Cellular and molecular basis of IR3535 perception in Drosophila.

BACKGROUND: IR3535 is among the most widely used synthetic insect repellents, particularly for the mitigation of mosquito-borne diseases such as malaria, yellow fever, dengue and Zika, as well as to control flies, ticks, fleas, lice and mites. These insects are well-known vectors of deadly diseases that affect humans, livestock and crops. Moreover, global warming could increase the populations of these vectors. RESULTS: Here, we performed IR3535 dose-response analyses on Drosophila melanogaster, a well-known insect model organism, using electrophysiology and binary food choice assays. Our fin…

Toxic and repellent effects of hemp leaf extracts and cannabidiol against adult female yellow fever mosquitoes, Aedes aegypti.

Hemp (Cannabis sativa) derived products have emerged as potential sources of novel biopesticides. We recently demonstrated that a hemp leaf extract was toxic to larvae of the yellow fever mosquito Aedes aegypti, and that cannabidiol (CBD) was the primary larvicidal component of the extract. Here we evaluated the toxic and repellent effects of a hemp leaf extract and CBD against adult female Ae. aegypti. Toxicity was assessed using an acute topical assay in which mosquitoes received a 500 nL application to the thorax and mortality was recorded at 24 h. Repellency was then evaluated using two co

Curcuma longa essential oils: toxicity and repellency against imported fire ants (Formicidae: Hymenoptera).

Curcuma longa L. (Zingiberales: Zingiberaceae) leaf and rhizome essential oils were evaluated for their toxicity and repellency against invasive fire ants: red imported fire ants (RIFA), Solenopsis invicta Buren, black imported fire ants (BIFA), Solenopsis richteri Forel, and a reproductively functional hybrid (HIFA). Ar-turmerone was the major constituent of leaf (42.4%) and rhizome (40.4%) essential oils. A range of concentrations starting from 156 µg/g until the failure of treatment were used. Removal of treated sand in digging bioassay was used as a criterion for repellency. Leaf essential

Prolonged Repellent Activity of Plant Essential Oils against Dengue Vector, Aedes aegypti.

Repellents are effective personal protective means against outdoor biting mosquitoes. Repellent formulations composed of EOs are finding increased popularity among consumers. In this study, after an initial screening of 11 essential oils (EOs) at the concentration of 33 μg/cm2, five of the most repellent EOs, Perovskia atriplicifolia, Citrus reticulata (fruit peels), C. reticulata (leaves), Mentha longifolia, and Dysphania ambrosioides were further investigated for repellent activity against Aedes aegypti mosquitoes in time span bioassays. When tested at the concentrations of 33 μg/cm2, 165 μg

Made-to-measure malaria vector control strategies: rational design based on insecticide properties and coverage of blood resources for mosquitoes.

Eliminating malaria from highly endemic settings will require unprecedented levels of vector control. To suppress mosquito populations, vector control products targeting their blood hosts must attain high biological coverage of all available sources, rather than merely high demographic coverage of a targeted resource subset, such as humans while asleep indoors. Beyond defining biological coverage in a measurable way, the proportion of blood meals obtained from humans and the proportion of bites upon unprotected humans occurring indoors also suggest optimal target product profiles for deliverin

Extracts of Calpurnia aurea leaves from southern Ethiopia attract and immobilise or kill ticks.

Calpurnia aurea extracts are used in southern Ethiopia to protect stock against ticks. Acetone, hexane and water leaf extracts of C. aurea collected in southern Ethiopia were tested for repellent/attractant and acaricidal properties on unfed adult Rhipicephalus pulchellus ticks. In contrast to many other plant species evaluated, C. aurea extracts did not have repellent properties, but rather had a slight attractant capacity. With 20% and 10% acetone extracts, all ticks were either killed or their mobility severely compromised after 1microl of extract was topically applied on the abdomen. At a

A comparison of topical formulations for the prevention of human schistosomiasis.

Recently, a dimeticone formulation has been shown to be effective at preventing Schistosoma cercariae infecting skin, while DEET (N,N-diethyl-m-toluamide), a highly effective insecticide, has been shown to have activity against cercariae. Seven formulations, 3 containing DEET, were prepared and applied to excised human skin in Franz cells for 1 h. Schistosoma cercariae were applied for 30 min at 1 and 24 h, and the number that penetrated the skin calculated (n = 9). DEET could not be incorporated into the dimeticone formulation, yet it remained the most effective at preventing cercarial penetr

Putting behaviors into context for vector-borne diseases: Examining behaviors that may reduce exposure to disease vectors.

Understanding why people adopt or ignore vector-borne disease (VBD) preventive measures is key to better risk assessment and control. However, little is known about why some households adopt preventive practices while others do not, which is particularly striking in this era of One Health's attention to human, environmental, and animal health. We examined what household factors are linked to practices that may reduce exposure to disease vectors, with a focus on Chagas disease (CD) and American Cutaneous Leishmaniasis (ACL) vectors. We surveyed 204 households in 12 rural communities in Coclé Pr

Non-targeted analysis and suspect screening of organic contaminants in temperate snowfall using liquid chromatography high-resolution mass spectrometry.

Contaminants released into the atmosphere that undergo regional and long-range transport can deposit back to Earth through snowfall. When snow melts, these contaminants re-enter the environment, sometimes far from their original emission sources. Here we present the first comprehensive characterization of organic contaminants in snow from North America. Fresh snowfall samples were collected in the central United States over a three-year period and measured by liquid chromatography high-resolution mass spectrometry for suspect screening and non-targeted analysis. The resulting data set was scre

[What primary prevention should be used to prevent Lyme disease?].

Arthropod-borne diseases are a real public health problem. One of these, Lyme disease, is a bacterial infection due to Borrelia spp., transmitted by a hard tick, Ixodes spp.. The infection is particularly prevalent in the Northern Hemisphere and primary prevention relies on the use of repellents for cloth impregnation (pyrethroids) or for skin application (DEET). Pyrethroids and DEET are the two most studied repellents. The concentration of the active principle is essential to get a real repellent efficiency. The most efficient are: DEET at 30-50%, picaridin or KBR3023 at 20%, citriodiol at 30

Morbidity assessment in sand flea disease (tungiasis).

Tungiasis, caused by the sand flea Tunga penetrans, is a health problem in many impoverished communities in Latin America, the Caribbean, and sub-Saharan Africa. Sand flea disease is associated with a broad spectrum of clinical and histological pathology. The factors determining the disease burden in endemic communities are not well understood, and severity of clinical pathology has never been assessed quantitatively. Thus, two severity scores were developed: one for acute disease and one for chronic sequels. These scores were evaluated in a cohort of 70 severely infested patients living in a

Maize terpene synthase 1 impacts insect behavior via the production of monoterpene volatiles β-myrcene and linalool.

Plant-derived volatiles are important mediators of plant-insect interactions as they can provide cues for host location and quality, or act as direct or indirect defense molecules. The volatiles produced by Zea mays (maize) include a range of terpenes, likely produced by several of the terpene synthases (TPS) present in maize. Determining the roles of specific terpene volatiles and individual TPSs in maize-insect interactions is challenging due to the promiscuous nature of TPSs in vitro and their potential for functional redundancy. In this study, we used metabolite GWAS of a sweetcorn diversi

Laboratory and field evaluation of neem (Azadirachta indica A. Juss) and Chinaberry (Melia azedarach L.) oils as repellents against Phlebotomus orientalis and P. bergeroti (Diptera: Psychodidae) in Ethiopia.

The study evaluated the efficacy of neem (Azadirachta indica A. Juss.) and Chinaberry (Melia azedarach L.) seed oils as repellents against laboratory and field populations of some sandflies in Ethiopia. In the laboratory, concentrations of 2% and 5% neem oil in coconut oil tested against Phlebotomus orientalis (vector of visceral leishmaniasis) provided 96.28% (95% CI=95.60-96.97) protection up to a mean time of 7h and 20 min and 98.26% (95% CI=93.46-104. 07) protection up to 9h, respectively. Similarly, M. azedarach oil at 2% concentration produced 95.13% (95% CI=90.74-99.52) protection for t

Potential of essential oils to prevent fly strike and their effects on the longevity of adult Lucilia sericata.

Lucilia sericata is a facultative ectoparasite causing fly strike or myiasis in warm-blooded vertebrates. It is controlled by traps or insecticides, but both have drawbacks and alternative ways of control are urgently needed. Essential oils (EOs) of vetiver (Chrysopogon zizanioides), cinnamon (Cinnamomum zeylanicum), and lavender (Lavandula angustifolia) and their blends (OBs); OB1 (2 ml of each EO plus 4 ml of sunflower oil as a carrier) and OB2 (2 ml of each EO) were tested. Oils were tested at 5% for deterrence assays, and a dose response assay 0.01-0.6%, was conducted to determine forced-c

Field trials of fatty acids and geraniol applied to cattle for suppression of horn flies, Haematobia irritans (Diptera: Muscidae), with observations on fly defensive behaviors.

Adult horn fly populations were tracked on cattle for 2-week periods before, during and after multiple treatments (every 3-4days) with two repellents in a mineral oil carrier. Cattle were sprayed four times in a two-week period either with 2% geraniol (125ml/cow) or a 15% mixture of short chain fatty acids (C8-C9-C10)(250ml/cow), and there were untreated control cattle. Trials were conducted in California and North Carolina for 3 summers. Short-term fly counts (same day) on treated cattle were reduced by 61-99%, depending on material and trial, and the fatty acid mixture provided better contro

Aedes aegypti (Diptera: Culicidae) biting deterrence: structure-activity relationship of saturated and unsaturated fatty acids.

In this study we evaluated the biting deterrent effects of a series of saturated and unsaturated fatty acids against Aedes aegypti (L), yellow fever mosquito (Diptera: Culicidae) using the K & Dbioassay module system. Saturated (C6:0 to C16:0 and C18:0) and unsaturated fatty acids (C11:1 to C14:1, C16:1, C18:1, and C18:2) showed biting deterrence index (BDI) values significantly greater than ethanol, the negative control. Among the saturated fatty acids, mid chain length acids (C10:0 to C13:0) showed higher biting deterrence than short (C6:0 to C9:0) and long chain length acids (C14:0 to C18:0
