The mechanisms of memory traces storage and interaction in snails: Role of DNA methylation and protein synthesis.

Understanding the molecular mechanisms underlying the storage and interaction of different memory traces remains an important, yet underexplored, topic in neurobiology. We addressed this using grape snails trained to reject two food-conditioned stimuli (CS). Our results indicate that memory storage mechanisms for distinct CS are independent, as selective impairment of memory reconsolidation with an NMDA receptor antagonist affected only one CS. When both CS reconsolidations were simultaneously disrupted, the snails developed amnesia, which manifested as anterograde amnesia on late amnesia stag

Acoustic enhancement of slow wave sleep improves memory retention by coordinating brain oscillatory rhythms and autonomic activity.

The interplay between sleep slow oscillations (SOs), spindles, and autonomic activity is thought to be crucial for sleep-dependent memory consolidation, yet the precise mechanisms remain unclear. In this randomized, single-blind, crossover study of 20 healthy young adults (25 ± 4 years, 75% female), we investigated whether one night of closed-loop acoustic stimulation of sleep slow waves during non-rapid eye movement sleep stages 2 and 3 enhanced memory retention at a paired word-associates task by modulating these neural and autonomic processes, compared to a sham condition. We found that mem…

Recent memory for socially transmitted food preferences in rats does not depend on the hippocampus.

The standard model of systems consolidation holds that the hippocampus (HPC) is involved only in the initial storage and retrieval of a memory. With time hippocampal-neocortical interactions slowly strengthen the neocortical memory, ultimately enabling retrieval of the memory without the HPC. Key support for this idea comes from experiments measuring memory recall in the socially-transmitted food preference (STFP) task in rats. HPC damage within a day or two of STFP learning can abolish recall, but similar damage five or more days after learning has no effect. We hypothesize that disruption of

Let's Shape Learning Into Lasting Memories.

Recent experiments in rats and humans have indicated that the effects of non-invasive electrical stimulation are primarily due to transcutaneous stimulation of peripheral nerves, specifically the greater occipital nerve. This stimulation pathway activates communication gateways from the periphery to the brain, impacting memory consolidation. In this invited commentary, I delve into and offer additional insights concerning the enhancement of episodic memory through transcutaneous electrical stimulation of the greater occipital nerve, building upon the findings published by my laboratory in both…

Motor skill learning and offline-changes in TGA patients with acute hippocampal CA1 lesions.

Learning and the formation of memory are reflected in various memory systems in the human brain such as the hippocampus based declarative memory system and the striatum-cortex based system involved in motor sequence learning. It is a matter of debate how both memory systems interact in humans during learning and consolidation and how this interaction is influenced by sleep. We studied the effect of an acute dysfunction of hippocampal CA1 neurons on the acquisition (on-line condition) and off-line changes of a motor skill in patients with a transient global amnesia (TGA). Sixteen patients (68 ±

Sleep enhances false memories depending on general memory performance.

Memory is subject to dynamic changes, sometimes giving rise to the formation of false memories due to biased processes of consolidation or retrieval. Sleep is known to benefit memory consolidation through an active reorganization of representations whereas acute sleep deprivation impairs retrieval functions. Here, we investigated whether sleep after learning and sleep deprivation at retrieval enhance the generation of false memories in a free recall test. According to the Deese, Roediger, McDermott (DRM) false memory paradigm, subjects learned lists of semantically associated words (e.g., "nig…

Circuit-informed modulation of traumatic memory in PTSD: integrating extinction, suppression, and reconsolidation.

Post-traumatic stress disorder (PTSD) persists when stress biases memory systems-broadening threat engrams, weakening context-appropriate inhibition, and rendering extinction fragile. Integrating preclinical and human evidence, we argue that these liabilities are tractable within a circuit-informed framework that aligns behavioral procedures with the biology of fear memory. Extinction can be stabilized by preferentially engaging hippocampal-prefrontal-amygdala pathways and their thalamic coordinators; targeted training and sleep-based reactivation strengthen top-down control over intrusions an

Sleep promotes illusory word compositions, a distinct form of false memory.

Extensive evidence supports the beneficial effects of sleep on memory and learning, including the consolidation and reorganization of memories and the extraction of regularities from encoded experiences. Nevertheless, some studies suggest that sleep may also increase false memories, potentially as a byproduct of regularities extraction. Physiologically, time-compressed memory replay in the hippocampus during non-rapid-eye-movement (nREM) sleep is believed to contribute to the consolidation process, although the functional significance of time compression remains elusive. Recently, we proposed

Unexpected awakenings in severe dementia from case reports to laboratory.

OBJECTIVE: Case report notions of unexpected memory retrieval in patients with severe dementia near to death are starting to alter the central "irreversible" paradigm of dementia and locate dementia as a problem of memory retrieval, not consolidation. We suggest that the most likely central tenet of this paradoxical memory retrieval is the fluctuation of neuromodulators projecting from the brain stem to the medial prefrontal cortex and the hippocampus. The neuromodulation-centric explanation of this phenomenon aims to open the "irreversible" paradigm of dementia up for discussion and suggest a

Stress and Memory: A Selective Review on Recent Developments in the Understanding of Stress Hormone Effects on Memory and Their Clinical Relevance.

Stress causes a neuroendocrine response cascade, leading to the release of catecholamines and glucocorticoids (GCs). GCs influence learning and memory by acting on mineralocorticoid (MR) and glucocorticoid (GR) receptors. Typically, GCs enhance the consolidation of memory processing at the same time as impairing the retrieval of memory of emotionally arousing experiences. The present selective review addresses four recent developments in this area. First, the role of the endocannabinoid system in mediating the rapid, nongenomic effects of GCs on memory is illustrated in rodents. Subsequently,

Targeted memory reactivation to augment treatment in post-traumatic stress disorder.

Post-traumatic stress disorder (PTSD) is a psychiatric disorder with traumatic memories at its core. Post-treatment sleep may offer a unique time window to increase therapeutic efficacy through consolidation of therapeutically modified traumatic memories. Targeted memory reactivation (TMR) enhances memory consolidation by presenting reminder cues (e.g., sounds associated with a memory) during sleep. Here, we applied TMR in PTSD patients to strengthen therapeutic memories during sleep after one treatment session with eye movement desensitization and reprocessing (EMDR). PTSD patients received e…

Intense training abolishes the two temporal windows of cycloheximide-induced amnesia in rats.

Several studies have identified two distinct time windows during memory consolidation that are sensitive to protein synthesis inhibitors, whereas only a single window is observed following enhanced training. These findings suggest that one or two waves of protein synthesis may be required for long-term memory formation, depending on the strength of the training. However, further intensifying the learning stimulus may counteract the disruptive effects of protein synthesis inhibitors. To test this hypothesis, we trained rats in an inhibitory avoidance task using either moderate or intense footsh…

Reorganization of CA1 dendritic dynamics by hippocampal sharp-wave ripples during learning.

The hippocampus plays a critical role in memory consolidation, mediated by coordinated network activity during sharp-wave ripple (SWR) events. Despite the link between SWRs and hippocampal plasticity, little is known about how network state affects information processing in dendrites, the primary sites of synaptic input integration and plasticity. Here, we monitored somatic and basal dendritic activity in CA1 pyramidal cells in behaving mice using longitudinal two-photon calcium imaging integrated with simultaneous local field potential recordings. We found immobility was associated with an in…

Investigation of T lymphocyte subsets in children with Mycoplasma pneumoniae pneumonia.

This study aims to characterize the majority of immune cell subsets in peripheral blood mononuclear cells in children with Mycoplasma pneumoniae pneumonia (MPP) by a 21-color flow cytometry panel. Patients who met the predetermined eligibility criteria for pneumonia diagnosis were recruited for the research study. Multi-color flow cytometry was conducted on the peripheral blood mononuclear cells of each patient group, which were then subjected to dimensionality reduction and cluster analysis. In our study, the proportion of activated CD4 + T cell and naïve CD8 + T in children with MPP was high

A Critical Time Window of Neocortex Modulation for Remote Fear Memory in Rats and Humans.

Persistent fear memories, central to trauma-related psychopathology, rely on long-term consolidation. While initial encoding involves subcortical circuits, the precise timing and necessity of neocortical engagement for remote fear memory expression remain elusive. In the present translational study from rats to humans, we identified a late consolidation phase in the medial prefrontal cortex (mPFC) that occurs around 18 h after fear conditioning, a phase essential for remote memory. Neuronal ensembles in the mPFC activated during this phase mediate the expression of remote fear memory in rats.

Forget me not: The effect of doxycycline on human declarative memory.

Investigations into neuroprotective drugs are in high demand for the treatment of neurodegenerative diseases, such as multiple sclerosis or … tive memory; instead, doxycycline improved declarative learning (p-value=0.022, Cohen's d=0.15) and memory consolidation (p=0.040, d=0.26). Contrarily, doxycycline slightly reduced motor learning (p=0.001, d=0.10) but subtly strengthened long-term motor memory (p=0.001, d=0.10). These results suggest that doxycycline can improve declarative learning and memory without …

Agmatine inhibits morphine-induced memory impairment in the mouse step-down inhibitory avoidance task.

The effect of agmatine on memory formation in morphine-treated mice on the step-down inhibitory avoidance test was examined. Pre-training and pre-test administration of agmatine (5, 10 and 20mg/kg, s.c.) facilitated memory formation and retrieval while post-training administration of agmatine (5, 10 and 20mg/kg, s.c.) had no effect on memory consolidation. Idazoxan (5mg/kg, i.p.) inhibited the effect of agmatine on memory formation and retrieval. Pre-training administration of morphine (1.25, 2.5 and 5mg/kg, s.c.) impaired memory formation while post-training and pre-test administration of mor…
