Interleukin-17A plays a key role in pulmonary fibrosis following Propionibacterium acnes-induced sarcoidosis-like inflammation.

Sarcoidosis is a granulomatous disease of unknown etiology, with limited therapeutic options. Chronic sarcoidosis can result in pulmonary fi … otic manner. Wild-type and IL-17A-knockout C57BL/6 mice were repeatedly challenged with heat-killed Propionibacterium acnes (PA) to induce sarcoidosis-like granulomata and sarcoidosis-related pulmonary fibrosis. Wild-type mice with granulomatosis were treated with anti-IL-17A antibody. Administration of PA enhanced the expression of IL-17A, granulomatosis, and fibrosis i …

Topical application of phenolic compounds suppresses Propionibacterium acnes-induced inflammatory responses in mice with ear edema.

Acne vulgaris (AV), a severe chronic inflammatory dermatosis, commonly treated with systemic or topical antibiotics that exacerbate bacterial resistance and pose adverse side effects, new approaches for suppressing or reducing Propionibacterium acnes-induced inflammatory responses and thereby treating AV remain necessary. In response, the goal of our study was to investigate the therapeutic potential of phenolic compounds in the in vivo inflammatory process induced by P. acnes. Mice were intradermally challenged with a suspension containing 1.0 × 107 CFU/mL of P. acnes per ear, after which gro…

Short lipopeptides specifically inhibit the growth of Propionibacterium acnes with dual antibacterial and anti-inflammatory action.

Источник пометил публикацию как отозванную

BACKGROUND AND PURPOSE: Propionibacterium acnes (P. acnes) is a Gram-positive bacterium associated with the skin disorder acne. In this study, we determined the importance of fatty acids in the life habitat of P. acnes; we tested our lipopeptide library in an attempt to create potent P. acnes-specific antimicrobial agents. EXPERIMENTAL APPROACH: Antimicrobial activity was determined by the minimal inhibitory concentration (MIC). Lipids from P. acnes were used to explore the mode of action. RAW264.7 cells respectively stimulated with LPS and P. acnes were used to measure the anti-inflammatory …

Control of Propionibacterium acnes by natural antimicrobial substances: Role of the bacteriocin AS-48 and lysozyme.

We report the high susceptibility of several clinical isolates of Propionibacterium acnes from different sources (skin, bone, wound exudates, abscess or blood contamination) to the head-to-tail cyclized bacteriocin AS-48. This peptide is a feasible candidate for further pharmacological development against this bacterium, due to its physicochemical and biological characteristics, even when it is growing in a biofilm. Thus, the treatment of pre-formed biofilms with AS-48 resulted in a dose- and time-dependent disruption of the biofilm architecture beside the decrease of bacterial viability. Furt…

Subclinical Propionibacterium acnes infection estimation in the intervertebral disc (SPInE-ID): protocol for a prospective cohort.

INTRODUCTION: Low back pain and vertebral endplate abnormalities are common conditions within the population. Subclinical infection caused by indolent pathogens can potentially lead to these findings, with differentiation between them notably challenging from a clinical perspective. Progressive infection of the intervertebral disc has been extensively associated with increasing low back pain, with Propionibacterium acnes specifically implicated with in relation to sciatica. The main purpose of this study is to identify if the presence of an infective pathogen within the intervertebral disc is …

Inhibitory effects of bee venom on Propionibacterium acnes-induced inflammatory skin disease in an animal model.

Propionibacterium acnes (P. acnes) is a major contributing factor to the inflammatory component of acne. The many prescription medications for acne allow for a large number of potential combination treatments. However, several antibiotics, apart from their antibacterial effects, exert side‑effects, such as the suppression of host inflammatory responses. Purified bee venom (BV) is a natural toxin produced by honeybees (Apis mellifera L.). BV has been widely used as a traditional medicine for various diseases. In the present study, to investigate the therapeutic effects of BV against P. acnes-in…

In vitro evaluation of the efficacy of chicken egg yolk antibodies (IgY) generated against Propionibacterium acnes.

UNLABELLED: Acne vulgaris driven by multiple factors appears in almost two-third of the population. Increased resistance of the bacterium to antibiotics and cost-effective treatments have made us resort to alternative therapy. OBJECTIVE: A novel method of targeting acne-causing bacterium Propionibacterium acnes with polyclonal chicken egg yolk antibodies was studied. METHODS: Anti-Propionibacterium acnes antibodies were generated in white leghorn chickens by immunizing with antigen. The antibodies extracted from the egg yolk by polyethylene glycol method were subjected to the analysis. The a…

Localization of propionibacterium acnes in granulomas supports a possible etiologic link between sarcoidosis and the bacterium.

Sarcoidosis likely results from the exposure of a genetically susceptible subject to an environmental agent, possibly an infectious one. Mycobacterial and propionibacterial organisms are the most commonly implicated potential etiologic agents. Propionibacterium acnes is the only microorganism, however, found in sarcoid lesions by bacterial culture. To evaluate the pathogenic role of this indigenous bacterium, we screened for the bacterium in sarcoid and non-sarcoid tissues using immunohistochemical methods with novel P. acnes-specific monoclonal antibodies that react with cell-membrane-bound l…

Correlation between Antimicrobial Resistant Propionibacterium acnes and Severity of Acne Vulgaris: Findings from a Tertiary Hospital.

The study of antimicrobial-resistant Propionibacterium acnes was not conducted regularly, especially in Indonesia. Conversely, regular monitoring of antibiotic efficacy through in vitro testing to assess the evolution of current resistance patterns is obligated; thus, filling the gap caused by a lack of appropriate antibiotic surveillance is required. Analyse the correlation between resistance patterns of P. acnes to doxycycline, clindamycin, erythromycin and azithromycin with the severity of acne vulgaris. This is an analytic observational laboratory study with a cross-sectional design of mil…

Analysis of complete genomes of Propionibacterium acnes reveals a novel plasmid and increased pseudogenes in an acne associated strain.

The human skin harbors a diverse community of bacteria, including the Gram-positive, anaerobic bacterium Propionibacterium acnes. P. acnes has historically been linked to the pathogenesis of acne vulgaris, a common skin disease affecting over 80% of all adolescents in the US. To gain insight into potential P. acnes pathogenic mechanisms, we previously sequenced the complete genome of a P. acnes strain HL096PA1 that is highly associated with acne. In this study, we compared its genome to the first published complete genome KPA171202. HL096PA1 harbors a linear plasmid, pIMPLE-HL096PA1. This is t…

[A comparison of biochemical and genetic classification of propionibacterium acnes strains isolated from skin lesions of patients with acne].

INTRODUCTION: Propionibacterium acnes is dominating group of resident bacteria in skin biocenosis. These bacteria participate in autosterilisation of skin with the process of decomposition of triglycerides into free fatty acids and by keeping the pH of skin on the level 5.5. When the process goes out of control the excess of fatty acids in sebaceous glands leads to necrosis and inflammation. Apart of the presence on the skin P acnes also are present in mucous membranes of intestinal tract, eyes, internal ears channels, and in upper respiratory tracts. Traditionally they are regarded as anaerob…

In vitro activities of azole antifungal agents against Propionibacterium acnes isolated from patients with acne vulgaris.

The Gram-positive bacterium Propionibacterium acnes is the causative agent of acne vulgaris. Antibiotics such as tetracycline and macrolide derivatives are used to treat this skin disease; however, the isolation frequency of antibiotic-resistant P. acnes has been increasing. The anti-P. acnes activity of imidazole antifungal agents was reported more than 20 years ago, and since then, new azole antifungal agents have been marketed. Thus, this study determined the in vitro activities of azole antifungal agents against P. acnes isolated from patients with acne vulgaris. Of the five agents tested,…

Resolution of inflammatory acne vulgaris may involve regulation of CD4+ T-cell responses to Propionibacterium acnes.

BACKGROUND: Propionibacterium acnes has been strongly implicated in inflammatory acne. However, its role in the disease is unclear. It has been hypothesized that an immune response to P. acnes and/or P. acnes heat shock proteins (HSPs) may play a role in the pathogenesis of inflammatory acne. OBJECTIVES: To compare the cell-mediated immune response to P. acnes and HSPs in acne patients, nonacne controls and individuals with resolved acne. METHODS: The proliferative response of peripheral blood mononuclear cells (PBMC) from acne patients, resolved acne donors and healthy controls to P. acnes,…

Effect of α-terpinolene on Propionibacterium acnes-induced hyperkeratosis and inflammation in vitro and in vivo.

Acne can impair patients' appearance and even cause psychological disorders such as anxiety and depression. Given the lipase inhibitory activity of the Pinus massoniana L. needles (PMLN) volatile oil, the present study explores the anti-P. acnes efficacy of the PMLN volatile essential oils and the related molecular mechanisms. Common ingredients in the PMLN volatile oil were screened and their anti-P. acnes effects were assessed by MIC test. P. acnes-induced HaCaT cells and mouse acne models validated the efficacy of an active ingredient with strong anti-P. acnes properties for acne treatment,
