Infusion of opiates into substantia nigra protects against maximal electroshock seizures in rats.

Microinfusion of morphine sulfate (50 nmol), [d-Ala2]-Met-enkephalin (35 nmol) or dynorphin A 1-13 (1 nmol) bilaterally into the substantia nigra significantly attenuated seizures induced by maximal electroshock in rats. This action was accompanied by stereotyped behavioral hyperactivity. These anticonvulsant and behavioral effects were antagonized by systemic naloxone administration; neither effect was observed after intranigral microinjection of dynorphin A 1-17 amide (1 nmol). These results are consistent with a mu opiate receptor-mediated inhibition of substantia nigra efferent neurons, an

The olivocochlear reflex strength and cochlear sensitivity are independently modulated by auditory cortex microstimulation.

In mammals, efferent projections to the cochlear receptor are constituted by olivocochlear (OC) fibers that originate in the superior olivary complex. Medial and lateral OC neurons make synapses with outer hair cells and with auditory nerve fibers, respectively. In addition to the OC system, there are also descending projections from the auditory cortex that are directed towards the thalamus, inferior colliculus, cochlear nucleus, and superior olivary complex. Olivocochlear function can be assessed by measuring a brainstem reflex mediated by auditory nerve fibers, cochlear nucleus neurons, and

Roles of orexin/hypocretin in regulation of sleep/wakefulness and energy homeostasis.

The finding of orexin (hypocretin) deficiency in patients with narcolepsy suggests that this hypothalamic neuropeptide plays a crucial role in regulating and maintaining sleep/wakefulness states and energy homeostasis. Orexin might be especially important for stabilization of behavioral states, because the major symptom in narcolepsy is instability of each behavioral state, which results in sleep/wakefulness fragmentation. The efferent and afferent systems of orexin neurons suggest interactions between these cells and arousal/sleep-wakefulness centers in the brainstem as well as important feed

Ultrastructure of hypothalamic paraventricular neurons.

The hypothalamic paraventricular nucleus (PVN) plays a pivotal role in the regulation of endocrine processes and the modulation of autonomic functions. The multi-channel outputs of the nucleus are directed toward the anterior and posterior lobes of the pituitary, autonomic centers, and limbic structures. Counterbalancing the wide spectrum of efferent projections, the nucleus receives humoral signals from endocrine glands and neuronal afferents from several loci of the brain. The multiple functions of the nucleus are executed by neurons that are organized in distinct subnuclei and display compl

Effects of contralateral sound on auditory-nerve responses. II. Dependence on stimulus variables.

The suppression by moderate-level contralateral sound of auditory-nerve-fiber responses to ipsilateral stimuli at the characteristic frequency (CF) was studied in barbiturate-anesthetized cats. The dependence of suppression strength on ipsilateral and contralateral stimulus variables, including level, frequency, bandwidth, and timing relationships, was investigated. The principal findings were: (1) Contralateral-sound suppression is greatest when the ipsilateral stimulus level is within the dynamic range of the unit. (2) When the contralateral stimuli are tones, suppression is greatest when th

Effects of contralateral sound on auditory-nerve responses. I. Contributions of cochlear efferents.

The response properties of single auditory-nerve fibers in barbiturate-anaesthetized cats were recorded with and without simultaneous presentation of sound to the contralateral ear. The tendons to the middle ear muscles on both sides were cut before all experiments, and contralateral stimuli were restricted to levels below the threshold for crosstalk to the ipsilateral ear. Contralateral tones and broad-band noise were found to suppress the responses of auditory-nerve afferents to ipsilateral tones at their characteristic frequency (CF), but not to tones off CF. The suppression due to contrala

Genital sensory field: enlargement by estrogen treatment in female rats.

Recordings of neuronal activity in the pudendal, genitofemoral, and pelvic nerves indicate that the sensory fields of these three nerves are the perineum, the caudal abdomen, and the vagino-cervical area and rectum, respectively. The sensory field of the pudendal nerve was significantly larger in estrogen-treated ovariectomized female rats than in uninjected controls. This effect of estrogen was not mediated by pudendal efferents.

Histochemical and biochemical studies of cholinesterases in the carotid labyrinth of amphibians.

Acetylcholinesterase activity was detected in the carotid labyrinth of amphibians by both biochemical and histochemical methods. The histochemical tests showed enzyme activity in surfaces of afferent and efferent nerve terminals in contact with type I cells, but none within the type I cells. Acetylcholinesterase activity occurred on some, though not all, nerve fibers in the extra-cellular spaces. These fibers might be parasympathetic cholinergic fibers innervating blood vessels.

Cholinergic modulation of stellate cells in the mammalian ventral cochlear nucleus.

The main source of excitation to the ventral cochlear nucleus (VCN) is from glutamatergic auditory nerve afferents, but the VCN is also innervated by two groups of cholinergic efferents from the ventral nucleus of the trapezoid body. One arises from collaterals of medial olivocochlear efferents, and the other arises from neurons that project solely to the VCN. This study examines the action of cholinergic inputs on stellate cells in the VCN. T stellate cells, which form one of the ascending auditory pathways to the inferior colliculus, and D stellate cells, which inhibit T stellate cells, are

An anterograde tracing study of the vagal innervation of rat liver, portal vein and biliary system.

In order to investigate the distribution and structure of the vagal liver innervation, abdominal vagal afferents and efferents were selectively labeled by injecting WGA-HRP or Dil into the nodose ganglia, and DiA into the dorsal motor nucleus, respectively. Vagal afferent fibers produced characteristic terminal-like structures at three locations in the liver hilus: 1. Fine varicose endings preferentially surrounding, but not entering, the numerous peribiliary glands in the larger intra and extrahepatic bile ducts 2. Large, cup-shaped terminals in almost all paraganglia 3. Fine varicose endings

Effects of acute graded strain on efferent conduction properties in the rabbit tibial nerve.

This study examines effects of acute stretch on electrophysiologic properties of rabbit tibial nerve. Compound motor action potentials (CMAP) were measured in 30 rabbits, during and after two hours of acute, graded strain. The rabbits were assigned to one of three groups defined by the degree of stretch (0%, 8%, or 15%) beyond original resting length. These groups were subdivided according to ankle position during nerve strain (ankle dorsiflexion or ankle plantar flexion). Two hours of 8% strain did not significantly decrease CMAP amplitude. Fifteen percent strain with the ankle in dorsiflexio

Oxaliplatin-Induced Damage to the Gastric Innervation: Role in Nausea and Vomiting.

Nausea and vomiting are common gastrointestinal side effects of oxaliplatin chemotherapy used for the treatment of colorectal cancer. However, the mechanism underlying oxaliplatin-induced nausea and vomiting is unknown. The stomach is involved in the emetic reflex but no study investigated the effects of oxaliplatin treatment on the stomach. In this study, the in vivo effects of oxaliplatin treatment on eating behaviour, stomach content, intrinsic gastric neuronal population, extrinsic innervation to the stomach, levels of mucosal serotonin (5-hydroxytryptamine, 5-HT), and parasympathetic vaga

Functional coupling between peripheral nociceptor terminals in vivo is enhanced during acute inflammation.

Primary nociceptors are essentially characterized as afferent neurons carrying noxious sensory information from the periphery to the central nervous system. However, the information flow on primary nociceptors is bidirectional. Nociceptor peripheral terminals release a variety of mediators to the target organ near the injured area. These mediators promote sensitization of adjacent sensory neurons, vasodilation, and edema, affecting innate and adaptive immunity, leading to hyperalgesia and inflammation that often expands beyond the injured areas. While many studies associate these phenomena wit

Anti-inflammatory effects of hunger are transmitted to the periphery via projection-specific AgRP circuits.

Caloric restriction has anti-inflammatory effects. However, the coordinated physiological actions that lead to reduced inflammation in a state of caloric deficit (hunger) are largely unknown. Using a mouse model of injury-induced peripheral inflammation, we find that food deprivation reduces edema, temperature, and cytokine responses that occur after injury. The magnitude of the anti-inflammatory effect that occurs during hunger is more robust than that of non-steroidal anti-inflammatory drugs. The effects of hunger are recapitulated centrally by activity in nutrient-sensing hypothalamic agout

The distribution of neurons containing delta sleep-inducing peptide in the hippocampal formation.

The distribution of neurons containing immunoreactive delta sleep-inducing peptide (IDSIP) in the hippocampal formation was investigated by immunocytochemistry. For this study, thirteen antisera to the peptide were used. Three antisera were found suitable for immunocytochemistry; the remainder gave only non-specific background staining. Neurons containing IDSIP were demonstrable in the subicular cortex immediately adjacent to CA1. No specific staining was found in other parts of the hippocampal formation including the pyramidal layer of Ammon's Horn and the dentate gyrus. These three antisera

Structure of cells and nerve endings in abdominal vagal paraganglia of the rat.

The abdominal vagal paraganglia of the rat consist of small groups of cells, interspersed by blood vessels and nerve bundles and lying close to, or within, the vagus nerve or its branches. Each cell group consists of 2-10 Type I cells incompletely invested by 1-3 satellite cells. Type I cells are characterised by the presence of numerous dense-cored vesicles in their cytoplasm and may exhibit 'synaptic'-like contact with each other. Small efferent nerve endings make synaptic contacts with Type I cells. Larger cup-shaped afferent nerve endings also make synaptic contacts of two kinds with Type

Feasibility of differentially measuring afferent and efferent neural activity with a single nerve cuff electrode.

OBJECTIVE: Advances in electrode technology have facilitated the development of neuroprostheses for restoring motor/sensory function in disabled individuals. Information extracted from a whole nerve, recorded using cuffs, can provide signals that control the operation of neuroprostheses. However, the amount of information that can be extracted from a tripolar cuff-which provides the highest signal-to-noise ratio (SNR)-is limited. The physical symmetry of the tripolar cuff results in neural recordings that cannot differentiate afferent versus efferent signals. In this study, we introduced a tet

New insights into peptidergic abnormalities in Hirschsprung's disease by wholemount immunohistochemistry.

In a pilot study previously reported, we showed that individual nerves could be traced in the different layers of the gut in Hirschsprung's disease (HD) using wholemount immunohistochemistry (WI). Little is known about the course of the important nonadrenergic, noncholinergic nerves containing neuropeptides in HD. Therefore, we studied the distribution of neuropeptides in 9 HD patients and 5 controls using WI. The new findings include the following: (1) there were two populations of substance P (SP) nerves--in aganglionic gut, SP-efferent nerves were decreased but SP-afferent fibres innervatin
