SARS-CoV-2-derived peptides define heterologous and COVID-19-induced T cell recognition.

T cell immunity is central for the control of viral infections. To characterize T cell immunity, but also for the development of vaccines, identification of exact viral T cell epitopes is fundamental. Here we identify and characterize multiple dominant and subdominant SARS-CoV-2 HLA class I and HLA-DR peptides as potential T cell epitopes in COVID-19 convalescent and unexposed individuals. SARS-CoV-2-specific peptides enabled detection of post-infectious T cell immunity, even in seronegative convalescent individuals. Cross-reactive SARS-CoV-2 peptides revealed pre-existing T cell responses in

Protection against lethal Neospora caninum infection in mice induced by heterologous vaccination with a mic1 mic3 knockout Toxoplasma gondii strain.

Neospora caninum and Toxoplasma gondii are closely related, obligate intracellular parasites infecting a wide range of vertebrate hosts and causing abortion and neonatal morbidity and mortality. Several lines of evidence suggest that cross immunity between these two pathogens could be exploited in the design of strategies for heterologous vaccination. We assessed the ability of an attenuated strain of T. gondii ("mic1-3KO strain") conferring strong protection against chronic and congenital toxoplasmosis to protect mice against lethal N. caninum infection. Mice immunized with mic1-3KO tachyzoit

In-life Chemical Inactivation of Plasmodium Parasites by Cabamiquine Induces Long Lasting Immune Protection.

BACKGROUND: Malaria remains a major global health challenge, and current vaccine strategies provide limited protection. Cabamiquine (CBQ), a Plasmodium translation elongation factor 2 inhibitor, arrests parasite growth during both the liver and blood stages of infection and may promote protective immune responses during controlled parasite infection. METHODS: We evaluated whether CBQ-based infection-and-treatment strategies could induce protective immunity in mouse models of liver- and blood-stage Plasmodium infection. Mice received a single oral dose of CBQ during liver-stage infection or CB

Immunization with an autotransporter protein of Orientia tsutsugamushi provides protective immunity against scrub typhus.

BACKGROUND: Scrub typhus is an acute febrile disease caused by Orientia tsutsugamushi infection. Recently, the rapid increase of scrub typhus incidence in several countries within the endemic region has become a serious public health issue. Despite the wide range of preventative approaches that have been attempted in the past 70 years, all have failed to develop an effective prophylactic vaccine. Currently, the selection of the proper antigens is one of the critical barriers to generating cross-protective immunity against antigenically-variable strains of O. tsutsugamushi. The impact of BCG dose and revaccination on trained immunity.

The innate immune system can display heterologous memory-like responses termed trained immunity after stimulation by certain vaccinations or infections. In this randomized, placebo-controlled trial, we investigated the modulation of Bacille Calmette-Guérin (BCG)-induced trained immunity by BCG revaccination or high-dose BCG administration, in comparison to a standard dose. We show that monocytes from all groups of BCG-vaccinated individuals exerted increased TNFα production after ex-vivo stimulation with various unrelated pathogens. Similarly, we observed increased amounts of T-cell-derived IF

BCG Vaccination Protects against Experimental Viral Infection in Humans through the Induction of Cytokines Associated with Trained Immunity.

The tuberculosis vaccine bacillus Calmette-Guérin (BCG) has heterologous beneficial effects against non-related infections. The basis of these effects has been poorly explored in humans. In a randomized placebo-controlled human challenge study, we found that BCG vaccination induced genome-wide epigenetic reprograming of monocytes and protected against experimental infection with an attenuated yellow fever virus vaccine strain. Epigenetic reprogramming was accompanied by functional changes indicative of trained immunity. Reduction of viremia was highly correlated with the upregulation of IL-1β,

T cell competence to heterologous and homologous thyroglobulins during the induction of experimental autoimmune thyroiditis.

The ability of T cells to respond to homologous vs. heterologous thyroglobulins (Tg) has been evaluated using different models for the induction of experimental autoimmune thyroiditis. Substantial levels of T cell activation could be demonstrated to heterologous Tg following immunization with heterologous Tg in complete Freund's adjuvant, whereas only minimal levels of T cell activation to homologous Tg could be obtained following immunization with homologous Tg in complete Freund's adjuvant. Using this immunization protocol, heterologous and homologous Tg induced equivalent levels of serum an

Inactivated Influenza Vaccine That Provides Rapid, Innate-Immune-System-Mediated Protection and Subsequent Long-Term Adaptive Immunity.

UNLABELLED: The continual threat to global health posed by influenza has led to increased efforts to improve the effectiveness of influenza vaccines for use in epidemics and pandemics. We show in this study that formulation of a low dose of inactivated detergent-split influenza vaccine with a Toll-like receptor 2 (TLR2) agonist-based lipopeptide adjuvant (R4Pam2Cys) provides (i) immediate, antigen-independent immunity mediated by the innate immune system and (ii) significant enhancement of antigen-dependent immunity which exhibits an increased breadth of effector function.

Induction of protective immunity to Haemophilus ducreyi in the temperature-dependent rabbit model of experimental chancroid.

The temperature-dependent rabbit model for chancroid, a sexually transmitted disease caused by the fastidious Gram-negative bacterium Haemophilus ducreyi, was used to investigate the abilities of previous infection and immunization with an acellular preparation of H. ducreyi to induce protective immunity. In the first set of experiments, animals were infected intradermally with either the 35000 or Cha-1 strains of H. ducreyi and then rechallenged 30 days later with both the homologous and heterologous strains. In animals infected with the 35000 strain, statistically significant protective immu

Non-specific effects of vaccines: Current evidence and potential implications.

Besides protection against specific microorganisms, vaccines can induce heterologous or non-specific effects (NSE). Epidemiological data suggest that vaccination with live-attenuated vaccines such as Bacillus Calmette-Guérin (BCG), measles vaccine, and oral polio vaccine results in increased overall childhood survival, and several of these observations have been confirmed in randomized trials. Immunological mechanisms mediating NSE include heterologous lymphocyte effects and induction of innate immune memory (trained immunity). Trained immunity induces long-term functional upregulation of inna

Vaccination against bovine babesiosis.

Bovine babesiosis is an important disease caused by Babesia bovis, B. bigemina, and B. divergens. Solid immunity develops after infection and this feature has been exploited with the use of live attenuated organisms as immunogens. Attributes of live vaccines include a durable immunity to heterologous challenge after one vaccination. To overcome disadvantages relating to poor quality control (risk of contamination and adverse reactions), production procedures have been modified to meet the requirements of codes of good manufacturing practice. This includes development of methods to allow produc

Development of clinical immunity to Plasmodium vivax following repeat controlled human malaria infection.

Clinical immunity to malaria can lead to asymptomatic infection, but the underlying mechanisms remain unclear. To examine the development of clinical immunity, we conducted a multi-cohort, repeat controlled human malaria infection (CHMI) study with Plasmodium vivax, and a heterologous rechallenge with P. falciparum (ClinicalTrials.gov NCT03797989). Malaria-naïve adults underwent CHMI up to three times, by administration of red blood cells infected with P. vivax PvW1 clone or P. falciparum 3D7 clone. Nineteen participants underwent primary CHMI with P. vivax, 12 returned for secondary homologou

Impact of prior flavivirus vaccination on immunogenicity and efficacy of an inactivated Zika vaccine in Indian rhesus macaques.

Flaviviruses are antigenically related. We evaluated the immunogenicity and efficacy of Takeda's purified inactivated Zika vaccine (PIZV) candidate in macaques previously vaccinated with several commercially available heterologous flavivirus vaccines. Heterologous flavivirus vaccination did not elicit Zika virus (ZIKV) neutralizing antibodies and did not impact neutralizing antibody titers after one dose of PIZV. After a second PIZV dose previous vaccination with flavivirus vaccines had variable impact on ZIKV neutralizing antibody titers. However, all macaques were protected against viremia a

Pre-existing YFV-17D immunity mediates T cell cross-protection against dengue virus serotype 2 infection in mice.

Widespread yellow fever virus (YFV) immunity in Sub-Saharan Africa may mitigate orthoflavivirus outbreaks. Here, we investigate whether pre-existing YFV-17D immunity confers cross-protection against dengue virus serotype 2 (DENV-2) infection in a murine model. IFNAR1-/- mice immunized with YFV-17D exhibited significantly reduced DENV-2 viremia, weight loss, and disease severity, with improved survival compared to naïve controls. Mechanistic studies revealed that cross-protection was mediated by heterologous T cell responses rather than cross-binding antibodies. Depletion of T cells in YFV-17D-

Trained immunity as a molecular mechanism for BCG immunotherapy in bladder cancer.

Intravesical BCG instillation is the gold-standard adjuvant immunotherapy for patients with high-risk non-muscle-invasive bladder cancer. However, the precise mechanism of action by which BCG asserts its beneficial effects is still unclear. BCG has been shown to induce a non-specific enhancement of the biological function in cells of the innate immune system, creating a de facto heterologous immunological memory that has been termed trained immunity. Trained immunity or innate immune memory enables innate immune cells to mount a more robust response to secondary non-related stimuli after being

Experimentally induced infectious bovine keratoconjunctivitis: resistance of vaccinated cattle to homologous and heterologous strains of Moraxella bovis.

In studies to determine whether vaccination with one strain of Moraxella bovis would protect against challenge with virulent homologous or heterologous strains, calves were intramuscularly inoculated 3 times with formalin-killed M bovis, with 14 days between inoculations. Fourteen days after the 3rd vaccinal dose was given, all calves were exposed to homologous or heterologous virulent cultures of M bovis. The results indicated that vaccination with one strain of M bovis may induce protective immunity against homologous and heterologous challenge exposure; however, because vaccinated cattle re

Primary ChAdOx1 vaccination does not reactivate pre-existing, cross-reactive immunity.

Currently available COVID-19 vaccines include inactivated virus, live attenuated virus, mRNA-based, viral vectored and adjuvanted protein-subunit-based vaccines. All of them contain the spike glycoprotein as the main immunogen and result in reduced disease severity upon SARS-CoV-2 infection. While we and others have shown that mRNA-based vaccination reactivates pre-existing, cross-reactive immunity, the effect of vector vaccines in this regard is unknown. Here, we studied cellular and humoral responses in heterologous adenovirus-vector-based ChAdOx1 nCOV-19 (AZ; Vaxzeria, AstraZeneca) and mRNA

Cross-protection between distantly related spotted fever group rickettsiae.

Two excellent C3H/HeN mouse models of spotted fever rickettsioses caused by the distantly related organisms, Rickettsia conorii and Rickettsia australis, were utilized to evaluate the possibility of the stimulation of broad cross-protective immunity. Sublethal infection stimulated complete immunity, that is absence of disease, after challenge with a dose of the heterologous Rickettsia that uniformly killed naïve mice. In contrast, heterologous immune sera did not protect mice against a lethal dose (two LD50) of rickettsiae in the mouse toxicity neutralization assay, the standard method for eva

New Insights Into an Old Vaccine: Potency and Breadth of Neutralizing Antibodies Elicited by Yellow Fever Vaccine 17D Are Boosted by Heterologous Orthoflavivirus Infection.

BACKGROUND: Yellow fever vaccine 17D has controlled yellow fever for almost 90 years and is considered highly successful. However, with recent outbreaks in South America and Africa, yellow fever continues to represent a significant threat to public and global health. Despite the long-standing success, few studies have characterized the capacity of 17D-immune sera to neutralize wild type (WT) yellow fever viruses (YFVs). METHODS: Using 17D and 13 WT YFVs, we conducted focus neutralization tests against sera from a unique cohort of 36 nonendemic vaccinees with diverse Orthoflavivirus histories.
