Absence of MMP2 mutation in idiopathic multicentric osteolysis with nephropathy.

The genetic basis of idiopathic multicentric osteolysis with nephropathy is unknown. This disorder is typically a sporadic, but sometimes an autosomal dominant, condition featuring carpal-tarsal destruction and nephropathy causing renal failure. Loss-of-function mutation within the gene encoding matrix metalloproteinase 2 (MMP2) causes the autosomal recessive disorder nodulosis-arthropathy-osteolysis syndrome characterized by carpal-tarsal destruction, subcutaneous nodules, and generalized osteoporosis. We questioned whether sporadic idiopathic multicentric osteolysis with nephropathy is allel…

The quality of osteolysis grafting with cementless acetabular component retention.

Periprosthetic osteolysis is a common cause for revision of total hip arthroplasty. When modular cementless acetabular components are stable, curettage and grafting of the osteo-lytic lesion while retaining the component are a good surgical option. Although the midterm outcome of this procedure is known, the quality of the surgical technique is not. We used preoperative and postoperative computed tomography to determine the percentage of periacetabular lesions that was grafted and the percentage of the lesion volume filled with an injectable bone graft substitute. We discovered, even with preo…

The value of anteroposterior pelvic radiographs for evaluating pelvic osteolysis.

We investigated the role of conventional radiographs and computed tomography scans for the routine followup of total hip arthroplasty patients. Among 92 total hip arthroplasties with a mean followup of 8.5 years, 94 acetabular lesions were detected among 63 hips using computed tomography and 42 of these hips had osteolysis diagnosed on radiograph. Using computed tomography as a gold standard, the sensitivity of anteroposterior pelvic radiographs for the detection of acetabular osteolysis was 67% and the specificity was 72%. Although smaller lesions were more frequently missed, osteolysis was d…

[Is osteolysis associated with a stable total hip replacement asymptomatic?].

PURPOSE OF THE STUDY: It has been reported that periprosthetic osteolysis in a stable cementless total hip prosthesis is often free from symptoms. The aim of this study is to check this assumption and provide details on the clinical presentation of this troublesome and worrying complication of hip arthroplasty. MATERIAL: Between March 1999 and June 2002, 76 hips in 72 patients underwent revision arthroplasty for periprosthetic osteolysis associated with stable ABG I prostheses. This patient group included 53 females and 19 males. The average age at the time of revision was 49 years (range, 32…

[Multifocal osteolysis as a result of delayed diagnosis for primary hyperparathyroidism].

Primary hyperparathyroidism caused by solitary adenomas occurs rarely (especially in children and adolescents). The clinical manifestations are usually subtle and that is why the mentioned disorder is usually late diagnosed and as an effect--late treated. We present the case of a 17 year old girl, an ambulatory patient who had been "observed" for over 12 months because of persistent ostealgia. She had not been properly diagnosed. The first diagnostic investigation of calcium and phosphate balance was provided only when multifocal osteolysis of tibias, hip and metacarpal bones was detected and …

Metacarpal fracture associated with lymphosarcoma-induced osteolysis in a horse.

A 19-year-old Appaloosa gelding was reluctant to move. Radiography revealed diffuse, permeative lysis of the cortex and subchondral bone of … vities were apparent in bone trabeculae contiguous with nodular foci of lymphosarcoma. Osteoclastic osteolysis was not evident at bone surfaces that were not directly adjacent to neoplastic cells. Although lymphosarcoma is the most frequent malignant neoplasia encountered in horses, diffuse neoplastic infiltration of cortical and subchondral bone of the appe …

[Contribution to the osteolysis in distal end of the clavicle (author's transl)].

A case of rare osteolysis of the distal clavicle end is reported. Our 36-years old male patients also suffered from a well-discernible Hyperlipoproteinemia, type II/a after Fredrickson. A short view of the various forms of clavicle osteolysis is given. Their differentialdiagnostical and ethiological relations are discussed.

Association of Somatic KRAS Variants with Osteolysis in Arteriovenous Malformations.

The genetic study of vascular anomalies provides a better understanding of their etiopathogenesis and allows the use of targeted therapies. Activating KRAS pathogenic variants promote cell proliferation by activating MAPK and PI3K signalling pathways, which have been associated with the pathogenesis of vascular anomalies, especially high-flow ones such as arteriovenous malformations (AVMs). AVMs' genomic landscape is extensive, and a genotype-phenotype correlation has not been shown. In this study, we aimed to prove an association between KRAS gene mutations and the presence of osteolysis in p…

Scoping review for treatment of osteolysis in total ankle arthroplasty.

BACKGROUND: Following total ankle replacement (TAR), osteolysis is a known complication. No consensus exists regarding management. METHODS: Applying the Arksey and O'Malley framework for scoping studies, a study protocol and a data extraction form was created. The research included all studies until March 2025. Study selection and data extraction were performed by two independent reviewers. RESULTS: Two hundred and eighty-seven studies were identified, of which 54 studies (case series and case reports) were included. 69% of the studies included below 10 cases. The most common reported treatm…

No osteolysis at the telescopic junction of 128 FITBONE lengthening nails.

BACKGROUND: Motorized lengthening nails are the treatment standard for bone lengthening of the lower limbs. However, bony changes namely osteolysis and periosteal hypertrophy have been described after certain type of magnetically driven lengthening nails. The aim of this study was to estimate the proportion rate of radiological bone abnormalities of Fitbone TAA femoral or tibial lengthening nails with a minimum follow-up time of 1 year. HYPOTHESIS: The bone surrounding the telescopic junction of Fitbone lengthening nails does not exhibit osteolysis or periosteal reactions. The bone surroundin…

KLF4 as a rheostat of osteolysis and osteogenesis in prostate tumors in the bone.

We previously showed that KLF4, a gene highly expressed in murine prostate stem cells, blocks the progression of indolent intraepithelial prostatic lesions into aggressive and rapidly growing tumors. Here, we show that the anti-tumorigenic effect of KLF4 extends to PC3 human prostate cancer cells growing in the bone. We compared KLF4 null cells with cells transduced with a DOX-inducible KLF4 expression system, and find KLF4 function inhibits PC3 growth in monolayer and soft agar cultures. Furthermore, KLF4 null cells proliferate rapidly, forming large, invasive, and osteolytic tumors when inje

Multifocal discrete osteolysis in a horse with silicate associated osteoporosis.

Silicate associated osteoporosis (SAO) was diagnosed post mortem in an adult horse with the shortest documented exposure to cytotoxic silica … SAO should be included as a differential diagnosis for horses with widespread, multifocal, discrete osteolysis and history of exposure to endemic regions with possible cytotoxic silicate inhalation. Exposure time of 2 years is potentially sufficient to develop SAO.

Clinical and mutation profile of multicentric osteolysis nodulosis and arthropathy.

Multicentric osteolysis nodulosis and arthropathy (MONA) is an infrequently described autosomal recessive skeletal dysplasia characterized by progressive osteolysis and arthropathy. Inactivating mutations in MMP2, encoding matrix metalloproteinase-2, are known to cause this disorder. Fifteen families with mutations in MMP2 have been reported in literature. In this study we screened thirteen individuals from eleven families for MMP2 mutations and identified eight mutations (five novel and three known variants). We characterize the clinical, radiographic and molecular findings in all individual…
