting the need for effective antifungal agents for the treatment of both localized and systemic C. albicans infections. Post-transcriptional modifications are key regulators of fungal virulence, modulating the production of virulence factors, yeast-to-hyphae transition, biofilm formation, and chemoresistance phenomena, whereas fungal histone deace

Global translational reprogramming in Trichophyton mentagrophytes-infected keratinocytes.

Dermatophytosis afflicts approximately 25% of the global population, representing a critical public health burden. Trichophyton mentagrophytes (T. mentagrophytes), a predominant zoonotic dermatophyte, is a significant contributor to disease morbidity. While the transcriptomic response of keratinocytes-the primary barrier of the skin-to T. mentagrophytes infection is well characterized, the genome-wide translational regulatory mechanisms remain unknown. Here, we employed parallel ribosome profiling (Ribo-seq) and RNA sequencing (RNA-seq) to dissect the genome-wide transcriptional and translatio

Understanding metabolic interactions between bacteria and fungi for cross-kingdom consortia applications.

Bacterial-fungal interactions represent fundamental ecological associations that shape microbial community structure across diverse environments. While traditionally framed through the lens of antagonism, bacteria and fungi engage in sophisticated metabolic dialogs extending far beyond simple warfare. Both primary and specialized metabolites function as context-dependent signals, nutrient resources, and modulators of cellular processes that fundamentally influence the physiology, development, and evolutionary trajectory of both fungi and bacteria. Primary metabolites mediate mutualistic relati

The regulatory pathway of transcription factor MYB36 from Trichoderma asperellum Tas653 resistant to poplar leaf blight pathogen Alternaria alternata Aal004.

In fungi, MYB transcription factors (TFs) mainly regulate growth, development, and resistance to stress. However, as major disease-resistance TFs, they have rarely been studied in biocontrol fungi. In this study, MYB36 of Trichoderma asperellum Tas653 (Ta) was shown to respond strongly to the stress caused by Alternaria alternata Aa1004. Compared with wild-type Ta (Ta-Wt), the inhibition rate of the MYB36 knockout strain (Ta-Kn) on Aa1004 decreased by 11.06%; the superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activities decreased by 82.15 U/g, 0.19 OD470/min/g, and 1631.2 μmo

The Biosynthesis of Penicillin and Cephalosporin C are Regulated by ROS at Transcriptional Level.

In a recent work we showed that, besides lovastatin, ROS also accumulate during the production phase in Pencillium chrysogenum and in Acremonium chrysogenum, and that these ROS regulate the biosynthesis of penicillin and cephalosporin C. In the present study, we investigated the level at which this positive regulation is exerted. Internal ROS levels were manipulated, i.e., increased or decreased, in the production phase of the respective fermentations. Penicillin production decreased by 51.2% when internal ROS concentration was diminished by 50%, while a 62% production increase was observed wh

Deregulation of CLN1 and CLN2 in the Saccharomyces cerevisiae whi2 mutant.

Wild-type cells of the budding yeast Saccharbmyces cerevisiae arrest in G1 upon nutrient exhaustion. Cell cycle arrest requires the WHI2 gene since whi2 mutants continue to divide and become abnormally small as nutrients are depleted. Here we show that CLN1 and CLN2 transcript levels in a whi2 strain are higher during exponential growth, and persist longer upon starvation, than in an isogenic wild-type strain. In contrast to CLN1 and CLN2, CLN3 levels declined only at very high cell density and were unaffected by the whi2 mutation. Elevated CLN expression is sufficient to explain the whi2 phen

A mechanosensitive ion channel is required for nematode predation in nematode-trapping fungi.

UNLABELLED: Mechanosensitive ion channels of small conductance-like channels (MSL) are conserved in cell-walled organisms and enable cellular responses to mechanical stimuli. Nematode-trapping fungi (NTF) rely on nematode-derived mechanical cues to form specialized predatory structures, including adhesive network and mechanically actuated constricting ring (CR) that rapidly inflate upon nematode contact. However, the sensory mechanisms underlying these processes remain unclear. Here, using the adhesive network-forming Arthrobotrys oligospora and the CR-forming Drechslerella dactyloides as repr

Recent progress on heterologous protein production in methylotrophic yeast systems.

Recombinant protein production technology is widely applied to the manufacture of biologics used as drug substances and industrial proteins such as recombinant enzymes and bioactive proteins. Various heterologous protein production systems have been developed using prokaryotic and eukaryotic hosts. Especially methylotrophic yeast in eukaryotic hosts is suggested to be particularly valuable because such systems have the following advantages: protein secretion into culture broth, eukaryotic quality control systems, a post-translational modification system, rapid growth, and established recombina

The morphotype heterogeneity in Cryptococcus neoformans.

Many environmental fungi have evolved exceptional abilities to overcome host defenses and to cause systemic infections. However, the evolutionary trajectory that gives rise to the remarkable pathogenic traits of otherwise saprophytic species is poorly understood. Recent studies suggest that social behaviors likely enhance fitness and augment virulence in the ubiquitous fungus Cryptococcus neoformans. In this regard, heterogeneity in morphotypes and the ability to switch morphotype offer flexibility and resilience for this fungus in disparate environmental and host niches. Here, we discuss the

Zinc acquisition: a key aspect in Aspergillus fumigatus virulence.

Zinc is an essential micronutrient required for the growth of all microorganisms. To grow in the lungs of a susceptible patient Aspergillus fumigatus must obtain zinc from the surrounding tissues. The concentration of Zn(2+) ions in living tissues is much lower than that required for optimal fungal growth in vitro because most of them are tightly bound to proteins at the physiological pH. However, A. fumigatus has several zinc transporters (ZrfA, ZrfB and ZrfC) that enable it to uptake zinc efficiently under the extreme zinc-limiting conditions provided by a susceptible host. The ZafA transcri

Signal transduction pathways regulating differentiation and pathogenicity of Cryptococcus neoformans.

The basidiomycetous yeast Cryptococcus neoformans is a human pathogen. Several phenotypes of this organism are defined as virulence traits including the polysaccharide capsule, melanin, and the ability to grow at 37 degreesC. The signaling pathways regulating the expression of these phenotypes and other important cellular processes are being defined on a molecular level. For example, the highly conserved signaling molecule calcineurin regulates high temperature growth in C. neoformans. An insertion mutation associated with constitutive expression of repressible acid phosphatase in Saccharomyces cerevisiae.

The PHO83 mutation in Saccharomyces cerevisiae, which had been detected on the basis of constitutive production of repressible acid phosphatase and mapped at the end of the PHO5 locus, was analysed by Southern hybridization with cloned DNA fragments of the PHO5 gene as probe. It was shown that this mutant has a DNA insertion of about 6 kilobase pairs, probably in the 5'-noncoding region of the PHO5 gene. Production of repressible acid phosphatase by the PHO83 mutant is partially independent of the function of the PHO2 and PHO4 genes, the positive regulatory genes whose functions are indispensa

Sexual recombination as a tool for engineering industrial Penicillium chrysogenum strains.

The recent discovery and functional characterization of opposite mating-type loci in the industrial penicillin producer Penicillium chrysogenum demonstrated their regulatory role in sexual as well as asexual development. Subsequent experiments further showed that a sexual life cycle can be induced in P. chrysogenum that was for long believed to reproduce exclusively by asexual propagation. Finally, crossing of wild type and production strains resulted in the generation of recombinant ascospore isolates. We predict from these recent findings that recombinant progeny for industrial applications
