Glucagon-like immunoreactivity in the forebrain and pituitary of the teleost, Clarias batrachus (Linn.).

The organization of glucagon-like immunoreactivity (GLI) in the olfactory system, forebrain, and pituitary was investigated in the teleost Clarias batrachus. Weak to moderate GLI was seen in some olfactory receptor neurons and basal cells of the olfactory epithelium. Intense GLI was seen in the olfactory nerve fascicles that ran caudally to the bulb, spread over in the olfactory nerve layer, and profusely branched in the glomerular layer to form tufts organized as spherical neuropils; some of the immunoreactive fibers seem to closely enfold the mitral cells. In the inner cell layer of the bulb

Topographic coding of odorant quality is maintained at different concentrations in the salamander olfactory epithelium.

In a recent study in the tiger salamander, Ambystoma tigrinum, were demonstrated topographic patterns of responsivity across the olfactory epithelium which were characteristic for each odorant. The present study was initiated to investigate whether these patterns remain constant when odorant concentration is varied. Odorant-induced electro- olfactograms were recorded from at least 12 sites on each epithelium. The odorants used were pinene, amyl acetate and propanol. Each epithelium was tested with one odorant, delivered at 3 concentrations. For comparison between animals, the epithelia were di

Mechanical obstruction of the olfactory cleft reveals differences between orthonasal and retronasal olfactory functions.

Following up on recent observations in patients with nasal polyposis (NP), the present study aimed to investigate whether a mechanical obstruction of the anterior olfactory cleft (OC) would produce differential effects on orthonasal and retronasal olfactory functions. To this end, we studied 33 healthy subjects in a randomized trial. Sponges with high content of saline were either placed in the OC or on the respiratory epithelium, such that this was blinded to both subject and observer. The results indicated that orthonasal (P = 0.04) but not retronasal (P = 0.15) olfactory identification abil

Expression of isoforms of the neural cell adhesion molecule (NCAM) and polysialic acid during the development of the Bufo arenarum olfactory system.

The neural cell adhesion molecule (NCAM), a member of the immunoglobulin superfamily that promotes Ca(2+)-independent cell-cell adhesion, is expressed as various isoforms generated by alternative splicing. In this study, the expression of the 180 kDa isoform (180-NCAM), total NCAM (180, 140 and 120 kDa isoforms) and the polysialic acid moiety of NCAM (PSA) were analyzed during the development of the olfactory system of the toad Bufo arenarum using specific antibodies and immunofluorescence light microscopy. NCAM and PSA were not found in the ectodermal thickening corresponding to the olfactory

Intracellular potentials of salamander mitral/tufted neurons in response to odor stimulation.

Intracellular responses of salamander mitral/tufted neurons to defined odor pulses are described. Responses to odor stimulation, which generally are more complex than responses to olfactory nerve or tract electrical stimulation, show periods of depolarization and hyperpolarization that are influenced by odor concentration and quality. The way these periods coincide with different types of spike patterns in individual cells supports the hypothesis that the temporal patterning of odor responses is generated by differential activation of bulbar circuits.

A parametric study of the stimulation variables affecting the magnitude of the olfactory nerve response.

The magnitude of olfactory responses can be related to three primary variables [number of odorant molecules (N), sniff volume (V), and sniff duration (T)] and three derived variables [concentration (C = N/V), flow rate (F = V/T), and delivery rate (D = N/T)]. To evaluate the effects of these interdependent variables upon the olfactory response, the summated multiunit discharges were recorded from the olfactory nerves of nine frogs in response to octane presented at two levels (in 2:1 ratio) of each primary variable. This presentation defined eight "sniff" combinations representing three levels

Olfaction and Executive Cognitive Performance: A Systematic Review.

Objective tests of olfaction are widely available to aid in the assessment of olfaction. Their clearest role is in the characterization of olfactory changes, either reported by or suspected in a patient. There is a rapidly growing literature concerned with the association of olfactory changes with certain neuropsychiatric conditions and the use of olfactory testing to supplement conventional assessments in clinical and research practice is evolving. Neural pathways important for olfactory processing overlap extensively with pathways important for cognitive functioning, and especially those imp

Monorhinal and Birhinal Odor Processing in Humans: an fMRI investigation.

The olfactory nerve, also known as cranial nerve I, is known to have exclusive ipsilateral projections to primary olfactory cortical structures. It is still unclear whether these projections also correspond to functional pathways of odor processing. In an olfactory functional magnetic resonance imaging (fMRI) study of twenty young healthy subjects with a normal sense of smell, we tested whether nostril specific stimulation with phenyl ethyl alcohol (PEA), a pure olfactory stimulant, asymmetrically activates primary or secondary olfactory-related brain structures such as primary olfactory corte

Development and migration of olfactory neurones in the nervous system of the neonatal opossum.

The neonatal opossum (Monodelphis domestica) was used to assess how different populations of cells are generated in the olfactory region, and how they migrate along pathways to the central nervous system. Developing nerve cells were immunocytochemically labelled using antisera directed against two specific markers of olfactory receptor neurones: olfactory marker protein (OMP) and the dipeptide carnosine. In new-born opossums both carnosine and OMP are already co-expressed in primary olfactory neurones and in those axons that extend towards the olfactory bulb. Expression of these markers in olf

Clonal analysis of olfaction in Drosophila: generation of flies with mosaic labeling.

Clonal analysis with the MARCM (mosaic analysis with a repressible cell marker) system can be used for studying cell lineage, development, and anatomy in the Drosophila olfactory system and other parts of the fly brain. This protocol gives a method for generating flies with mosaic labeling. It describes how to establish a mating cage for MARCM in PNs (projection neurons) of the fly antennal lobe and then select appropriate flies for dissection and staining using immunohistochemistry. The protocol can be adapted to determine the birth order of neuroblast lineages or individual cells. Alternativ

Connections of the terminal nerve and the olfactory system in two galeomorph sharks: an experimental study using a carbocyanine dye.

In elasmobranchs the terminal nerve courses separately from the olfactory nerve. This characteristic makes elasmobranchs excellent models to study the anatomy and function of these two systems. Here we study the neural connections of the terminal nerve and olfactory system in two sharks by experimental tracing methods using carbocyanine dyes. The main projections from the terminal nerve system (consisting of three ganglia in Scyliorhinus canicula) course ipsilaterally to the medial septal nucleus and bilaterally to the ventromedial telencephalic pallial region. Minor terminal nerve projections

Pattern of calretinin immunoreactivity in the main olfactory system and the vomeronasal system of the tree shrew, Tupaia belangeri.

The distribution of the calcium-binding protein calretinin was studied in peripheral and central parts of the main olfactory system (MOS) and the vomeronasal system (VNS) of adult tree shrew Tupaia belangeri. The calretinin immunoreaction was carried out with a peroxidase-coupled polyclonal antibody. In the VNS, complete labeling of all receptor cells and vomeronasal nerve fibers was observed, whereas only a subset of the somata and dendrites of receptor cells and of the olfactory nerve fibers of the MOS was immunoreactive. From the immunoreactive dendritic clubs of vomeronasal receptor cells,

Glycobiology of the olfactory system.

The olfactory system is a highly plastic region of the nervous system. Continuous remodeling of neuronal circuits in the olfactory bulb takes place throughout life as a result of constant turnover of primary sensory olfactory neurons in the periphery. Glycoconjugates are very important in olfactory development, regeneration and function. This article deals with different aspects of glycobiology relevant for the olfactory system. Various anatomical, developmental and functional subdivisions of the olfactory system have been labeled with exogenous lectins. The application of reverse lectin histo

Odor responses of the vomeronasal system in Reeve's turtle, Geoclemys reevesii.

The number of vomeronasal receptor cells and olfactory receptor cells were determined in several turtles and tortoises. Using semi-aquatic turtles with developed vomeronasal organs, neural responses to various odor stimuli were recorded as extracellular unitary spike responses of vomeronasal receptor cells, compound action potentials from vomeronasal nerve twigs, induced wave responses on the accessory olfactory bulb (AOB) surface and unitary responses of AOB neurons. The vomeronasal system in turtles responded to various volatile and non-volatile chemicals. These results yield insights concer

Early development of the olfactory and terminalis systems in baleen whales.

The development of the olfactory and terminalis systems was studied in tissue from eight embryonic and early fetal specimens belonging to three species of baleen whales. In contrast to toothed whales, baleen whales, particularly in these ontogenetic stages, are much less specialized in nasal organ morphology. The nasal cavity and peripheral olfactory system are well developed and do not show signs of reduction. However, as in toothed whales, there is no trace of a vomeronasal organ or nerve. The terminalis neuroblasts can already be distinguished from the olfactory material in the embryonic pe

Effects of carnosine on olfactory bulb EEG, evoked potentials and DC potentials.

Carnosine is a dipeptide found in great quantities in the primary olfactory nerve and has been suggested to be the neurotransmitter of the olfactory receptor axons. The aim of the present study was to describe some of its electrophysiological actions in the olfactory bulb (OB) of rabbits under anesthesia. Carnosine as a 10% solution in amounts of 2-5 microliter was injected to the OB at the level of the glomerular layer by means of a pipette attached to a Hamilton syringe. Average evoked potentials (AEPs) on the stimulation of the lateral olfactory tract (LOT-AEP), electroencephalographic (EEG
