Bone is one of the most adaptable tissues in the body as it is continuously subjected to load bearing. In fact, mechanical loading is an important regulator of bone mass. The skeleton adjusts to load by changing its mass, shape and microarchitecture, depending on the magnitude of the strain. Mechanical stimulation is necessary for the development of the skeleton, whereas in adults physiological levels of strain help maintain bone mass by reducing bone resorption. On the other hand, an excessive level of strain or bone disuse induces bone loss. Osteocytes are long-lived cells comprising more th

Chronic inflammatory diseases of the spine, typified by axial spondyloarthritis (axSpA) and intervertebral disc degeneration (IDD), impose a substantial burden through refractory pain and irreversible structural remodelling (pathological ossification or fibrosis). Although current anti-inflammatory therapies can alleviate symptoms, their capacity to arrest structural deterioration remains limited, underscoring the constraints of approaches that target inflammatory cytokines in isolation. This therapeutic intractability is rooted in the distinctive spinal microenvironment: high mechanical loadi

Recently, it has been recognized that physical abnormalities (e.g. elevated solid stress, elevated interstitial fluid pressure, increased stiffness) are associated with tumor progression and development. Additionally, these mechanical forces originating from tumor cell environment through mechanotransduction pathways can affect metabolism. On the other hand, mitochondria are well-known as bioenergetic, biosynthetic, and signaling organelles crucial for sensing stress and facilitating cellular adaptation to the environment and physical stimuli. Disruptions in mitochondrial dynamics and function

Mechanotransduction is pivotal in the maintenance of homeostasis in different tissues and involves multiple cell signaling pathways. In bone, mechanical stimuli regulate the balance between bone formation and resorption; osteocytes play a central role in this regulation. Dysfunctions in mechanotransduction signaling or in osteocytes response lead to an imbalance in bone homeostasis. This alteration is very relevant in some conditions such as osteoporosis and aging. Both are characterized by increased bone weakness due to different causes, for example, the increase of osteocyte apoptosis that c

BACKGROUND: Mutations in PIEZO1 (Piezo type mechanosensitive ion channel component 1) cause human lymphatic malformations. We have previously uncovered an ORAI1 (ORAI calcium release-activated calcium modulator 1)-mediated mechanotransduction pathway that triggers lymphatic sprouting through Notch downregulation in response to fluid flow. However, the identity of its upstream mechanosensor remains unknown. This study aimed to identify and characterize the molecular sensor that translates the flow-mediated external signal to the Orai1-regulated lymphatic expansion.
METHODS: Various mutant mous

Nuclear Envelope Membrane Protein 1 (NEMP1) is crucial for metazoan fertility; loss of Nemp1 causes death of primordial oocytes that reside in the mechanically challenging ovarian cortex. Here, we show that softening the ovary rescues oocyte loss and restores fertility in Nemp1 knockout (KO) mice. In cell culture, NEMP1 depletion on stiff substrates leads to death, while cells remain viable on soft substrates. We further show that NEMP1 regulates YAP nuclear translocation, essential for mechanotransduction. Mechanistically, Nemp1-depleted cells on stiff substrates or subjected to stretching ex

PURPOSE OF REVIEW: Osteocytes are the most abundant cells in bone and serve as central skeletal regulators. Recent findings from our and other groups demonstrate that beyond soluble mediators, osteocytes communicate by releasing extracellular vesicles (EVs). This review provides a systematic overview of osteocyte-derived EVs and highlights their importance in diagnostic and therapeutic innovation.
RECENT FINDINGS: Accumulating evidence has linked osteocyte mechanotransduction to EV biogenesis and secretion. Mechanically induced calcium oscillations and signaling pathways impact load-induced E

Tendons are specialized connective tissues that transmit mechanical forces from muscle to bone, ensuring joint stability and efficient locomotion. Their homeostasis and regenerative capacity depend on the interplay between extracellular matrix (ECM), resident and recruited cell populations, and immune-mediated signaling. This review provides an overview of tendon structure and composition, emphasizing the collagen-based organization and the functional role of non-collagenous matrix components in mechanotransduction and cell signaling. The heterogeneity of tendon-resident cells, including tenoc

Mechanical cues shape bone regeneration, but treatments for delayed union, nonunion, and mechanically mismatched repair often still treat them as static constraints. In this review, we use mechanical intelligence to describe the time-dependent capacity of biomaterials, cells, and therapeutic devices to store, transform, and transmit mechanical history during repair. Two forms of memory are central to this view. Scaffolds and implants can preserve or release previous mechanical states through relaxation, residual stress, shape recovery, evolving stiffness, and architecture, whereas cells can ca

Osteoarthritis (OA) is a complex disease associated with genetic, biological, and mechanical risk factors that act, in part, to alter chondrocyte homeostasis. Our recent exome sequencing studies identified a damaging genetic variant in COL6A3, a monomeric unit of collagen type VI and a distinguishing component of the pericellular matrix (PCM) of articular cartilage, a transducer of mechanical and biochemical signals for the chondrocyte. To study the effect of this genetic variant, human induced pluripotent stem cell (hiPSC)-derived chondrocytes, genetically edited to harbor the COL6A3 mutation

Pregnancy is a highly coordinated process that relies on the precise regulation of the extracellular matrix, cells, and various molecules at the maternal-fetal interface. As the fetus grows and amniotic fluid increases, the uterus undergoes adaptive remodeling, a process in which biomechanics plays an indispensable role. This review aims to systematically explore the biomechanical characteristics of the uterus and specifically elucidate the crucial regulatory roles of mechanical signals such as stiffness, shear stress, tension, and compression force in both physiological and pathological pregn

AIM: The glomerulus is a specialized microvascular unit that filters plasma through the coordinated function of podocytes and parietal epithelial cells (PECs). From this perspective, the glomerulus functions like a living hydrogeological filtration system. This review aims to integrate mechanobiology and hydrogeology, reframing podocytes and PECs as active regulators in a pressure-driven network, with Piezo1 central to glomerular homeostasis, adaptation, and pathology.
METHODS: This review integrates existing literature on glomerular biology, mechanosensitive signaling, and epithelial cell fu

TMC1 and TMC2 are mechanosensory ion channels of the vertebrate inner ear that mediate hearing and balance. How these channels open in response to mechanical force remains unresolved. Through comparative analyses of TMCs across eukaryote species, we find that TMC1 and TMC2 arose in vertebrates by gene duplication and evolved elaborate extracellular loops. Structural models demonstrate that the loop between transmembrane domains 1 and 2 arches over the channel pore and lies near TMIE, an auxiliary protein essential for function. In mammals, this loop shows signatures of positive selection and c

Interactions between glial cells and the central nervous system (CNS) extracellular matrix (ECM) maintain neural homeostasis and plasticity. In contrast to traditional reviews focusing on biochemistry, this work summarizes the structural and mechanical properties of major ECM subtypes, and illustrates how mechanical and biochemical signals jointly regulate glial activity via key mechanosensitive pathways. Glia in turn dynamically remodel ECM, forming a reciprocal regulatory network. This paper also analyzes characteristic mechanical disorders in Alzheimer's disease, multiple sclerosis and trau

Mechanical forces influence a range of cellular behaviors; however, how these forces are sensed and converted into biochemical changes remains incompletely understood. A key aspect of mechanotransduction is the regulation of subcellular protein localization. Here, we present a protocol describing the engineering of cell lines with tunable actomyosin contractility combined with a proximity biotinylation strategy confined to the nucleus followed by liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis. This approach allows the identification of proteins whose nuclear localization is

Bone is a highly mechanosensitive tissue, where mechanical signaling plays a central role in maintaining skeletal homeostasis. Mechanotransduction regulates the balance between bone formation and resorption through coordinated interactions among bone cells. Key mechanosensing structures-including the extracellular/pericellular matrix (ECM/PCM), integrins, ion channels, connexins, and primary cilia, translate mechanical cues into biochemical signals that drive bone adaptation. Disruptions in mechanotransduction are increasingly recognized as an important factor in osteoporosis. Under pathologic

Osteoarthritis (OA) is a degenerative condition of bone characterized by loss of cartilage, subchondral bone sclerosis, osteophyte growth, synovial inflammation, and influenced by biomechanical loading. The loss of articular cartilage is the primary concern of OA, while subchondral bone plays a crucial role in the integrity of articular cartilage, providing mechanical support and nutrition supply and constantly adapting to the changing biomechanical environment in weight-bearing joints. Osteocytes, abundant in subchondral bones, sense mechanical loading and control bone adaptive remodeling. Th

The sense of touch is crucial for cognitive, emotional, and social development and relies on mechanically activated (MA) ion channels that transduce force into an electrical signal. Despite advances in the molecular characterization of these channels, the physiological factors that control their activity are poorly understood. Here, we used behavioral assays, electrophysiological recordings, and various mouse strains (males and females analyzed separately) to investigate the role of the calmodulin-like Ca2+ sensor, caldendrin, as a key regulator of MA channels and their roles in touch sensatio

It is well understood that the balance of bone formation and resorption is dependent on both mechanical and biochemical factors. In addition to cell-secreted cytokines and growth factors, sex hormones like estrogen are critical to maintaining bone health. Although the direct osteoprotective function of estrogen and estrogen receptors (ERs) has been reported extensively, evidence that estrogen signaling also has a role in mediating the effects of mechanical loading on maintenance of bone mass and healing of bone injuries has more recently emerged. Recent studies have underscored the role of est
