Microscopic and Molecular Tracing of Cryptosporidium Oocysts: Identifying a Possible Reservoir of Infection in Red Grouse.

Infection by Cryptosporidiumbaileyi causes respiratory cryptosporidiosis in red grouse Lagopuslagopusscotica. First diagnosed in 2010, it has since been detected across half of moors managed for grouse shooting in northern England. We hypothesised that contaminated grouse faeces within communal trays visited by grouse containing grit coated with flubendazole, provided to control Trichostrongylustenuis parasites of grouse, is a reservoir of infection. To establish the basis to this hypothesis, contents of 23 trays from a grouse moor were examined for Cryptosporidium oocysts. Contents were subje…

An improved stool concentration procedure for the detection of Cryptosporidium oocysts in Orang Asli stool samples.

To improve the stool concentration procedure, we modified different steps of the standard formalin-ether concentration technique and evaluated these modifications by examining stool samples collected in the field. Seven samples were found positive by the modified formalin-ether concentration technique (M-FECT). Therefore, the M-FECT procedure provides enhanced detection of Cryptosporidium oocysts.

Detection and genotyping of oocysts of Cryptosporidium parvum by real-time PCR and melting curve analysis.

Several real-time PCR procedures for the detection and genotyping of oocysts of Cryptosporidium parvum were evaluated. A 40-cycle amplification of a 157-bp fragment from the C. parvum beta-tubulin gene detected individual oocysts which were introduced into the reaction mixture by micromanipulation. SYBR Green I melting curve analysis was used to confirm the specificity of the method when DNA extracted from fecal samples spiked with oocysts was analyzed. Because C. parvum isolates infecting humans comprise two distinct genotypes, designated type 1 and type 2, real-time PCR methods for discrimin…

Detection of Cryptosporidium oocysts in human fecal specimens by an indirect immunofluorescence assay with monoclonal antibodies.

An indirect fluorescence assay (IFA) with monoclonal antibodies developed for the detection of Cryptosporidium oocysts in fecal smears (Meridian Diagnostics, Inc., Cincinnati, Ohio) was compared with the Zeihl-Neelsen-modified acid-fast stain (MAFS) in 119 human fecal specimens collected between 1984 and 1987. The sensitivity of the IFA was 100%; all 56 specimens positive by MAFS exhibited fluorescence. There were 63 specimens negative for Cryptosporidium sp. by MAFS; of these, 61 were negative by IFA (97% specificity). This discrepancy may reflect an increased sensitivity of the IFA to detect…

The occurrence of Cryptosporidium oocysts in faecal samples submitted for routine examination at Kenyatta National Hospital.

One hundred and thirty-three loose or diarrhoeic stool specimens from patients admitted at Kenyatta National Hospital were examined for cryptosporidiosis. Oocysts were detected in 5(3.8%) of the total in which Cryptosporidium was the sole pathogenic agent. In addition to Cryptosporidium, other pathogens isolated included bacteria, protozoa and helminths. We therefore report that Cryptosporidium is a cause of diarrhoea and should be looked for in the absence of other pathogenic organisms.

The development and excretion of Toxoplasma gondii oocyst manipulate the gut microbiota in its definitive host.

BACKGROUND: Oocysts serve as the primary source of Toxoplasma infection. Therefore, understanding oocyst development and exploring effective strategies to prevent oocyst excretion are crucial for controlling toxoplasmosis. METHODS: In this study, shotgun metagenomics was employed to characterize the functional and compositional changes in the gut microbiota of cats during oocyst development. The Spearman correlation test was utilized to analyze the correlation between differential Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways and carbohydrate-active enzymes (CAZymes) in key bacteria…

EtMIC2 overexpression attenuates pathogenicity while enhancing oocyst production and immunogenicity in Eimeria tenella.

Avian coccidiosis, caused by Eimeria parasites, is a devastating birds disease with global economic impacts. Despite its significance, the pathogenesis of E. tenella remains unclear due to genetic manipulation challenges. Previous studies have identified E. tenella microneme protein 2 (EtMIC2) as a critical factor for host cell invasion, but its precise role during developmental stages in vivo remains poorly characterized. To address this knowledge gap, we generated a genetically edited strain stably overexpressing EtMIC2 (EtMIC2-OE) and investigated the impact of EtMIC2 overexpression on E. t

Detection of Toxoplasma gondii oocysts in soil and risk mapping in an island environment in the Northeast of Brazil.

Toxoplasmosis is an emerging and re-emerging infectious disease that can be transmitted through a contaminated environment. Environmental contamination is an emergency health issue, and determining its occurrence is fundamental to a One Health approach. In this study, we addressed the extent of environmental contamination and viability of Toxoplasma gondii oocysts in soil in different environments on Fernando de Noronha Island, Brazil. In addition, we performed species distribution modelling to predict the environmental suitability for coccidia persistence in the studied area. Soil samples wer…

A combined miRNA-piRNA signature in the serum and urine of rabbits infected with Toxoplasma gondii oocysts.

BACKGROUND: Increasing evidence has shown that non-coding RNA (ncRNA) molecules play fundamental roles in cells, and many are stable in body fluids as circulating RNAs. Study on these ncRNAs will provide insights into toxoplasmosis pathophysiology and/or help reveal diagnostic biomarkers. METHODS: We performed a high-throughput RNA-Seq study to comprehensively profile the microRNAs (miRNAs) and PIWI-interacting RNAs (piRNAs) in rabbit serum and urine after infection with Toxoplasma gondii oocysts during the whole infection process. RESULTS: Total RNA extracted from serum and urine samples of…

A Simple Alcohol-based Method of Oocyst Inactivation for Use in the Development of Detection Assays for Cryptosporidium.

Cryptosporidium spp. are obligate, intracellular parasites that cause life-threatening diarrhea among children and immunocompromised adults. Transmission occurs by the fecal-oral route following ingestion of thick-walled oocysts that can contaminate, persist, and resist disinfection in water and food. Sodium hypochlorite, peroxides, ozone, formaldehyde, and ammonia are suitable disinfectants against Cryptosporidium oocysts. Effective concentrations of these chemicals can be toxic and not practical for downstream research use of non-viable oocysts. Oocyst inactivation approaches such as UV ligh…
