Effectiveness of topical steroids in the control of radiation dermatitis: a randomised trial using 1% hydrocortisone cream and 0.05% clobetasone butyrate (Eumovate).

Radiation dermatitis often presents as a problem for patients and radiotherapists during treatment. Topical corticosteroids have been shown to have an anti-inflammatory effect in the treatment of many skin diseases and are commonly prescribed during a course of radiation treatment. A comparison of two different steroid creams, 1% hydrocortisone cream and 0.05% clobetasone butyrate (Eumovate), in a double blind trial was carried out in 54 patients undergoing radiation therapy for breast cancer. 'The cream' was administered when patients reached a given dose of 2000 rad (or earlier if required)

[Aspirin against cancer].

PURPOSE: That aspirin as an anti-aggregative and anti-inflammatory compound might prevent tumor spread is an old concept that is still not of clinical relevance. To date, aspirin has been shown in several epidemiologic studies to be linked with a reduction of colorectal cancer incidence, as well as the incidence of lung and breast cancer. In this issue, we have summarized the mechanisms that support this hypothesis, and we have analysed the main clinical studies. RESULTS: Only case-control studies and most of the prospective cohort studies showed a reduction of colorectal cancer incidence in

Does the metastatic lymph node ratio influence the disease-free survival of patients with breast cancer: single-center experiences.

BACKGROUND: Axillary lymph nodes (ALNs) are the most important prognostic factor for survival in breast cancer. Pathological evaluation can affect the number of involved lymph nodes. In the current study, we evaluated whether the metastatic lymph node ratio (n ratio) is important in predicting disease-free survival (DFS) for breast cancer patients. MATERIAL AND METHODS: From 802 breast cancer cases, 427 patients with ALN metastasis were analyzed retrospectively. The n ratio was categorized as n ratio 1 (1-10%), n ratio 2 (10.01-50%) and n ratio 3 (>50%). DFS was established according to the K

Oral contraceptives: current status.

During the past four decades, oral contraceptives have remained a safe and effective method of birth control. Reductions in the estrogen and progestin dosages have significantly decreased the incidence of cardiovascular complications. The association between oral contraceptives and breast cancer appears to be primarily because of detection bias or possibly a promotional effect. Despite the changes in formulation, the problems related to side effects have not been totally solved. Because compliance and successful use is strongly affected by side effects, improvement in this area is probably the

Detection and treatment of akathisia in advanced cancer patients during adjuvant analgesic therapy with tricyclic antidepressants: case reports and review of the literature.

OBJECTIVE: There is substantial evidence that tricyclic antidepressants are effective in the management of chronic pain, including cancer pain. In oncological settings, these agents are used as adjuvant analgesic drugs. However, cases of akathisia due to tricyclic antidepressants used as adjuvant analgesic therapy have not previously been reported. CASE REPORTS: Two cancer patients experiencing chronic pain who were refractory to nonsteroidal anti-inflammatory drugs and opioids were prescribed amoxapine as an adjuvant analgesic therapy for neuropathic pain. These patients developed inner rest

Aspirin use and risk of breast cancer in African American women.

BACKGROUND: Use of aspirin and other non-steroidal anti-inflammatory drugs (NSAIDs) has been hypothesized to be associated with reduced risk of breast cancer; however, results of epidemiological studies have been mixed. Few studies have investigated these associations among African American women. METHODS: To assess the relation of aspirin use to risk of breast cancer in African American women, we conducted a prospective analysis within the Black Women's Health Study, an ongoing nationwide cohort study of 59,000 African American women. On baseline and follow-up questionnaires, women reported

Prognostic impact of genetic variants of CYP19A1 and UGT2B17 in a randomized trial for endocrine-responsive postmenopausal breast cancer.

Polymorphisms of genes involved in estrogen synthesis have been linked to breast cancer risk, prognosis, and treatment response. We investigated the prognostic impact of a deletion spanning the entire UGT2B17 gene (UGT2B17*2) and genetic variants of the aromatase CYP19A1 and estrogen receptor α (ESR1) in 125 postmenopausal women with ER-positive breast cancer enrolled in a randomized pre-surgical trial. The UGT2B17*2 was estimated by copy number variation assays and the CYP19A1 rs10046/rs4646 and ESR1 rs2077647/rs2234693/rs9340799 by TaqMan allelic discrimination assays. Serum exemestane/17-hy

Use of nonsteroidal anti-inflammatory drugs and reduced breast cancer risk among overweight women.

Chronic inflammation is associated with increased risk of multiple cancers, including breast cancer. Adipose tissues produce proinflammatory cytokines, and obesity is a risk factor for postmenopausal breast cancer. We evaluated the association of regular use of nonsteroidal anti-inflammatory drugs (NSAIDs) with breast cancer risk, overall and by body mass index (BMI) and tumor subtypes defined by estrogen receptor, progesterone receptor, and human epidermal growth factor receptor 2 status. We conducted a population-based, case-control study involving 5,078 women aged 25-75 years who were recru

Hiding in plain view: the potential for commonly used drugs to reduce breast cancer mortality.

Many medications have been developed for one purpose but then are found to have other clinical activities. There is tremendous interest in whether non-cancer medications may potentially have effects on breast cancer survival. In this review article, we have presented and evaluated the evidence for several commonly used over-the-counter and prescription medications - including aspirin (and other non-steroidal anti-inflammatory drugs), beta-blockers, angiotensin-converting enzyme inhibitors, statins, digoxin, and metformin - that have been evaluated among breast cancer survivors in prospective s

Vesiculo-bullous dermatomyositis: report of three cases.

Dermatomyositis (DM) presents with various types of skin lesions, but vesicle formation is rare. We herein present three female patients with DM who formed vesicles on the trunk and extremities. In all cases the bullous lesions appeared in parallel with increased disease activity of DM. In one case, only one large bulla within an erythematous background was found on the abdomen; the other two cases presented with a number of vesicles. Histological examination revealed marked subepidermal edema with mucin deposition in the upper dermis in all cases. Breast carcinoma and malignant lymphoma were

Association of frequency and duration of aspirin use and hormone receptor status with breast cancer risk.

CONTEXT: Use of aspirin and other nonsteroidal anti-inflammatory drugs (NSAIDs) has been associated with a decrease in the risk of several cancers, including breast cancer. NSAIDs inhibit cyclooxygenase activity and thereby reduce prostaglandin synthesis; prostaglandins stimulate aromatase gene expression and thereby stimulate estrogen biosynthesis. Given the importance of estrogen in the pathogenesis of breast cancer, the ability of aspirin and other NSAIDs to protect against breast cancer could vary according to hormone receptor status. OBJECTIVES: To determine the association between the f

Quantitative MALDI imaging of aspirin metabolites in mouse models of triple-negative breast cancer.

Rationale: The non-steroidal anti-inflammatory drug aspirin is currently being developed as activatable contrast agent for chemical exchange saturation transfer (CEST) magnetic resonance imaging (MRI), for detection of its CEST MRI active metabolite salicylic acid (SA). This study refines and develops quantitative matrix-assisted laser desorption/ionization (QMALDI) imaging to investigate the distribution of aspirin metabolites including SA in triple-negative breast cancer (TNBC) models in mice. Method: In this study, we established QMALDI imaging with norharmane (nH) matrix and assisted by th

Integrative analysis of bulk and single-cell transcriptomics identifies factors related to immunosuppressive microenvironment to predict unfavorable prognosis in inflammatory breast cancer.

INTRODUCTION: Inflammatory breast cancer (IBC) is a rare invasive tumor and characterized by the formation of tumor emboli within dermal lymphatic vessels. The tumor microenvironment (TME) is a key factor in IBC aggressiveness, but its heterogeneity and intercellular role remain incompletely understood. METHODS: Weighted gene coexpression network analysis (WGCNA) was performed to identify immune infiltration-associated genes. We also depicted cellular communication networks and specific ligand-receptor signaling pathways using the single-cell transcriptomics analysis of IBC and non-IBC sample

Design and synthesis of stilbene analogs based on resveratrol as NF-κB inhibitors for the treatment of breast cancer.

NF-κB is a critical signaling molecule connecting inflammation and tumors, involved in numerous cellular processes, including inflammation, cell transformation, tumor cell survival, proliferation, invasion, angiogenesis, and metastasis by regulating immune, growth, and inflammatory gene expression. Inhibition of the NF-κB signaling pathway in tumor cells can effectively reduce inflammation levels, potentially providing antitumor benefits. Resveratrol, a natural polyphenolic compound known for its anti-inflammatory properties, has been shown both anti-inflammatory and anticancer effects in brea

The role of body mass index at diagnosis in patients with inflammatory breast cancer.

INTRODUCTION: Inflammatory Breast Cancer (IBC) is an aggressive presentation of BC present in 1-5 % of all patients with BC. While obesity has been consistently associated with worse prognosis in patients with BC, it is understudied in patients with IBC. PATIENTS AND METHODS: We retrospectively evaluated the association of body mass index (BMI) at diagnosis with clinicopathological characteristics, pathological complete response (pCR) to chemotherapy and survival in a multicentric cohort of patients with IBC treated with pre-operative chemotherapy. RESULTS: Of the 542 patients, 6 were underw

Lifetime use of nonsteroidal anti-inflammatory drugs and breast cancer risk: results from a prospective study of women with a sister with breast cancer.

BACKGROUND: Nonsteroidal anti-inflammatory drugs (NSAIDs) have been shown to inhibit several pathways in experimental models of breast carcinogenesis, but epidemiological evidence remains insufficient to support their use for breast cancer prevention. We examined the association between use of NSAIDs and breast cancer risk in a prospective cohort. METHODS: The Sister Study is a prospective cohort study of women who had a sister(s) with breast cancer. As of December 2013, 2118 incident breast cancers were ascertained from 50,884 women enrolled between 2003 and 2009. Lifetime history of NSAID u
