Disease associated with Nipah virus infection causes a devastating and often fatal spectrum of syndromes predominated by both respiratory and neurologic conditions. Additionally, neurologic sequelae may manifest months to years later after virus exposure or apparent recovery. In the two decades since this disease emerged, much work has been completed in an attempt to understand the pathogenesis and facilitate development of medical countermeasures. Here we provide detailed organ system-specific pathologic findings following exposure of four African green monkeys to 2.41×105 pfu of the Malaysia

The continuing outbreaks of ebola virus disease highlight the ongoing threat posed by filoviruses. Fortunately, licensed vaccines and therapeutics are now available for Zaire ebolavirus. However, effective medical countermeasures, such as vaccines for other filoviruses such as Sudan ebolavirus and the Marburg virus, are presently in early stages of development and, in the absence of a large outbreak, would require regulatory approval via the U.S. Food and Drug Administration (FDA) Animal Rule. The selection of an appropriate animal model and virus challenge isolates for nonclinical studies are

Smallpox is a human disease caused by infection with variola virus, a member of the genus Orthopoxvirus. Although smallpox has been eradicated, concern that it might be reintroduced through bioterrorism has therefore led to intensive efforts to develop new vaccines and antiviral drugs against this disease. Because these vaccines and therapeutics cannot be tested in human trials, it is necessary to test such medical countermeasures in different animal models. Although several orthopoxviruses cause disease in laboratory animals, only rabbitpox virus (RPXV) infection of rabbits shows patterns of

Marburgviruses cause severe hemorrhagic disease in humans, yet characterization of rodent models for pathogenesis and medical countermeasure development remains limited. Here, we investigated clinical course, blood chemistry, viral dissemination, and mucosal viral detection in inbred BALB/c and outbred CD-1 mice infected with mouse-adapted (MA)-Marburg virus (MARV)/Angola or MA-Ravn virus (RAVV) to further mouse model development. Regardless of virus or mouse strain, infection resulted in widespread viral dissemination in animals sampled up to 5 days post-infection. However, in animals followe

Mpox, formerly known as monkeypox, has shifted from a regional concern to a global health priority. Progress toward targeted antivirals and vaccines, however, still hinges on a complex array of animal models developed over the last sixty years. This review distills all literature regarding Mpox animal models - from rodent to non-human primate species - into a clear, comparative narrative. We outline how host species, viral clade, and inoculation route shape clinical course and immune responses, and we identify which models best address questions of pathogenesis, transmission, and medical count

AEOL-10150 is a broad-spectrum metalloporphyrin superoxidase dismutase (SOD) mimic specifically designed to neutralize reactive oxygen and nitrogen species. Research has shown that AEOL-10150 is a potent medical countermeasure against national security threats including sulfur mustard (SM), nerve agent exposure and radiation pneumonitis following a radiological/nuclear incident sufficient to cause acute radiation syndrome (ARS). AEOL-10150 performed well in animal safety studies, and two completed phase 1 safety studies in patients demonstrated that the drug was safe and well tolerated, indica

Long work hours, overnight call duty, and rotating shifts are implicit features of hospital medical practice. Rigorous schedules have been deemed necessary to fulfill the professional obligation of patient beneficence, to optimize trainee learning, and to respond to economic realities. However, the resultant disruption and restriction of physicians' sleep produce demonstrable neurobehavioral impairments that may threaten other fundamental professional mandates, such as that of primum non nocere ("first, do no harm"). This article provides a basic overview of sleep/wake regulatory processes, ex

Francisella tularensis, the causative agent of tularemia, is capable of causing disease in a multitude of mammals and remains a formidable human pathogen due to a high morbidity, low infectious dose, lack of a FDA approved vaccine, and ease of aerosolization. For these reasons, there is concern over the use of F. tularensis as a biological weapon, and, therefore, it has been classified as a Tier 1 select agent. Fluoroquinolones and aminoglycosides often serve as the first line of defense for treatment of tularemia. However, high levels of resistance to these antibiotics has been observed in gr

This report provides CDC recommendations to U.S. health care providers and preparedness personnel regarding treatment and postexposure prophylaxis (PEP) of tularemia, an uncommon but potentially serious disease caused by the gram-negative coccobacillus, Francisella tularensis. Tularemia occurs naturally in the United States and other Northern Hemisphere regions. Because F. tularensis has a low infectious inoculum, it is classified as a potential bioterrorism agent that could infect thousands of persons if intentionally released, requiring rapid, informed decision-making by public health agenci

Autonomic neural functions are important to regulate vital functions in the living body. There are different methods to evaluate indirectly and directly autonomic, sympathetic and parasympathetic, neural functions of human body. Among various methods, microneurography is a technique to evaluate directly sympathetic neural functions in humans. Using this technique sympathetic neural traffic leading to skeletal muscles (muscle sympathetic nerve activity; MSNA) can be recorded from human peripheral nerves in situ. MSNA plays essentially important roles to maintain blood pressure homeostasis again

Gastrointestinal acute radiation syndrome (GI-ARS) poses a critical public health concern, necessitating the development of effective medical countermeasures (MCM). Here, we evaluated the therapeutic potential of a commercially available probiotic formulation using a human gut-on-a-chip model that mimics the physiodynamic intestinal microenvironment. Intestinal epithelial Caco-2 cells were subjected to 8 Gy gamma radiation, targeting the epithelial layer, the culture medium, or both. The irradiated epithelial cells challenged to the irradiated medium resulted in significant DNA damage quantifi

Emerging and re-emerging viruses represent a major global health challenge. To address this, the WHO encourages the development of broad-spectrum medical countermeasures. This study evaluated the efficacy of repurposed broad-spectrum molecules against Bunyaviricetes viruses and more specifically Rift Valley fever virus (RVFV). In vitro, favipiravir and nitazoxanide showed low effective doses with an additive effect in combination against several viruses, including RVFV, Crimean-Congo hemorrhagic fever virus and Hantaan virus. In a murine model of severe RVFV infection, combination treatment re

Q fever, caused by Coxiella burnetii, leads to debilitating human infections that are often recalcitrant to antibiotics. The development of new medical countermeasures is critical, as the only available vaccine has significant limitations that prevent its widespread or rapid use. To address this, we characterized aerosol C. burnetii infection in 16 cynomolgus macaques (Macaca fascicularis), providing the first detailed pathological description of Q fever pneumonia in a nonhuman primate model. Following exposure, all animals developed clinical signs consistent with human Q fever, including sust

Vesicants, powerful chemical weapons of mass destruction, are agents that cause blistering of the skin and blindness upon accidental exposure or during warfare. In addition, their exposure induces a wide range of other symptoms, including those affecting the respiratory tract, digestive system, skin, and ocular tissues. The ocular tissue can be exposed through both direct and indirect routes, such as direct contact with the affected corneal tissue and systemic blood circulation. Investigating eye injuries caused by vesicants is a critical and growing field of research in ophthalmology and ocul

Topical nonsteroidal anti-inflammatory drugs (NSAIDs) are commonly used to treat sports injuries, but evidence-based medical guidance for their standardized and rational use is lacking. This guideline working group identified clinically important issues, obtained the full opinions of patients and clinical staff, and discussed them with the expert group. Based on evidence from the literature, the "clinical practice guidelines for topical NSAIDs in the treatment of sports injuries" were formulated following the methods and principles of international guidelines. According to these guidelines, 7

Poxviruses comprise pathogens that are highly pathogenic to humans and animals, causing diseases such as smallpox and mpox (formerly monkeypox). The family also contains members developed as vaccine vectors and oncolytic agents to fight other diseases. Vaccinia virus is the prototype poxvirus and the vaccine used to eradicate smallpox. Poxvirus genes follow a cascade temporal expression pattern, categorized into early, intermediate, and late stages using distinct transcription factors. This review comprehensively summarized the temporal expression classification of over 200 vaccinia virus gene

Marburg virus disease (MVD) is a severe infectious disease caused by the Marburg virus (MARV), posing a significant threat to humans. MARV needs to be operated under strict biosafety Level 4 (BSL-4) laboratory conditions. Therefore, accessible and practical animal models are urgently needed to advance prophylactic and therapeutic strategies for MARV. In this study, we constructed a recombinant vesicular stomatitis virus (VSV) expressing the Marburg virus glycoprotein (VSV-MARV/GP). Syrian hamsters infected with VSV-MARV/GP presented symptoms such as thrombocytopenia, lymphopenia, haemophilia,

Bioluminescent images of viral replication in live animals (in vivo) reveal disease dynamics and effects of medical countermeasures over time. After selecting an appropriate orthopoxvirus animal model for the study, a recombinant virus with the firefly luciferase gene inserted in the genome is used to infect the animals. On the day of bioluminescent imaging, the substrate, D-luciferin, is prepared; animals are sedated and injected with the substrate and IVIS imager is utilized; various bioluminescent images are acquired; then animals recover and are able to continue in the study. Ex vivo imagi
