Acetaminophen inhibits hemoprotein-catalyzed lipid peroxidation and attenuates rhabdomyolysis-induced renal failure.

Hemoproteins, hemoglobin and myoglobin, once released from cells can cause severe oxidative damage as a consequence of heme redox cycling between ferric and ferryl states that generates radical species that induce lipid peroxidation. We demonstrate in vitro that acetaminophen inhibits hemoprotein-induced lipid peroxidation by reducing ferryl heme to its ferric state and quenching globin radicals. Severe muscle injury (rhabdomyolysis) is accompanied by the release of myoglobin that becomes deposited in the kidney, causing renal injury. We previously showed in a rat model of rhabdomyolysis that …

Dietary trans-resveratrol bioavailability and effect on CCl4-induced liver lipid peroxidation.

BACKGROUND AND AIM: Several in vitro studies have demonstrated the ability of pure trans-resveratrol (t-Res) to act as an anti-oxidant, but the scientific literature is lacking in in vivo studies dealing with dietary t-Res bioavailability in oxidative stress models. Our aim was to investigate the bioavailability of t-Res from dietary sources and its effect on an animal model of carbon tetrachloride (CCl4)-induced liver lipid peroxidation. METHODS: Ten rats were intragastrically administered for 14 days with a grape-stalk extract determining a daily t-Res dosage of 3 mg/kg. The control group (…

Effect of carnitine on stress-induced lipid peroxidation in rat gastric mucosa.

PURPOSE: Carnitine is an essential cofactor in the mitochondrial transfer of fatty acids, but is also a scavenger of oxygen free radicals in mammalian tissues. It has been shown that cold-restraint stress (CRS) produces gastric mucosal injury due to oxygen free radicals. The aim of this study was to determine the effect of L-carnitine on lipid peroxidation induced by CRS in rat stomachs. METHODS: Rats pretreated with L-carnitine (50 mg/kg per day for 10 days) were restrained in a wire cage for 4 h at 4 degrees C. At the end of the experimental period, the lesion index in gastric mucosa was de…

Increased circulating products of lipid peroxidation in patients with alcoholic liver disease.

F2-isoprostanes (F2-IP) and 4-hydroxynonenal (4-HNE), peroxidation products of polyunsaturated fatty acids (PUFA), are considered the most reliable indicators of endogenous lipid peroxidation in vivo. To determine to what extent these are also altered in patients with alcoholic liver disease, plasma free and esterified F2-IP as well as 4-HNE were measured by GC/MS in 49 fasting subjects who underwent diagnostic percutaneous needle biopsies of the liver. Compared to patients with mild steatosis and no fibrosis, free F2-IP and 4-HNE were strikingly increased in individuals with alcoholic hepatit…

In situ detection of lipid peroxidation by-products in chronic liver diseases.

Lipid peroxidation is an autocatalytic mechanism leading to oxidative destruction of cellular membranes. The deleterious consequences of this mechanism are related in part to the formation of reactive aldehydic products that bind to intra- or extracellular molecules to form adducts. Specific antibodies directed against malondialdehyde (MDA) and 4-hydroxynonenal (HNE) adducts, major aldehydic metabolites of lipid peroxidation, allowed us to investigate in situ, with an immunohistochemical procedure, the occurrence of lipid peroxidation in a panel of different chronic liver diseases. Intracellul…

Increased lipid peroxidation and antioxidant activity in methionine-induced hepatitis in rabbits.

The liver is especially susceptible to the toxic effects of methionine due to its role in sulfur amino acid metabolism. Therefore, the excessive amounts of this amino acid may induce liver damage. To test the mechanisms of methionine-related hepatotoxicity, liver thiobarbituric acid reactive substances (TBARS), and activities of superoxide dismutase, catalase and selenium-dependent glutathione peroxidase were measured in rabbits fed a methionine-enriched diet for 6 or 9 mo. Morphological studies of livers were also made. Feeding rabbits the methionine-enriched diet for 9 mo resulted in a signi

Lipid peroxidation in neural tissues and fractions from vitamin E-deficient rats.

A severe and chronic deficiency of vitamin E results in a characteristic neurological syndrome in both man and experimental animals. This is presumed to result from increased oxidative stress arising from a reduction in antioxidant capacity. In this study we have examined parameters of endogenous lipid peroxidation and susceptibility to in vitro oxidative stress of neural tissues and fractions, and some non-neural tissues from 1-year-old vitamin E-deficient and control rats. We have shown: (1) an increase in endogenous lipid peroxidation (thiobarbituric acid reactive substances and malondialde…

Role of lipid peroxidation in acetaminophen-induced hepatotoxicity: comparison with carbon tetrachloride.

The effect of acetaminophen on lipid peroxidation in vivo and in vitro was studied in rat liver and the data were compared with those with carbon tetrachloride. Carbon tetrachloride increased diene conjugates in vivo and thiobarbituric acid reactive substance production in vitro in the liver microsomal incubation. These changes were further enhanced by ethanol that has previously been shown to increase carbon tetrachloride-induced hepatotoxicity. On the other hand, acetaminophen did not increase diene conjugates in vivo and inhibited thiobarbituric acid reactive substance production in vitro. …
