Comparative quantitative ultrastructural studies of the choroidal epithelium of hydrocephalic (hpy/hpy) and normal mice, and the effect of stress induced by water deprivation.

Animals homozygous for the recessive, pleiotropic, mutation hpy(hydrocephalic-polydactyl) develop hydrocephalus early in the postnatal period. The condition develops in the apparent absence of any overt indications of obstruction in the cerebrospinal fluid (CSF) drainage system suggesting a continued, inappropriate, secretion of CSF. Electron microscope investigations were undertaken to both characterize and quantify the cell types present in the choroidal epithelium of mutants and their wild-type littermates and to gauge their response to a prolonged (24 h) deprivation of water, which has sev

Pathways of transcytosis in the fenestrated endothelium of pancreatic capillaries.

A procedure which covalently attaches an electron dense tracer to cell surface sialoglycoconjugates was used in order to label the luminal surface of capillary endothelium in rat pancreas. Mild oxidation of the vasculature by perfusion with 1 mM sodium periodate, followed by a 30-90 min incubation with ferritin hydrazide (FH) at 37 degrees C, led to a characteristic decoration of the endothelial surface. FH was taken up by the endothelium in coated vesicles and, to a much lesser extent, in plasmalemmal vesicles or vacuoles. Though rarely, FH was found at the abluminal front of the endothelium

KRAS-MEK Signaling Controls Ago2 Sorting into Exosomes.

Secretion of RNAs in extracellular vesicles is a newly recognized form of intercellular communication. A potential regulatory protein for microRNA (miRNA) secretion is the critical RNA-induced silencing complex (RISC) component Argonaute 2 (Ago2). Here, we use isogenic colon cancer cell lines to show that overactivity of KRAS due to mutation inhibits localization of Ago2 to multivesicular endosomes (MVEs) and decreases Ago2 secretion in exosomes. Mechanistically, inhibition of mitogen-activated protein kinase kinases (MEKs) I and II, but not Akt, reverses the effect of the activating KRAS muta

miR-27b-5p targets RAB27A to regulate exosome secretion and migration of pancreatic cancer cells.

BACKGROUND: Exosomes are the key mediators of intercellular communication within the tumor microenvironment. Pancreatic cancer (PC) exhibits an abnormal increase in exosome secretion, and this abnormal secretion is closely associated with the invasive migration and metastatic progression of PC. Although the role of exosomes in PC metastasis has been confirmed, the precise molecular mechanism regulating this process remains unclear. This study aims to explore the regulatory mechanism of the exosome secretion cascade and its correlation with malignant migration in PC. METHODS: By comparing the

The P2X4 purinergic receptor controls the autophagy-related release of small extracellular vesicles from mammary cancer cells under hypoxia.

BACKGROUND: Small extracellular vesicles (sEVs) are critical mediators of cell communications and are influenced by cellular stress. This study investigates the role of P2X4 and P2X7 purinoreceptors in the biogenesis and release of sEVs from triple-negative mammary cancer cells under hypoxia. METHODS: In this study, we used CRISPR/Cas9 knockdown models from the triple-negative mammary cancer cell line 4T1 and selective pharmacology, to knock-down or inhibit either P2X4 or P2X7 receptors. We analysed intracellular endo-lysosomal compartments with transmission electron microscopy and epifluorece

Distinct roles of ascorbic acid in extracellular vesicles and free form: Implications for metabolism and oxidative stress in presymptomatic Huntington's disease.

Huntington's disease (HD) is a neurodegenerative disorder caused by a CAG trinucleotide repeat expansion in the first exon of the huntingtin gene. The huntingtin protein (Htt) is ubiquitously expressed and localized in several organelles, including endosomes, where it plays an essential role in intracellular trafficking. Presymptomatic HD is associated with a failure in energy metabolism and oxidative stress. Ascorbic acid is a potent antioxidant that plays a key role in modulating neuronal metabolism and is highly concentrated in the brain. During synaptic activity, neurons take up ascorbic a

Exosomes Exploit the Virus Entry Machinery and Pathway To Transmit Alpha Interferon-Induced Antiviral Activity.

Alpha interferon (IFN-α) induces the transfer of resistance to hepatitis B virus (HBV) from liver nonparenchymal cells (LNPCs) to hepatocytes via exosomes. However, little is known about the entry machinery and pathway involved in the transmission of IFN-α-induced antiviral activity. In this study, we found that macrophage exosomes uniquely depend on T cell immunoglobulin and mucin receptor 1 (TIM-1), a hepatitis A virus (HAV) receptor, to enter hepatocytes for delivering IFN-α-induced anti-HBV activity. Moreover, two primary endocytic routes for virus infection, clathrin-mediated endocytosis

Dendritic cells and macrophages in the uveal tract of the normal mouse eye.

BACKGROUND/AIMS: Dendritic cells (DC) and macrophages are components of the immune cell populations in the uveal tract whose density, distribution, turnover, and function may play a role in the maintenance of immunological homeostasis in the eye. Little is known of these cells in the mouse eye despite this being the predominant experimental model in many studies of ocular immune responses and immunoinflammatory mediated eye diseases. The aim of the present study was to obtain further immunophenotypic data on resident tissue macrophages and DC populations in the mouse uveal tract. METHODS: Pie

Thioredoxin peroxidase from Onchocerca volvulus: a major hydrogen peroxide detoxifying enzyme in filarial parasites.

Random screening of an Onchocerca volvulus third-stage (L3) cDNA library identified a highly abundant cDNA encoding a newly discovered antioxidant enzyme, thioredoxin peroxidase (TPx), a member of the peroxidoxin superfamily. This TPx cDNA (Ov-tpx-2) encodes a polypeptide of 199 amino acid residues with a calculated molecular weight of 21,890 Da. The Ov-tpx-2 cDNA represents roughly 2.5% of the total cDNAs from the L3 cDNA library. The gene was expressed in Escherichia coli and the protein product was shown to have antioxidant activity. Antiserum raised against Ov-TPX-2 recognized a native pro

Basic fibroblast growth factor (FGF-2) internalization through the heparan sulfate proteoglycans-mediated pathway: an ultrastructural approach.

Biochemical studies have shown that basic fibroblast growth factor (bFGF or FGF-2) is internalized by two pathways, after binding to either FGF tyrosine kinase receptors or to heparan sulfate proteoglycans (HSPG). To get insights on the HSPG-mediated pathway, we have examined by electron microscopy the intracellular route of bFGF-HRP, a monovalent conjugate of bFGF and horseradish peroxidase which was found to bind to HSPG only and was detectable by electron microscopy. bFGF-HRP association to adult bovine aortic endothelial (ABAE) cells or baby hamster kidney (BHK) cells was inhibited by a hi

The intestine is a site of passage for potato leafroll virus from the gut lumen into the haemocoel in the aphid vector, Myzus persicae Sulz.

Four detection techniques, three of which gave reliable identification of the virus particles, were used to locate potato leafroll virus (PLRV) in the alimentary canal of its main aphid vector, Myzus persicae Sulz: immunofluorescence on cryostat sections, conventional transmission electron microscopy on ultrathin sections and immune electron microscopy with gold labeling, either prior to or after fixation-embedding. Each method clearly showed the presence of the virus in the intestine epithelium and its absence in cells of the other parts of the alimentary canal. Under the experimental conditi

The effects of colchicine on the ultrastructure of odontogenic cells in the common skate, Raja erinacae.

Ultrastructural alterations induced by colchicine were investigated to determine the secretory activities of odontogenic cells during formation of tooth enameloid matrix in skates. Treated skate inner dental epithelial (IDE) cells did not display dilated cisternae of the granular endoplasmic reticulum (GER) nor accumulate Golgi-associated secretory granules at any dose level or time interval examined. This response was markedly different from that observed in teleost IDE cells synthesizing the enameloid collagen matrix. Treated skate IDE cells did show increased accumulations of glycogen-conta

Transferrin receptors on the surfaces of retinal pigment epithelial cells are associated with the cytoskeleton.

Retinal pigment epithelial cells, derived from human donor eyes, have been grown in culture as monolayers on membrane filters or plastic surfaces and shown to possess transferrin receptors with a monomeric molecular mass of 93,000. These receptors internalize 125I-labelled transferrin and recycle it to the surrounding medium in a similar manner to other cell types. Scatchard analyses show that there are about 100,000 high-affinity receptors on the surface of each cell and most of these receptors are associated with the cytoskeleton. In total cell extracts, there are additional low-affinity bin

Specific binding sites for albumin restricted to plasmalemmal vesicles of continuous capillary endothelium: receptor-mediated transcytosis.

The interaction of homologous and heterologous albumin-gold complex (Alb-Au) with capillary endothelium was investigated in the mouse lung, heart, and diaphragm. Perfusion of the tracer in situ for from 3 to 35 min was followed by washing with phosphate-buffered saline, fixation by perfusion, and processing for electron microscopy. From the earliest time examined, one and sometimes two rows of densely packed particles bound to some restricted plasma membrane microdomains that appeared as uncoated pits, and to plasmalemmal vesicles open on the luminal front. Evidence for cytotoxic T-lymphocyte-target cell interaction in brains of mice infected intracerebrally with lymphocytic choriomeningitis virus.

Murine lymphocytic choriomeningitis is a T-cell-mediated pathologic immune phenomenon. The name of this experimental illness was derived from the principal histopathologic alterations of the central nervous system (CNS) of adult mice infected intracerebrally with lymphocytic choriomeningitis virus, i.e., lymphocytic infiltrations of plexus choroidei and meninges. The general assumption that the main event in the pathogenesis is damage to virus-infected target cells by cytotoxic T-lymphocytes is plausible but direct evidence is scarce. We have studied the ultrastructural alterations of both typ

The ESCRT pathway and HIV-1 budding: A paradigm of viral exploitation of cellular regulation.

The ESCRT (Endosomal Sorting Complexes Required for Transport) pathway is a conserved cellular machinery essential for reverse-topology membrane fission events, such as multivesicular body biogenesis and cytokinesis. Human Immunodeficiency Virus Type 1 (HIV-1) co-opts this machinery to mediate the final scission step of virion budding from the host cell membrane. This review presents our current understanding of how HIV-1 hijacks the ESCRT system, moving beyond a simple recruitment model to a dynamic interplay with host cell regulation. We explore the spatial organization of the budding machin

The proteogenomic landscape of the human kidney and implications for cardio-kidney-metabolic health.

Nearly one-third of the global population is affected by cardio-kidney-metabolic (CKM) diseases; however, the molecular mechanisms underlying CKM diseases are poorly understood. Here we show that tissue proteomics provide critical insights not captured by tissue gene expression or blood proteomics information by performing whole-genome and RNA sequencing and proteomics analysis of human kidney samples (n = 337), and we generated a publicly available database. Via Bayesian co-localization and Mendelian randomization analyses of kidney protein quantitative trait loci and 36 CKM genome-wide assoc

SINAT E3 ligases regulate the stability of the ESCRT component FREE1 in response to iron deficiency in plants.

The endosomal sorting complex required for transport (ESCRT) machinery is an ancient, evolutionarily conserved membrane remodeling complex that is essential for multivesicular body (MVB) biogenesis in eukaryotes. FYVE DOMAIN PROTEIN REQUIRED FOR ENDOSOMAL SORTING 1 (FREE1), which was previously identified as a plant-specific ESCRT component, modulates MVB-mediated endosomal sorting and autophagic degradation. Although the basic cellular functions of FREE1 as an ESCRT component have been described, the regulators that control FREE1 turnover remain unknown. Here, we analyzed how FREE1 homeostasi

Cutting Edge: Regulation of Exosome Secretion by the Integral MAL Protein in T Cells.

Exosomes secreted by T cells play an important role in coordinating the immune response. HIV-1 Nef hijacks the route of exosome secretion of T cells to modulate the functioning of uninfected cells. Despite the importance of the process, the protein machinery involved in exosome biogenesis is yet to be identified. In this study, we show that MAL, a tetraspanning membrane protein expressed in human T cells, is present in endosomes that travel toward the plasma membrane for exosome secretion. In the absence of MAL, the release of exosome particles and markers was greatly impaired. This effect was
