Co-existence of blue-sky catastrophe and bistability in a coupled Leech-heart inter-neuron model with both chemical and electrical coupling.

Neuronal activity is characterized by diverse firing patterns that underlie both normal physiological processes and pathological states of the brain. In this study, we explore diverse firing activities, including tonic, bursting and spiking in a coupled Leech Heart inter-neuron model incorporated with both chemical and electrical synapses. These rich dynamics arise due to the emergence of multiple bifurcations, including codimension-one bifurcations (Hopf, saddle-node, and branch point) and codimension-two bifurcations (Bogdanov-Takens and cusp) in the model. Further, due to the changes of che

Clinical Efficacy of Leech Therapy (Ta'līq al-'Alaq) in Eczema (Nār Fārsī): An Open-Label Single-Arm Clinical Study.

BACKGROUND: Eczema, known as Nār Fārsī in Unani medicine, is a chronic skin disorder characterized by intense pruritus, dryness, and scaling of skin, along with periodic flares and remissions. Unani literature recognizes it as a disorder caused by safrāwī (bilious) humor. Conventional treatment relies on emollients, humectants, and anti-inflammatory drugs. Unani medicine recommends the expulsion of morbid matter by local blood-letting or oral therapies. Leech therapy, used in many traditional medicine systems for centuries, is a method of expulsion of morbid humors from a disease site. It is p

Production, Purification, and Characterization of a Novel Cysteine-Rich Anticoagulant from the Medicinal Leech and the Functional Role of Its C-Terminal Motif.

The saliva of the medicinal leech Hirudo medicinalis contains a wide range of biologically active compounds, including multiple anticoagulants. Previously, we identified a novel cysteine-rich anticoagulant protein (CRA) from leech saliva and produced it recombinantly in Escherichia coli, demonstrating its potential as a basis for new anticoagulant drugs. In this study, we developed an optimized procedure for scalable production and purification of recombinant CRA. The purified protein was analyzed for common contaminants originating from E. coli, such as endotoxins, bacterial proteins, and DNA

Transcriptomic profiling and the first spatial expression analysis of candidate genes in the salivary gland of the East Asian medicinal leech, Hirudo nipponia.

Hirudo nipponia, a blood-sucking leech native to East Asia, possesses a rich repertoire of active ingredients in its saliva, showcasing significant medical potential due to its anticoagulant, anti-inflammatory, and antibacterial effects against human diseases. Despite previous studies on the transcriptomic and proteomic characteristics of leech saliva, which have identified medicinal compounds, our knowledge of tissue-specific transcriptomes and their spatial expression patterns remains incomplete. In this study, we conducted an extensive transcriptomic profiling of the salivary gland tissue i

Life without blood: Molecular and functional analysis of hirudins and hirudin-like factors of the Asian non-hematophagous leech Whitmania pigra.

BACKGROUND: Several leech species of the genera Hirudo, Hirudinaria, and Whitmania are widely used in traditional Chinese medicine (TCM) for the oral treatment of disorders associated with blood stasis. Among them, the non-hematophagous leech Whitmania pigra expresses a variety of components that have the potential to act on the vertebrate blood coagulation system. OBJECTIVE: Whether the thrombin inhibitor hirudin, probably the most prominent leech-derived anticoagulant, is actually present in Whitmania pigra, is still a matter of debate. To answer that open question was the aim of the study.

Incomplete versus Sustained Innate-Immune Responses in the Medicinal Leech following Chronic, Environmentally Relevant Per- and Polyfluoroalkyl Substance Exposure.

Per- and polyfluoroalkyl substances (PFAS) persist in aquatic systems, yet hazard evaluations still rely largely on short-term tests. We chronically exposed the medicinal leech Hirudo verbana for two months to four PFAS congeners at 1 nmol L-1 aligned with the EU Drinking Water Directive thresholds (PFAS total = 0.50 μg L-1): a legacy long-chain acid (PFOA), two short-chain ether replacements (HFPO-DA and PFMoBa), and an intermediate methoxy analogue (PFMOPrA). Histology, enzymatic histochemical staining, immunofluorescence, and quantitative polymerase chain reaction tracked the canonical infl

Trophic transfer of chromium from Bellamya aeruginosa drives edema in medicinal leech Whitmania pigra via the ROS-FoxO-ENaC signaling.

Environmental accumulation of chromium in aquatic food webs presents a growing threat to animal and human health. While previous studies established a link between intestinal dysbiosis and Whitmania pigra edema disease (WPE), the underlying mechanisms remain poorly understood. In this study, we demonstrate that Bellamya aeruginosa collected from polluted freshwater habitats bioaccumulates Cr and transmits it through the food chain, inducing WPE under the conditions of this experiment. Cr (VI) exposure via feeding or immersion led to disrupted gut microbiota, elevated reactive oxygen species (R

Two new leech species of the genus Haemadipsa Tennent, 1859 from Tibet, China and characterization of their mitochondrial genomes (Clitellata: Hirudiniformes: Haemadipsidae).

This paper presents two novel terrestrial leech species, Haemadipsa nigrilineata sp. nov. and Haemadipsa medogensissp. nov., collected from forested regions in Tibet, China. On the basis of morphological observations, mitogenomic sequence analysis and the mitochondrial COI gene sequence phylogenetic inference, both species are herein described as new within the genus Haemadipsa Tennent, (Haemadipsidae). These two species are in line with the general diagnosis of the genus Haemadipsa in terms of living environment, annulation pattern, having five pairs of eyespots, and the position of the gonop

Multidrug resistant Aeromonas infection following medical leech therapy: A case report and development of a joint antimicrobial stewardship and infection prevention protocol.

OBJECTIVE: Aeromonas sp. infections are a recognized complication of medical leech therapy (MLT). In patients requiring MLT, ciprofloxacin or trimethoprim-sulphamethoxazole are commonly used to prevent such nosocomial infections. After a patient at our institution developed a MLT-associated multi-drug resistant (MDR) Aeromonas infection, we developed and evaluated a joint antimicrobial stewardship and infection prevention protocol for MLT at our institution. METHODS: We describe a case of a surgical site infection with MDR Aeromonas following MLT that was resistant to typically prescribed pro

Oropharyngeal leech infestation as an unusual cause of severe anemia and hypovolemic shock in a 2-year-old child: A case report from Ethiopia.

BACKGROUND: Leech infestations can cause significant, often unrecognized, blood loss. This type of infestation occurs most commonly in young children living in rural settings who consume unfiltered water from rivers. CASE PRESENTATION: A two-year-old female patient presented to a general hospital from a rural area of Ethiopia with a five-day history of epistaxis, blood-stained saliva, fatigue, and poor appetite. On examination, hemoglobin was 3 g/dL. A dark brown, mobile foreign body - subsequently identified as a leech - was found attached to the posterior oropharynx. The patient (weight 11

From Maggots to Recovery: Exploring an Integrative Medicine Approach to Chronic Non-Healing Ulcer with Leech Therapy and Panchvalkal Ointment: A Case Study.

BACKGROUND: Chronic, non-healing ulcers, referred to as dushtavrana in Ayurveda present a challenge in medical care since ancient times. Ayurvedic literature, notably Acharya Susrutha Chikithsa Sthana, dedicated a chapter to the treatment of dushtavrana, encompassing sixty procedures (shashti upakramas). Chronic ulcers, influenced by diverse factors, resist a singular therapeutic approach. The inflammatory stage is critical, and disruptions in the order of stages of healing can lead to chronicity. Despite conventional treatments, many ulcers remain resistant, causing physical, financial, and e

Couplet medicines of leech and centipede granules improve erectile dysfunction via inactivation of the CaSR/PLC/PKC signaling in streptozotocin-induced diabetic rats.

Erectile dysfunction (ED) is one of the significant complications of diabetes mellitus (DM), and CASR plays an important role in cellular antiapoptosis and NO production in the vascular endothelium by activating PKC. The present study was aimed to investigate the efficacy of Leech and Centipede Granules (LCG) through the CaSR/PLC/PKC signaling. Fifty male Sprague-Dawley rats were treated with streptozotocin to induce the DM model. After 10 weeks, an apomorphine test was used to confirm DMED. Rats with DMED were administrated with LCG and U73122 for 4 weeks. Fasting blood glucose, body weight,

[Infections Associated with Treatment with Hirudo medicinalis - Report of Two Cases and Review of the Literature].

Medicinal leeches are well-established for promoting venous drainage in transplants/flaps and analgesia in osteoarthritis. Although medicinal leeches are bred and kept under controlled conditions, they are colonised by a genuine species-specific bacterial flora. Therapeutic application of leeches implies skin penetration carrying an a priori risk of infection. We report 2 cases with different indications for medicinal leech therapy. In both cases wound infection occurred in close temporal and spatial correlation or with evidence of a leech-associated germ that could be treated successfully. An

Single and combined effects of cadmium, microplastics, and their mixture on whole-body serotonin and feeding behaviour following chronic exposure and subsequent recovery in the freshwater leech, Nephelopsis obscura.

Microplastics and metals are contaminants detected in many freshwater systems globally. Interactions of microplastics with other contaminants including cadmium poses potential threats to the health of aquatic organisms including Nephelopsis obscura, a predatory leech species that is widespread and serves important ecological and economic roles. The feeding biology of N. obscura has been well-described, including that serotonin regulates feeding behaviour. Further, exposure to cadmium has been found to cause decrease whole-body concentrations of serotonin. The influence that microplastic contam

Hirudiniasis in Cattle in Mpwapwa District, Dodoma Region of Tanzania.

Hirudiniasis is a condition caused by infestation by leeches. Despite the annoyance, clinical signs, and associated lesions caused by leeches in both humans and animals, the extent of the problem in Tanzania is not well elucidated. Individual animals and people become infested when they drink, bath, or walk through contaminated water sources. Leech infestations are more common in rural areas where water is scarce especially during the dry season. In this report, clinical cases of hirudiniasis in twenty-seven (27) cattle in Mpwapwa, Tanzania, are presented. The report further reviews the morpho
