Immunogenomics of gastrointestinal nematode infection in ruminants - breeding for resistance to produce food sustainably and safely.

Gastrointestinal nematode (GIN) infection of ruminants represents a major health and welfare challenge for livestock producers worldwide. The emergence of anthelmintic resistance in important GIN species and the associated animal welfare concerns have stimulated interest in the development of alternative and more sustainable strategies aimed at the effective management of the impact of GINs. These integrative strategies include selective breeding using genetic/genomic tools, grazing management, biological control, nutritional supplementation, vaccination and targeted selective treatment. Human Pulmonary Infection by the Zoonotic Metastrongylus salmi Nematode. The First Reported Case in the Americas.

Pulmonary metastrongylosis, a zoonotic disease found primarily in pigs, is caused by eight different species of the cosmopolitan nematode Metastrongylus genus. To date, only four human cases have been reported, all from Europe. Herein, a severe case of pulmonary infection caused by Metastrongylus salmi in an Ecuadorian man, with successful treatment with ivermectin, is described.

Effect of gastro-intestinal nematode infection on sheep performance: a systematic review and meta-analysis.

BACKGROUND: Gastrointestinal nematode (GIN) infections are common in domestic sheep and impact directly and indirectly on the health of infected animals as well as on the associated economic production. In this study, we aim at summarizing the current knowledge on the influence of GIN infections on sheep production by conducting a systematic review. A subsequent meta-analysis of relevant studies was performed to provide an estimate of the effect of GIN infections on weight gain, wool production and milk yield. METHODS: A literature search was performed on the CAB, Pubmed and Web of Science da

Immune responses in sheep naturally infected with Oestrus ovis (Diptera: Oestridae) and gastrointestinal nematodes.

This study was carried out to evaluate the immune response in young Ile de France (IF) and Santa Ines (SI) sheep naturally infected by Oestrus ovis and gastrointestinal nematodes (GIN). Mast cells, eosinophils and globule leucocytes were enumerated in the upper respiratory tract (septum, middle meatus and ventral nasal conchae) and in the mucosa of abomasum and small intestine. Immunoglobulin G (IgG) levels in serum samples and immunoglobulin A (IgA) levels in mucus from the nasal, abomasum and small intestinal mucosae were determined against O. ovis, Haemonchus contortus and Trichostrongylus

Diversity and prevalence of metastrongyloid nematodes infecting the red panda (Ailurus fulgens) in European zoos.

Metastrongyloid induced pneumonia has been described sporadically in the red panda (Ailurus fulgens). Early descriptions in pandas recently imported to the USA from China involved parasites morphologically similar to Angiostrongylus spp. and Crenosomatidae. More recently, four cases of severe verminous pneumonia associated with Angiostrongylus vasorum have been reported from European zoos. A coprological survey of the red panda population within European zoos was conducted in 2008. Faecal samples from 115 pandas originating from 54 zoos were collected on 3 consecutive days. Using Baermann tech

Challenges to the development of new tests for diagnosis of infection and prediction of resistance of sheep to gastrointestinal nematodes.

Strategies for control of gastrointestinal nematode (GIN) infections in sheep require information on the severity of infection and species (or genus) of parasite present. Tests for diagnosis of GIN fall into 3 classes. Current tests measure either: 1) the presence of eggs, worm antigens or worms themselves; 2) components of host immunity (e.g. antibodies, eosinophils, other immune mediators); or 3) components of host pathology (e.g. wool growth, body growth, appetite, blood loss, digestive enzymes, anaemia, hypoproteinaemia, odours). To offer advantages over current diagnostic methods, new tes

Value of Haemonchus longistipes purified antigens in diagnosis of gastro-intestinal nematodes infection in camels.

Fractionated Haemonchus longistipes crude antigen revealed 7 protein fractions at molecular weights of 93, 74, 67, 56, 32, 26 and 16 kDa. The bands at 56, 32 and 26 kDa reacted specifically with H. longistipes hyperimmune sera prepared in rabbit (1:100 dilution) using EITB technique. The eluted concentrated fractions as well as crude worm antigens were used in diagnosis of natural infection of Gastro-Intestinal Nematodes (GIN) only or associated with other parasites in camels using ELISA technique. H. longistipes crude antigen and fraction of 26 kDa induced high sensitivity in diagnosis of inf

Identification of novel genes in intestinal tissue that are regulated after infection with an intestinal nematode parasite.

Infection of resistant or susceptible mice with Trichuris muris provokes mesenteric lymph node responses which are polarized towards Th2 or Th1, respectively. These responses are well documented in the literature. In contrast, little is known about the local responses occurring within the infected intestine. Through microarray analyses, we demonstrate that the gene expression profile of infected gut tissue differs according to whether the parasite is expelled or not. Genes differentially regulated postinfection in resistant BALB/c mice include several antimicrobial genes, in particular, intele

Allergen-specific IgE and IgG4 are markers of resistance and susceptibility in a human intestinal nematode infection.

IgG4 has been proposed to act as a 'blocking antibody' due to its ability to compete for the same epitopes as IgE thus preventing IgE-dependent allergic responses. IgG4 and IgE are both elevated in helminth infections and strong anti-parasite IgE responses are associated with resistance to infection. We wished to determine the relationship between anti-parasite IgG4 and IgE and Ascaris lumbricoides infection status. We examined anti-parasite responses, including antibody levels to recombinant Ascaris allergen-1A (rABA-1A), a target of serum IgE in endemic populations. Worm burden was indirectl

The sequential analysis of local inflammatory cells during abomasal nematode infection in periparturient sheep.

A technique to take sequential tissue biopsy samples in multiparous, periparturient ewes from the abomasal mucosa is described, developed in parallel in Scotland and New Zealand. Samples were extracted via abomasal cannulae inserted into the wall of the abomasum and exteriorised through dorso-ventral laparotomy. Animals recovered quickly post-surgery, and tolerated the cannula and sampling without any adverse signs of pain or discomfort. The technique was deployed in two pilot studies to investigate the sequential mucosal inflammatory cell responses in well-defined parasitological models, duri

Cytokine response profiles predict species-specific infection patterns in human GI nematodes.

This study investigated associations between pre-treatment cytokine expression and infection patterns, before and after de-worming, in humans exposed to two gastrointestinal nematode species. Quantitative measures of Ascaris lumbricoides and Trichuris trichiura infection (based on faecal egg counts) were estimated immediately before and 8-9 months after treatment in a Cameroonian population. Whole blood cytokine responses to parasite-derived antigens were assayed immediately pre-treatment. An overall measure of the tendency towards species-specific infection (increasing with A. lumbricoides fa

Inflammation-induced impairment of enteric nerve function in nematode-infected mice is macrophage dependent.

Trichinella spiralis infection in rodents is associated with suppression of ACh release from myenteric plexus that can be mimicked by macrophage-derived cytokines. We verified the presence of a macrophage infiltrate in the intestine during T. spiralis infection and determined the extent to which this cell type is responsible for the neural changes. C57BL/6 mice were infected with 375 T. spiralis larvae by gavage, and the presence of macrophages (F4/80 positive) in the jejunum was determined immunohistochemically. In another experiment, infected mice were treated intravenously with liposomes co

Functional alterations in jejunal myenteric neurons during inflammation in nematode-infected guinea pigs.

Intracellular recordings of jejunal myenteric neurons with an afterspike hyperpolarization (AH) from Trichinella spiralis-infected animals showed enhanced excitability on days 3, 6, and 10 postinfection (PI) compared with uninfected animals. Lower membrane potential, increased membrane input resistance, decreased threshold for action potential discharge, decreased AH amplitude and duration, and increased fast excitatory postsynaptic potential amplitude and duration were characteristic of neuronal recordings from infected animals. Concurrent with electrophysiological changes during T. spiralis

Effects of preventive anthelmintic treatment on acquired resistance to gastrointestinal nematodes in naturally infected cattle.

The objective of this study was to investigate the influence of different types of chemoprophylaxis in first season grazing calves on their resistance against a natural reinfection with Ostertagia ostertagi and Cooperia oncophora in the second grazing season. Thirty helminth-naive crossbred calves were randomly divided in three groups of 10 animals. The animals of group B received an ivermectin sustained release bolus on day 0. The calves of group D were treated on days 0 and 56 with a subcutaneous injection of doramectin (0.2 mg kg(-1) BW). Group C was the untreated control group ('immune' co

Chitinase genes expressed by infective larvae of the filarial nematodes, Acanthocheilonema viteae and Onchocerca volvulus.

State-specific products of 220 and 75 kDa were identified by metabolic labelling of infective larvae of the filarial nematode Acanthocheilonema viteae in ticks. Synthesis was temperature sensitive, occurring at 27 degrees C but not at 37 degrees C. These products were secreted 3-6 days after leaving the vector during post-infective development, but subsequent expression was not detected. The smaller protein with a pI of 6.2, was purified by two-dimensional electrophoresis and the N-terminal amino acid sequence was derived. This provisionally identified the protein as a chitinase, which was con

Heterogeneity amongst infected children in IgE antibody repertoire to the antigens of the parasitic nematode Ascaris.

Plasmas from a sample of 6-year-old children chronically infected with Ascaris lumbricoides were examined by immunoblotting for IgE antibody to the pseudocoelomic fluid of adult parasites. This revealed a high degree of heterogeneity in the antibody repertoire to putative allergens of the parasite, such that none of those screened had identical recognition patterns. This heterogeneity occurred despite the fact that infection with A. lumbricoides involves both tissue-invasive and intestinal phases of the infection, which should result in close contact between the immune system and potentially a

Seasonal occurrence of infective nematode larvae in California high Sierra pastures grazed by cattle.

Worm-free Holstein calves were used to assess the seasonal variation in numbers of nematode infective larvae on herbage at 2 commercial cow-calf ranches in the Sierra Nevada mountains. Both ranches used Herefords as their basic breed. One ranch near Sattley, California, is at an elevation of approximately 1,500 m above sea level, whereas the study area on the 2nd ranch near Susanville is at an elevation of approximately 1,250 m above sea level. Worm-free tracer calves were allowed to graze for approximately 30 days and were then kept free of further nematode infection until euthanasia and necr

Studies on the effect of treatment with anthelmintics on weight gain in calves, naturally infected with gastrointestinal nematodes.

To avoid disease outbreaks caused by gastro-intestinal nematodes different prophylactic measures are recommended. In this connection pasture hygienic achievements and anthelmintics are used. In practice it is often difficult to realize pasture hygienic steps as move the animals to parasite free areas and to avoid that they come back to the infected. In many cases the parasite control is limited to routine deworming. Our intention was to study if anthelmintic treatment without other measures had any effect on weight gain and number of eggs in faeces in calves naturally infected with Ostertagia

Anthelmintic Effect of Chitosan-Encapsulated Bromelain on Gastrointestinal Nematodes in Naturally Infected Goats in Kenya.

The emergence of anthelmintic resistance calls for the development of alternatives, including medicinal plant extracts. The present study was aimed at assessing the efficacy of chitosan-encapsulated bromelain (EB) as an oral solution drug against GIN in goats. Standard methods were followed to extract bromelain from the pineapple peels and the conventional manufacturing procedure used to make an oral solution of EB. The in vivo study was performed on 20 healthy male goats that were naturally infected with GIN. The goats were randomly divided into four groups, each of which consisted of three t
