Cortical responses to amplitude modulation in guinea pigs and the effects of general anesthesia by pentobarbital.

Test of amplitude modulation detection threshold against modulation frequency (Mf) is one of the major measures in behavior studies for expl … idth of peripheral auditory channels. We further evaluated the effects of the general anesthesia by pentobarbital on temporal acuity and found that, in addition to the suppressions of response amplitude, this anesthesia significantly decreases the -3dB cutoff frequencies of TMTF.

Comparison of cardiovascular effects of tiletamine-zolazepam, pentobarbital, and ketamine-xylazine in male rats.

We allocated 35 male Sprague-Dawley rats into 7 groups and anesthetized each by using one of the following regimens: ketamine 50 mg+xylaxine 5 mg; ketamine 75 mg+xylazine 5 mg; pentobarbital 45 mg; and Telazol 30, 40, 50, and 60 mg/kg; supplemental doses were used as required. Respiratory rate, heart rate, mean arterial pressure, cardiac index, and stroke index were measured every 30 min for 4 h. The Telazol groups showed a dose-dependent increase in duration of anesthesia. Duration of anesthesia was significantly shorter for the ketamine and pentobarbital groups than for any of the Telazol do…

Effects of isoflurane, pentobarbital, and urethane on apoptosis and apoptotic signal transduction in rat kidney.

BACKGROUND: Renal cell apoptosis contributes significantly to the pathogenesis of acute renal failure. Anesthetic agents have been shown to modulate apoptotic signal transduction in various tissues. We examined the effects of 6 h of different general anesthetic techniques on renal cell apoptosis in rat kidneys. METHODS: Twenty-one male Sprague-Dawley rats were randomly allocated into four groups: (i) control, non-anesthetized rats (n= 3) and rats anesthetized with (ii) inhaled isoflurane (n= 6), (iii) intraperitoneal pentobarbital (n= 6), and (iv) intraperitoneal urethane (n= 6). Animals were…

Postmortem effect of pentobarbital anesthetic on survival of adult cortical neurons in primary culture.

We determined whether pentobarbital anesthetic is required to culture postmortem adult rat neurons. Pentobarbital treatment resulted in two-fold increases in neuron survival in culture after 2 and 4 h postmortem compared to non-anesthetic controls, but was not as effective as simple postmortem treatment on ice and therefore not essential.

The inhibitory effects of ketamine and pentobarbital on substance p receptors expressed in Xenopus oocytes.

UNLABELLED: Substance P receptors (SPR) modulate nociceptive transmission within the spinal cord. The effects of IV anesthetics on SPR are not clear. In this study, we investigated the effects of IV anesthetics on SPR expressed in Xenopus oocytes. We examined the effects of ketamine, pentobarbital, propofol, and tramadol on SP-induced Ca(2+)-activated Cl(-) currents mediated by SPR expressed in Xenopus oocytes using a whole-cell voltage clamp. Ketamine and pentobarbital inhibited the SPR-induced currents at pharmacologically relevant concentrations, but propofol and tramadol had little effect …

Effect of pentobarbital, d-amphetamine, and nicotine on two models of sustained attention in pigeons.

RATIONALE: The animal literature examining the effects of drugs of abuse on sustained attention has provided conflicting results. One reason for these inconsistencies could be the different type of tasks used to measure sustained attention. OBJECTIVE: Acute effects of pentobarbital (0.3, 1, 3, 5.6, 10, and 13 mg/kg), d-amphetamine (0.03, 0.1, 0.3, 1, 3, and 5.6 mg/kg), and nicotine (0.03, 0.1, 0.3, 1, and 3 mg/kg) were compared in two models of sustained attention. METHODS: Dose-response curves were compared in eight male, white Carneaux pigeons trained under a continuous-trial attention pro…

Method for determination of pentobarbital in dry dog food by gas chromatography/mass spectrometry.

A procedure has been developed and validated for measuring the concentration of pentobarbital residues in dry, extruded animal feed in the range of 3-200 ng/g (ppb) with an estimated limit of quantitation of 2 ppb. The method was developed for surveillance purposes: to measure the concentration of euthanizing agent which might be present in feeds incorporating rendered products which themselves might include some fraction of euthanized animals. A previously published qualitative procedure was modified by adding isotopically labelled pentobarbital as an internal standard. Dry feed was ground an…

Structural domains of the human GABAA receptor 3 subunit involved in the actions of pentobarbital.

This study was conducted to search for the residues of the beta3 subunit which affect pentobarbital action on the gamma-aminobutyric acid type A (GABAA) receptor. Three chimeras were constructed by joining the GABAA receptor beta3 subunit to the rho1 subunit. For each chimera, the N-terminal sequence was derived from the beta3 subunit and the C-terminal sequence from the rho1 subunit, with junctions located between the membrane-spanning regions M2 and M3, in the middle of M2, or in M1, respectively. In receptors obtained by the coexpression of alpha1 with the chimeric subunits, in contrast wit…

Cocaine- and alcohol-mediated expression of inducible transcription factors is blocked by pentobarbital anesthesia.

Identifying the neurocircuitry involved in behavioral responses to drugs of abuse is an important step towards understanding the mechanisms of drug addiction. The present study sought to distinguish brain regions involved in pharmacological effects of cocaine and ethanol from secondary effects by administering these drugs in the presence or absence of pentobarbital anesthesia. Changes in neuronal activity were assessed by immunohistochemical analysis of expression of an inducible transcription factor (ITF), c-Fos, in the brain of rats habituated to repeated pentobarbital anesthesia or saline a…

Pentobarbital-morphine anesthetic interactions in terms of intensity of noxious stimulation required for arousal.

BACKGROUND: Previous reports suggest that the outcome (synergism, antagonism, summation) of opioid-barbiturate interactions may depend on the depth of anesthesia. One aim of the present study was to determine whether pentobarbital, alone and in combination with morphine, blocks awakening caused by noxious stimulation in a dose-related manner: the more intense the noxious stimulation, the more pentobarbital is required to suppress the response. A second aim of the study was to determine whether the pentobarbital-morphine anesthetic interaction depends on the depth of anesthesia measured in term…

Interactions of GABAA antagonists with deltamethrin, diazepam, pentobarbital, and SKF100330A in the rat dentate gyrus.

Activation of dentate gyrus granule cells leads to a period of depressed excitability mediated primarily by recurrent inhibition. This response can be increased in intensity and duration by drugs that enhance GABAA-mediated inhibition including GABA uptake blockers (SKF100330A), benzodiazepines (diazepam), and barbiturates (pentobarbital). It can be attenuated or abolished by administration of the GABAA antagonists picrotoxin and bicuculline. Type II pyrethroids, such as deltamethrin, produce effects in vitro which lead one to expect that they would decrease GABA-mediated inhibition. However, …

Effects of pentobarbital on proopiomelanocortin opioid products of neonatal piglets during normoxia and hypoxia.

Abstract Endogenous opioids have been shown to suppress physiological functions in the neonate. It has been suggested that anesthesia with barbiturates might enhance this suppression by influencing opioid systems directly. To explore this possibility, naive piglets, 2.2+/-0.8 (X+/-SD) days old, underwent one of five protocols: 1) normoxia (control); 2) 10% 0(2)/90% N(2) (hypoxia); 3) saline injection ip during normoxia (sham anesthesia); 4) pentobarbital sodium, 25 mg/kg ip, during normoxia (barbiturate anesthesia); and 5) pentobarbital sodium, 25 mg/kg ip, during hypoxia (combined hypoxia and…

Effects of thiopental, pentobarbital, and ketamine on endothelin-induced constriction of porcine cerebral arteries.

Contractile mechanisms of endothelin, a newly isolated vasoactive substance from endothelium, were evaluated in anterior cerebral arteries (ACA). Furthermore, the effects of thiopental, pentobarbital, ketamine, and diltiazem on the endothelin-induced cerebral vasoconstriction were also studied. Endothelin induced cerebral arterial contractions in concentrations above 3 X 10(-10) M. The median effective concentration (ED50: X10(-9) M) of endothelin was 2.1 +/- 0.7 (n = 6). Endothelin did not elicit contractions in preparations soaked in Ca2(+)-free solution, but addition of 2.5 mM Ca2+ to the b…

Pentobarbital and EEG burst suppression in treatment of status epilepticus refractory to benzodiazepines and phenytoin.

Seven patients with complex partial or secondarily generalized tonic-clonic status epilepticus (SE) refractory to benzodiazepines (BZDs) and phenytoin (PHT) were treated with pentobarbital (PTB) coma with an EEG burst suppression (BSP) pattern. PTB administered by continuous intravenous (i.v.) infusion pump at a loading dose of 6-8 mg/kg in 40-60 min was usually sufficient to produce BSP activity and seizure control. PTB was continued 0-24 h at 1-4 mg/kg/h, adjusted to maintain blood pressure (BP) and BSP. Infusion rate was decreased if systolic BP (SBP) was less than 90 mm Hg. Normal saline f…

Differential generalization to pentobarbital in rats trained to discriminate lorazepam, chlordiazepoxide, diazepam, or triazolam.

In drug discrimination studies benzodiazepine-trained animals have typically responded on the drug lever when tested with barbiturates. In a recent study, greater specificity appeared to be shown when lorazepam was used as a training drug. The generality and limits of this finding were explored in the present set of experiments. The asymmetrical cross-generalization found in lorazepam- and pentobarbital-trained baboons was replicated in rats and was shown not to be a function of either lorazepam (0.1., 0.32, or 1.0 mg/kg) or pentobarbital (10 or 25 mg/kg) training dose (i.e., pentobarbital-tra…

Differential effects of phenobarbital and pentobarbital on stimulus train-induced bursting in the hippocampal slice.

Stimulus train-induced bursting (STIB) in the hippocampal slice is an in vitro model of epileptiform activity that is analogous to kindling … es and anticonvulsants. In this study we assessed the effects of the barbiturates phenobarbital and pentobarbital on both spontaneous and stimulus-triggered bursts after STIB. Concentration-response studies indicated that both drugs had inhibitory effects on stimulus-triggered bursting, although at significantly different concentrations. The rate of spontaneous …

Comparison of the effects of valproate, ethosuximide, phenytoin, and pentobarbital on cerebral energy metabolism in the rat.

The acute effects of valproate (200 and 400 mg/kg), ethosuximide (200 and 400 mg/kg), phenytoin (25 and 50 mg/kg), and pentobarbital (30 and 60 mg/kg) on cerebral energy metabolism of rats were studied by measuring the cerebral content of energy metabolites and by evaluating the rate of metabolite utilization following decapitation. The treatments did not affect the levels of phosphocreatine (PCr), ATP, ADP, and AMP, but did enhance the glycogen or glucose stores. Pentobarbital induced a decrease in lactate, whereas valproate led to a decrease in pyruvate and an increase in lactate. Calculatio…

Facilitatory action of etomidate and pentobarbital on recurrent inhibition in rat hippocampal pyramidal neurons.

Biochemical studies have shown that the non-barbiturate anesthetic etomidate can interact in a stereoselective, barbiturate-like fashion with the GABA/benzodiazepine receptor complex, enhancing both benzodiazepine and GABA binding, but its electrophysiological effects upon the mammalian CNS are largely unknown. The present investigations were designed to characterize the electrophysiological effects of etomidate on the recurrent GABAergic inhibitory pathway in the CA1 region of the rat in vitro hippocampal slice and to compare the actions of etomidate to those of pentobarbital. Electrical stim…
