Terpenes: natural compounds found in plants as potential senotherapeutics targeting senescent mesenchymal stromal cells and promoting apoptosis.

BACKGROUND: Senescence in stem cells and progenitor cells can be particularly detrimental because these cells are essential for tissue renewal and overall organismal homeostasis. In mesenchymal stromal cells (MSCs), which comprise a heterogeneous mix of stem cells, progenitors, fibroblasts, and other stromal cells, senescence poses a significant challenge, as it impairs their ability to support tissue repair and maintenance. This decline in regenerative capacity can contribute to aging-related diseases, impaired wound healing, and degenerative disorders. One hallmark of senescence is resistanc

Crude Plant Extracts and Their Anti-Inflammatory Potential in Oral Inflammatory Cell Models: A Systematic Review of In Vitro Studies.

Plants are a rich source of bioactive compounds with broad pharmaceutical potential, particularly for their anti-inflammatory properties. Oral inflammation underlies many local and systemic diseases, yet conventional anti-inflammatory drugs have adverse effects. Crude plant extracts offer promising, safer alternatives. This systematic review synthesizes evidence on the anti-inflammatory activity of whole plant extracts in vitro oral models of inflammation. It also highlights methodological considerations for improved reproducibility. PubMed searches following PRISMA guidelines identified studi

A culture method with berbamine, a plant alkaloid, enhances CAR-T cell efficacy through modulating cellular metabolism.

Memory T cells demonstrate superior in vivo persistence and antitumor efficacy. However, methods for manufacturing less differentiated T cells are not yet well-established. Here, we show that producing chimeric antigen receptor (CAR)-T cells using berbamine (BBM), a natural compound found in the Chinese herbal medicine Berberis amurensis, enhances the antitumor efficacy of CAR-T cells. BBM is identified through cell-based screening of chemical compounds using induced pluripotent stem cell-derived T cells, leading to improved viability with a memory T cell phenotype. Humoral and cell-mediated immune responses to plant-produced African horse sickness virus VP7 quasi-crystals.

African horse sickness (AHS) is a devastating viral disease affecting equines and has resulted in many disastrous epizootics. To date, no successful therapeutic treatment exists for AHS, and commercially used live-attenuated vaccines have various undesirable side effects. Previous studies have shown that mice inoculated with insoluble African horse sickness virus (AHSV) VP7 crystals are protected from live challenge with a lethal dose of AHSV. This study investigates the humoral and cell-mediated immune responses in guinea-pigs to a safer monovalent vaccine alternative based on AHSV-5 VP7 quas

The coordination of guard-cell autonomous ABA synthesis and DES1 function in situ regulates plant water deficit responses.

INTRODUCTION: Drought stress triggers the synthesis and accumulation of the phytohormone abscisic acid (ABA), which regulates stomatal aperture and hence reducing plant water loss. Hydrogen sulfide (H2S), which is produced by the enzyme L-cysteine desulfhydrase 1 (DES1) that catalyzes the desulfuration of L-cysteine in Arabidopsis, also plays a critical role in the regulation of drought-induced stomatal closure. However, little is known about the regulation of DES1 or the crosstalk between H2S and ABA signaling in response to dehydration. OBJECTIVES: To demonstrate the potential crosstalk bet

Cell-length heterogeneity: a population-level solution to growth/virulence trade-offs in the plant pathogen Dickeya dadantii.

Necrotrophic plant pathogens acquire nutrients from dead plant cells, which requires the disintegration of the plant cell wall and tissue structures by the pathogen. Infected plants lose tissue integrity and functional immunity as a result, exposing the nutrient rich, decayed tissues to the environment. One challenge for the necrotrophs to successfully cause secondary infection (infection spread from an initially infected plant to the nearby uninfected plants) is to effectively utilize nutrients released from hosts towards building up a large population before other saprophytes come. In this s

Plant-derived antiviral drugs as novel hepatitis B virus inhibitors: Cell culture and molecular docking study.

Despite high anti-HBV efficacies, while the nucleoside analogs (e.g., lamivudine) lead to the emergence of drug-resistance, interferons (e.g., IFN-α causes adverse side-effects. Comparatively, various natural or plant products have shown similar or even better efficacy. Hence, new antiviral strategies must focus not only on synthetic molecules but also on potential natural compounds. In this report, we have combined the in vitro cell culture and in silico molecular docking methods to assess the novel anti-HBV activity and delineate the inhibitory mechanism of selected plant-derived pure compou

Characterization of a chemically modified plant cell culture expressed human α-Galactosidase-A enzyme for treatment of Fabry disease.

Fabry disease is an X-linked recessive disorder caused by the loss of function of the lysosomal enzyme α-Galactosidase-A. Although two enzyme replacement therapies (ERTs) are commercially available, they may not effectively reverse some of the Fabry pathology. PRX-102 is a novel enzyme for the therapy of Fabry disease expressed in a BY2 Tobacco cell culture. PRX-102 is chemically modified, resulting in a cross-linked homo-dimer. We have characterized the in-vitro and in-vivo properties of PRX-102 and compared the results with the two commercially produced α-Galactosidase-A enzymes. Results sho

Plant defense responses in opium poppy cell cultures revealed by liquid chromatography-tandem mass spectrometry proteomics.

Opium poppy (Papaver somniferum) produces a diverse array of bioactive benzylisoquinoline alkaloids, including the narcotic analgesic morphine and the antimicrobial agent sanguinarine. In contrast to the plant, cell cultures of opium poppy do not accumulate alkaloids constitutively but produce sanguinarine in response to treatment with certain fungal-derived elicitors. The induction of sanguinarine biosynthesis provides a model platform to characterize the regulation of benzylisoquinoline alkaloid pathways and other defense responses. Proteome analysis of elicitor-treated opium poppy cell cult

Production of glucocerebrosidase with terminal mannose glycans for enzyme replacement therapy of Gaucher's disease using a plant cell system.

Gaucher's disease, a lysosomal storage disorder caused by mutations in the gene encoding glucocerebrosidase (GCD), is currently treated by enzyme replacement therapy using recombinant GCD (Cerezyme) expressed in Chinese hamster ovary (CHO) cells. As complex glycans in mammalian cells do not terminate in mannose residues, which are essential for the biological uptake of GCD via macrophage mannose receptors in human patients with Gaucher's disease, an in vitro glycan modification is required in order to expose the mannose residues on the glycans of Cerezyme. In this report, the production of a r

Engineered genomic attachment sites for site-specific recombinases enable high-efficiency integration in plants and human cells.

Here we developed a DNA-centric strategy for optimizing site-specific recombination by rationally engineering chimeric attachment sites. The high-activity att variants enhance Bxb1-mediated integration efficiency in human cells and plants. Among these att variants, the engineered attB(V111) site achieved 51.9% integration efficiency in HEK293T cells (1.7-fold versus wild-type attB) and 35.6% in rice protoplasts (4.4-fold versus wild-type attB). When paired with an engineered single protein mutant in the Bxb1 catalytic domain, the optimized system achieved targeted integration efficiencies of 3

Single-cell RNA-seq uncovers landscape of tomato metabolic rewiring mediated by plant growth-promoting rhizobacteria.

Tomato contains various classes of metabolites conferring benefits to human health and plant defense. Metabolic reprogramming of tomato induced by plant growth-promoting rhizobacteria (PGPR), such as Streptomyces spp., provides a feasible solution to enhance fruit quality and plant performance. However, the spatiotemporal and intensity characteristics of plant metabolism rewired following PGPR inoculation remain largely uncovered. Here, we characterized the landscape of PGPR-mediated metabolic reprogramming in whole plants of tomato. Large-scale single-cell transcriptomic and metabolomic analy

Cell-specific-resolved coordination of mitochondrial metabolism and DNA damage responses underpins aluminium responses in plant roots.

Aluminium (Al) toxicity is a major constraint on crop productivity in acidic soils (pH < 5.5), severely impairing root growth and function. While organic acid (OA)-mediated chelation and DNA damage response (DDR)-mediated control of cell division are recognized as key tolerance mechanisms, their integration at the cellular level remains poorly understood. Here, we employed Translating Ribosome Affinity Purification (TRAP) to achieve cell-type-specific gene expression profiling of Arabidopsis thaliana roots, enabling the dissection of molecular responses in whole roots, the quiescent centre, an

Extracellular vesicles released by in vitro plant cell cultures: emerging systems for vesicle-mediated communication and biomedical translation.

This article summarizes how extracellular vesicles are emerging as important means through which cells communicate with each other in plants and their potential medical applications. Extracellular vesicles are emerging as important mediators of intercellular communication in plants, with roles in defense responses, stress adaptation, and cross-kingdom molecular signaling. Although plant-derived extracellular vesicles have attracted increasing interest, most studies have focused on vesicles isolated from whole tissues or edible plant materials, whereas extracellular vesicles released by in vitr
