Lipidomic profiling reveals metabolic signatures in psoriatic skin lesions.

Psoriasis is a chronic immune-mediated inflammatory disease. Lipids play an important role in regulating the inflammatory response. However, the alteration of lipids involved in psoriasis particular in skin lesions remain unclear. Here, we performed the lipidomics to investigate lipid profiling in the skin lesions of the imiquimod-induced psoriasis-like dermatitis and psoriasis patients. The findings showed that ceramides phosphate (CerP) and ceramides were enriched in psoriatic lesions compared with controls from both psoriasis patients and psoriasis-like mouse model. Psoriasis patients were

Lipidome profile predictive of disease evolution and activity in rheumatoid arthritis.

Lipid mediators are crucial for the pathogenesis of rheumatoid arthritis (RA); however, global analyses have not been undertaken to systematically define the lipidome underlying the dynamics of disease evolution, activation, and resolution. Here, we performed untargeted lipidomics analysis of synovial fluid and serum from RA patients at different disease activities and clinical phases (preclinical phase to active phase to sustained remission). We found that the lipidome profile in RA joint fluid was severely perturbed and that this correlated with the extent of inflammation and severity of syn

Diet and Life Stage-Associated Lipidome Remodeling in Atlantic Salmon.

Salmon is an important source of long-chain highly unsaturated fatty acids (LC-HUFAs) such as 22:6n-3 [docosahexaenoic acid (DHA)]. In the present study, we conducted two identical experiments on salmon in freshwater (FW) and seawater (SW) stages, with a diet switch from fish oil (high in LC-HUFA) to vegetable oil (low in LC-HUFA) and vice versa. Our aim was to investigate the diet and life stage-specific features of lipid uptake (gut), processing (liver), and deposition (muscle). The lipid composition changed much faster in the gut of SW fish relative to FW fish, suggesting that the former ha

Nuclear lipidome is altered in amyotrophic lateral sclerosis: A pilot study.

Nucleocytosolic transport, a membrane process, is impaired in motor neurons in amyotrophic lateral sclerosis (ALS). This study analyzes the nuclear lipidome in motor neurons in ALS and examines molecular pathways linked to the major lipid alterations. Nuclei were obtained from the frozen anterior horn of the lumbar spinal cord of ALS patients and age-matched controls. Lipidomic profiles of this subcellular fraction were obtained using liquid chromatography and mass spectrometry. We validated the mechanisms behind presumable lipidomic changes by exploring ALS surrogate models including human mo

Comprehensive-targeted lipidomic analysis in Niemann-Pick C disease.

Niemann-Pick C disease (NPC) is a lysosomal disease caused by mutations in NPC1 or NPC2 genes responsible for intracellular accumulation of free cholesterol and glycosphingolipids in a variety of tissues. We collected plasma samples from 15 NPC1 patients and 15 age-matched controls to analyze the impairment of lipid metabolism. Comprehensive-targeted quantitative lipidomic analysis was per-formed by Ion Mobility Mass Spectrometry, while oxysterols and lyso-sphingolipids, the classical NPC biomarkers, were analyzed by LC-MS/MS. Lipidomic analysis allowed the quantitation of ~1100 lipid species,

Lipidomic approaches to dissect dysregulated lipid metabolism in kidney disease.

Dyslipidaemia is a hallmark of chronic kidney disease (CKD). The severity of dyslipidaemia not only correlates with CKD stage but is also associated with CKD-associated cardiovascular disease and mortality. Understanding how lipids are dysregulated in CKD is, however, challenging owing to the incredible diversity of lipid structures. CKD-associated dyslipidaemia occurs as a consequence of complex interactions between genetic, environmental and kidney-specific factors, which to understand, requires an appreciation of perturbations in the underlying network of genes, proteins and lipids. Modern

Lipidome association with vascular disease and inflammation in HIV+ Ugandan children.

OBJECTIVE: HIV infection and antiretroviral therapy (ART) have both been linked to dyslipidemia and increased cardiovascular disease (CVD). The relationships among the lipidome, immune activation, and subclinical vascular disease in children with perinatally acquired HIV (PHIV) have not been investigated. METHODS: Serum lipid composition, including 13 lipid classes constituting 850 different lipid species were measured by direct infusion-tandem mass spectrometry in samples from 20 ART-treated PHIV and 20 age-matched and sex-matched HIV- Ugandan children. All participants were between 10 and 1

Lipidome-wide characterization of phosphatidylinositols and phosphatidylglycerols on CC location level.

Phosphatidylglycerol (PG) and phosphatidylinositol (PI) are two essential classes of glycerophospholipids (GPs), playing versatile roles such as signalling messengers and lipid-protein interaction ligands in cell. Although a majority of PG and PI molecular species contain unsaturated fatty acyl chain(s), conventional tandem mass spectrometry (MS/MS) methods cannot discern isomers different in carbon-carbon double bond (CC) locations. In this work, we paired phosphate methylation with acetone Paternò-Büchi (PB) reaction, aiming to provide a solution for sensitive and structurally informative an

A detailed lipidomic study of human pathogenic fungi Candida auris.

The present study is an attempt to determine the lipid composition of Candida auris and to highlight if the changes in lipids can be correlated to high drug resistance encountered in C. auris. For this, the comparative lipidomics landscape between drug-susceptible (CBS10913T) and a resistant hospital isolate (NCCPF_470033) of C. auris was determined by employing high throughput mass spectrometry. All major groups of phosphoglycerides (PGL), sphingolipids, sterols, diacylglycerols (DAG) and triacylglycerols (TAG), were quantitated along with their molecular lipid species. Our analyses highlight

Lipoxygenase drives lipidomic and metabolic reprogramming in ischemic heart failure.

BACKGROUND: After myocardial infarction (MI), delayed progression or reversal of cardiac remodeling is a prime target to limit advanced chronic heart failure (HF). However, the temporal kinetics of lipidomic and systemic metabolic signaling is unclear in HF. There is no consensus on metabolic and lipidomic signatures that influence structure, function, and survival in HF. Here we use genetic knock out model to delineate lipidomic, and metabolic changes to describe the role of lipoxygenase in advancing ischemic HF driven by leukocyte activation with signs of non-resolving inflammation. Dysregulation of soluble epoxide hydrolase and lipidomic profiles in anorexia nervosa.

Individuals with anorexia nervosa (AN) restrict eating and become emaciated. They tend to have an aversion to foods rich in fat. Because epoxide hydrolase 2 (EPHX2) was identified as a novel AN susceptibility gene, and because its protein product, soluble epoxide hydrolase (sEH), converts bioactive epoxides of polyunsaturated fatty acid (PUFA) to the corresponding diols, lipidomic and metabolomic targets of EPHX2 were assessed to evaluate the biological functions of EPHX2 and their role in AN. Epoxide substrates of sEH and associated oxylipins were measured in ill AN, recovered AN and gender-

Serum Lipidomics Profiling Identify Novel Biomarkers of Distal Symmetrical Polyneuropathy in Type 1 Diabetes.

BACKGROUND: Distal symmetrical polyneuropathy (DSPN) is a common complication of type 1 diabetes (T1D), yet validated biomarkers for early detection or prognosis are lacking. Metabolic disturbances driven by chronic hyperglycemia and dyslipidemia contribute to DSPN development and progression. METHODS: We used untargeted serum liquid chromatography-mass spectrometry lipidomics to identify DSPN biomarkers. The discovery cohort included 153 individuals with T1D (109 with and 44 without DSPN) and 50 non-diabetic controls. The independent validation cohort included 99 individuals with T1D with es

Spatial lipidomics of the human brain: systematic review of current state and future perspectives.

BACKGROUND: Lipids represent a significant component of the human brain, exerting crucial functions in both physiological and pathological conditions. Mapping brain lipids distribution is an emerging area of research, with mass spectrometry imaging allowing the detection of lipid species and their localization within tissue sections. However, comprehensive spatial mapping of lipids in the human brain remains to be achieved. This systematic review addresses this gap by critically synthesizing the available literature in the field. METHODS: A bibliographic search on PubMed, Scopus and Web of Sc

Integrated metabolomics and lipidomics reveal fatty acid dysregulation in aortic dissection independent of hypertension.

Aortic dissection (AD), characterized by rupture of the aortic wall, presents significant diagnostic challenges due to its rapid onset and nonspecific clinical symptoms. Although conventional imaging techniques are clinically useful, they typically lack sensitivity for early AD detection, are often unavailable in resource-limited settings, and cannot elucidate the mechanistic basis of AD pathogenesis. In this study, we used metabolomics and lipidomics to analyze serum samples from healthy controls, hypertensive AD patients, and normotensive AD patients, revealing distinct metabolic and lipidom

Lipidomics-Guided Ultrasound-Assisted Ethanol Extraction of Avocado Pulp and Evaluation of Bioactive Properties.

Avocados are a valuable source of lipids with potential applications in food, nutraceutical, and cosmetic products, all requiring food-grade and environmentally friendly extraction methods. Though often enjoyed fresh, avocado pulp is also processed for oil. However, conventional extraction techniques can compromise pulp lipid composition, triggering the need for alternative methods that preserve pulp lipid integrity and meet industrial sustainability standards. Ultrasound probe-assisted extraction using ethanol, a green solvent, could be a sustainable alternative to extract lipids from avocado
