Imipramine improves motor impairments in a rat model of Parkinson's disease induced by 6-hydroxydopamine; the role of oxidative stress and neurotrophic factors.

INTRODUCTION: Parkinson's disease (PD) is a progressive neurological disorder characterized by the loss of dopaminergic neurons in the substantia nigra and is associated with neuroinflammation, apoptosis, oxidative stress, and motor impairment. Imipramine, a tricyclic antidepressant, has a wide range of biological effects such as anti-inflammatory, anti-apoptotic, and free radical scavenging activities. The present study was designed to investigate the neuroprotective effect of imipramine in a rat model of PD induced by 6-hydroxydopamine (6-OHDA). METHODS: Male Wistar rats were treated with d

Oral sodium butyrate alleviates motor deficits and dopaminergic neuronal loss in 6-hydroxydopamine-induced Parkinson's disease rat model.

BACKGROUND: Short-chain fatty acids regulate intestinal homeostasis and modulate the gut-brain axis, suggesting they may be applicable in adjunctive treatments for Parkinson's disease (PD). However, the therapeutic efficacy of sodium butyrate (NaB) in PD remains unclear. Using appropriate PD animal models may aid in clarifying the therapeutic effects of NaB and facilitate the development of NaB-based PD treatment strategies. We comprehensively evaluated the therapeutic effects of oral NaB in rats with 6-hydroxydopamine (6-OHDA)-induced PD. METHODS: We induced hemiparkinsonism in rats by admin

Comparison of the Efficiency of Fractional Anisotropy or Relaxation Time T2* in the Early Diagnosis of 6-Hydroxydopamine-Induced Rat Model of Parkinson's Disease.

Assessment of fractional anisotropy (FA) in diffusion tensor imaging (DTI) and measurement of transverse relaxation time T2* in T2*-weighted imaging (T2*WI) are the basic MRI methods used in the diagnosis of Parkinson's disease (PD). However, it remains unclear which of these methods is more effective. We compared the sensitivity and specificity of DTI and T2*WI methods to determine which one is more effective in the early diagnosis of PD. For PD modeling, male Sprague-Dawley rats (n = 20) were injected with 6-hydroxydopamine (6-OHDA) into the right substantia nigra (SN). MRI was performed and

Expression of Galectin 3 and Activating Transcription Factor 3 in Nigral Dopaminergic Neurons of 6-Hydroxydopamine Induced Parkinsonian Rat Model.

BACKGROUND/AIM: Parkinson's disease (PD) is an age-related neurodegenerative disease marked by the relatively progressive dopaminergic neuronal loss in the substantia nigra (SN). Retrograde degeneration of the nigrostriatal dopaminergic neurons by 6-hydroxydopamine (6-OHDA) has been widely used as a PD animal model, while endogenous 6-OHDA promotes the progression of PD pathology. Galectin 3 (Gal3) and activating transcription factor 3 (ATF3) have been implicated in neurodegenerative processes. The aim of this study was to investigate the expression pattern and roles of Gal3 and ATF3 in a Park

Paeony root prevents 6-hydroxydopamine-induced neurotoxicity by regulating autophagic dysfunction via AKT signaling in Parkinson's disease models.

Parkinson's disease (PD) is a neurodegenerative disease that primarily affects motor function. Dopaminergic cell death occurs progressively in the PD brain and autophagic dysfunction is critically implicated in the neuronal death. Therefore, modulating impaired autophagic flux is a promising strategy to treat dopaminergic neurodegeneration, addressing the unmet needs in PD. This study aimed to investigate efficacy and the underlying mechanism of Paeonia lactiflora extract (PLE) focusing on autophagic dysfunction in 6-hydroxydopamine (6-OHDA)-induced PD models in vitro and in vivo for the first

Effects of Unilateral High Frequency Stimulation of the Subthalamic Nucleus on Risk-avoidant Behavior in a Partial 6-hydroxydopamine Model of Parkinson's Disease.

BACKGROUND: Deep brain stimulation (DBS) of the subthalamic nucleus (STN) is a well-established treatment for the motor symptoms of Parkinson's disease (PD). While PD is primarily characterized by motor symptoms such as tremor, rigidity, and bradykinesia, it also involves a range of non-motor symptoms, and anxiety is one of the most common. The relationship between PD and anxiety is complex and can be a result of both pathological neural changes and the psychological and emotional impacts of living with a chronic progressive condition. Managing anxiety in PD is critical for improving the patie

Acupuncture Alleviates Levodopa-Induced Dyskinesia via Melanin-Concentrating Hormone in Pitx3-Deficient aphakia and 6-Hydroxydopamine-Lesioned Mice.

Although L-3,4-dihydroxyphenylalanine (L-DOPA) is currently the most effective medication for treating Parkinson's disease (PD) motor symptoms, its prolonged administration causes several adverse effects, including dyskinesia. To identify the mechanisms underlying the effects of acupuncture on L-DOPA-induced dyskinesia (LID), antidyskinetic effects of acupuncture were investigated in two mouse models of PD. Acupuncture stimulation at GB34 alleviated abnormal involuntary movements (AIMs) in Pitx3-deficient aphakia mice (ak/ak) following L-DOPA administration and these effects were reproduced in

Botanical Drug Puerarin Attenuates 6-Hydroxydopamine (6-OHDA)-Induced Neurotoxicity via Upregulating Mitochondrial Enzyme Arginase-2.

Inhibition of nitric oxide synthases (NOSs) shows promise to halt the progression of neurodegenerative diseases. The present study was designed to explore whether botanical isoflavone puerarin could attenuate nitric oxide (NO)-mediated neurotoxicity via modulating the enzymes in the L-arginine-NO pathway. Neurotoxin 6-hydroxydopamine (6-OHDA) is well known to induce neurodegeneration via a NO-dependent mechanism. We first validated that puerarin protected rat dopamingeric PC12 cells against 6-OHDA-induced neurotoxicity in a concentration-dependent manner. We subsequently profiled the cellular

Colour-coded pellets increase the sensitivity of the staircase test to differentiate skilled forelimb performances of control and 6-hydroxydopamine lesioned rats.

The Montoya staircase test has previously been used to study the skilled forelimb performance of mice and rats following lesions and cell implants in different parts of the central nervous system. Here we describe a modification of the original test design which introduces differently coloured food pellets for each step, and present the results of the new and modified method. In this study unilaterally 6-hydroxydopamine (6-OHDA) lesioned rats and healthy control rats were used. The new evaluation of reaching and grasping movements takes into consideration the various levels of reaching difficu

Nitric oxide prevents 6-hydroxydopamine-induced apoptosis in PC12 cells through cGMP-dependent PI3 kinase/Akt activation.

Nitric oxide (NO) functions not only as an important signaling molecule in the brain by producing cGMP, but also regulates neuronal cell apoptosis. The mechanism by which NO regulates apoptosis is unclear. In this study, we demonstrated that NO, produced either from the NO donor S-nitroso-N-acetyl-d,l-penicillamine (SNAP) or by transfection of neuronal NO synthase, suppressed 6-hydroxydopamine (6-OHDA)-induced apoptosis in PC12 cells by inhibiting mitochondrial cytochrome c release, caspase-3 and -9 activation, and DNA fragmentation. This protection was significantly reversed by the soluble gu

6-hydroxydopamine lesion in the A8 cell group of cats produces a short-lasting decreased accuracy in goal-directed forepaw-movements.

Recently, feline studies have shown that a lesion in the retrorubral area, which includes the dopaminergic A8 cell group, produces motor programming deficits inherent to a hypofunction of the A9 system. A hypofunction in the striatal terminal area of A9 fibers, in turn, is known to produce a hypofunction of its first-order output station, namely the substantia nigra pars reticulata (SNR). The integrity of the SNR allows animals to execute (1) 'postural adjustments that rely on proprioceptive stimuli that originate in body parts at rest' and (2) 'non-externally guided' targeting movements. Dopamine D1 receptor modulation of glutamate receptor messenger RNA levels in the neocortex and neostriatum of unilaterally 6-hydroxydopamine-lesioned rats.

The effect of treatment with the D1 dopamine receptor agonist SKF 38393 on the expression of metabotropic glutamate receptor 1, 3, 4 and 5 receptor subtypes and of the glutamate N-methyl-D-aspartate ionotropic receptor subunits NRI, NR2A and NR2B was analysed using in situ hybridization. We studied the neocortex and neostriatum of normal rats and of rats unilaterally treated with 6-hydroxydopamine, a neurotoxin that, after intracerebral injection into the ventral tegmental area, causes selective degeneration of the ascending dopamine pathway. In the 6-hydroxydopamine-lesioned rats, metabotropi

Effect of 6-hydroxydopamine lesions of the medial prefrontal cortex on intravenous cocaine self-administration under a progressive ratio schedule of reinforcement.

Rats were trained to self-administer intravenous cocaine under a progressive ratio (PR) schedule of reinforcement. Under this schedule, an increasing number of lever responses had to be made to obtain each subsequent reinforcement (1.5 mg/kg per injection). Once stable responding was achieved with this schedule, bilateral 6-hydroxydopamine (6-OHDA) or vehicle-only injections were delivered into the medial prefrontal cortex (mPFC). Following recovery from surgery, the animals were given access to cocaine under the PR schedule. The effect of the lesion on self-administration behaviour was examin

Levodopa replacement therapy alters enzyme activities in striatum and neuropeptide content in striatal output regions of 6-hydroxydopamine lesioned rats.

The effects of striatal dopamine denervation and levodopa replacement therapy on neuronal populations in the rat striatum were assessed by measurement of glutamic acid decarboxylase (GAD) and choline acetyltransferase (CAT) activities in the striatum, dynorphin and substance P concentrations in the substantia nigra, and enkephalin concentration in the globus pallidus. Rats with a unilateral 6-hydroxydopamine (6-OHDA) lesion of the nigrostriatal pathway were treated for 21 days with levodopa (100 mg/kg/day, i.p., with 25 mg/kg benserazide) on either an intermittent (b.i.d.) or continuous (osmot

Chemogenetic activation of CaMKⅡα positive neurons in the ventral hippocampus produces antidepressant and anxiolytic effects in unilateral 6-hydroxydopamine-lesioned rats.

Comorbidity of depression and anxiety is quite common in Parkinson's disease (PD). The ventral hippocampus (vHIP) has been widely implicated in depression and anxiety. However, the roles of CaMKⅡα-positive neurons in the vHIP in depression- and anxiety-like behaviors associated with PD in rats remain unclear. Here, we used chemogenetic techniques, such as designer receptors exclusively activated by designer drugs (DREADDs), to activate or inhibit CaMKⅡα-positive neurons in the vHIP of rats. In the present study, the unilateral 6-hydroxydopamine (6-OHDA) lesions of the medial forebrain bundle (

Ginkgolide B Attenuates 6-Hydroxydopamine-Induced Cellular Damage and Modulates Oxidant-Evoked Ca2+ Signalling in SH-SY5Y Cells.

Oxidative stress and disruption of intracellular Ca2+ homeostasis contributes to dopaminergic neuronal damage in Parkinson's disease. In this study, the protective effects of Ginkgolide B (GKB) against 6-hydroxydopamine (6-OHDA)-induced SH-SY5Y cell damage were investigated, with particular attention to oxidant-evoked intracellular Ca2+ signalling. Cells were pre-treated with GKB (20 µg/mL) for 2 h, followed by exposure to 6-OHDA (200 µM) for 24 h. CCK-8-derived metabolic activity, oxidative stress, cell death, PARP-1, caspase-3, and intracellular Ca2+ responses were evaluated using biochemica

The Inhibition of Fatty Acid Amide Hydrolase-4 Affords Neuroprotection in a Toxic Model Induced by 6-Hydroxydopamine in Caenorhabditis elegans Nematodes.

Parkinson's disease (PD) is a neurodegenerative disorder characterized by progressive loss of dopaminergic neurons. Drugs modulating the endocannabinoid system (ECS) emerge as promising tools for reducing symptomatology and/or progression of this disease. Here, the protective potential of ECS regulation against 6-hydroxydopamine (6-OHDA)-induced neuronal damage was evaluated in the nematode, Caenorhabditis elegans. First, the effect of the endocannabinoid anandamide (AEA; 1 to 100 µM) was tested in adult nematodes. As this eCB had no protective effects on the 6-OHDA-induced increased neurodege
