Biological effects of low-dose ionizing radiation exposure on interventional cardiologists.

BACKGROUND: Interventional cardiologists (ICs) are likely to receive high radiation exposure as a result of procedures they undertake. AIMS: To assess the effects of low-dose X-ray radiation exposure on chromosomal damage and on selected indices of cellular and humoral immunity in ICs. METHODS: The study population consisted of 37 ICs and 37 clinical physicians as the control group with similar age, sex and duration of employment, without any work-related exposure to ionizing radiation. Cytogenetic studies were performed by chromosome aberration analysis and immunological studies by flow cyt

Ionizing radiation downregulates ASPM, a gene responsible for microcephaly in humans.

Microcephaly is a malformation associated with in utero exposed atomic bomb survivors and can be induced in mice by fetal exposure to ionizing radiation (IR). The pathogenesis of IR-induced microcephaly, however, has not been fully understood. Our analyses of high-coverage expression profiling (HiCEP) demonstrated that the abnormal spindle-like microcephaly associated gene (ASPM) was down-regulated in irradiated human diploid fibroblasts. ASPM was recently reported as the causative gene for MCPH-5, the most common type of congenital microcephaly in humans. Here, we show that the expression of

Effects of ionizing radiation on cellular structures, induced instability and carcinogenesis.

Ionizing radiation is perhaps the most extensively studied human carcinogen. There have been a number of epidemiological studies on human populations exposed to radiation for medical or occupational reasons, as a result of protracted environmental exposures due to radiation accidents, or after atomic bombings. As a result of these studies exposure to ionizing radiation has been unambiguously linked to cancer causation. While cancer induction is the primary concern and the most important somatic effect of exposure to ionizing radiation, potential health risks do not only involve neoplastic dise

NF-kappaB activation elicited by ionizing radiation is proapoptotic in testis.

The transcription factor NF-kappaB modulates apoptotic machinery following activation by the IkappaB kinase (IKK) complex. Inhibiting activity of one of the catalytic subunits of the IKK complex, IKKbeta (also known as IKBKB and IKK2) severely inhibits NF-kappaB nuclear translocation in response to most stimuli, including ionizing radiation. Doubly floxed Ikkbeta(F/F) mice (control) were compared to haplo-insufficient Ikkbeta(F/)(delta) mice (NF-kappaB knockdown) to examine the in vivo apoptotic role of NF-kappaB in the testis. Although Ikkbeta(F/F) control adult mice had spermatid head counts

Biological dosimetry -- cytogenetics findings at persons occupationally exposed to ionizing radiation.

A large number of physical and chemical agents are capable to course chromosomal aberrations. Ionizing radiation is frequent and well known course of chromosomal aberrations. If deoxyribonucleic acid (DNA) is irradiated before synthesis chromosomal-type aberrations are caused. Chromatid-type aberrations are results of DNA damages occurred during or after synthesis. Some of these changes could exist at patients several years after exposition. Biological dosimetry-cytogenetics analysis of persons occupational exposed to ionizing radiation in Federation of Bosnia and Herzegovina have been carried

Possible biomarkers for ionizing radiation exposure in human peripheral blood lymphocytes.

Biomarkers to indicate past exposure to radiation have not been entirely satisfactory. Using cDNA microarray hybridization to find new potential biomarkers, we identified highly expressed genes in human peripheral blood lymphocytes (PBLs) after irradiation 1 Gy ex vivo. The present set of radiation markers in PBLs was identified 12 h after radiation. A total of 44 genes were identified. However, when RT-PCR was performed with mRNA from the PBLs of five individuals, only four genes, including TRAIL receptor 2, DRAL (now known as FHL2), cyclin G, and cyclin protein gene, showed greater than 50%

Metallothionein induction in human CNS in vitro: neuroprotection from ionizing radiation.

PURPOSE: There have been extensive studies on the regulation of metallothionein (MT) synthesis, and its biological role in liver and kidney. Although there are few reports on brain MT, there is a growing interest in the role of MT in brain. There have been no publications to date on MT synthesis in the human central nervous system (CNS) following exposure to ionizing radiation. In the present study, primary human CNS cultures were used to examine the effect of ionizing radiation on MT mRNA and protein synthesis. In the same cultures, the neuroprotective effects of zinc (Zn) and cadmium (Cd)-in

Role of DNA mismatch repair in the cytotoxicity of ionizing radiation.

The DNA mismatch repair (MMR) system in mammalian cells not only serves to correct base mispairs and other replication errors, but it also influences the cellular response to certain forms of DNA damage. Cells that are deficient in MMR are relatively resistant to alkylation damage because, in wild-type cells, the MMR system is thought to promote toxicity via futile repair of alkylated mispairs. Conversely, MMR-deficient cells are sensitive to UV light, possibly due to the requirement for MMR factors in transcription-coupled repair of active genes. MMR deficiency has been associated with famili

Pulse exposure to ionizing radiation elicits rapid changes in cellular radiosensitivity.

A linear electron accelerator, operated in a recurrent chopped mode, was used for time-resolved investigation of split-dose radiation recovery in 3 mammalian cell lines in vitro. The time intervals separating the sequential radiation exposures in this study ranged from fractions of a second to a few minutes. The primary pulse brought about rapid, synchronous oscillations of cellular radiosensitivity giving rise to a tetraphasic, W-shaped time-dependent profile whose first phase was accomplished by a large decrease of cell survival. Only the last phase correlated with sub-lethal damage repair d

Recent advances in the study of ionizing radiation damage and repair.

This workshop, organized under the auspices of the EC Concerted Action Programme on DNA Repair and Cancer, was held at the CRC Gray Laboratory, Northwood, Middlesex, UK, 23-25 October 1991. The 42 participants were drawn mainly from laboratories participating in the EC Concerted Action, with a few visitors from elsewhere. The discussions centred on the increasing convergence of classical radiobiology and biophysics with molecular biology and mammalian cell genetics to study mechanisms of DNA strand break accumulation and repair following exposure to ionizing radiation. There was a strong empha

[Dosage loads on the bone marrow in exposure to ionizing radiation].

The authors made calculations of the expected dose loads on the bone marrow in the population of several regions due to the disaster release of radionuclides at the Chernobyl Atomic Station as well as in professional groups subjected to ionizing radiation, in particular, those working with radium light-compounds of permanent effect. The dose loads were largest in the population of the Byelorussian ecological region. Persons working in conditions of ionizing radiation showed a functional insufficiency of hemopoiesis in correspondence with the doses but there was no synchronicity in the response

Protective effect of selenium against ionizing radiation-induced malformations in mice.

A single dose of sodium selenite (0.5 mg Se/kg b.w.) was injected intraperitoneally into mice on day 9 of pregnancy, either 30 min or 2 h before 1.75 Gy whole body irradiation. Administration of selenite 2 h but not 30 min before irradiation resulted in a significant decrease in the number of malformed foetuses (p less than 0.005). The decrease in foetal malformations occurred proportionally for all the major malformations observed, i.e. short or kinked tail, rib and vertebral malformations, coloboma and deformation of retina and iris. In addition, selenium pretreatment also protected against

Adrenoreceptor development in rat cerebrum and the effects of ionizing radiation.

Previous studies in the rat have shown that neonatal radiation alters certain aspects of CNS monoaminergic systems including altering cerebral norepinephrine levels [17]. To determine if cerebral adrenoreceptor development is also altered by neonatal ionizing radiation, a single dose of whole body gamma-radiation was administered to rats on postnatal day two. This treatment did not alter (3H)-dihydroalprenolol (beta adrenoreceptor) or (3H)-WB4101 (alpha1 adrenoreceptor) binding when cerebra from irradiated animals were compared to age-matched sham-irradiated controls on postnatal days 9, 15 an
