Population genomics and connectivity of Vazella pourtalesii sponge grounds of the northwest Atlantic with conservation implications of deep sea vulnerable marine ecosystems.

Sponges are key ecosystem engineers that shape, structure and enhance the biodiversity of marine benthic communities globally. Sponge aggregations and reefs are recognized as vulnerable marine ecosystems (or VMEs) due to their susceptibility to damage from bottom-contact fishing gears. Ensuring their long-term sustainability, preservation, and ecosystem functions requires the implementation of sound scientific conservation tools. Here, the genetic diversity, structure, and connectivity of the deep-sea glass sponge, Vazella pourtalesii (Schmidt, 1870), was investigated using 1,102 neutral SNPs

Enriching cancer pharmacology with drugs of marine origin.

Marine natural products have proven, over the last half-century, to be effective biological modulators. These molecules have revealed new targets for cancer therapy as well as dissimilar modes of action within typical classes of drugs. In this scenario, innovation from marine-based pharmaceuticals has helped advance cancer chemotherapy in many aspects, as most of these are designated as first-in-class drugs. Here, by examining the path from discovery to development of clinically approved drugs of marine origin for cancer treatment-cytarabine (Cytosar-U®), trabectedin (Yondelis®), eribulin (Hal

Diacarnusins A-F, nor-Terpene Endoperoxides of the Red Sea Sponge Diacarnus erythraeanus.

Six new nor-terpene endoperoxides, diacarnusins A-F (1-6), along with five known ones (7-11), were isolated from the extract of the Red Sea sponge Diacarnus erythraeanus, which presented cytotoxic activity against the colon carcinoma cell line. The structure elucidation was performed by analysis of mass spectrometric and 1D and 2D NMR data. The relative and absolute configurations of the stereocenters of the compounds were deduced based on the Capon and Macleod empirical rule and comparison of the sign of their specific rotations with those of endoperoxides with proven absolute configurations.

Antimicrobial activity of some bacteria isolated from marine sponge in Neom region, Saudi Arabia.

Antimicrobial resistance reduces the possibility of avoiding and curing infectious diseases triggered by bacteria, viruses, parasites, and fungi. Therefore, searching for new antimicrobials from prolific producers of novel natural products, including marine sponges, is necessary to address this resistance. Marine sponges are known to host diverse microbial communities, many of which are prolific producers of bioactive secondary metabolites. Several biological activities are exhibited by bioactive metabolites extracted from marine sponges. Mounting research demonstrates that these secondary met

Design, Synthesis, and Antibacterial Activity of Long-Chain Alkyl-Substituted DemethyloxyAaptamine Derivatives.

Demethyloxyaaptamine, isolated from the marine sponge Aaptos aaptos, features a 1H-benzo[de][1,6]naphthyridine core and exhibits potent antibacterial activity. To systematically investigate its underexplored antibacterial properties and facilitate structural optimization, we constructed a focused library of 28 C-3 alkylamino-substituted derivatives of demethyloxyaaptamine via regioselective functionalization. In vitro evaluation against Staphylococcus aureus revealed that several derivatives possess minimum inhibitory concentrations (MICs) superior to vancomycin. Structure-activity relationshi

Exploring Antibacterial Properties of Marine Sponge-Derived Natural Compounds: A Systematic Review.

The rise in multidrug-resistant (MDR) bacteria has prompted extensive research into antibacterial compounds, as these resistant strains compromise current treatments. This resistance leads to prolonged hospitalization, increased mortality rates, and higher healthcare costs. To address this challenge, the pharmaceutical industry is increasingly exploring natural products, particularly those of marine origin, as promising candidates for antimicrobial drugs. Marine sponges, in particular, are of interest because of their production of secondary metabolites (SM), which serve as chemical defenses a

Ammonium nitrogen and phosphorus removal by bacterial-algal symbiotic dynamic sponge bioremediation system in micropolluted water: Operational mechanism and transformation pathways.

Construct a bacteria-algae symbiotic dynamic sponge bioremediation system to simultaneously remove multiple pollutants under micro-pollution conditions. The average removal efficiencies of NH4+-N, PO43--P, total nitrogen (TN), and Ca2+ were 98.35, 78.74, 95.64, and 84.92 %, respectively. Comparative studies with Auxenochlorella sp. sponge and bacterial sponge bioremediation system confirmed that NH4+-N and TN were mainly removed by bacterial heterotrophic nitrification - aerobic denitrification (HN-AD). PO43--P was removed by algal assimilation and the generation of Ca3(PO4)2 and Ca5(PO4)3OH,

Natural Product Neopeltolide as a Cytochrome bc1 Complex Inhibitor: Mechanism of Action and Structural Modification.

The marine natural product neopeltolide was isolated from a deep-water sponge specimen of the family Neopeltidae. Neopeltolide has been proven to be a new type of inhibitor of the cytochrome bc1 complex in the mitochondrial respiration chain. However, its detailed inhibition mechanism has remained unknown. In addition, neopeltolide is difficult to synthesize because of its very complex chemical structure. In the present work, the binding mode of neopeltolide was determined for the first time by integrating molecular docking, molecular dynamics simulations, and molecular mechanics Poisson-Boltz

The presynaptic effect of fractions isolated from the sponge Tedania ignis.

Two fractions from the sponge Tedania ignis which have presynaptic effects were isolated. In neuromuscular junctions of Rana pipiens, the crude fraction decreases the amplitude of the evoked endplate potential and increases the frequency of miniature endplate potentials (MEPP), without effects on their amplitude or shape. Elution with 1 M acetic acid through Sephadex G15 produces three peaks, only one of which contains the biological activity. The compounds in this peak possess a molecular weight close to 900. Elution with 1M acetic acid through BioGel P2 produces 9 peaks, only 2 of which are

Isolation and Characterization of Glycosylated Fatty Acid Amides from the Norwegian Deep-Sea Sponge Phakellia sp.

Marine organisms from deep-sea environments have attracted considerable attention in drug discovery because they produce structurally diverse natural products. However, Norwegian deep-sea ecosystems remain largely unexplored in terms of natural product chemistry and bioactivity. In this study, a deep-sea sponge belonging to the genus Phakellia was selectively collected using a low-impact, remotely operated vehicle approach from the Mohn's Treasure area in the Norwegian Sea at a depth of 2858 m, representing one of the deepest sponge samples investigated in Norwegian waters to date. Chemical in

Identification of marine methylotrophic bacteria cultured from cold-water coral reefs.

Cold-water coral reefs host diverse microbial communities that play key roles in nutrient cycling and deep-sea productivity. Among them, methylotrophic bacteria metabolize and cycle one-carbon (C1) compounds such as methane, methanol, methylated nitrogen and sulphur, linking food webs and having a central role in the global cycling of climate-active trace gases. To date, few methylotrophic bacteria have been cultured from cold-water coral reefs. In this study, we enriched methylotrophs from seawater, sediment and sponge samples collected in cold-water coral reefs off mid-Norway. Cultivation at
