IFN Signaling and ICB Resistance: Time is on Tumor's Side.

Activation of antitumor immunity upon immune checkpoint blockade (ICB) is one of the most promising strategies in cancer therapy. However, ICB resistance is frequently observed in cancer preclinical models and patients. A recent report in Cell reveals that sustained interferon (IFN) signaling confers tumor resistance to ICB by inducing the expression of an immunosuppressive multigenic program.

Deciphering mechanisms of brain metastasis in melanoma - the gist of the matter.

Metastasis to distant organs and particularly the brain still represents the most serious obstacle in melanoma therapies. Melanoma cells acquire a phenotype to metastasize to the brain and successfully grow there through complex mechanisms determined by microenvironmental than rather genetic cues. There do appear to be some prerequisites, including the presence of oncogenic BRAF or NRAS mutations and a loss of PTEN. Further mediators of the brain metastatic phenotype appear to be the high activation of the PI3K/AKT or STAT3 pathway or high levels of PLEKHA5 and MMP2 in metastatic cells. A yet

LIMCH1 in Cancer: A Context-Dependent Modulator Beyond the Tumor Suppressor-Oncogene Dichotomy.

LIM and calponin homology domain-containing protein 1 (LIMCH1) has recently gained attention as a functionally perplexing factor in human cancers, with studies attributing both tumor-suppressive and tumor-promoting properties that defy simple categorization. Originally described as an actin-associated cytoskeletal protein that promotes non-muscle myosin-IIA (NM-IIA) activity and thereby curtails cell migration, LIMCH1 has more recently been linked to diverse oncogenic pathways across a range of tumor types. In lung adenocarcinoma and clear-cell renal cell carcinoma, diminished LIMCH1 expressio

Development of 5-FU-modified tumor suppressor microRNAs as a platform for novel microRNA-based cancer therapeutics.

MicroRNA (miRNAs) are pleiotropic post-transcriptional modulators of gene expression. Their inherently pleiotropic nature makes miRNAs strong candidates for the development of cancer therapeutics, yet despite their potential, there remains a challenge to deliver nucleic acid-based therapies into cancer cells. We developed a novel approach to modify miRNAs by replacing the uracil bases with 5-fluorouracil (5-FU) in the guide strand of tumor suppressor miRNAs, thereby combining the therapeutic effect of 5-FU with tumor-suppressive effect of miRNAs to create a potent, multi-targeted therapeutic m

Disruption of chromatin folding domains by somatic genomic rearrangements in human cancer.

Chromatin is folded into successive layers to organize linear DNA. Genes within the same topologically associating domains (TADs) demonstrate similar expression and histone-modification profiles, and boundaries separating different domains have important roles in reinforcing the stability of these features. Indeed, domain disruptions in human cancers can lead to misregulation of gene expression. However, the frequency of domain disruptions in human cancers remains unclear. Here, as part of the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium

Expression of p53 in a malignant melanoma of a goat.

A six-year-old brown haired goat was presented due to recurring tissue growth at the base of the right horn. The tumour recurred after repeated surgical excision and the animal was finally euthanized because of poor prognosis. Necropsy revealed a malignant, partly amelanotic melanoma originating from the right horn base with metastases in both frontal sinuses, urinary bladder, pleura, pericardium, second thoracic vertebra, kidney and mesenteric lymph nodes. Diagnosis was confirmed by histopathology and immunohistochemistry. A strong expression of p53, a suppressor of cell proliferation and act

Expression profiling of key androgen-metabolizing enzymes in canine sebaceous gland tumors.

This study investigated the expression profiles of key enzymes involved in androgen metabolism in canine sebaceous gland tumors and compared them with those in normal and hyperplastic sebaceous glands. Based on histopathological evaluation, lesions were classified as hyperplasia, adenoma, epithelioma, or carcinoma. Tissue sections from each case were examined for expression of the androgen receptor (AR); the de novo steroidogenic enzymes StAR, CYP11A1, CYP17A1, and 3β-HSD; the steroid-precursor-activating enzyme STS; the potent-androgen biosynthetic enzymes 17β-HSD3 and SRD5A1; the androgen-in

Expression profiling of inhibitory immune checkpoints in colorectal cancer stem cells and their association with tumor immunity and immunotherapy biomarkers.

BACKGROUND: Colorectal cancer stem cells (CSCs) represent a rare subpopulation within tumors endowed with self-renewal capabilities and are recognized as key drivers of tumor initiation, metastasis, and therapeutic resistance. These cells also display immunomodulatory properties that enable them to evade immune surveillance. However, the mechanisms underlying their immune evasion, including the role of immune checkpoints (ICPs), remain poorly understood. Therefore, this study aimed to characterize the inhibitory ICP expression landscape in colorectal CSC-enriched models and to evaluate its ass

Spheroid cultures reveal cancer stem cell enrichment and distinct chemotherapeutic response in triple-negative breast cancer.

BACKGROUND: Three-dimensional (3D) cancer spheroids mimic key morphological and biological features of solid tumors, providing a valuable model for evaluating drug responses. Here, we investigated whether intrinsic cancer stem cell composition and tumor subtype determine spheroid formation capacity, transcriptional profiling, and therapeutic response in breast cancer cell lines. METHODS: Using the liquid overlay technique, we optimized conditions for reproducible spheroid generation using 10 human cancer cell lines derived from breast cancer, glioblastoma, and malignant melanoma. Non-coding RNA repertoires in malignant pleural mesothelioma.

Malignant pleural mesothelioma (MPM) is a rare malignancy, with extremely poor survival rates. There are limited treatment options, with no second line standard of care for those who fail first line chemotherapy. Recent advances have been made to characterise the underlying molecular mechanisms of mesothelioma, in the hope of providing new targets for therapy. With the discovery that non-coding regions of our DNA are more than mere junk, the field of research into non-coding RNAs (ncRNAs) has exploded in recent years. Non-coding RNAs have diverse and important roles in a variety of cellular pr

Current concepts and challenges to unravel the role of iodothyronine deiodinases in human neoplasias.

Thyroid hormones (THs) are essential for the regulation of several metabolic processes and the energy consumption of the organism. Their action is exerted primarily through interaction with nuclear receptors controlling the transcription of thyroid hormone-responsive genes. Proper regulation of TH levels in different tissues is extremely important for the equilibrium between normal cellular proliferation and differentiation. The iodothyronine deiodinases types 1, 2 and 3 are key enzymes that perform activation and inactivation of THs, thus controlling TH homeostasis in a cell-specific manner.

Identification of tumor initiating cells and early marker genes in histologically normal colonic mucosa that lead to neoplastic transformation.

BACKGROUND & AIMS: Colorectal cancer (CRC) remains a leading cause of cancer‑related morbidity and mortality worldwide. Although the adenoma-carcinoma sequence and its genetic drivers are well described, the earliest cellular and molecular events initiating tumorigenesis within histologically normal colonic epithelium remain poorly defined. This study aims to identify tumor‑initiating cells (TICs), distinguish them from normal stem‑like cells (nSTMs), and delineate early transcriptional and signaling programs using single‑cell RNA sequencing (scRNA‑seq) from paired normal‑appearing and transfo

Homeobox genes and cancer: new OCTaves for an old tune.

In this issue of Cancer Cell, Gidekel et al. demonstrate that Oct-4, a member of the POU class of homeobox genes, is a critical player in the genesis of testicular germ cell tumors. This study provides further evidence that deregulated expression of homeobox genes, which occurs in many solid tumors, is functionally relevant for carcinogenesis and highlights unique features that distinguish homeobox genes from other cancer-promoting genes.

Acetate Metabolism in Thyroid Cancer Progression.

In recent years, several studies have highlighted the ability of malignant cells to use acetate as an alternative energy and biosynthetic source to glucose. In this context, the present study aimed at characterizing the expression profile of genes involved in acetate metabolism in thyroid carcinomas. To this end, we analyzed molecular and clinical data from 496 papillary thyroid cancers (PTCs) and 59 normal thyroid tissues from The Cancer Genome Atlas (TGCA). In addition, we examined 57 PTCs and matched normal tissues, and six anaplastic thyroid carcinomas (ATCs) collected in our institutions,
