Novel therapeutic agents for relapsed classical Hodgkin lymphoma.

Hodgkin Lymphoma (HL) is a B-cell lymphoproliferative disorder with an excellent prognosis for the majority of patients who are treated with multi-agent chemotherapy, with or without radiation. A subset of patient, however, does not respond to therapy or relapses after accomplishing an initial response. Their outcome is poor and new treatment strategies are urgently needed for this patient population. Recent advances in the understanding of the tumour biology of HL and its microenvironment have ushered a new era of targeted- and immuno-therapies with remarkable activity. Most notably, the anti

Use of pyrrolobenzodiazepines and related covalent-binding DNA-interactive molecules as ADC payloads: Is mechanism related to systemic toxicity?

Antibody-drug conjugates (ADCs) consist of monoclonal antibodies (mAbs) or antibody fragments conjugated to biologically active molecules (usually highly cytotoxic small molecules) through chemical linkers. Although no ADCs containing covalent-binding DNA-interactive payloads have yet been approved (although two containing the DNA-cleaving payload calicheamicin have), of those in clinical trials systemic toxicities are beginning to emerge. This article discusses the observed toxicities in relation to the structures and mechanisms of action of payload type.

CTLA4Ig and the therapeutic potential of T cell co-stimulation blockade.

It is now generally accepted that CD4 T cells are critical players in the initiation of adaptive immune responses by contributing to the terminal differentiation of effector B cells or CD8+ T cells. It is therefore not surprising that CD4+ T cell activation is tightly controlled through the concerted action of a large number of molecular interactions. Activation requires not only the recognition of the appropriate antigen within a MHC molecule by the T cell receptor TCR but also the delivery of co-stimulatory signals by the antigen presenting cell APC . As a consequence therapeutic modulation

Facile Gram-Scale Solution Phase Synthesis of a Universal Cathepsin B-Cleavable Antibody-Drug Conjugate Linker.

Antibody drug conjugates (ADCs) are a key therapy in the cancer treatment arsenal. Critical to the success of the ADC is the release of the drug cargo at the desired site, and thus, enzymatically or chemically responsive linkers, connecting the drug to the antibody, play a central role. Linkers containing the peptide sequence Glu-Val-Cit are particularly important as cleavage of this peptide occurs by both murine and human cathepsin B enzymes. Additional linker design considerations include components for free drug release via the self-immolative moiety para-aminobenzyl alcohol (PABOH), copper

Antibody-drug conjugates in gastric cancer: clinical advances and resistance mechanisms.

Gastric cancer remains a major clinical challenge owing to its insidious onset and marked heterogeneity, which contribute to poor prognosis and limit the effectiveness of chemotherapy‑based systemic therapy. Antibody‑drug conjugates (ADCs) deliver potent cytotoxins to tumor cells with antigen specificity and have emerged as a therapeutic class that can enhance efficacy while reducing off‑target toxicity. Recent trials of ADCs targeting HER2, CLDN18.2, TROP2, CEACAM5/6, and HER3 demonstrate substantial therapeutic activity. Notably, the HER2‑directed agents trastuzumab deruxtecan and disitamab

Immune checkpoint targeted drug conjugate therapies: Bridging preclinical and clinical innovation for solid tumor therapy.

Solid tumors remain a significant therapeutic challenge due to their complex biology, heterogeneous tumor microenvironment (TME), and frequent development of resistance to standard treatments. Although immune checkpoint inhibitors (ICIs) have redefined tumor therapy by restoring antitumor immunity, their clinical benefit in Solid tumors is often constrained by low response rates (LRR), immune-related toxicities, and adaptive resistance. Immune checkpoint targeting drug conjugates (IDCs) have recently emerged as an innovative approach that combines checkpoint blockade with the targeted delivery

Process Development of Site-Specific Antibody-Drug Conjugates.

Antibody-drug conjugates (ADCs) combine the targeting specificity of monoclonal antibodies with cytotoxic small-molecule payloads, enabling selective delivery of highly potent therapeutics. As the field has advanced, conjugation strategy has become a primary driver of product quality, manufacturability, and development risk. Site-specific approaches include engineered cysteine residues, non-canonical amino acid (ncAA) incorporation, and enzyme-mediated conjugation. These platforms enable defined drug-to-antibody ratios (DAR) and improved product homogeneity, shifting control to molecular desig

Circulating tumour cells for guiding antibody‒drug conjugate therapy: Role of artificial intelligence.

Antibody‒drug conjugates (ADCs) represent a major advance in precision oncology, yet their effectiveness depends critically on adequate target antigen expression. Tumour heterogeneity and dynamic antigen expression during treatment pose major challenges for conventional tissue biopsies used in patient selection. Liquid biopsy components such as circulating tumour cells (CTCs) enable real-time, minimally invasive monitoring of tumour burden, molecular characteristics and target antigen status. This review evaluates the potential of CTC analysis to overcome the key challenges in ADC therapy, inc

[Corneal events with mirvetuximab soravtansine : A review of ocular surface events associated with the tubulin-acting antibody-drug conjugate, mirvetuximab soravtansine].

Antibody-drug conjugates (ADC) are a class of targeted anticancer therapy that consist of a monoclonal antibody (linker) and a cytotoxic substance. Ocular adverse events (AEs) are common among ADCs with tubulin-targeted active agents, such as belantamab mafodotin, tisotumab vedotin and mirvetuximab soravtansine (MIRV). The substance MIRV targets folate receptor alpha (FRalpha) and has a tubulin-acting agent (the maytansinoid DM4). The use of MIRV received full approval in the USA (March 2024) for FRalpha-positive platinum-resistant ovarian cancer (PROC) and is the first novel agent to demonstr

Unmet Potential of Antibody-Drug Conjugates: An Evaluation of the Past, Present, and Future of Antibody-Drug Conjugate Development in Advanced Urothelial Carcinoma.

Antibody-drug conjugates (ADCs) represent an emerging class of therapeutics across solid tumor oncology and are already positioned as a cornerstone therapy for patients with urothelial carcinoma (UC). In recent years, there has been a paradigm shift in the therapeutic landscape as frontline treatment of UC formerly relied on the use of platinum-based agents and has now evolved to include combination strategies with ADCs. These dramatic changes are due in part to our improved understanding of the molecular features of bladder tumors and the identification of tumor-associated antigens specific t

Antibody-Drug Conjugates (ADCs): current and future biopharmaceuticals.

Antibody-drug conjugates (ADCs) represent a novel class of biopharmaceuticals comprising monoclonal antibodies covalently conjugated to cytotoxic agents via engineered chemical linkers. This combination enables targeted delivery of cytotoxic agents to tumor site through recognizing target antigens by antibody while minimizing off-target effects on healthy tissues. Clinically, ADCs overcome the limitations of traditional chemotherapy, which lacks target specificity, and enhance the therapeutic efficacy of monoclonal antibodies, providing higher efficacy and fewer toxicity anti-tumor biopharmace

Antibody-Drug Conjugates to Treat Bacterial Biofilms via Targeting and Extracellular Drug Release.

The treatment of implant-associated bacterial infections and biofilms is an urgent medical need and a grand challenge because biofilms protect bacteria from the immune system and harbor antibiotic-tolerant persister cells. This need is addressed herein through an engineering of antibody-drug conjugates (ADCs) that contain an anti-neoplastic drug mitomycin C, which is also a potent antimicrobial against biofilms. The ADCs designed herein release the conjugated drug without cell entry, via a novel mechanism of drug release which likely involves an interaction of ADC with the thiols on the bacter

Comparison of methods generating antibody-epitope conjugates for targeting cancer with virus-specific T cells.

Therapeutic antibody-epitope conjugates (AECs) are promising new modalities to deliver immunogenic epitopes and redirect virus-specific T-cell activity to cancer cells. Nevertheless, many aspects of these antibody conjugates require optimization to increase their efficacy. Here we evaluated different strategies to conjugate an EBV epitope (YVL/A2) preceded by a protease cleavage site to the antibodies cetuximab and trastuzumab. Three approaches were taken: chemical conjugation (i.e. a thiol-maleimide reaction) to reduced cysteine side chains, heavy chain C-terminal enzymatic conjugation using
