Identification of Malus halliana R2R3-MYB gene family under iron deficiency stress and functional characteristics of MhR2R3-MYB4 in Arabidopsis thaliana.

Iron (Fe) is an essential element for plant growth and development. Fe deficiency can trigger leaf chlorosis and reduce fruit yield. Therefore, it is necessary to explore transcription factors in response to Fe deficiency stress. A total of 29 MhR2R3-MYB transcription factors were identified based on the transcriptome of Malus halliana under Fe deficiency stress. A comprehensive analysis of physical and chemical properties, gene structures, conserved motif composition, evolutionary relationship and chromosome distribution was performed. Subsequently, based on the transcriptome, 14 genes with t

The MdSLR1b-MdKNOX15-MdGA20ox2/MdGA2ox7 module regulates dwarfing by mediating the gibberellin pathway in apple.

Dwarf and high-density cultivation is a major trend in the apple industry, yet the regulatory mechanisms remain largely unexplored. In this study, we identified MdKNOX15, a KNOX transcription factor highly expressed in spur-type and dwarfing rootstock varieties, as a critical regulator of plant height. Overexpression of MdKNOX15 caused a gibberellin (GA)-deficient dwarf phenotype, whereas VIGS-mediated knockdown increased endogenous GA content and plant height. Exogenous GA alleviated, and uniconazole exacerbated, the dwarfism. Using DAP-seq and RNA-seq, we identified MdGA2ox7 and MdGA20ox2 as

TaEPFL1 gene controls the development of wheat pistils and stamens by regulating ethylene synthesis.

Proper development of floral organs is essential for reproductive success and grain yield in wheat. However, the molecular mechanisms regulating wheat floral organ development remain largely unknown. In this study, we characterized the role of the wheat TaEPFL1 gene in floral organ development and its association with ethylene signaling. TaEPFL1 was highly expressed in immature spikes of the pistillody mutant HTS-1, particularly during the pistil and stamen specification stages. Its expression was responsive to both exogenous ethylene and the ethylene inhibitor 1-Methylcyclopropene (1-MCP). Ov

Transcriptomic and Metabolomic Investigation on Leaf Necrosis Induced by ZmWus2 Transient Overexpression in Nicotiana benthamiana.

WUSCHEL (WUS) is a crucial transcription factor in regulating plant stem cell development, and its expression can also improve genetic transformation. However, the ectopic expression of WUS always causes pleiotropic effects during genetic transformation, making it important to understand the regulatory mechanisms underlying these phenomena. In our study, we found that the transient expression of the maize WUS ortholog ZmWus2 caused severe leaf necrosis in Nicotiana benthamiana. We performed transcriptomic and non-target metabolomic analyses on tobacco leaves during healthy to wilted states aft

Root-specific CLE3 expression is required for WRKY33 activation in Arabidopsis shoots.

This study focused on the role of CLE1-7 peptides as defense mediators, and showed that root-expressed CLE3 functions as a systemic signal to regulate defense-related gene expression in shoots. In the natural environment, plants employ diverse signaling molecules including peptides to defend themselves against various pathogen attacks. In this study, we investigated whether CLAVATA3/EMBRYO SURROUNDING REGION-RELATED (CLE) genes (CLE1-7) respond to biotic stimuli. CLE3 showed significant up-regulation upon treatment with flg22, Pep2, and salicylic acid (SA). Quantitative real-time PCR (qRT-PCR)

Comparative transcriptome analysis of differentially expressed genes related to the physiological changes of yellow-green leaf mutant of maize.

Chlorophylls, green pigments in chloroplasts, are essential for photosynthesis. Reduction in chlorophyll content may result in retarded growth, dwarfism, and sterility. In this study, a yellow-green leaf mutant of maize, indicative of abnormity in chlorophyll content, was identified. The physiological parameters of this mutant were measured. Next, global gene expression of this mutant was determined using transcriptome analysis and compared to that of wild-type maize plants. The yellow-green leaf mutant of maize was found to contain lower contents of chlorophyll a, chlorophyll b and carotenoid

Identification of genes associated with sex expression and sex determination in hemp (Cannabis sativa L.).

Dioecy in flowering plants has evolved independently many times, and thus the genetic mechanisms underlying sex determination are diverse. In hemp (Cannabis sativa), sex is controlled by a pair of sex chromosomes (XX for females and XY for males). In an attempt to understand the molecular mechanism responsible for sex expression in hemp plants, we carried out RNA sequencing of male and female plants at different developmental stages. Using a pipeline involving differential gene expression analysis and weighted gene co-expression network analysis, we identified genes important for male and fema

Exogenous ABA regulates stress tolerance of Leymus chinensis via modulation of PYL‑mediated signaling and antioxidant enzymes.

As an important species for ecological restoration and high-quality forage in northern China, Leymus chinensis has drawn considerable research interest regarding its stress tolerance regulatory mechanisms. Exogenous abscisic acid (ABA), a key stress-signaling hormone, can effectively pre-activate the antioxidant defense system in L. chinensis. To elucidate the ABA response mechanism, this study combined physiological assessments-including antioxidant enzyme activities-with transcriptome sequencing of ABA-treated plants. The results demonstrated that ABA treatment enhanced the activities of per

Hydrogen sulphide alleviates iron deficiency by promoting iron availability and plant hormone levels in Glycine max seedlings.

BACKGROUND: Hydrogen sulphide (H2S) is involved in regulating physiological processes in plants. We investigated how H2S ameliorates iron (Fe) deficiency in soybean (Glycine max L.) seedlings. Multidisciplinary approaches including physiological, biochemical and molecular, and transcriptome methods were used to investigate the H2S role in regulating Fe availability in soybean seedlings. RESULTS: Our results showed that H2S completely prevented leaf interveinal chlorosis and caused an increase in soybean seedling biomass under Fe deficiency conditions. Moreover, H2S decreased the amount of roo

[Enhancement of tropane alkaloids production in transgenic hair roots of Atropa belladonna by overexpressing endogenous genes AbPMT and AbH6H].

Atropa belladonna L. is the officially medicinal plant species and the main commercial source of scopolamine and hyoscyamine in China. In this study, we reported the simultaneous overexpression of two functional genes involved in biosynthesis of scopolamine, which respectively encoded the upstream key enzyme putrescine N-methyltransferase (PMT; EC 2.1.1.53) and the downstream key enzyme hyoscyamine 6beta-hydroxylase (H6H; EC 1.14.11.11) in transgenic hair root cultures of Atropa belladonna L. HPLC results suggested that four transgenic hair root lines produced higher content of scopolamine at

Research Progress on miRNAs and Artificial miRNAs in Insect and Disease Resistance and Breeding in Plants.

MicroRNAs (miRNAs) are small, non-coding RNAs that are expressed in a tissue- and temporal-specific manner during development. They have been found to be highly conserved during the evolution of different species. miRNAs regulate the expression of several genes in various organisms, with some regulating the expression of multiple genes with similar or completely unrelated functions. Frequent disease and insect pest infestations severely limit agricultural development. Thus, cultivating resistant crops via miRNA-directed gene regulation in plants, insects, and pathogens is an important aspect o

The SlDUF789 Gene Family in Tomato: Genome-Wide Identification and Responsiveness to Hormonal and Abiotic Stress Signals.

The DUF789 protein family comprises conserved domain proteins in plants, most of which remain functionally uncharacterized despite their involvement in growth, development, and stress responses. Moreover, systematic identification of the DUF789 family in tomato (Solanum lycopersicum L.) has not been reported. In this study, 11 SlDUF789 members were identified from the tomato genome, unevenly distributed across seven chromosomes, with members within the same subgroup sharing similar exon-intron structures and conserved motifs. Promoter analysis revealed the enrichment of cis-acting elements ass

XsiAMT1.1a was identified as a novel ammonium uptake functional gene and its overexpression combined with GA4 application significantly increased yield in Arabidopsis thaliana.

Nitrogen (N) is a key limiting factor for plant yield. Ammonium is one of the main N forms absorbed by plants. Overexpression of ammonium uptake functional genes, such as ammonium transporter (AMT), can increase yield. However, the AMTs reported to enhance yield significantly is still limited. No researches have focused on the effect of overexpressing AMT combined with hormone application on yield improvement. In this study, we first investigated the role of XsiAMT1.1a, a potential ammonium uptake functional gene in an ammonium preference plant Xanthium sibiricum, in ammonium uptake by the ana

Role of the 4-phosphopantetheinyl transferase of Trichoderma virens in secondary metabolism and induction of plant defense responses.

Trichoderma virens is a ubiquitous soil fungus successfully used in biological control due to its efficient colonization of plant roots. In fungi, 4-phosphopantetheinyl transferases (PPTases) activate enzymes involved in primary and secondary metabolism. Therefore, we cloned the PPTase gene ppt1 from T. virens and generated PPTase-deficient (?ppt1) and overexpressing strains to investigate the role of this enzyme in biocontrol and induction of plant defense responses. The ?ppt1 mutants were auxotrophic for lysine, produced nonpigmented conidia, and were unable to synthesize nonribosomal peptid

Beta-cyanoalanine synthase as a molecular marker for induced resistance by fungal glycoprotein elicitor and commercial plant activators.

ABSTRACT The biocontrol agent Pythium oligandrum produces glycoprotein elicitor in the cell wall fraction, designated CWP, and induces resistance to a broad range of pathogens. To understand the mechanism of CWP-induced resistance to pathogens, gene expression at the early stage of CWP treatment in tomato roots was analyzed using a cDNA array. At 4 h after CWP treatment, 144 genes were up-regulated and 99 genes were down-regulated. In the 144 up-regulated genes, nine genes exhibited about eightfold increased expression. Analysis of the response of these nine genes to three commercial plant act

Transcriptomic analysis of response to high-temperature stress in cotton.

High temperature stress (HT) is a major abiotic factor limiting global crop production. Cotton (Gossypium hirsutum L.) is particularly susceptible to heat stress, yet the underlying molecular mechanisms remain poorly understood. Here, we integrated physiological, transcriptomic, and functional analyses to elucidate the response of cotton material ZhongMianSuo087 (ZMS087) to HT. Field-based assays revealed that heat stress compromised cotton productivity by impairing pollen viability, reducing chlorophyll biosynthesis and leaf area, and altering biomass partitioning, leading to significant yiel
