Altered brain serotonin 5-HT1A receptor expression and function in juvenile Fmr1 knockout mice.

There are no approved pharmacotherapies for fragile X syndrome (FXS), a rare neurodevelopmental disorder caused by a mutation in the FMR1 promoter region that leads to various symptoms, including intellectual disability and auditory hypersensitivity. The gene that encodes inhibitory serotonin 1A receptors (5-HT1ARs) is differentially expressed in embryonic brain tissue from individuals with FXS, and 5-HT1ARs are highly expressed in neural systems that are disordered in FXS, providing a rationale to focus on 5-HT1ARs as targets to treat symptoms of FXS. We examined agonist-labeled 5-HT1AR densi

In utero exposure to valproic acid disrupts ascending projections to the central nucleus of the inferior colliculus from the auditory brainstem.

Prenatal exposure to the antiepileptic valproic acid (VPA) is associated with an increased risk of autism spectrum disorder (ASD) in humans. Accordingly, in utero exposure to VPA is a validated and biologically relevant animal model of ASD. The majority of individuals with ASD exhibit some degree of auditory dysfunction, ranging from deafness to hypersensitivity. Animals exposed to VPA in utero have abnormal tonotopic maps and responses in the cerebral cortex and hyperactivation, hypoplasia, abnormal neuronal morphology and reduced calcium binding protein expression throughout the auditory bra

Functional magnetic resonance imaging confirms forward suppression for rapidly alternating sounds in human auditory cortex but not in the inferior colliculus.

Forward suppression at the level of the auditory cortex has been suggested to subserve auditory stream segregation. Recent results in non-streaming stimulation contexts have indicated that forward suppression can also be observed in the inferior colliculus; whether this holds for streaming-related contexts remains unclear. Here, we used cardiac-gated fMRI to examine forward suppression in the inferior colliculus (and the rest of the human auditory pathway) in response to canonical streaming stimuli (rapid tone sequences comprised of either one repetitive tone or two alternating tones). The fir

Age-related changes in the expression of NMDA, serotonin, and GAD in the central auditory system of the rat.

CONCLUSIONS: We suggest that age-induced changes of serotonin, N-methyl-d-aspartate receptor (NMDAR), and glutamate decarboxylase (GAD) expression in rats are dependent on the specific location in the central auditory system (CAS). OBJECTIVES: Despite the importance of understanding changes in neurotransmitters during presbycusis, only a few studies have assessed age-associated changes in neurotransmitter at each level of the CAS. We therefore evaluated effects of aging on neurotransmission in the CAS of rats. METHODS: The concentrations of serotonin, NMDAR, and GAD were assayed immunohistoc

Responses of inferior colliculus neurons to sounds presented at different rates in anesthetized albino mouse.

We recorded extracellular activity from 402 single units located in the inferior colliculus (IC) of barbiturate-anesthetized albino mice. The stimuli were pure tones at characteristic frequency (CF) with durations of 10, 40 and 100 ms and intensities ranged from 5 to 25 dB above unit's minimum threshold (MT). The tones were presented with different repetition rates (RRs) ranging from 0.2 to 20.0 Hz. At low intensities (5 dB above MT, determined at RR of 0.5 Hz) the great majority of units exhibited a strong decline of their responses when the stimulus RR was increased. About one-half of the un

General anesthesia changes gap-evoked auditory responses in guinea pigs.

CONCLUSION: General anesthesia induced by sodium pentobarbital reduces temporal resolution as represented by an increase in the threshold of gap-evoked auditory responses in guinea pigs. OBJECTIVES: To explore the potential impact of general anesthesia by sodium pentobarbital (PB), a common anesthetic used in animal research, on gap-evoked responses. MATERIALS AND METHODS: The evoked potentials in response to gaps formed by bursts of broadband noise were recorded from electrodes implanted in the inferior colliculus (IC) and the auditory cortex (AC) of guinea pigs. The gap responses were comp

Inputs from the cochlea and the inferior colliculus converge on olivocochlear neurones.

Medial olivocochlear (MOC) neurones, located in the superior olivary complex, can suppress cochlear gain by their action on the cochlear outer hair cells. Inputs from the contralateral cochlea and the inferior colliculus (IC) have been separately shown to increase activity of MOC neurones. In this study we have investigated in guinea-pigs under barbiturate anaesthesia the interactions between these two inputs by combining electrical stimulation of the IC with acoustic stimulation of the contralateral cochlea. Electrical stimulation of the IC resulted in a significant suppression of the amplitu

A novel approach to investigate subcortical and cortical sensitivity to temporal structure simultaneously.

Hearing loss is associated with changes at the peripheral, subcortical, and cortical auditory stages. Research often focuses on these stages … well as adaptation and neural recovery time in subcortical structures (probably cochlear nuclei and inferior colliculi). Subcortical adaptation to inter-click intervals was reflected in longer latencies. Cortical responses to the 3.5 Hz modulation included phase-locking, probably originating from auditory cortex, and sustained activity likely originating from higher …

Controlled scanpath variation alters fusiform face activation.

We investigated the influence of experimentally guided saccades and fixations on fMRI activation in brain regions specialized for face and object processing. Subjects viewed a static image of a face while a small fixation cross made a discrete jump within the image every 500 ms. Subjects were required to make a saccade and fixate the cross at its new location. Each run consisted of alternating blocks in which the subject was guided to make a series of saccades and fixations that constituted either a Typical or an Atypical face scanpath. Typical scanpaths were defined as a scanpath in which the

Divergence of dim-light vision among bats (order: Chiroptera) as estimated by molecular and electrophysiological methods.

Dim-light vision is present in all bats, but is divergent among species. Old-World fruit bats (Pteropodidae) have fully developed eyes; the eyes of insectivorous bats are generally degraded, and these bats rely on well-developed echolocation. An exception is the Emballonuridae, which are capable of laryngeal echolocation but prefer to use vision for navigation and have normal eyes. In this study, integrated methods, comprising manganese-enhanced magnetic resonance imaging (MEMRI), f-VEP and RNA-seq, were utilized to verify the divergence. The results of MEMRI showed that Pteropodidae bats have

General anesthetic induced differential changes in latency of auditory evoked potential in the central nucleus of inferior colliculus of mouse.

Confirming the effect of general anesthetic on brainstem auditory evoked potential (BAEP) is important to interpret BAEP data, elucidate the neuroanatomical sites of action of general anesthetic and monitor the effect of general anesthetic. However, the effect of general anesthetic on BAEP is not thoroughly understood, which may be due to unreasonable acoustic stimulation scheme. This study aimed to redesign acoustic stimulation scheme and attempted to test our hypothesis that general anesthetic induces differential changes in BAEP latency in mouse. Auditory evoked potential in the central nuc

Neural coding of the sound envelope is changed in the inferior colliculus immediately following acoustic trauma.

Sensorineural hearing loss is often accompanied by difficulties with understanding speech in fluctuating backgrounds, suggesting that neural coding of complex sound features, such as the sound envelope, is impaired. Here, we studied how temporal and rate coding of the envelope is affected in the inferior colliculus immediately after acoustic trauma. Neural activity in response to amplitude-modulated noise was recorded from the inferior colliculus of the guinea pig, before and immediately after a 1-hr 11-kHz acoustic trauma. Units with a characteristic frequency (CF) below the trauma frequency

Testing the Central Gain Model: Loudness Growth Correlates with Central Auditory Gain Enhancement in a Rodent Model of Hyperacusis.

The central gain model of hyperacusis proposes that loss of auditory input can result in maladaptive neuronal gain increases in the central auditory system, leading to the over-amplification of sound-evoked activity and excessive loudness perception. Despite the attractiveness of this model, and supporting evidence for it, a critical test of the central gain theory requires that changes in sound-evoked activity be explicitly linked to perceptual alterations of loudness. Here we combined an operant conditioning task that uses a subject's reaction time to auditory stimuli to produce reliable mea

Tinnitus: Maladaptive auditory-somatosensory plasticity.

Tinnitus, the phantom perception of sound, is physiologically characterized by an increase in spontaneous neural activity in the central auditory system. However, as tinnitus is often associated with hearing impairment, it is unclear how a decrease of afferent drive can result in central hyperactivity. In this review, we first assess methods for tinnitus induction and objective measures of the tinnitus percept in animal models. From animal studies, we discuss evidence that tinnitus originates in the cochlear nucleus (CN), and hypothesize mechanisms whereby hyperactivity may develop in the CN a

Neuronal activity evoked in the inferior colliculus of the cat by surface macroelectrodes and penetrating microelectrodes implanted in the cochlear nucleus.

Persons lacking functional auditory nerves cannot benefit from cochlear implants, but an auditory brainstem implant (ABI) utilizing stimulating electrodes adjacent to or on their cochlear nucleus (CN) can restore some hearing. We are investigating the feasibility of supplementing these surface electrodes with penetrating microstimulating electrodes within the ventral CN (VCN), and how the two types of electrodes can be used synergistically. Multiunit neuronal responses evoked by VCN electrical stimulation with surface electrodes and microelectrodes were recorded in the inferior colliculus (ICC

Preservation of synapses on principal cells of the central nucleus of the inferior colliculus with aging in the CBA mouse.

The light and electron microscopic features of principal neurons of the central nucleus of the inferior colliculus were quantitated in the CBA mouse. Three age groups of mice were examined, including young (3 months), middle-aged (8 months) and old (25 months). No changes were noted in the size of the principal neurons over the age range examined. At the ultrastructural level, synapses on the somata of the principal neurons showed no change in the number or type of synapses, the length of synaptic apposition nor the size of synaptic terminal area. These results are in contrast with the moderat

Age-related changes in GABA(A) receptor subunit composition and function in rat auditory system.

A decline in the ability to discriminate speech from noise due to age-related hearing loss (presbycusis) may reflect impaired auditory information processing within the central nervous system. Presbycusis may result, in part, from functional loss of the inhibitory neurotransmitter GABA. The present study assessed age-related changes of the GABA(A) receptor in the inferior colliculus of young-adult, middle-aged, and aged rats related to: (i) receptor subunit composition and (ii) receptor function. Western blotting was used to measure protein levels of selected GABA(A) receptor subunits in prepa

Effects on noise exposure on the auditory functions.

Noise induced hearing loss is accompanied by a wide variety of hearing deficits, including threshold and suprathreshold deficits, which often prevent effective rehabilitation even when hearing aids are used. A study of the neural mechanisms responsible for deficits associated with sensorineural loss is based on neurophysiological and anatomical research. In this paper, we intend to review the basic physiological characteristics of the peripheral auditory system.

Comparison of the auditory sensitivity of neurons in the cochlear nucleus and inferior colliculus of young and aging C57BL/6J and CBA/J mice.

Thresholds of neurons to sounds were compared as a function of central auditory structure [ventral cochlear nucleus (VCN), dorsal cochlear nucleus (DCN), and inferior colliculus (IC)] in young and middle-aged C57BL/6J mice (multiple- and single-unit recordings) and in young and old CBA/J mice (single-unit recordings). Middle-aged C57 mice show progressive loss of sensitivity to high frequencies and noise due to cochlear pathology; CBA mice show little loss of sensitivity through most of their lifespan. Multiple-unit threshold curves (MTCs) for tones indicated that neurons in the C57 VCN suffer

[Anatomy of the central auditory pathway. Demonstration with horseradish peroxidase in the guinea pig].

The neurones of the afferent and efferent auditory pathways were labelled by axonally transported horseradish peroxidase. The first neurone of the ascending system is the acoustic nerve with the spiral ganglion. These fibres terminate mainly in the ventral cochlear nucleus (VCN). The neurones of the latter chiefly run to the contralateral superior olivary complex (SOC), whereas the neurones of the dorsal cochlear nucleus (DCN) terminate mainly in the central nucleus of the contralateral inferior colliculus (IC). The nerve cells of the superior olivary complex and nuclei of the lateral lemniscu
