The current model for virulence gene regulation in Bacillus anthracis involves several trans-acting factors, the most important of which appears to be the anthrax toxin activator encoded by the atxA gene. AtxA is a positive regulator of the toxin genes pagA, cya and lef, and of a number of other plasmid- and chromosome-encoded genes. The AtxA protein (56 kDa) possesses a predicted winged-helix DNA-binding domain and phosphotransferase system-regulated domains, but the mechanism for positive regulation of AtxA target genes is not known. Sequence similarities in the promoter regions of AtxA-regu

The gene cut-1 from Neurospora encodes a protein of the haloacid dehalogenase (HAD) family, a group of enzymes usually associated to phosphatase or phosphotransferase activities. Loss of cut-1 function results in an osmosensitive phenotype and its transcription is induced by high osmotic pressure or heat-shock, but not by other stressing conditions tested. Unexpectedly, cut-1 transcript levels markedly decrease following illumination in the wild type and in some mutants tested, such as bd and vvd. The reduction was less pronounced in wc-1 and wc-2 mutants, indicating that this photoresponse is

We have used a host cell reactivation system to study the effect of 8-methoxypsoralen (8-MOP) reaction on CAT (chloramphenicol acetyltransferase) and NEO (aminoglycoside phosphotransferase) expression in normal human cells, as well as two cell lines with possible DNA repair-processing defects. Plasmid DNA was treated with psoralen plus near-ultraviolet (NUV) irradiation. The reacted plasmids, pSV2cat and pSV2neo, were transfected into Fanconi anemia (FA), xeroderma pigmentosum (XP), and normal human fibroblast cells for transient or stable assay. The cells were assayed for CAT activity at vari

We have studied the incorporation and interconversion of purines into nucleotides by freshly isolated Toxoplasma gondii. They did not synthesize nucleotides from formate, glycine, or serine. The purine bases hypoxanthine, xanthine, guanine, and adenine were incorporated at 9.2, 6.2, 5.1, and 4.3 pmol/10(7) cells/h, respectively. The purine nucleosides adenosine, inosine, guanosine, and xanthosine were incorporated at 110, 9.0, 2.7, and 0.3 pmol/10(7) cells/h, respectively. Guanine, xanthine, and their respective nucleosides labeled only guanine nucleotides. Inosine, hypoxanthine, and adenine l

Change of lipid metabolism in FL cells derived from human amniotic membrane during fusion from within induced by measles virus was studied. The following results were obtained. (1) Incorporation of [32P]orthophosphate and [methyl]-14C]choline into the lipid fraction of infected cells was higher than that of control cells. In this case, no detectable alteration of labeled phospholipid composition was observed. (2) Radioactivity of the lipids in subcellular fractions of infected-cells labeled with [1-14C]oleic acid for a short term also increased. (3) It was confirmed that specific enzyme activi

Previously, it has been found that phenobarbital inhibited protein kinase C (PKC) and the enzymes of the metabolism of polyphosphoinositide, especially phosphatidylinositol 4-phosphate (PIP) kinase (PIP-kinase). As a continuation of these studies, a number of barbiturates (barbituric acid, barbital, butabarbital, pentobarbital, amobarbital, phenobarbital, secobarbital and hexobarbital) were tested for inhibition of these enzymes and also of phosphatidylinositol (PI) kinase (PI-kinase), in a synaptosomal preparation at pH 7.8 from the brain of rat. All compounds, except barbituric acid (and Na-

The bacterial PASTA kinase, IreK, is required for intrinsic cephalosporin resistance in the Gram-positive opportunistic pathogen, Enterococcus faecalis. IreK activity is enhanced in response to cell wall stress, such as cephalosporin exposure. The downstream consequences of IreK activation are not well understood in E. faecalis, but recent work in other low-GC Gram-positive bacteria demonstrated PASTA kinase-dependent regulation of MurAA, an enzyme that performs the first committed step in the peptidoglycan synthesis pathway. Here, we used genetic suppressor selections to identify MurAA as a d

PMM2-CDG is the most common defect among the congenital disorders of glycosylation. In order to investigate the effect of hypoglycosylation on important cellular pathways, we performed extensive biochemical studies on skin fibroblasts of PMM2-CDG patients. Among others, acylcarnitines, amino acids, lysosomal proteins, organic acids and lipids were measured, which all revealed significant abnormalities. There was an increased expression of acylcarnitines and amino acids associated with increased amounts of calnexin, calreticulin and protein-disulfid-isomerase in combination with intensified amo

Receptor-like kinases (RLKs) constitute the largest receptor family involved in the regulation of plant immunity and growth, but small-molecule inhibitors that target RLKs to improve agronomic traits remain unexplored. The RLK member FERONIA (FER) negatively regulates plant resistance to certain soil-borne diseases that are difficult to control and cause huge losses in crop yields and economy. Here, we identified 33 highly effective FER kinase inhibitors from 1494 small molecules by monitoring FER autophosphorylation in vitro. Four representative inhibitors (reversine, cenisertib, staurosporin

The aim of this study was to investigate the clinical features and DGUOK gene mutations of an infant with mitochondrial DNA depletion syndrome (MDS). The patient (more than 7 months old) manifested as hepatosplenomegaly, abnormal liver function, nystagmus and psychomotor retardation. Genetic DNA was extracted from peripheral blood samples of the patient and her parents. Targeted Exome Sequencing was performed to explore the genetic causes. Sanger sequencing was carried out to confirm the detected mutations. The sequencing results showed that the patient was a compound heterozygote for c.679G>A

Poly- and perfluoroalkyl substances (PFAS) are pollutants disrupting lipid metabolism and cell membrane integrity with unclear molecular mechanisms. Molecular docking and molecular dynamics (MD) simulations were used to investigate whether the perfluorinated analogue of palmitic acid (PFHxDA, parametrized through SwissParam) could enter membrane lipid metabolism and get embedded in cell membranes, possibly interfering with their properties. The possible role of PFHxDA as substrate for enzymes involved in the biosynthesis of membrane lipids was studied by investigating its interaction with two

Tunicamycins are potent inhibitors of dolichyl-phosphate N -acetylglucosamine phosphotransferase (DPAGT1) but are unsuitable for therapeutic development due to non-selective cytotoxicity, acid-labile glycosidic linkages, and poor physicochemical properties. Although prior structural modifications reduced the promiscuous toxicity of tunicamycins, the intrinsic 11'-β-1″-α trehalose-type glycosidic linkage remains chemically unstable, limiting biological durability. Here, we report a rationally engineered scaffold-stabilization strategy in which the acid-labile linkage is replaced with a chemical

Skin wound infections persist through a self-reinforcing "infection-inflammation" cycle that compromises tissue repair. Here, we demonstrate that surface chirality of nanomaterials serves as an independent parameter to disrupt this loop through targeted metabolic interference. Using gold nanoparticles (GNPs) as a model, we show that D-chiral GNPs (GNP-D) exhibit superior efficacy over their L-enantiomers (GNP-L) in repressing bacterial basal metabolism and impeding near-surface motility. Integration of transcriptomic profiling and molecular dynamics identifies the mannitol-specific phosphotran

The treatment of orthopedic implant-associated infections (IAIs) presents significant challenges due to increasing drug resistance. This study reports a virulence-selective trap-capture-kill antibacterial system for effectively treating persistent infections while promoting local bone tissue regeneration by regulating disordered immune and metabolic microenvironments caused by IAIs. Here, this work fabricates a nanostructured antibacterial system (F-14-TNA) based on a TiO2 nanotube array modified with a cationic pentacyclic coplanar backbone, fascaplysin derivative 14 (F-14). The modified surf

Staphylococcus aureus is an opportunistic pathogen, a member of the ESKAPE group, associated with nosocomial infections and foodborne illnesses due to its production of various toxins. This study conducted a comprehensive genomic characterization of S. aureus isolates producing toxic shock syndrome toxin (TSST-1) using a comparative genomics and bioinformatics approach. A total of 166, including 3 bovine mastitis isolates and 163 public genomes, were analyzed. Twenty-eight distinct sequence types (STs) were identified, with ST30 and ST5 being the most prevalent, corresponding to the clonal com

Mycobacterium tuberculosis (Mtb) is a crucial and destructive intracellular pathogen responsible for causing tuberculosis (TB), a disease of substantial morbidity and mortality. Mtb capsular polysaccharides can misdirect the host's immune response pathways, resulting in additional challenges in TB treatment. These capsule polysaccharides are biosynthesized by a series of stealth proteins including CpsY. Our prior investigations elucidated the structural and functional information of the central domain (aa 201-520) of CpsY within Mtb. However, within the host milieu, it is the full-length itera

Listeria monocytogenes, a significant foodborne pathogen, is known for its remarkable adaptability to diverse environments through a genomic regulatory network that coordinates metabolic activities and stress responses. However, many of these genomic elements remain poorly understood. This study investigates the role of deoRF, a previously understudied member of the DeoR-family, in oxidative tolerance, intracellular infection, and virulence. Interestingly, the F2365ΔdeoRF strain showed no significant growth defects in minimal media with glucose, fructose, or sucrose, suggesting that DeoRF does

The selenoprotein family includes 25 members, many of which are antioxidant or redox regulating enzymes. A unique member of this family is Selenoprotein I (SELENOI), which does not catalyze redox reactions, but instead is an ethanolamine phosphotransferase (Ept). In fact, the characteristic selenocysteine residue that defines selenoproteins lies far outside of the catalytic domain of SELENOI. Furthermore, data using recombinant SELENOI lacking the selenocysteine residue have suggested that the selenocysteine amino acid is not directly involved in the Ept reaction. SELENOI is involved in two di
