Opposite sex-specific effects of Wolbachia and interference with the sex determination of its host Ostrinia scapulalis.

In the adzuki bean borer, Ostrinia scapulalis, the sex ratio in most progenies is 1 : 1. Females from Wolbachia-infected matrilines, however, give rise to all-female broods when infected and to all-male broods when cured of the infection. These observations had been interpreted as Wolbachia-induced feminization of genetic males into functional females. Here, we show that the interpretation is incorrect. Females from both lines have a female karyotype with a WZ sex-chromosome constitution while males are ZZ. At the time of hatching from eggs, WZ and ZZ individuals are present at a 1 : 1 ratio i

Visualization, fate, and pathogenicity of antigen-specific CD8+ T cells in the graft-versus-host reaction.

To follow the fate of alloreactive T cell effectors in graft-vs-host disease, Ld-specific CD8+ T cells from C57BL/6 2C TCR-transgenic donors were transplanted into sublethally irradiated (750 cGy) Ld+ or Ld- recipients. In Ld- C57BL/6 or (BALB/c-dm2 x C57BL/6)F1 recipients, naive 2C T cells engrafted and survived long term, but did not acquire effector function. In Ld+ (BALB/c x C57BL/6)F1 recipients, 2C T cells engrafted, expanded, became cytolytic, destroyed host B cells and double-positive thymocytes, and later disappeared. Despite marked damage to lymphoid and hemopoietic cells by 2C T cel

Immunotherapy of graft versus host disease with specific alloantiserum: restoration of cerebellar development in infant rats.

Fischer (FI) rats were stimulated to produce high-titre alloantiserum against DA antigens for alleviation of the symptoms of graft versus host disease (g.v.h.d.) in FI neonates. G.v.h.d. was procured by intravenous injection of DA lymph node cells on the day of birth. Daily injections of alloantiserum on postnatal days 11-13 not only prevented the progress of the disease in the whole animal but reversed the altered neuronal histogenesis that was present in diseased animals at day 11. Specifically, by 3 days after onset of alloantiserum treatment, the severely depressed DNA synthesis had return

Genomic comparison of contemporaneous dog and human clinical E. coli isolates in Denmark reveals lineage-specific host associations and limited genomic relatedness across hosts.

Escherichia coli causes extraintestinal infections in humans and dogs, but the genomic relatedness of clinical lineages circulating in both hosts within the same region remains unclear. This study assessed the genomic relatedness between canine and human clinical isolates in the Zealand Region, Denmark, during 2022-2023. Ninety-five canine clinical E. coli isolates obtained from routine veterinary diagnostics were sequenced using Illumina MiSeq. Among 3832 contemporaneous human clinical genomes from the same region, all 1263 isolates belonging to the three most prevalent canine sequence types

Synergistic effects of two bacteriophages with distinct infection patterns and broad host specificity against multidrug-resistant Salmonella Typhimurium and their potential applications in the poultry industry.

Controlling Salmonella Typhimurium in poultry requires effective alternatives to conventional antimicrobials. This study evaluated the efficacy of a two-phage cocktail composed of S. Typhimurium-infecting phages SLAM_phiST45 and SLAM_phiST56 and investigated its mechanistic basis. Host-range profiling of 61 phages against 10 S. Typhimurium strains revealed two major host-associated clusters (swine-human vs. poultry). Notably, the cocktail exhibited broad host ranges that crossed these clusters. The cocktail showed clear synergy, supported by complementary receptor usage and reduced emergence o

Diversity and host specificity of Borrelia burgdorferi's outer surface protein C (ospC) alleles in synanthropic mammals, with a notable ospC allele U absence from mixed infections.

Interactions among pathogen genotypes that vary in host specificity may affect overall transmission dynamics in multi-host systems. Borrelia burgdorferi, a bacterium that causes Lyme disease, is typically transmitted among wildlife by Ixodes ticks. Despite the existence of many alleles of B. burgdorferi's sensu stricto outer surface protein C (ospC) gene, most human infections are caused by a small number of ospC alleles ["human infectious alleles" (HIAs)], suggesting variation in host specificity associated with ospC. To characterize the wildlife host association of B. burgdorferi's ospC alle…

New insights into the evolution of host specificity of three Penicillium species and the pathogenicity of P. Italicum involving the infection of Valencia orange (Citrus sinensis).

UNLABELLED: Blue and green molds, the common phenotypes of post-harvest diseases in fruits, are mainly caused by Penicillium fungal species, including P. italicum, P. digitatum, and P. expansum. We sequenced and assembled the genome of a P. italicum strain, which contains 31,034,623 bp with 361 scaffolds and 627 contigs. The mechanisms underlying the evolution of host specificity among the analyzed Penicillium species were associated with the expansion of protein families, genome restructuring, horizontal gene transfer, and positive selection pressure. A dual-transcriptome analysis following t…

Molecular characterization of Cryptosporidium in wild rodents from the Inner Mongolian Autonomous Region and Liaoning Province, China: assessing host specificity and the potential for zoonotic transmission.

INTRODUCTION: Wild rodents are key hosts for Cryptosporidium transmission, yet there is a dearth of information regarding their infection status in the Inner Mongolian Autonomous Region and Liaoning Province of China. Therefore, the present study was conducted to determine the prevalence and genetic characteristics of Cryptosporidium among wild rodents residing in these two provinces. METHODS: A total of 486 rodents were captured, and fresh feces were collected from each rodent's intestine for DNA extraction. Species identification of rodents was performed through PCR amplification of the ver

Scope of Microbial Transglutaminase for Site-Specific and Oriented Immobilization of Native Antibodies from Various Host Species.

Modification of antibodies to chemically couple labels or immobilization reagents is essential for developing biosensors. Typically, conjugation occurs through chemical methods that leverage reactive amines and thiols on native antibodies; however, this nonspecific approach can interfere with antibody function. Microbial transglutaminase (mTG) is an enzyme that has been used for site-specific conjugation of chemical modifiers to the Fc region of native antibodies, but thus far mTG-mediated conjugation has been limited to production of antibody-drug conjugates with human IgGs. Here, we assessed

Pseudorabies virus infection reshapes the host epitranscriptome by globally suppressing but selectively elevating specific RNA modifications.

RNA modifications are ubiquitous in host cells and hold crucial roles in regulating diverse biological processes. Although the m6A modification during viral infections has garnered significant attention, the involvement of other modifications in such infections remains underexplored. This study was aimed to map out the comprehensive RNA modification profiles in porcine kidney PK-15 cells infected with pseudorabies virus (PRV) at various time points. Throughout the PRV infection, we identified a total of 31 distinct RNA modifications, among which 25 were detected in significantly higher abundan

Specific host translation perturbations promote Tn Smu 1 loss independently of the canonical ImmA regulator.

Integrative and conjugative elements (ICEs) are major drivers of horizontal gene transfer and bacterial genome evolution. Although ICE-encoded regulatory circuits have been extensively characterized, the impact of host physiology on the stability of integrated ICEs remains poorly understood. Here, we identify a host-dependent pathway that links specific host translation perturbations to loss of the ICE Tn Smu1 in Streptococcus mutans . Analysis of host-gene deletion mutants revealed that disruption of fmt , rnjA , or rnjB -three translation-associated host genes-reproducibly promoted Tn Smu1 l

Mitochondrial cytochrome b motif architecture predicts host-specific genomic vulnerability across vertebrate species.

Mitochondrial DNA (mtDNA) plays a pivotal role in cellular bioenergetics, immune regulation, and evolutionary adaptation, making it a uniquely informative genomic system for studying host-pathogen interactions. The cytochrome b (cyt b) gene, one of the most conserved mitochondrial protein-coding loci, is widely recognized for its utility in species identification, phylogenetics, and molecular evolution [1-3]. However, its potential role in shaping comparative host susceptibility to infectious diseases remains underexplored. In this study, we re-analyzed PCR-RFLP-derived mitochondrial cyt b res

Identification of an Almond Cultivar Resistant to Xylella fastidiosa subsp. fastidiosa and Insights into Host-Specific Bacterial Behavior.

Xylella fastidiosa (Xf) is a xylem-limited bacterial pathogen causing disease in a variety of important agricultural crops, including almonds. Almond leaf scorch (ALS) disease was first identified in Israel in 2017 and is associated with the subspecies X. fastidiosa subsp. fastidiosa (Xff) sequence type 1. Currently, there is no effective means to cure infected almond trees, and ALS management relies primarily on uprooting infected trees and reducing vector populations, yet these methods are only moderately effective. Thus, identifying resistant almond genotypes is a critical goal for developi

Characterization of the role of putative Aeromonas caviae-specific virulence factor, flgB, in virulence and host-pathogen interactions.

Aeromonas caviae, Gram-negative bacteria ubiquitous in the environment, are an emerging human pathogen associated with various infectious diseases, particularly gastroenteritis. Despite recent studies demonstrating A. caviae is the most predominant Aeromonas species underlying human infection, A. caviae remains understudied, and no A. caviae-specific virulence factors associated with human disease have been identified. To identify A. caviae-specific putative virulence factors, we conducted comparative genomic analyses among clinical Aeromonas isolates (n = 431), which identified a variant of f

Comparative Patho-Genomics of Salmonella enterica Serovar Enteritidis Reveal Potential Host-Specific Virulence Factors.

Salmonella enterica serovar Enteritidis (S. Enteritidis) is one of the most common causes of bacterial foodborne infections worldwide. It has an extensive host range, including birds and humans, making it one of the most adaptable Salmonella serovars. This study aims to define the virulence gene profile of S. Enteritidis and identify genes critical to its host specificity. Currently, there is limited understanding of the molecular mechanisms that allow S. Enteritidis to continue as an important foodborne pathogen. To better understand the genes that may play a role in the host-specific virulen…
