Endogenous oxalate synthesis in mouse models of metabolic dysfunction-associated steatotic liver disease.

Metabolic dysfunction-associated steatotic liver disease (MASLD) may lead to increased urinary oxalate excretion, a risk factor for calcium oxalate kidney stone formation, and progression of chronic kidney disease. We used mouse models of diet-induced obesity to examine the impact of MASLD on endogenous oxalate synthesis and urinary oxalate excretion. Mice were either fed a high-fat diet to induce mild MASLD, or a high-fat, sucrose, and cholesterol diet (Western diet) to produce metabolic dysfunction-associated steatohepatitis (MASH), a more severe form of MASLD. Liver tissue was collected to

Acute interstitial nephritis with oxalate deposition after hair‑straightening treatment: a case report.

BACKGROUND: Acute interstitial nephritis (AIN) is a recognized cause of acute kidney injury (AKI), most commonly associated with medications. Increasing evidence also implicates exposure to environmental chemicals, including hair‑straightening products marketed as "formaldehyde‑free," which may still contain or release nephrotoxic compounds. Reports have linked these products to oxalate nephropathy that may occur concurrently with interstitial nephritis, underscoring the need for awareness of their renal risks. CASE PRESENTATION: We describe a 26-year-old healthy woman who presented with naus

Enhanced bioethanol Production from Wheat Bran Feedstock by a Mild Oxalic Acid Pretreatment.

Organic acid pretreatment is one of the most promising methods to pretreat lignocellulose biomass due to its mild condition and weak corrosion to reactor. In this study, five different organic acids were used for wheat bran pretreatment, and the enzymatic hydrolysis yields of pretreated feedstocks were evaluated. Among these organic acids, oxalic acid pretreatment demonstrated the highest glucose yield of 87.93%. Subsequently, the optimization of oxalic acid pretreatment parameters was conducted including temperature, time, solid-liquid ratio, and acid usage. A high glucose yield of 96.92% was

Single-Atom Catalysts for CO2 Reduction to Oxalate: Theoretical Design and Reaction Condition Prediction.

The electrochemical conversion of carbon dioxide (CO2) into high-value-added products under mild conditions is crucial for achieving carbon neutrality. Oxalate (C2O42-) is one of the most important industrial raw materials and is widely used as a reducing agent in the fields of medicine, dyeing, and plastics yet faces challenges in efficient C-C bond formation under mild conditions. In this study, we investigate the reduction of CO2 to C2O42- using single-atom catalysts (SACs) with M-Nx-C configurations, employing density functional theory (DFT) to assess their catalytic performance under vary

Inhibition of NLRP3 alleviates calcium oxalate crystal-induced renal fibrosis and crystal adhesion.

Kidney stones are gaining attention as one of the most common urological diseases. In this study, we first constructed a mouse model of calcium oxalate (CaOx) crystal deposition by intraperitoneal injection of glyoxalate (Gly) and found that the levels of NLRP3, CASP1 and ASC, which constitute the NLRP3 inflammasome, as well as the level of its downstream product, IL-1β, were elevated in the kidneys of the model group of mice, as determined by RNA-seq. We then examined NLRP3 expression via immunohistochemistry, immunofluorescence, qPCR, and Western blotting in human samples, calcium oxalate mo

Metabolic Profile of Calcium Oxalate Stone Patients with Enteric Hyperoxaluria and Impact of Dietary Intervention.

This study investigated the risk profile and the impact of dietary intervention in calcium oxalate stone formers with enteric hyperoxaluria under controlled, standardized conditions. Thirty-seven patients were included in the study. Dietary and 24-h urinary parameters were obtained on the self-selected diet and a balanced, standardized diet. Tests for [13C2]oxalate absorption, calcium- and ammonium chloride-loading were performed. Mean [13C2]oxalate absorption was 18.8%. A significant positive association was observed between urinary oxalate excretion and intestinal oxalate absorption. In addi

Bidirectional Impact of Varying Severity of Acute Kidney Injury on Calcium Oxalate Stone Formation.

INTRODUCTION: Acute kidney injury (AKI) is a prevalent renal disorder. The occurrence of AKI may promote the formation of renal calcium oxalate stones by exerting continuous effects on renal tubular epithelial cells (TECs). We aimed to delineate the molecular interplay between AKI and nephrolithiasis. METHODS: A mild (20 min) and severe (30 min) renal ischemia-reperfusion injury model was established in mice. Seven days after injury, calcium oxalate stones were induced using glyoxylate (Gly) to evaluate the impact of AKI on the formation of kidney stones. Transcriptome sequencing was performe

Oxalate Oxidase for In Situ H2 O2 -Generation in Unspecific Peroxygenase-Catalysed Drug Oxyfunctionalisations.

H2 O2 -driven enzymes are of great interest for industrial biotransformations. Herein, we show for the first time that oxalate oxidase (OXO) is an efficient in situ source of H2 O2 for one of these biocatalysts, which is known as unspecific peroxygenase (UPO). OXO is reasonably robust, produces only CO2 as a by-product and uses oxalate as a cheap sacrificial electron donor. UPO has significant potential as an industrial catalyst for selective C-H oxyfunctionalisations, as we confirm herein by testing a diverse drug panel using miniaturised high-throughput assays and mass spectrometry. 33 out o

A Sodium Oxalate-Rich Diet Induces Chronic Kidney Disease and Cardiac Dysfunction in Rats.

Chronic kidney disease (CKD) is a worldwide public health issue affecting 14% of the general population. However, research focusing on CKD mechanisms/treatment is limited because of a lack of animal models recapitulating the disease physiopathology, including its complications. We analyzed the effects of a three-week diet rich in sodium oxalate (OXA diet) on rats and showed that, compared to controls, rats developed a stable CKD with a 60% reduction in glomerular filtration rate, elevated blood urea levels and proteinuria. Histological analyses revealed massive cortical disorganization, tubula

Urinary versus serum microRNAs in human oxalic acid poisoning: Contrasting signals and performance.

MicroRNAs are key regulators of the normal kidney function and development, and altered in acute kidney injury (AKI). However, there is a lack of studies comparing serum and urine miRNA expression in toxic AKI in humans. We aimed to compare the global signature of urinary and serum microRNAs, with and without kidney injury, after human oxalic acid poisoning. We profiled urinary microRNAs in patients who ingested oxalic acid and developed no injury (No AKI n = 3), moderate injury (AKIN2 n = 3) or severe injury (AKIN3 n = 3) and healthy controls (n = 3). We validated a signature of 30 urinary mi

Oxalic acid-mediated biochemical and physiological changes in the common bean-Sclerotinia sclerotiorum interaction.

The success of Sclerotinia sclerotiorum infection relies mainly on the production of the non-host selective toxin named oxalic acid (OA). This toxin is known to play multiple roles in a host infected by the fungus, but its effect on photosynthesis and the antioxidant system of common bean plants remain elusive. Therefore, we performed detailed analysis of leaf gas exchange, chlorophyll a fluorescence, activities of antioxidant enzymes, concentrations of reactive oxygen species and photosynthetic pigments to investigate the OA's role during the S. sclerotiorum pathogenesis. To achieve this goal

[Remediation of Cd/Ni Contaminated Soil by Biochar and Oxalic Acid Activated Phosphate Rock].

Taking soil contaminated with a combination of Cd and Ni as the research objective, biochar, and oxalic acid activated phosphate rock (APR) were applied both together and separately for the remediation of this contaminated soil. The effects of different ratios of amendments on the remediation of Cd and Ni heavy metals in the soil and on inorganic nitrogen and microbial biomass nitrogen (MBN) in the soil were compared. The results show that an increasing amount of biochar and APR, increases the soil pH gradually and acid-extractable Cd and Ni are gradually transformed into reducible, oxidable a

Calcium oxalate in the sputum may aid in the diagnosis of pulmonary aspergillosis: A report of two cases.

We present two cases of pulmonary aspergillosis in which calcium oxalate crystals in the sputum proved to be a useful diagnostic clue. In case 1, Aspergillus hyphae was not identified; however, calcium oxalate crystals were present, and chronic necrotizing pulmonary aspergillosis was diagnosed. In case 2, calcium oxalate was detected and Aspergillus fumigatus was identified later. Thus, the presence of calcium oxalate in the sputum may be an important indicator for an A. fumigatus infection.

Calcium oxalate nephrolithiasis: effect of renal crystal deposition on the cellular composition of the renal interstitium.

Urinary calcium oxalate (CaOx) crystals and crystal agglomerates are normally harmlessly excreted, but in nephrolithiasis they are retained by tubular epithelial cells and shifted into the renal interstitium. This crystalline material induces an inflammatory response consisting of an increase in the number of interstitial cells and an expansion of the extracellular matrix. The newly arrived cells either derive from the blood or the connective tissue or they are formed by local proliferation. Identification of the cells that surround the interstitial crystals is a first step in investigating th

Immunochemical detection of oxalate monoalkylamide, an ascorbate-derived Maillard reaction product in the human lens.

Carbohydrates with reactive aldehyde and ketone groups can undergo Maillard reactions with proteins to form advanced glycation end products. Oxalate monoalkylamide was identified as one of the advanced glycation end products formed from the Maillard reaction of ascorbate with proteins. In these experiments, we have analyzed human lens proteins immunochemically for the presence of oxalate monoalkylamide. Oxalate monoalkylamide was absent in most of the very young lenses but was present in old and cataractous lenses. The highest levels were found in senile brunescent lenses. Incubation experimen

[Indirect determination of trace amount of oxalic acid radical by the I2-CCl4 extraction spectrophotometry].

The article studies that in dilute H2SO4 dielectric substance IO3(-) reacts with C2O4(2-) to release certain amount of iodine under certain reaction condition: 2IO3(-) + 5C2O4(2-) + 12H+ = I2 + 10CO2 increasing + 6H2O therefore the method of indirect determination with I2-CCl4 extraction spectrophotometry is established. The reaction temperature is controlled in 90 degrees C; the reaction time 3 minutes; the reaction acidity pH1.0-2.0. The common ion basically does not affect the determination. The lowest detection limit of C2O4(2-) is 0.08 microg x mL(-1). After comparing the method of the re

Damage attributable to extracorporeal shockwave lithotripsy and deposition of calcium oxalate crystals on ureteral stents.

Ureteral stents have been widely used during and after SWL depending on the size of the stones and the condition of the urinary tracts. The use of stents can help to reduce complications and contributes to stone passage. However, some reports note complications that were attributed to indwelling ureteral stents. We attended to the role of obstruction of ureteral stents in those complications. The aim of this study was to analyze factors that influence ureteral stent obstruction and damage caused by SWL. Crystal deposition and the damage by SWL were examined in all ureteral stents by the use of

Calcium and oxalate uptake by the renal brush border membrane vesicles in magnesium-deficient rats.

Calcium and oxalate uptake by renal brush border membrane vesicles (BBMV) was examined in magnesium-deficient and pair-fed control rats. Uptake studies were carried out by rapid filtration technique and rate of influx of calcium and oxalate as a function of extravesicular concentration (0.1 nM--1.0 mM) examined. Calcium uptake by renal BBMV exhibited saturable kinetics while oxalate uptake followed a biphasic transport mechanism showing saturable kinetics at low oxalate concentrations and passive diffusion at higher concentrations. In magnesium deficiency the kinetics of calcium and oxalate up

Oxalate binding to rat intestinal brush-border membrane in pyridoxine deficiency: a kinetic study.

Oxalate bound specifically to the intestinal brush-border membrane (BBM) of pyridoxine-deficient rats, but not to BBM of control rats. The binding of oxalate to intestinal BBM of pyridoxine-deficient rats was rapid, reversible, dependent on concentration of oxalate, temperature sensitive and competitively inhibited by oxalate analogues. Kinetic analysis of the oxalate binding data revealed induction of two distinct classes of receptor site for oxalate. The high-affinity oxalate binding sites, reached saturation at 60-70 nM oxalate, had a Kd of 24.29 nM and the number of binding sites were 30 p
