Clinical Mechanistic Insights Into Polymorphic Ventricular Tachycardia: Infection-Triggered Unmasking of Long QT Syndrome.

A 63-year-old woman presented with fever, cough, and recurrent presyncope following a lower respiratory tract infection. Initial telemetry demonstrated intermittent complete atrioventricular block with ventricular premature complexes. Despite correction of electrolyte abnormalities and withdrawal of QT-prolonging medications (prescribed for cough), she developed repetitive episodes of polymorphic ventricular tachycardia triggered by R-on-T phenomena, requiring electrical cardioversion and suppression with overdrive pacing after magnesium and phenytoin proved ineffective. A nasopharyngeal bio f

The evolutionary origin of psychosis.

Imagination, the driving force of creativity, and primary psychosis are human-specific, since we do not observe behaviors in other species that would convincingly suggest they possess the same traits. Both these traits have been linked to the function of the prefrontal cortex, which is the most evolutionarily novel region of the human brain. A number of evolutionarily novel genetic and epigenetic changes that determine the human brain-specific structure and function have been discovered in recent years. Among them are genomic loci subjected to increased rates of single nucleotide substitutions

Collision tumor, a metastatic melanoma within a meningioma: a case report.

BACKGROUND: Collision tumors, involving two distinct neoplasms in a single anatomical site, are rare. Among these, the metastasis of melanoma into an intracranial meningioma is particularly uncommon, with only four previously reported cases. Melanoma, known for its aggressive metastatic potential, contrasts sharply with the small number of collision tumor reports. The coexistence of these tumors poses diagnostic and therapeutic challenges, particularly in patients with a stable meningiomas. RESULTS: We present the fifth documented case of melanoma metastasizing to a meningioma, the first to i

Chromosome-level reference genome of the European wasp spider Argiope bruennichi: a resource for studies on range expansion and evolutionary adaptation.

BACKGROUND: Argiope bruennichi, the European wasp spider, has been investigated intensively as a focal species for studies on sexual selection, chemical communication, and the dynamics of rapid range expansion at a behavioral and genetic level. However, the lack of a reference genome has limited insights into the genetic basis for these phenomena. Therefore, we assembled a high-quality chromosome-level reference genome of the European wasp spider as a tool for more in-depth future studies. FINDINGS: We generated, de novo, a 1.67 Gb genome assembly of A. bruennichi using 21.8× Pacific Bioscien

Measles virus in the brain.

Measles virus can give three different forms of infections in the central nervous system. These are acute postinfectious encephalitis, acute progressive infectious encephalitis, and subacute sclerosing panencephalitis (SSPE). The postinfectious acute disease is interpreted to reflect an autoimmune reaction. The acute progressive form of brain disease, also referred to as inclusion body encephalitis, reflects a direct attack by the virus under conditions of yielding cellmediated immunity. The late progressive form of encephalitis (SSPE) has been extensively analyzed. Recent molecular genetic st

Evolutionary genomics, tumor dynamics, and mathematical modeling of cancer progression: Implications for precision oncology.

Cancer is a disease of somatic evolution driven by the sequential acquisition of genetic alterations that confer reproductive advantage. Understanding how tumors grow, diversify, and adapt to therapeutic pressure requires an integrated framework spanning evolutionary genomics, population genetics, and dynamical systems theory. This review synthesizes current knowledge on clonal dynamics, intratumor heterogeneity (ITH), and the mathematical tools used to model cancer progression, with direct implications for the design of precision oncology strategies. Neutral evolution and Darwinian selection

Spinocerebellar ataxias in Venezuela: genetic epidemiology and their most likely ethnic descent.

Dominantly inherited ataxias (spinocerebellar ataxias, SCAs) are a genetically heterogeneous group of neurologic diseases characterized by progressive cerebellar and spinal tract degeneration with ataxia and other signs, common to all known subtypes. Several types are relatively frequent worldwide, but in several countries, one specific SCA may show a higher prevalence owing to founder phenomena. In Venezuela, genetic epidemiological features of SCAs have been assessed during the last 30 years; mutations in ATXN1 (SCA1), ATXN2 (SCA2), ATXN3 (SCA3), CACNA1A (SCA6), ATXN7 (SCA7), ATXN8 (SCA8), A

Epidermolysis bullosa and the partnership with autoimmunity: what should we assimilate?

Bullous skin diseases are characterized by genetic abnormalities related to structural epidermal proteins or organ-specific autoantibodies against the same proteins and are revealed by blister formation on skin or mucous membranes, with differences in blister depth, morphology, and topography. Both inherited and autoimmune forms of these disorders can be framed in the context of epidermolysis bullosa. Their clinical spectrum varies from early lethal to mild variants with normal life expectancy, and several distinct phenotypes differ for age of onset, extent, location and depth of skin and muco

Computational biology in the study of cardiac ion channels and cell electrophysiology.

The cardiac cell is a complex biological system where various processes interact to generate electrical excitation (the action potential, AP) and contraction. During AP generation, membrane ion channels interact nonlinearly with dynamically changing ionic concentrations and varying transmembrane voltage, and are subject to regulatory processes. In recent years, a large body of knowledge has accumulated on the molecular structure of cardiac ion channels, their function, and their modification by genetic mutations that are associated with cardiac arrhythmias and sudden death. However, ion channe

Murine Model of Tuberculosis. In vitro and in vivo Study.

Animal models that mimic the whole spectrum of susceptibility and resistance of human populations to tuberculosis would be useful for studying pathogenesis of tuberculosis, mechanisms of the host defense, and for testing prospective prophylactic and therapeutic tuberculosis vaccines and drugs. Inbred mouse strains are broadly and successfully used to model human tuberculosis and study mechanisms of host-mycobacteria interaction. Here we consider numerous aspects of murine models of tuberculosis. We analyze the course of tuberculosis depending on route and dosage of mycobacterial infection, and

Progress and challenges in development of animal models for dengue virus infection.

ABSTRACTThe severity of the dengue epidemic is on the rise, with its geographic range had expanded to southern Europe by 2024. In this August, the WHO updated the pathogens that could spark the next pandemic, dengue virus was on the list. Vaccines and drugs serve as powerful tools for both preventing dengue infections and treating patients. Animal models play a pivotal role in vaccine development and drug screening. Available potential susceptible animals, including non-human primates, rodents, pigs, and tree shrews, have been extensively explored to establish animal models of dengue disease.

Evolution in the clinic: Maladaptive units and "minor anomalies".

It is here argued that the application of the term "minor anomalies" is often imprecise and likely outdated. In the past, the designation was used indiscriminately to refer to a great variety of unrelated morphogenetic phenomena. Also, the term does not discriminate between mild qualitative defects of development (mild malformations) and quantitative variants of normal structure. The human face was formed by natural and sexual selection. Morphological and morphogenetic analyses have shown that the human face with its skin, muscles, nerves, arteries, veins, glands, and lymphatics is a complex s…

Permanent diaphragmatic deficits and spontaneous respiratory plasticity in a mouse model of incomplete cervical spinal cord injury.

High spinal cord injuries (SCI) lead to permanent respiratory insufficiency, and the search for new therapeutics to restore this function is essential. To date, the most documented preclinical model for high SCI is the rat cervical C2 hemisection. However, molecular studies with this SCI model are limited due to the poor availability of genetically modified specimens. The aim of this work was to evaluate the pathophysiology of respiratory activity following a cervical C2 injury at different times post-injury in a C57BL/6 mouse model. No significant spontaneous recovery of diaphragmatic activit

Forensic molecular pathology: its impacts on routine work, education and training.

The major role of forensic pathology is the investigation of human death in relevance to social risk management to determine the cause and process of death, especially in violent and unexpected sudden deaths, which involve social and medicolegal issues of ultimate, personal and public concerns. In addition to the identification of victims and biological materials, forensic molecular pathology contributes to general explanation of the human death process and assessment of individual death on the basis of biological molecular evidence, visualizing dynamic functional changes involved in the dying

Comparable low-level mosaicism in affected and non affected tissue of a complex CDH patient.

In this paper we present the detailed clinical and cytogenetic analysis of a prenatally detected complex Congenital Diaphragmatic Hernia (CDH) patient with a mosaic unbalanced translocation (5;12). High-resolution whole genome SNP array confirmed a low-level mosaicism (20%) in uncultured cells, underlining the value of array technology for identification studies. Subsequently, targeted Fluorescence In-Situ Hybridization in postmortem collected tissues demonstrated a similar low-level mosaicism, independently of the affected status of the tissue. Thus, a higher incidence of the genetic aberrati

Hypopituitarism and anemia: effect of replacement therapy with hydrocortisone and/or levothyroxine.

Wiskott-Aldrich Syndrome (WAS) is a rare X-linked immunodeficiency characterized by microthrombocytopenia, eczema, and recurrent infections. In addition to this classical triad, recent studies have emphasized the roles of immune dysregulation, autoimmunity, chronic inflammation, endothelial dysfunction, and increased susceptibility to infections. These factors contribute to a broader disease spectrum that may involve not only bleeding complications but also, in rare instances, thrombotic events. In this review, we present three siblings from the same family affected by WAS. Two of them were ge

Pediatric dilated cardiomyopathy and the progression to heart failure: mechanisms and interventions focusing on the "gene-inflammation-metabolism" axis.

Dilated cardiomyopathy (DCM) is the most common cardiomyopathy in children and a leading indication for heart transplantation. Despite considerable progress in understanding its genetic architecture, translating these insights into improved outcomes for children remains challenging. The pathophysiology of pediatric dilated cardiomyopathy is highly complex, extending beyond a simple monogenic model to encompass intricate interactions between genetic predisposition, inflammatory triggers, and metabolic dysregulation. This review systematically synthesizes current evidence on the mechanisms drivi

[Developmental psychopathological characteristics of obsessive-compulsive disorder].

According to the currently available research data obsessive-compulsive disorder (OCD)is a disorder of multifactorial etiology, the causes of which include biological, genetic and environmental-social factors alike. Based on an etiology of that kind, it is justifiable to conduct a developmental psychopathological review of OCD, which may lead, through an exploration of the different factors involved, to a deeper understanding of the disorder's overall nature and specific characteristics, as well as to the development of the most efficient therapies possible. The main objective of the present c

Mutagenic selectivity of carcinogenic nitroso compounds: III. N,alpha-acetoxymethyl-N-methylnitrosamine.

A comparative genetic study was undertaken on the testicular tissue of Drosophila with N-alpha-acetoxymethol-N-METHYLNITROSAMINE (AcODMN) and its unsubstituted parent N,N-dimethylnitrosamine (DMN), to assess the role of intracellular metabolism on their mutagenicities. The relative genetic potencies of the two compounds were deduced from regression studies of the dose effect on the metabolically inert sperm and the metabolizing early germ cells (spermatocytes and spermatogonia) with respect to the induction of the non-specific X-chromosome recessives (lethals and visibles) and the specific eff
