Cuproptosis in Cerebrovascular Diseases: Mechanisms and Advances in Targeted Therapeutic Strategies.

Cuproptosis is a recently identified form of regulated cell death. It is primarily driven by abnormal mitochondrial copper accumulation. Ferredoxin 1-dependent copper reduction promotes the pathological aggregation of lipoylated proteins involved in the tricarboxylic acid cycle and the loss of iron-sulfur cluster proteins. This process induces proteotoxic stress, ultimately causing the collapse of mitochondrial energy metabolism. Evidence from transcriptomic analyses, single-cell multi-omics studies, and in vivo and in vitro models suggests that copper dyshomeostasis may be involved in cerebro

IRF8 Suppresses Hepatocellular Carcinoma Progression Through LIAS-Dependent IFN Production and Cuproptosis Activation.

BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most lethal malignancies worldwide. Cuproptosis is a novel form of regulated cell death closely linked to tumor progression. Although interferon regulatory factors (IRFs) exert tumor-suppressive effects in various cancers, its precise regulatory mechanisms concerning cuproptosis in HCC remain largely elusive. METHODS: IRF8 expression was evaluated in clinical HCC specimens and cell lines. Gain- and loss-of-function assays were conducted. Underlying transcriptional mechanisms were elucidated using ChIP-qPCR and dual-luciferase reporter a

Elesclomol-copper combination synergistically targets mitochondrial metabolism in cancer stem cells to overcome chemoresistance in PDAC.

Pancreatic ductal adenocarcinoma (PDAC) is an aggressive cancer with a poor prognosis, partly due to cancer stem cells (CSCs) that drive progression and treatment resistance. We explored the therapeutic potential of inducing cuproptosis, a copper-dependent, regulated cell death, in CSC-enriched PDAC models. Using human and murine PDAC models, we evaluated elesclomol, a copper transport enhancer. Elesclomol alone had minimal effects; combined with copper chloride (CuCl2), it significantly and irreversibly reduced CSC phenotypes without affecting non-transformed cells. Mechanistically, only the

Harnessing cuproptosis for pancreatic cancer therapy: From molecular insights to clinical prospects.

Pancreatic cancer (PC) remains a high-fatality malignancy with limited clinical progress, characterized by aggressive biology, marked resistance to standard therapies, and dismal outcomes. Even with state-of-the-art resection, radiotherapy, and multidrug chemotherapy, median survival benefits are modest, highlighting an urgent need for mechanism-based interventions. Cuproptosis, a newly delineated modality of regulated cell death initiated by intracellular copper accumulation and mitochondrial stress, presents a biologically coherent therapeutic avenue. Distinct from apoptosis, necroptosis, an

Hepatocyte cuproptosis promotes the progression of hepatic fibrosis: Emerging a potential therapeutic target.

Copper (Cu)-dependent aggregation of lipoylated proteins in mitochondria triggers cuproptosis, a newly discovered modality of regulated cell death marked by impaired mitochondrial respiration. During the progression of hepatic fibrosis (HF), hepatocytes, responsible for Cu storage and metabolism, exhibit vigorous mitochondrial activity and are supposed to be cuproptosis-susceptible. However, the cellular landscape of cuproptosis in fibrotic livers remains unclear, and the therapeutic potential of cuproptosis suppression in resolving HF has yet to be explored. In this study, single-cell RNA seq

Identification and analysis of the major M2 autoantigens in primary biliary cirrhosis.

Primary biliary cirrhosis (PBC) is a chronic cholestatic liver disease characterized by the presence of antimitochondrial antibodies in the serum. It is possible that the PBC-specific immunoreactive trypsin-sensitive antigens on the inner mitochondrial membrane, termed M2, are important in the pathogenesis of this autoimmune disease. We have previously shown that a major M2"a" antigen is the E2 component of the pyruvate dehydrogenase multienzyme complex located within mitochondria. Analysis of the primary structure of the E2 components of all three 2-oxo acid dehydrogenase complexes reveals a

Ferroptosis, cuproptosis, and metal ion-modulated cytotoxicity in osteosarcoma: regulatory mechanisms and emerging therapeutic strategies.

Osteosarcoma is characterized by early metastatic potential, frequent postoperative recurrence, and resistance to conventional chemotherapy, and therapeutic progress remains limited. Osteosarcoma may have exploitable vulnerabilities due to metal homeostasis and redox control dysregulation. However, metal-associated cytotoxic processes do not constitute a single class of regulated cell death. This review examines ferroptosis and cuproptosis as mechanistically defined metal-dependent regulated cell-death modalities and distinguishes them from broader metal ion-modulated cytotoxic processes. Electric-Field-Driven Ferredoxin 1-Independent Cuproptosis Induction Overcomes Therapy-Induced Resistance in Glioblastoma.

Cuproptosis presents a potential therapeutic avenue for glioblastoma (GBM), yet its efficacy is severely limited by intrinsic and adaptive resistance mechanisms. Here, we identify a critical therapy-induced barrier where standard-of-care interventions, including Temozolomide, radiotherapy, and Tumor Electric-Field Therapy (TEFT), consistently induce a profound downregulation of essential cuproptosis-execution genes such as Ferredoxin 1 (FDX1) and Dihydrolipoamide S-Acetyltransferase (DLAT). This transcriptomic remodeling reveals a universal mechanism of acquired cuproptosis resistance in recur

A TIGIT nanotrapping-guided STING-activatable immunometabolic strategy overcomes innate immune silence and T cell exhaustion in breast cancer.

Breast cancer exhibits a profoundly immunosuppressive tumor microenvironment (TME), where innate immune silence prevents antigen sensing and persistent T cell exhaustion limits effector responses, rendering most immunotherapies ineffective. Clinical profiling of 1093 The Cancer Genome Atlas (TCGA) cases identified a glucose-fueled glutathione (GSH)-glutathione peroxidase 4 (GPX4)-dihydrolipoamide S-acetyltransferase (DLAT) axis as a dominant metabolic shield that suppresses oxidative stress, and thereby enforces both stimulator of interferon genes (STING) silence and CD8+ T cell exclusion. Cuproptosis in age-related orthopedic diseases: mechanisms and therapeutic prospects.

Cuproptosis is an emerging copper-dependent and mitochondria-associated form of regulated cell death (RCD) that links intracellular labile copper accumulation to metabolic vulnerability. Mechanistically, cuproptosis is triggered by intracellular copper accumulation, leading to aggregation of lipoylated tricarboxylic acid (TCA) cycle proteins, destabilization of iron-sulfur cluster proteins, and proteotoxic stress-induced bioenergetic collapse. Aging-associated orthopedic disorders-including osteoarthritis (OA), osteoporosis (OP) and intervertebral disc degeneration (IVDD)-represent a rapidly e

Copper depletion ameliorates neuronal damage after intracerebral hemorrhage.

BACKGROUND: Intracerebral hemorrhage (ICH) is a severe subtype of stroke. There are currently no specific treatment strategies for secondary brain injury and neurological deficits following ICH. Copper (Cu) is an essential cofactor for all living organisms. Cytotoxicity can occur when copper ion concentration exceeds the homeostatic threshold, leading to cell death. However, the relationship between copper and ICH is unclear. METHODS: In vivo, an ICH model was established in male Sprague-Dawley rats by stereotactically injecting autologous blood into the right basal ganglia. In vitro, we empl

A metallic metabolic nano-regulator reprograms the PKM2/HIF-1α/DLAT axis to amplify tumor-specific cuproptosis.

Cuproptosis represents a novel form of mitochondria-dependent cell death, demonstrating a unique antitumor potential. However, tumor cells have evolved robust metabolic compensation mechanisms that mitigate the cytotoxic effects of copper ions; this barrier must be disrupted to trigger cuproptosis. In this study, a metallic metabolic nano-regulator (MC@BSA; MC, manganese-copper nanocomposite; BSA, bovine serum albumin) is designed, which efficiently accumulates in tumor tissues and disrupts copper homeostasis by inhibiting copper efflux and promoting the generation of reactive oxygen species (

Copper-Oxamate Coordination Polymers Reverse Cuproptosis Resistance in Nonsmall Cell Lung Cancer via Glycolysis Reprogramming.

Cuproptosis, a recently identified regulated cell death pathway, is emerging as a promising therapeutic target for cancer. However, nonsmall cell lung cancer (NSCLC) exhibits inherent resistance to copper-induced toxicity, which is primarily due to enhanced glycolytic activity. Herein, a coordination polymer (Cu-Ox@HA) is designed by the chelation of copper ions with the glycolysis inhibitor oxamate (Ox), using hyaluronic acid (HA) as a biocompatible template. In the tumor microenvironment, Cu-Ox@HA disassembles in response to glutathione (GSH), enabling the synchronous release of copper ions

Biomimetic Copper Nanozyme Reprograms Cold Tumor via Cuproptosis-Pyroptosis Crosstalk for Potent Renal Carcinoma Immunotherapy.

Immune checkpoint blockade (ICB) therapy is an emerging strategy for renal cell carcinoma (RCC). However, its clinical efficacy remains constrained by its inherently poor immunogenicity and insufficient cytotoxic T lymphocyte (CTL) infiltration. Herein, we engineer a biomimetic copper nanozyme (Cu2O-OMV) by integrating Cu2O nanoparticles with bacterial outer-membrane vesicles (OMVs) to activate the antitumor immune response and synergize with ICB therapy. The Cu2O-OMV nanozyme exhibits peroxidase (POD)-like catalytic activity and releases Cu+ to exert Fenton-like activity, generating cytotoxic

Overexpression of MCL1 attenuates irritable bowel syndrome by regulating cuproptosis: Screening and validation.

Irritable bowel syndrome (IBS) is a type of chronic bowel disorder with a poorly understood pathophysiology. Recently, the imbalance of copper has been reported to influence the progression of IBS, suggesting cuproptosis, a new type of copper-induced cell death, may play a role in IBS. This study found 17 cuproptosis-related differentially expressed genes in IBS through bioinformatic analysis. Six hub genes were identified after the protein-protein interaction network analysis, namely myeloid cell leukemia 1 (MCL1), epidermal growth factor receptor 2, cadherin-associated protein beta 1, solute

Structures of Ostα/β reveal a unique fold and bile acid transport mechanism.

Bile acid and steroid hormone homeostasis are critical for human health, with disruptions linked to metabolic and endocrine disorders1,2. The organic solute transporter Ostα/β, essential for bile acid efflux in enterohepatic circulation3, has long defied mechanistic elucidation. Here we present cryogenic electron microscopy structures of human Ostα/β in apo and substrate-bound states at 2.6-3.1 Å resolution, revealing a distinctive membrane protein architecture that defines a new transporter class. Ostα/β forms a symmetric tetramer of heterodimers, with each Ostα subunit showing a new seven-tr

COMMD1 Induces Copper Deficiency of SOD1 by Inhibiting the Palmitoylation of CCS in ALS.

Mutations in superoxide dismutase 1 (SOD1) compromise its metal-binding capacity, resulting in protein misfolding and aggregation, which ultimately induces cellular apoptosis in amyotrophic lateral sclerosis (ALS). Copper metabolism domain containing 1 (COMMD1), a gene implicated in copper homeostasis, has not been thoroughly characterized in the context of ALS pathogenesis. In this study, we identified elevated COMMD1 expression in ALS, potentially contributing to diminished copper incorporation into SOD1. Knockdown of COMMD1 enhanced palmitoylation of the copper chaperone for SOD1 (CCS), fac

ZDHHC9 and spermine metabolism: a palmitoylation-driven pathway to prostate carcinogenesis.

BACKGROUND: Spermine is a natural metabolite that has been shown to hinder the progression of prostate cancer. Protein palmitoylation, a recently identified post-translational lipid modification, is catalyzed by a family of 23 enzymes known as zinc finger DHHC-type palmitoyltransferases (ZDHHCs). This study primarily investigates the role of ZDHHC9 in spermine metabolism and its potential association with prostate cancer. METHODS: We employed immunohistochemistry, Western blotting, and RT-qPCR techniques to assess ZDHHC9 expression in prostate cancer tissues and cell lines. The connection betw
