Copper ions dynamically regulate beta3 integrin subunit expression in Ishikawa cells.

Copper intrauterine device (IUD) users have a reduced expression of beta3 integrin subunit during the implantation window. This integrin has been found to be a uterine marker for implantation. In order to verify if copper ions are involved in this reduction, beta3 integrin subunit expression was assessed in an in vitro model. The beta3 integrin subunit expression was determined by flow cytometry in the presence of copper D-gluconate and/or epidermal growth factor (EGF). The mean percentages of positive beta3 integrin subunit cells under different culture conditions were: 4.7 +/- 0.29 (mean +/-…

Organization-dependent effects of toxic bivalent ions microtubule assembly and glycolysis.

The effects of bivalent ions on tubulin dynamics and the upper phase of glycolysis were investigated at different organization levels in vitro. Cu2+, Cd2+, Hg2+ and CrO4(2-) inhibit the tubulin polymerization at an IC50 of 14-24 microM with high cooperativity and also induce microtubule disassembly. The apparent binding constants of the ions to tubulin, estimated by fluorescence quenching, vary between 6 and 28 microM. BIAcore measurements for tubulin-tubulin interaction suggest that the presence of Cu2+ affects neither koff nor kon, but the amount of the bound tubulin. While the inhibitory ef…

Removal of calcium ions triggers a novel type of intercadherin interaction.

Depletion of Ca(2+) ions from epithelial cell cultures has been shown to result in the rapid destruction of intercellular junctions. To understand the mechanism of this effect we have examined how removal of calcium ions from the culture medium of A-431 epithelial cells affects complexes incorporating the cell-cell adhesive receptors, E-cadherin, desmoglein or desmocollin. Sedimentation and biochemical analysis demonstrated that calcium removal triggers a rapid formation of a novel type of complex formed via direct lateral E-cadherin-desmoglein, E-cadherin-desmocollin and desmoglein-desmocolli…

The importance of calcium ions for in vitro malignant hyperthermia testing.

Intracellular Ca++ levels in skeletal muscle are elevated during the in vitro contracture response of muscle from subjects with malignant hyperthermia. The role of Ca++ in the bathing medium and the consequences of substitution of Sr++ for Ca++ in the response to agents associated with malignant hyperthermia were examined. When Ca++ was omitted from the bathing medium the contractures induced in human vastus lateralis by halothane (three per cent) or succinylcholine (50 mM) were reduced by 80 and 100 per cent, respectively, while contractures induced by caffeine (8 mM) were only reduced by 50

Inhibition of glycogen synthase (casein) kinase-1 by divalent metal ions.

The specificity of glycogen synthase (casein) kinase-1 (CK-1) for different divalent metal ions was explored in this study. Of nine metal ions (Mg2+, Mn2+, Zn2+, Cu2+, Ca2+, Ba2+, Ni2+, Co2+, Fe2+) tested, only Mg2+ supported significant kinase activity. Several of the other metals, however, inhibited the Mg2+-stimulated kinase activity. Half-maximal inhibitions by Mn2+, Zn2+, Co2+, Fe2+, and Ni2+ were observed at 55, 65, 110, 125, and 284 microM, respectively. Kinetic analyses indicate that the metal ions are acting as competitive inhibitors of CK-1 with respect to the protein substrate (case…

pH shifts and precipitation associated with metal ions in tissue culture.

Recently there has been interest in tests to assess the physiological response to surgical implant corrosion products. Both cell culturing in metal-bearing solutions and intramuscular injection of such solutions have been carried out. This paper examines the effects of the constraints of multicomponent equilibrium conditions on the characteristics of the solutions used in these biocompatibility tests. It is demonstrated that, unbuffered, they will have pH values shifted from neutral and that, in the buffered state, these solutions may contain both dissolved metal ions and insoluble hydroxides.…

Effects of heavy ions on rabbit tissues: damage to the forebrain.

As part of a study of progressive radiation effects in normal tissues, the forebrains of New Zealand white rabbits (Oryctolagus cuniculus) (about 6 weeks old) were irradiated locally with single acute doses of 60Co gamma-photons (LET infinity = 0.3 keV/micron), Ne ions (LET infinity = 35 +/- 3 keV/microns) or Ar ions (LET infinity = 90 +/- 5 keV/microns). Other rabbits received fractionated doses of 60Co gamma-photons according to a standard radiotherapeutic protocol. Irradiated rabbits and appropriately aged controls were sacrificed at selected intervals, and whole sagittal sections of their …

Calcium ions modulate hormonally stimulated progesterone production in isolated ovarian cells.

Swine granulosa-luteal cells incubated in Ca2+-deficient medium (5 muM final Ca2+ concentration) for short time periods produced diminished quantities of progesterone in response to lutropin. Maximally stimulating effects of prostaglandin E2 and L-adrenaline were also impaired significantly. Diminished progesterone production could not be attributed to alterations in protein synthesis or cell viability. Under Ca2+-deprived conditions, the stimulatory actions of cholera toxin, 3-isobutyl-1-methylxanthine and 8-bromo cyclic AMP were also significantly impeded. Administration of a presumptive ant…

Magnesium ions and the control of the cell cycle in yeast.

A study has been made of the role of magnesium ions in cell division cycle control in the fission yeast, Schizosaccharomyces pombe, and the budding yeast, Kluyveromyces fraglis. Synchronization of cell division in these organismms can be induced by restoring magnesium to magnesium-exhausted cultures. In S. pombe, a correlation exists between the time taken for cells to enter the first synchronous division and the period of magnesium exhaustion. During short-term incubation in magnesium-deficient media, S. pombe cells are observed to continue growth in length, but they fail to make a cell plate…

Migration of metallic ions from screwposts into dentin and surrounding tissues.

Previous investigations have shown htat corrosion and other electrochemical processes occure when different alloys or metals are found together in the same mouth. In the present report, when teeth were restored using non-noble metallic posts, the metals diffused out to surrounding hard and soft connective tissues. The material consisted of extracted teeth with screw posts and surrounding discolored connective tissues. The screwposts had been cemented to the teeth 3--10 years earlier. The distribution of metal ions was determined by means of energy-dispersive X-ray microanalysis. Copper and zin…

Movements of labelled sodium ions in isolated rat superior cervical ganglia.

1. Isolated rat superior cervical ganglia were incubated in Krebs solution containing (24)Na and carbachol for 4 min at 25 degrees C. They were then washed at 3 degrees C for 15 min to remove extracellular (24)Na and the efflux of residual intracellular (24)Na stimulated by warming to 25 degrees C.2. During the 15 min wash at 3 degrees C desaturation curves became exponential with a rate constant of 0.012 +/- 0.001 min(-1) (n = 24). This was assumed to represent loss of intracellular (24)Na, and initial uptake of (24)Na was calculated therefrom by back-extrapolation to zero wash-time. After 4

Intrinsic Reactivity of the Elusive Cyclic N,N-Diacyliminium Ions in Mannich Reactions: The Complete Reactivity Order for Non, N-Mono and N,N-Diacyliminium Ions.

We report the first intrinsic reactivity measurements of four cyclic N,N-diacyliminium ions in Mannich-type reactions. Using MSn experiments (n = 1, 2) performed in the unique, convenient, and gaseous environment of a pentaquadrupole mass spectrometer, we reacted four gaseous five- or six-membered cyclic N,N-diacyliminium ions with allyltrimethylsilane and have established a complete reactivity order, demonstrating that N,N-diacyliminium ions surpass their N-monoacyl counterparts in reactivity. The study also enabled the isolation and characterization of key Mannich intermediates, confirming t…

Correlation of precursor and product ions in single-stage high resolution mass spectrometry. A tool for detecting diagnostic ions and improving the precursor elemental composition elucidation.

Monitoring of common diagnostic fragments is essential for recognizing molecules which are members of a particular compound class. Up to now … us precursor selection. Thus, to address this limitation, an in silico approach to locate precursor ions, based on diagnostic fragments, was developed. Implemented as an Excel macro, the algorithm rapidly assembles and surveys exact mass data to provide a list of feasible precursor candidates according to the correlation of the chromatographic peak sha …

Electrochemically tailored ion-trapping nanoarchitectures in COF membranes for selective monovalent/divalent ion sieving.

The efficient separation of monovalent and divalent ions remains a core challenge in desalination and ion resource recovery. Here, we propose an electrochemical confinement strategy to spatially organize EDTA into aligned arrays within the one-dimensional transport channels of TFP-Tag membrane with a pore size of 1.1 nm. This structure establishes a biomimetic, gradient-functionalized conical nanochannel. The negatively charged upper channel regions capture and block divalent ions, while the lower regions with their larger pore size and an anion-affinitive local environment facilitate monovale…

Solvent and Ion-Mediated Behavior of a Thermoresponsive Brush: Specific Ion Effects in Methanol-Water Electrolytes.

In this study, specific ion effects are explored in methanol-water mixtures, which play a critical role in a diverse range of applications, including protein solubilization and supercapacitors. Spectroscopic ellipsometry and neutron reflectometry are employed to investigate the solvent- and ion-mediated behavior of a poly(N-isopropylacrylamide) (PNIPAM) brush, a well-known thermoresponsive polymer. In the absence of ions and at low methanol mole fractions (xM), PNIPAM displays lower critical solution temperature (LCST) type behavior, with the thermotransition temperature decreasing as xM incre…

Supramolecular Ion Channels to Engineer Zn2+ Ion Transport Mediated Chemical-to-Optical Signal Transduction.

Artificial ion channels have significant potential for various applications, including molecular communication, bio-sensing, and constructing artificial cells. In this report, we demonstrated the development of a molecular communication system that transports ions across the lipid bilayers via the formation of supramolecular ion channels and harnesses chemical reactions to overcome the challenges of executing signal-processing functions at the molecular level. The potent ionophore self-assembles into nanochannels within the lipid bilayers and selectively transports Zn2+ ions. The movement of Z…

Redox-Regulated Synthetic Channels: Enabling Reversible Ion Transport by Modulating the Ion-Permeation Pathway.

Natural redox-regulated channel proteins often utilize disulfide bonds as redox sensors for adaptive regulation of channel conformations in response to diverse physiological environments. In this study, we developed novel synthetic ion channels capable of reversibly switching their ion-transport capabilities by incorporating multiple disulfide bonds into artificial systems. X-ray structural analysis and electrophysiological experiments demonstrated that these disulfide-bridged molecules possess well-defined tubular cavities and can be efficiently inserted into lipid bilayers to form artificial…

Prediction of the Be2B2 monolayer: an ultrahigh capacity anode material for Li-ion and Na-ion batteries.

In the recent past, beryllium-boron compounds have been the focus of attention as a consequence of their extremely rich structures and potential applications. Nevertheless, their complex phase structures greatly hinder further exploration of the physical and chemical properties. To this end, we propose a novel beryllium-boron compound based on a two-dimensional (2D) structure, namely a Be2B2 monolayer. Using first-principles methods, we first prove that the structure of the 2D Be2B2 monolayer is energetically and thermodynamically stable. Subsequently, we systematically explored its potential …
