We report a case of Burkholderia mallei causing glanders in a 73-year-old patient from the Northeast Region of Brazil. The patient was hospitalized with severe pneumonia. PCR and genomic sequencing confirmed B. mallei in pleural drainage. Genotyping revealed a novel genotype, emphasizing the need for genetic surveillance in zoonotic infections.

Burkholderia mallei and Burkholderia pseudomallei are Gram-negative bacteria that cause glanders and melioidosis, respectively. Inhalational infection with either organism can result in severe and rapidly fatal pneumonia. Inoculation by the oral and cutaneous routes can also produce infection. Chronic infection may develop after recovery from acute infection with both agents, and control of infection with antibiotics requires prolonged treatment. Symptoms for both meliodosis and glanders are non-specific, making diagnosis difficult. B. pseudomallei can be located in the environment, but in the

Molecular diagnostic kits for detection and identification of agents of melioidosis and glanders on environmental objects and in clinical material are described. It was demonstrated that PCR with use of specific primers on the basis of different genetic targets could be useful for determination of generic, inter- and intraspecies belonging of pathogenic Burkholderia as well as for epidemiologic inspection of territories where melioidosis is enzootic.

Little is known about the virulence factors of Burkholderia mallei, the etiologic agent of glanders. We employed subtractive hybridization to identify genetic determinants present in B. mallei but not in Burkholderia thailandensis, a non-pathogenic soil microbe. Three subtractive hybridization products were mapped to a genetic locus encoding proteins involved in the biosynthesis, export and translocation of a capsular polysaccharide. We identified an insertion sequence (IS 407 A) at one end of the capsule gene cluster and demonstrated that it was functional in B. mallei. Mutations were introdu

UNLABELLED: Burkholderia mallei are Gram-negative bacteria, responsible for the disease glanders. B. mallei has recently been classified as a Tier 1 agent owing to the fact that this bacterial species can be weaponised for aerosol release, has a high mortality rate and demonstrates multi-drug resistance. Furthermore, there is no licensed vaccine available against this pathogen. Lipopolysaccharide (LPS) has previously been identified as playing an important role in generating host protection against Burkholderia infection. In this study, we present gold nanoparticles (AuNPs) functionalised with

Glanders is a zoonotic disease caused by Burkholderia mallei (Bm). Bm is phylogenetically similar to B. pseudomallei (Bp), the causative agent of melioidosis, making it difficult to differentiate between them. Recently, the PCR method recommended by the World Organisation for Animal Health (WOAH) failed to detect a Kuwait strain of Bm, and the WOAH has cautioned that ongoing Bm evolution may lead to the emergence of variants that escape standard assays. Therefore, this study aimed to develop a novel real-time PCR method to discriminate between Bm and Bp. Local and web-based BLAST searches iden

Burkholderia pseudomallei and B. mallei are two highly pathogenic bacteria, responsible for melioidosis and glanders, respectively. The two are closely related and can also be mistaken for B. thailandensis, a nonpathogenic species. To improve their differential identification, we describe a hydrolysis probe-based real-time PCR method using the uneven distribution of type III secretion system genes among these three species.

A compilation of conditions deemed unusual from previous eras serves to highlight now obsolete jobs and remind that even relatively prevalent occupational disorders of today will be viewed retrospectively as strange in the proximate future. Discussed are coal miners' nystagmus, scrotal cancer in chimney sweeps, phossy jaw, hatters' shakes, painters' colic, potters' rot, chauffeurs' knee, glanders, caisson disease, and others.

Melioidosis, caused by the Gram-negative bacterium Burkholderia pseudomallei, is a severe infectious disease that is responsible for a significant amount of morbidity and mortality in endemic areas. While the majority of melioidosis cases occur in Southeast Asia, South Asia and Northern Australia, the disease is being increasingly recognized across tropical and subtropical regions worldwide. Due to diagnostic and treatment challenges as well as the potential misuse of B. pseudomallei as a biothreat agent, an effective vaccine is critically needed. Over the years, numerous different strategies

Burkholderia mallei is an aerobic, Gram-negative, non-motile bacillus. As an obligate mammalian pathogen, it primarily affects solipeds. Although rarely transmitted to humans, the disease it causes, glanders, is classified as a zoonosis. The bacterium was officially eradicated in Brazil in 1969; however, it reemerged after three decades. This study aims to assess the virulence of a specific B. mallei strain, isolated in Brazil, in BALB/c mice through intranasal infection. The strain, B. mallei BAC 86/19, was obtained from the tracheal secretion of a young mare displaying positive serology but

Treatment with an oligodeoxynucleotide (ODN) containing CPG motifs (CpG ODN 7909) was found to protect BALB/c mice from lung infection or death after aerosol challenge with Burkholderia mallei. Protection was associated with enhanced levels of gamma interferon (IFN-gamma)-inducible protein 10, interleukin-12 (IL-12), IFN-gamma, and IL-6. Preexposure therapy with CpG ODNs may protect victims of a biological attack from glanders.

Glanders, caused by a bacterium called B. mallei, is primarily an infectious horse and human disease. Although its incidence is rare in developed countries, it is nonetheless prevalent in several geographical areas of the world. There is a lack of cost-effective, rapid and specific molecular typing tools for epidemiological tracing of glanders cases. We previously reported an SNP-based typing method that categorizes global B. mallei strains into three lineages (L1 to L3), as well as additional branches, sub-branches and groups. However, further discrimination of the Indian and Pakistani isolat

is a highly pathogenic bacterium that causes the fatal zoonosis glanders. The organism specifies multiple membrane proteins, which represent prime targets for the development of countermeasures given their location at the host-pathogen interface. We investigated one of these proteins, Pal, and discovered that it is involved in the ability of B. mallei to resist complement-mediated killing and replicate inside host cells in vitro, is expressed in vivo and induces antibodies during the course of infection, and contributes to virulence in a mouse model of aerosol infection. Burkholderia pseudomallei and Burkholderia mallei are closely related gram-negative bacteria that can cause serious diseases in humans and animals. This review summarizes the current and rapidly expanding knowledge on the specific virulence factors employed by these pathogens and their roles in the pathogenesis of melioidosis and glanders. In particular, the contributions of recently identified virulence factors are described in the context of the intracellular lifestyle of these pathogens. Throughout this review, unique and shared virulence features of B. pseudomallei and B. mallei are discus

The specific features of the submicroscopic structure of the causative agents of melioidosis and glanders, grown in culture media and at the initial stages of experimental infection in guinea pigs and golden hamsters, were studied with the use of electronic cytochemistry and immunocytochemistry. P. pseudomallei and P. mallei were found to be capsule-forming bacteria. According to the data obtained with the use of electronic cytochemistry, the basis of the capsular substance P. pseudomallei and P. mallei is formed by polysaccharide biopolymers protecting these bacteria from phagocytosis.

An antigen, common for the infective agents of glanders , tularemia, plague, pseudotuberculosis, cholera and brucellosis, has been obtained from the aqueous saline extract of P. pseudomallei by means of affinity chromatography on antibody sorbent. The isolated antigen has proved to be of a protein-polysaccharide-lipid nature, to have insignificant cathode mobility and to constitute a component of O-antigen. It has also been found to be antigenically related to the tissues of melioidosis-sensitive animals and localized in the surface structure of the bacterial cell.

A hypothesis on the Achaean disease. Analyzing the text in the light of the modern medical knowledge, the article traces a profile of the mortal illness described in the first book of Homer's Iliad. The disease was a severe, acute epidemic zoonosis, rapidly bringing death. First, it affected mules and dogs, then it involved men; the infection spread quickly and the number of deaths was high. These elements suggest that the illness occurred among the Achaeans can be identified with glanders, an infectious disease affecting primarily mules and horses that can be transmitted to man by contact wit
