HLA antigens in Japanese patients with narcolepsy. All the patients were DR2 positive.

A surprisingly high association between an HLA-DR locus antigen and narcolepsy was revealed in the Japanese. All 40 Japanese patients with narcolepsy (22 males and 18 females) were confirmed to be DR2 positive. However, the DR2 of the patients was found to associate negatively with Bw52, whereas Bw52-DR2 is the commonest haplotype in normal Japanese population. These data suggested that a "disease" allele predisposing to narcolepsy was inherited with relatively rare haplotypes with DR2 in the Japanese.

[Cadaveric kidney transplantation and HLA typing in Switzerland 1977-1981. Combined analysis of Swisstransplant and EDTA Registry].

Graft survival for cadaveric kidneys transplanted in Switzerland between 1977 and 1981 was analysed according to HLA matching grade. Most if not all recipients had received blood transfusions before grafting. One year survival for first cadaveric grafts averaged 72 +/- 4% and differed little between groups of transplants with widely varying tissue matching grades both at the HLA A/B as well as DR loci. Hyperimmunized recipients with cytotoxic antibodies to over 50% of a random blood donor panel had equally satisfactory graft survival, particularly when matched according to Swisstransplant kidn

Histocompatibility antigens in primary biliary cirrhosis.

Primary biliary cirrhosis (PBC) is characterized by marked perturbation of the immune system. In addition, there are familial associations which suggest genetic background to the disease. We have studied HLA-A, B, and D loci in 15 Japanese women with PBC, and compared them to controls. Although HLA-A and B loci in PBC patients showed similar prevalence to the control population, HLA-DHO was significantly more common, occurring in 69% of patients with PBC and 21% of controls. This suggests a genetic predisposition to PBC, and the HLA-DHO locus association may reflect a genetic abnormality of th

The Jo-1 antibody system in myositis: relationships to clinical features and HLA.

Antibodies to Jo-1, a saline extractable nuclear antigen, were found in 11/47 (23%) patients with myositis and in 0/35 controls with systemic lupus erythematosus or progressive systemic sclerosis (p less than or equal to 0.01). Within myositis subgroups, anti-Jo-1 occurred in 6/20 (30%) with polymyositis, 2/16 (13%) with dermatomyositis and 3/7 (43%) with overlap. There was an association between Jo-1 antibodies and HLA-DR3, irrespective of race or clinical subgroups (p less than or equal to 0.05). All anti-Jo-1 positive patients had either HLA-DR3, HLA-DRW6 or both (p less than or equal to 0.

Major histocompatibility complex and central nervous system involvement by paracoccidioidomycosis.

Paracoccidioidomycosis (PCM) is a chronic granulomatous infectious disease, whose etiologic agent is the fungus Paracoccidioides brasiliensis. The central nervous system (CNS) involvement with paracoccidioidomycosis (NPCM) occurs more frequently than has been admitted in the past. There are some major histocompatibility complex antigen association studies with systemic paracoccidioidomycosis. Some indicate a positive association with HLA antigens, but there is no study with the involvement of the CNS. To investigate why not all cases of systemic PCM show the involvement of the CNS and whether

Case report: HLA-haploidentical HSCT rescued with donor lymphocytes infusions in a patient with X-linked chronic granulomatous disease.

Chronic granulomatous disease is an inborn error of immunity due to disrupted function of the nicotinamide adenine dinucleotide phosphate (NADPH) oxidase complex. This results in impaired respiratory burst of phagocytes and insufficient killing of bacteria and fungi. Patients with chronic granulomatous disease are at increased risk for infections, autoinflammation and autoimmunity. Allogeneic hematopoietic stem cell transplantation (HSCT) is the only widely available curative therapy. While HSCT from human leukocyte antigen (HLA) matched siblings or unrelated donors are standard of care, trans

The use of cellular reagents as an adjunct to typing for HLA-DRw10.

Two human T cell lines recognizing DRw10 were generated. One line was directly alloreactive, while a second cloned line recognized mumps virus in the context of a DRw10-like determinant. In panel studies, the lines responded only to DRw10 cells, and not to DR1 (DRw10-) cells. When compared to 5 anti-DRw10 sera, the cellular reagents gave virtually identical results. The generation and use of such cellular reagents may be of particular value for the identification of HLA-D-region specificities that are rare, poorly defined, or for which serologic typing reagents are limited.

Implantation site in normal pregnancy. A study with monoclonal antibodies.

In the present study, the presence of major histocompatibility complex antigens (MHC) and the degree and nature of inflammatory response in the human placenta were determined by staining frozen tissue sections with monoclonal antibodies and an immunoperoxidase technique. Although class I (HLA-A, B, and C) and Class II (HLA-DR, Ia-like) MHC antigens were not demonstrated in the syncytiotrophoblast, Class I antigens were found in trophoblast of the placental septum, shell, and implantation site and in the chorionic villous stroma. There was no staining for Ia-like antigens in the fetal component

Determining the Quantitative Principles of T Cell Response to Antigenic Disparity in Stem Cell Transplantation.

Alloreactivity compromising clinical outcomes in stem cell transplantation is observed despite HLA matching of donors and recipients. This has its origin in the variation between the exomes of the two, which provides the basis for minor histocompatibility antigens (mHA). The mHA presented on the HLA class I and II molecules and the ensuing T cell response to these antigens results in graft vs. host disease. In this paper, results of a whole exome sequencing study are presented, with resulting alloreactive polymorphic peptides and their HLA class I and HLA class II (DRB1) binding affinity quant

Corneal allograft rejection.

Corneal allograft rejection remains the major impediment to the success of the most commonly performed solid tissue transplant. Immunologic mechanisms, particularly cellular immunity, appear to be responsible for the majority of cases of graft failure. While the prognosis for dystrophic and noninflammatory conditions is highly favorable, the failure rate in vascularized corneas can be as high as 60%. Considerable interest in the HLA system has led to a greater understanding of the distributions of these major histocompatibility antigens in the cornea. It is generally believed that tissue typin

Hepatitis C virus and ethanol alter antigen presentation in liver cells.

Alcoholic patients have a high incidence of hepatitis C virus (HCV) infection. Alcohol consumption enhances the severity of the HCV disease course and worsens the outcome of chronic hepatitis C. The accumulation of virally infected cells in the liver is related to the HCV-induced inability of the immune system to recognize infected cells and to develop the immune responses. This review covers the effects of HCV proteins and ethanol on major histocompatibility complex (MHC) class I- and class II-restricted antigen presentation. Here, we discuss the liver which functions as an immune privilege o

Association of HLA class I and class II alleles with myositis in Japanese patients.

OBJECTIVE: To investigate the correlation of HLA class I and class II antigens and alleles with various forms of myositis in Japanese patients. METHODS: Eighty-four Japanese patients with myositis [22 with polymyositis (PM), 46 with dermatomyositis (DM), 16 with myositis overlapping with other collagen vascular diseases] were typed serologically for HLA-A, B, C antigens. HLA-DRB1, DQA1, and DQB1 alleles were determined by polymerase chain reaction dependent DNA typing methods. Fifty-eight Japanese controls were typed serologically while HLA-DRB1, DQA1, and DQB1 allele typing was carried out i

HLA-A*9, a probable secondary susceptibility marker to ankylosing spondylitis in Basque patients.

HLA-B27 is strongly associated to ankylosing spondylitis (AS). The objective of our study was to analyze HLA-B27 association, B27 subtype distribution and frequency of other HLA class I and DR antigens in a group of Basque AS patients. HLA class I antigens were typed serologically and HLA-B27 and A9 subtypes were determined by DNA typing in samples from 46 patients with AS, 54 B27-positive spondyloarthropathies, 82 healthy subjects and 20 B27-positive controls. A class I HLA 9.2 kb PvuII restriction fragment length polymorphism (RFLP), previously associated with AS, was analyzed in a represent

3D genome contributes to MHC-II neoantigen prediction.

Reliable and ultra-fast DNA and RNA sequencing have been achieved with the emergence of high-throughput sequencing technology. When combining the results of DNA and RNA sequencing for tumor cells of cancer patients, neoantigens that potentially stimulate the immune response of either CD4+ or CD8+ T cells can be identified. However, due to the abundance of somatic mutations and the high polymorphic nature of human leukocyte antigen (HLA) it is challenging to accurately predict the neoantigens. Moreover, comparing to HLA-I presented peptides, the HLA-II presented peptides are more variable in le

Human Leukocyte Antigen Markers for Distinguishing Pustular Psoriasis and Adult-Onset Immunodeficiency with Pustular Reaction.

Pustular skin diseases, with pustular psoriasis (PP) being the prototype, are immune-mediated diseases characterized by the presence of multiple pustules, resulting from neutrophil accumulation in the layer of epidermis. Sterile skin pustular eruption, like PP, is also observed in 20-30% of patients with adult-onset immunodeficiency syndrome (AOID) and anti-interferon γ autoantibodies (IFN-γ), leading to challenges in classification and diagnosis. While the mechanism underlying this similar phenotype remains unknown, genetic factors in relation to the immune system are suspected of playing an

Class II major histocompatibility complex typing across the ethnic barrier in pemphigoid gestationis. A study in Mexicans.

BACKGROUND: Pemphigoid gestationis (PG), also called herpes gestationis, is a rare autoimmune disease of pregnancy or puerperium (estimated 1 out of 50,000 pregnancies among Caucasians). A previous series has demonstrated an association of PG with human leukocyte antigen (HLA)-DR3 or HLA-DR4 haplotypes. While these haplotypes are most commonly found in individuals of European ancestry, they have also been found in African-American patients affected with PG. PG has rarely been reported in other ethnic groups, and the HLA association in non-Europeans has not been examined. METHODS: We have char
