N-hydroxy-2-thiopyridone (1), an established source of the hydroxyl radical (HO., Boivin, J., Crepon, E., and Zard, S. Z. (1990) Tetrahedron Lett. 31, 6869-6872), produced HO. under conditions directly applicable to biological studies. Generation of HO. by subjecting 1 to irradiation with visible light was monitored in the following "HO." assays: deoxyribose degradation, addition to dimethyl sulfoxide, and hydroxylation of salicylate and phenol. All four assays demonstrated the production of HO. from 1 (added as a sodium salt) under mild conditions in aqueous buffer systems. An improved analys

Natural colloidal particles (NCPs) are ubiquitous in lake ecosystems, whereas information on the photochemical production, distribution, and formation mechanism for hydroxyl radicals (•OH) from NCPs remains unknown. Herein, NCPs from different lake waters were extracted, with source- and concentration-dependent heterogeneities in formation potential, microheterogeneous distribution, and mechanism for •OH being explored. NCPs were composed of inorganic colloidal particles (ICPs) and organic colloidal particles (OCPs), and NCPs-mediated •OH was divided into free •OH and bound •OH. Results showed

Despite the importance of the oxidation of reduced sulfur compounds in the biogeochemical sulfur cycle and climate, the mechanism of the transformation of methanethiol (CH3SH), the second highest gas-phase marine sulfur emission besides dimethyl sulfide, remains weakly explored. This work presents an investigation of the reaction of CH3SH with hydroxyl radicals (˙OH) and chlorine atoms (Cl˙) using the DLPNO-CCSD(T)/aug-cc-pVTZ//M06-2X/aug-cc-pV(T+d)Z method, both in the absence and presence of water. The results indicate that the hydrogen abstraction by ˙OH/Cl˙ from the -SH group is more favor

A photochemical system combining iron (hydr)oxides and oxalate (Ox) shows application prospects in wastewater treatment due to the abundance of reactive oxygen species (ROS) generation. Nevertheless, it is a challenge to the investigate photochemical activity of iron (hydr)oxides/Ox systems with varying structural properties. Herein, the photochemical behaviors of Ox on goethite (Gt) surface from the view of structural dependent activity, containment degradation, and ROS generation were explored in detail. Results confirmed that bidentate mononuclear was formed on Gt surface after complexing O

Frequent cycles of flooding and drainage in paddy soils lead to the reductive dissolution of iron (Fe) minerals and the reoxidation of Fe(II) species, all while generating a robust and consistent output of reactive oxygen species (ROS). In this study, we present a comprehensive assessment of the temporal and spatial variations in Fe species and ROS during the flooding-drainage process in a representative paddy soil. Our laboratory column experiments showed that a decrease in dissolved O2 concentration led to rapid Fe reduction below the water-soil interface, and aqueous Fe(II) was transformed

The antioxidant properties of condensed tannins (CTs) are closely related to the mean degree of polymerization (mDP), and CTs with low mDP show stronger antioxidant effects. Therefore, obtaining CTs with a low mDP are very meaningful in improving their antioxidant properties and utilization. In this study, hydroxyl radicals generated by the decomposition of hydrogen peroxide under UV irradiation were used to degrade bayberry tannins in a clean and controllable manner. Taking the formaldehyde reactivity as an index to control the mDP of the degradation product, the changes in antioxidant proper

Fe-based materials are currently considered for manufacturing biodegradable coronary stents. Here we show that Fe has a strong potential to generate hydroxyl radicals (HO) during corrosion. This HO generation, but not corrosion, can be inhibited by catalase. Oxidative stress was observed (increased HO-1 expression) in aortic rings after direct exposure to Fe, but not in the presence of catalase or after indirect exposure. This oxidative stress response induced an uncoupling of eNOS in, and a consequent reduced NO production by endothelial cells exposed to Fe. In isolated rat aortic rings NO pr

BACKGROUND: Rhododendrol (4-(4-hydroxyphenyl)-2-butanol) has been used as a lightening/whitening cosmetic but was recently reported to induce leukoderma. Although rhododendrol has been shown to be transformed by tyrosinase to hydroxyl-rhododendrol, which is cytotoxic to melanocytes, its detailed mechanism of action including the involvement of reactive oxygen species is not clearly understood.
OBJECTIVE: To confirm the relationship of hydroxyl radical generation to melanocyte cytotoxicity induced by rhododendrol, this study was performed.
METHODS: An electron spin resonance method with a hig

Photooxidation of arsenite(As(III)) in a suspension of natural montmorillonite under the irradiation of metal halide lamp (λ ≥ 313 nm)has been investigated. The results showed that the natural montmorillonite induced the photooxidation of As(III) by generating hydroxyl radicals (HO·) and hydroperoxyl/superoxide radicals (HO₂·/O₂⁻·). HO· which was responsible for the As(III) photooxidation. Approximately 38% of HO· was generated by the photolysis of ferric ions, and the formation of the remaining 62% was strongly dependent on the HO₂·/O₂⁻·. The presence of free ironions (Fe(2+) and Fe(3+)), mad

BACKGROUND AND OBJECTIVE: Isoflurane has been shown to mimic ischaemic preconditioning (IPC). The protective effect of IPC, or applying isoflurane or perfusion with the 'push-pull' complex zinc-desferrioxamine (Zn-DFO) in the canine heart, was investigated.
METHODS: Thirty minutes after salicylate administration (100 mg kg(-1)) the heart was exposed. All dogs were subjected to a 10 min left anterior descending artery occlusion followed by 2 h of reperfusion. In Group I (n = 9) isoflurane (2.5%) was administered 10 min prior to and during ischaemia. In Group II (n = 8), IPC was elicited by 5 m

We have used microdialysis to establish an experimental model to characterize mechanisms whereby released substances cause secondary damage in spinal cord injury. We use this model here to characterize damaging effects of the hydroxyl radical (OH.) in vivo in the spinal cord. OH. was generated in vivo by pumping H2O2 and FeCl2/EDTA through parallel microdialysis fibers inserted into the spinal cord. These agents mixed in the tissue to produce OH. by Fenton's reaction. Two types of control experiments were also conducted, one administering only 5 mM H2O2 and the other only 0.5 mM FeCl2/0.82 mM

Tissue damage in many diseases is caused by hydroxyl radicals, generated during single electron reduction of oxygen. The first step is usually the formation of the superoxide radical. This radical is constantly formed in all living cells, and in particular during activation of phagocytes or during reoxygenation following ischaemia. Damage, however, only occurs in the presence of catalytic transition metals of which iron is the most important in human pathology. Oxygen-radical-mediated damage can be prevented by iron chelators, as has been demonstrated in numerous in vitro and in vivo experimen

2,2'-Dithiodipyridine (PySSPy) is a typical pyridine disulfide, which has been widely used in industry and biomedical research, and frequently identified as the photo-degradation product of ubiquitous metal pyrithiones. Although many disulfides can moderately react with physiological oxidants such as H2O2, it was rarely reported that the highly-reactive hydroxyl radicals (•OH) could be produced from the reaction of disulfides with H2O2. Here we unexpectedly found that PySSPy can readily react with H2O2 over a wide pH range to continuously produce •OH and lead to appreciable DNA damage. Compare

Reactive oxygen species (ROS) are critical in tumor therapy, but the antioxidant tumor microenvironment severely restricts the ROS-modulated antitumor efficacy. In this study, a photothermal-promoted cascade nanozyme (MnO2@Lap/PAH/L-Arg@Au) was constructed by modifying β-lapachone (β-Lap), poly(allylamine hydrochloride) (PAH), and L-arginine (L-Arg) and depositing gold nanoparticles (AuNPs) on hollow manganese dioxide (MnO2), enhancing ROS-modulated catalytic/gas combination therapy. The pH- and glutathione-responsive hollow MnO2 degradation released β-Lap to elevate the generation of hydrogen

Charcot-Marie-Tooth disease type 2Z (CMT2Z) is an inherited axonal neuropathy caused by haploinsufficiency of microrchidia CW-type zinc finger protein 2 (MORC2), which leads to elevated hydroxyl radical levels, reduced ATPase activity, and apoptosis-mediated neuromuscular degeneration. CMT2Z presents with severe clinical manifestations, yet no widely applicable and affordable treatment has been developed. While gene therapy presents a theoretical solution, its feasibility remains constrained by prohibitive costs and delivery challenges. We observed sex-specific differences in muscle function i

BACKGROUND: In the past decades, antimicrobial resistance (AMR) has been a major threat to global public health. Long-term, chronic otitis media is becoming more challenging to treat, thus the novel antibiotic alternative agents are much needed.
METHODS: ZnO@TiO2@AMP (ATZ NPs) were synthesized through a solvothermal method and subjected to comprehensive characterization. The in vitro and in vivo antibacterial effect and biocompatibility of ATZ NPs were evaluated. For the antibacterial mechanism exploration, we utilized the Electron Paramagnetic Resonance (EPR) Spectrometer to detect and analy

PURPOSE: Radiotherapy (RT) is currently recognized as an important treatment for glioblastoma (GBM), however, it is associated with several challenges. One of these challenges is the radioresistance caused by hypoxia, whereas the other is the low conversion efficiency of the strongly oxidized hydroxyl radical (•OH), which is produced by the decomposition of water due to high-energy X-ray radiation. These factors significantly limit the clinical effectiveness of radiotherapy.
RESULTS: To address these limitations, we developed a highly stable and efficient nanoplatform (MnO2/Pt@BSA). Compared

1,2-dibromo-4-(1,2-dibromoethyl)cyclohexane (TBECH), as an emerging brominated flame retardant (EBFR) pollutant, has been often observed in the air, and to comprehend its fate in the environment is still challenging due to the diversity of its stereoisomers. In this work, the environmental transformation behavior and potential toxicological implications of TBECH stereoisomers under the oxidation of OH· in the gas phase were investigated by computational chemistry. Our results indicate the complexity of the TBECH transformation reactions and the diversity of its transformation products in the a

Excessive oxidative stress has been recognized as an important cause of the adverse health effects associated with exposure to ambient particulate matter (PM). Transition metals (TMs) (e.g., iron (Fe) and copper (Cu)) are known catalysts in the formation of reactive oxygen species (ROS) in surrogate lung fluid containing antioxidants. Humic-like substances (HULIS), extracted from atmospheric aerosols, retain the compositional complexity of real-world samples. It contains mixtures of organics that chelate TMs and was used in this work to examine the roles of atmospheric organics in affecting RO
