Modes of Action, Resistance and Toxicity of Insecticides Targeting Nicotinic Acetylcholine Receptors.

BACKGROUND: Nicotinic acetylcholine receptors (nAChRs) of insects play a key role in fast excitatory neurotransmission. Several classes of insecticides target insect nAChRs, which are composed of subunit members of a family of multiple subunit encoding genes. Alternative splicing and RNA A-to-I editing can add further to receptor diversity. Native and recombinant receptors have been explored as sites of insecticide action using radioligands, electrophysiology and site-directed mutagenesis. METHODS: We have reviewed the properties of native and recombinant insect nAChRs, the challenges of func

Differential expression of glycosomal and mitochondrial proteins in the two major life-cycle stages of Trypanosoma brucei.

Label-free semi-quantitative differential three-dimensional liquid chromatography coupled to mass spectrometry (3D-LC-MS/MS) was used to compare the glycosomal and mitochondrial proteomes of the bloodstream- and insect-form of Trypanosoma brucei. The abundance of glycosomal marker proteins identified in the two life-cycle stages corresponded well with the relative importance of biochemical pathways present in the glycosomes of the two stages and the peptide spectral count ratios of selected enzymes were in good agreement with published data about their enzymatic specific activities. This appro

Lipophorin: The Lipid Shuttle.

Insects need to transport lipids through the aqueous medium of the hemolymph to the organs in demand, after they are absorbed by the intestine or mobilized from the lipid-producing organs. Lipophorin is a lipoprotein present in insect hemolymph, and is responsible for this function. A single gene encodes an apolipoprotein that is cleaved to generate apolipophorin I and II. These are the essential protein constituents of lipophorin. In some physiological conditions, a third apolipoprotein of different origin may be present. In most insects, lipophorin transports mainly diacylglycerol and hydroc

Intestinal Morphometry, Enzymatic and Microbial Activity in Laying Hens Fed Different Levels of a Hermetia illucens Larvae Meal and Toxic Elements Content of the Insect Meal and Diets.

To evaluate the effects of feeding a Hermetia illucens (HI) larvae meal on the different intestinal traits of hens, and to determine the toxic elements' concentration in the insect meal and diets, 162 hens were randomly allotted to three groups. The control received a corn-soybean meal-based diet (SBM); the HI25 and HI50 groups received two diets in which the 25% and 50% of the dietary protein were replaced by the HI protein, respectively. The duodenal and jejunal villi height and villi/crypt were higher (p < 0.01) in the SBM than in the HI groups. The ileal villi height was higher (p < 0.05)

Intestinal Taste Receptor Expression and Its Implications for Health: An Integrative Analysis in Female Rats after Chronic Insect Supplementation.

Taste receptors are found in the gastrointestinal tract, where they are susceptible to dietary modulation, a key point that is crucial for diet-related responses. Insects are sustainable and good-quality protein sources. This study analyzed the impact of insect consumption on the modulation of taste receptor expression across various segments of the rat intestine under healthy or inflammatory conditions. Female Wistar rats were supplemented with Tenebrio molitor (T) or Alphitobius diaperinus (B), alongside a control group (C), over 21 days under healthy or LPS-induced inflammation. The present

NlCYP417A1 is a critical regulator of molting by governing ecdysone homeostasis in the brown planthopper.

BACKGROUND: The brown planthopper (BPH), Nilaparvata lugens, is a destructive rice pest across Asia. Suppressing insect development is enduring goals in insect pest management, underpinning the action of existing insecticides and guiding the design of new ones. Insect molting, which is tightly regulated by ecdysteroid signaling, is essential for insect growth and development. Cytochrome P450 (CYP) enzymes play critical roles in ecdysteroid biosynthesis, and identifying P450 genes involved in this process may provide new insights into insect developmental regulation and potential targets for pe

Cross-kingdom RNA interference mediated by insect salivary microRNAs may suppress plant immunity.

Communication between insects and plants relies on the exchange of bioactive molecules that traverse the species interface. Although proteinic effectors have been extensively studied, our knowledge of other molecules involved in this process remains limited. In this study, we investigate the role of salivary microRNAs (miRNAs) from the rice planthopper Nilaparvata lugens in suppressing plant immunity. A total of three miRNAs were confirmed to be secreted into host plants during insect feeding. Notably, the sequence-conserved miR-7-5P is specifically expressed in the salivary glands of N. lugen

Development and characterization of Rift Valley fever virus-like particles.

Rift Valley fever (RVF) is an acute, febrile zoonotic disease that is caused by the RVF virus (RVFV) and spread by arthropod vectors. RVF is currently prevalent in Africa and the Arabian Peninsula, and causes substantial economic losses. Furthermore, this disease poses a serious threat to animal and human health in regions worldwide, making it a serious public health concern. However, RVFV vaccines for human use are still unavailable, and hence there is an urgent need for novel efficient vaccines against RVFV. Vaccine preparation techniques have become a crucial factor in developing new vaccin

First In Silico Screening of Insect Molecules for Identification of Novel Anti-Parasitic Compounds.

Schistosomiasis is a neglected tropical disease caused by blood flukes of the genus Schistosoma. In silico screenings of compounds for the identification of novel anti-parasitic drug candidates have received considerable attention in recent years, including the screening of natural compounds. For the first time, we investigated molecules from insects, a rather neglected source in drug discovery, in an in silico screening approach to find novel antischistosomal compounds. Based on the Dictionary of Natural Products (DNP), we created a library of 1327 insect compounds suitable for molecular dock

Purification and cloning of the salivary nitrophorin from the hemipteran Cimex lectularius.

Cimex lectularius and Rhodnius prolixus contain salivary nitric oxide (NO) that may help them to feed on their vertebrate hosts by promoting vasodilation and inhibiting platelet aggregation. Salivary NO is associated with heme proteins (nitrophorins) that store and transport NO from the insect salivary glands to the skin of the host. In this study, the salivary nitrophorin of Cimex lectularius was purified by DEAE chromatography and reverse-phase high-performance liquid chromatography. The purified nitrophorin had a molecular mass of 32.9 kDa. The DEAE-purified hemoprotein was able to bind NO,

Identification and initial characterisation of Trypanosoma Cox17 copper metallochaperone.

Copper plays an essential role in organisms as a catalytic co-factor for key enzymes like cytochrome c oxidase. Copper importation, distribution and secretion is carried out by copper transport and copper-binding proteins known as copper chaperones. Cox17 is a chaperone that conveys copper to Cox11 and Sco1 for metalation of the CuB and CuA of Cox1 and Cox2 respectively in eukaryotes. Cox17 from Trypanosoma brucei brucei and T. congolense were recombinantly expressed and affinity purified as MBP-fusion proteins. Immunization with recombinant surface antigens p26 with Freund's adjuvants against Babesia rodhaini infection.

The surface proteins of Babesia rodhaini have previously been shown to induce a high degree of protective immunity. In the present study, one of those proteins, B. rodhaini antigen p26 was expressed in Escherichia coli and in insect cells infected with a recombinant baculovirus. These proteins were recognized by immune serum from a drug-cured BALB/c mouse. While BALB/c mice immunized with both recombinant antigens and Freund's adjuvants showed 40-100% survival rate against challenge infection with B. rodhaini, saponin failed to induce protection, although significant levels of B. rodhaini-spec

A testis-specific heat shock protein gene plays a critical role in male fertility in the fall armyworm.

Heat shock proteins (Hsps) are critical for stress responses and multiple physiological processes, yet their function in insect reproduction remains underexplored. In this study, we identified 36 Hsp genes in the fall armyworm (Spodoptera frugiperda), of which LOC118269601 (designated as SfHsp68A) exhibited robust testis-specific expression, peaking during pupal testis development. CRISPR/Cas9-mediated knockout of SfHsp68A resulted in recessive male sterility, significantly reduced testis size, sperm counts, and egg hatch rates. Transcriptomic analysis of mutant testes revealed substantial dow

Tying pest insects in knots: the deployment of spider-venom-derived knottins as bioinsecticides.

Spider venoms are complex chemical arsenals that contain a rich variety of insecticidal toxins. However, the major toxin class in many spider venoms is disulfide-rich peptides known as knottins. The knotted three-dimensional fold of these mini-proteins provides them with exceptional chemical and thermal stability as well as resistance to proteases. In contrast with other bioinsecticides, which are often slow-acting, spider knottins are fast-acting neurotoxins. In addition to being potently insecticidal, some knottins have exceptional taxonomic selectivity, being lethal to key agricultural pest

Discovery of a novel alphavirus related to Eilat virus.

Most alphaviruses are transmitted by arthropods and infect vertebrate hosts. An exception is Eilat virus (EILV), the only described alphavirus with a host range restricted to insects. We established a new generic reverse transcription PCR assay for alphaviruses and tested 8860 tropical mosquitoes. We detected a novel alphavirus, tentatively named Taï Forest alphavirus (TALV), in Culex decens mosquitoes collected in Ivory Coast. The full genome was sequenced, and closest similarity was found to EILV. Pairwise amino acid identities to EILV ranged between 67 and 88 % for the corresponding protein

Direct complement restriction of flavivirus infection requires glycan recognition by mannose-binding lectin.

An intact complement system is crucial for limiting West Nile virus (WNV) dissemination. Herein, we define how complement directly restricts flavivirus infection in an antibody-independent fashion. Mannose-binding lectin (MBL) recognized N-linked glycans on the structural proteins of WNV and Dengue virus (DENV), resulting in neutralization through a C3- and C4-dependent mechanism that utilized both the canonical and bypass lectin activation pathways. For WNV, neutralization occurred with virus produced in insect cells, whereas for DENV, neutralization of insect and mammalian cell-derived virus

Are Sulfates of the Hydroxyl Amino Acids Threonine and Serine Truly Extremely Rare or Do They Simply Escape Detection?

Sulfonation is important for bioactivity across species-substrates range from small molecules to proteins, in which almost exclusively tyrosine sulfonation (sTyr) has been investigated. Although sulfoalkyl-hydroxyl amino acid residues in peptides and proteins have rarely been found, we will argue that it is highly likely that sSer and sThr are much more abundant than is currently evident. In the context of the recent detection of sThr-containing adipokinetic hormones in insects, the literature was screened to answer questions about the relative stability (lability) of the sulfate moiety at Tyr

The zona pellucida protein piopio regulates the metamorphosis and reproduction in Tribolium castaneum.

The zona pellucida domain protein piopio (Pio) was only reported to mediate the adhesion of the apical epithelial surface and the overlying apical extracellular matrix in Drosophila melanogaster, but the developmental roles of Pio were poorly understood in insects. To address this issue, we comprehensively analyzed the function of Pio in Tribolium castaneum. Phylogenetic analysis indicated that pio exhibited one-to-one orthologous relationship among insects. T. castaneum pio had a 1236-bp ORF and contained eight exons. During development pio was abundantly expressed from larva to adult and low
