Oral vaccine of Lactococcus lactis harbouring pandemic H1N1 2009 haemagglutinin1 and nisP anchor fusion protein elevates anti-HA1 sIgA levels in mice.

OBJECTIVE: An oral lactococcal-based vaccine which haboured the haemagglutinin1 (HA1) antigen fused to nisP anchor protein for the purpose of surface displaying the HA1 antigen was developed against H1N1 virus. RESULTS: Recombinant L. lactis strains expressed HA1-nisP fusion proteins when induced with nisin, as confirmed through western blotting. However, immunofluorescense did not detect any surface-displayed proteins, suggesting that the protein was either unsuccessfully translocated or improperly displayed. Despite this, oral administration of recombinant L. lactis strains to BALB/c mice r

Engineered invasive Lactococcus lactis as an oral vaccine delivery platform is associated with compartmentalized immune modulation and enhances control of Leishmania infantum infection.

Visceral leishmaniasis caused by Leishmania infantum remains a major challenge for vaccine development due to the intracellular localization of the parasite and the need for effective antigen delivery and cellular immune responses. Genetically engineered lactic acid bacteria have emerged as promising platforms for mucosal vaccination because of their genetic tractability and capacity to interact with the host immune system. Here, we evaluated an engineered invasive Lactococcus lactis strain expressing fibronectin-binding protein A (FnBPA) and carrying the chimeric TryPan antigen-encoding plasm…

Comparison of Postbiotics and Probiotics of Lactococcus lactis NZ9000 in Alleviating Liver Injury Induced by Bisphenol A via Binding and Removing Bisphenol A in Mouse Model.

Studies have demonstrated the toxic effects of inevitable exposure of bisphenol A (BPA), especially liver injury via gut-liver axis. Probiotics are increasingly used as adjunctive therapy to ameliorate adversely effects induced by BPA. In view of safety concerns of probiotics, postbiotics are proposed as alternatives to probiotics in clinical applications. At present, no study has directly compared the ameliorative effect of probiotics and postbiotics. This study compares the effect of postbiotics and probiotics derived from Lactococcus lactis NZ9000 employed for BPA-induced liver injury in a …

Response to intranasal Lactococcus lactis W136 probiotic supplementation in refractory CRS is associated with modulation of non-type 2 inflammation and epithelial regeneration.

JUSTIFICATION: We have previously documented that in individuals with chronic rhinosinusitis (CRS) refractory to surgery, intranasal application of live Lactococcus lactis W136, a probiotic bacterium, improves sinus-specific symptoms, SNOT-22, and mucosal aspect on endoscopy, accompanied by a reduction in sinus pathogens and an increase in protective bacteria. The present work explores the molecular mechanisms underpinning these observations using transcriptomics of the sinus mucosa. METHOD: Epithelial brushings collected prospectively as a sub-study of the L. lactis W136 clinical trial were …

Whole-Genome Sequencing Reveals Virulence and Antimicrobial Resistance Determinants of Lactococcus garvieae Causing Lactococcosis in Cage-Cultured Nile Tilapia (Oreochromis niloticus) in Thailand.

Lactococcosis is an important bacterial disease affecting farmed fish worldwide and is primarily associated with Lactococcus garvieae, Lactococcus petauri, and Lactococcus formosensis. In Thailand, information on L. garvieae infection in tilapia remains limited, particularly regarding genome-based identification, virulence determinants, and antimicrobial resistance profiles. This study characterized two L. garvieae isolates, AAHM-LG2501 and AAHM-LG2509, recovered from a lactococcosis outbreak in cage-cultured Nile tilapia (Oreochromis niloticus) in Ubon Ratchathani province, Thailand. Both iso…

High GABA-producing Lactococcus lactis ZFM559 and its derived GABA alleviate chronic unpredictable mild stress (CUMS)-induced depression via the "microbiota-brain" axis.

γ-Aminobutyric acid (GABA) is a crucial inhibitory neurotransmitter with recognized benefits in alleviating depression. However, the ability of exogenous GABA to cross the blood-brain barrier remains debated, and the underlying anti-depression mechanisms via the "microbiota-brain" axis remain unclear. Herein, a high GABA-producing strain (L. lactis ZFM559) was optimized via whole-cell catalysis, and its derived GABA (pGABA, purity of 99.47%) was also prepared, followed by uncovering the underlying anti-depression mechanisms using a chronic unpredictable mild stress (CUMS) model. Results indica

Therapeutic efficacy of oral immunization with a non-genetically modified Lactococcus lactis-based vaccine CUE-GEM induces local immunity against Helicobacter pylori infection.

The gastric bacterial pathogen Helicobacter pylori persistently colonizes the gastric mucosa of humans and plays a critical role in the development of gastritis, peptic ulceration and gastric adenocarcinoma. Consequently, the eradication of H. pylori might contribute to the prevention of H. pylori-associated gastric diseases. In this study, a multi-epitope vaccine CTB-UE (CUE) was displayed on the surface of non-genetically modified Lactococcus lactis particles (GEM) to enhance immunogenicity. This particulate vaccine CUE-GEM induced serum and mucosal specific antibody responses against native…

Production and targeting of the Brucella abortus antigen L7/L12 in Lactococcus lactis: a first step towards food-grade live vaccines against brucellosis.

Brucella abortus is a facultative intracellular gram-negative bacterial pathogen that infects humans and animals by entry mainly through the digestive tract. B. abortus causes abortion in pregnant cattle and undulant fever in humans. The immunogenic B. abortus ribosomal protein L7/L12 is a promising candidate antigen for the development of oral live vaccines against brucellosis, using food-grade lactic acid bacteria (LAB) as a carrier. The L7/L12 gene was expressed in Lactococcus lactis, the model LAB, under the nisin-inducible promoter. Using different signals, L7/L12 was produced in cytoplas…

Polymorphism in the Yersinia LcrV Antigen Enables Immune Escape From the Protection Conferred by an LcrV-Secreting Lactococcus Lactis in a Pseudotuberculosis Mouse Model.

Yersinioses caused by Yersinia pestis, Yersinia pseudotuberculosis, and Yersinia enterocolitica are significant concerns in human and veterinary health. The link between virulence and the potent LcrV antigen has prompted the latter's selection as a major component of anti-Yersinia vaccines. Here, we report that (i) the group of Yersinia species encompassing Y. pestis and Y. pseudotuberculosis produces at least five different clades of LcrV and (ii) vaccination of mice with an LcrV-secreting Lactococcus lactis only protected against Yersinia strains producing the same LcrV clade as that of used…

Experimental and numerical study of heterogeneous pressure-temperature-induced lethal and sublethal injury of Lactococcus lactis in a medium scale high-pressure autoclave.

The present contribution is dedicated to experimental and theoretical assessment of microbiological process heterogeneities of the high-pressure (HP) inactivation of Lactococcus lactis ssp. cremoris MG 1363. The inactivation kinetics are determined in dependence of pressure, process time, temperature and absence or presence of co-solutes in the buffer system namely 4 M sodium chloride and 1.5 M sucrose. The kinetic analysis is carried out in a 0.1-L autoclave in order to minimise thermal and convective effects. Upon these data, a deterministic inactivation model is formulated with the logistic…

Recombinant Lactococcus lactis-based oral vaccine expressing non-toxic alpha toxin domains of Clostridium septicum can offer protection against Clostridial dermatitis in turkeys.

Clostridial dermatitis (CD), caused by the anaerobic spore-forming Clostridium septicum bacteria, is an important emerging disease of turkeys. Despite its economic burden on the poultry industry, there are no efficacious vaccines currently available for CD control in turkeys. We recently identified two non-toxic domains of C. septicum alpha toxin (ntATX), namely ntATX-D1 and ntATX-D2, and showed that subcutaneous immunization of turkeys with purified recombinant subunit ntATX proteins can offer protection against CD. In the present study, we used the pNZ8124-NICE vector®-based Lactococcus lact…
